Variations in Phenotypic Characteristics, Pathogenicity and Fungicides Resistance of Botrytis cinerea, Gray Mold Rot Fungus, Isolated from Various Host Plants (잿빛곰팡이 병원균 Botrytis cinerea 균주의 분리기주별 표현형적 특성, 병원성 및 약제저항성 변이)

  • Phenotypic characteristics, pathogenicity and fungicides resistance of fifty one isolates of Botrytis cinerea obtained from various host plants were observed and determined. The relationships between these characteristics were also investigated on the basis of isolation host plants. The isolates of B. cinerea varied in the capacity of sclerotia formation and sporulation. The pathogenicity of 44 isolates from tomato, cucumber, and strawberry was significantly stronger with 3.2 cm in average diameter of necrotic lesions on cucumber leaves than that of seven isolates from other host plants such as orange, gerbera, ginseng, kiwi, grape, pear and from butter with 1.8 cm in average diameter of necrotic lesions. Benomyl resistance of 12 isolates from tomato plants was much higher with the $EC_{50}$, 562 ppm than that of 19 isolates from various host plants. Diethofencarb resistance, however, of 11 isolates from strawberry plants was highest with the $EC_{50}$, 210 ppm among isolates from other host plants. Polygalacturonase activity varied among isolates in the range of 0 to 103 unit and that of isolates from tomato, cucumber and strawberry was slightly lower than that of isolates from other host plants. No significant relationship between pathogenicity and fungicides resistance, polygalacturonase activity was found among 51 isolates of B. cinerea. Isozyme patterns of polygalacturonase produced from two strongly and weakly pathogenic isolates (FC122, KC6) were slightly different depending upon carbon sources during cultivation.

Establishment of Analytical Method for Chlorophyll Using the N,N-Dimethylformamide and Dimethylsulfoxide in Citrus Leaves (N,N-Dimethylformamide와 Dimethylsulfoxide를 이용한 감귤 잎의 엽록소 측정방법 확립)

  • BACKGROUND: Purpose of the study was to establish the extinction coefficients of chlorophyll a and b in N,N-dimethylformamide(DMF) and Dimethylsulfoxide(DMSO) solvents and to find out the conditions of optimal extraction temperature and time in citrus leaves. METHODS AND RESULTS: Chlorophyll a and b standards were dissolved in DMF and DMSO. Extinction coefficients of chlorophyll pigments were determined and their contents were quantified using spectrophotometer. Chlorophyllous pigments of citrus(Citrus unshiu Marc. cv. Okitsu wase) leaves were extracted at 25, 40, 60 and $80^{\circ}C$ for 4, 6, 8, 24 and 48 hours to determine the optimal extraction condition. CONCLUSION: The extinction coefficients of chlorophyll a(Chl a) and chlorophyll b(Chl b) of DMF extracts for high extinction wavelength were 663.8 and 647.2 nm. Similarly, the high extinction wavelength of DMSO extracts were 665.8 and 649.0 nm for chl a and b respectively. Chl a, Chl b and total chlorophyll content of DMF extracts were Chl a = $12.10A_{663.8}-2.74A_{647.2}$, Chl b = $21.94A_{647.2}-5.06A_{663.8}$ and total $Chl=19.193A_{647.2}+7.04A_{663.8}$. Similarly, Chl a, Chl b and total Chl of DMSO extracts were Chl a = $14.53A_{665.8}-5.40A_{649.0}$, Chl b = $26.98A_{649.0}-7.11A_{665.8}$ and total $Chl=21.58A_{649.0}+7.43A_{665.8}$. The chlorophyll extracts of DMF and DMSO were very stable in dark. High chlorophyll contents of citrus leaves were found at $80^{\circ}C$ extraction for 6 hours in DMF and at $80^{\circ}C$ extraction for 24 hours in DMSO. However, the chlorophyll content was decreased significantly after 8 hours in DMF extraction while it was remained up to 30 hours in DMSO extraction.

Frequency and Growth Characteristics of Polyploids Occurred Spontaneously in Some Mandarin Hybrids (만다린 잡종에서 자연발생 배수체의 발생 빈도와 생장 특성)

  • The study was conducted to determine the efficiency in producing spontaneous polyploids in some mandarin hybrids with different seed embryony. Seed formation by open pollination, frequency of spontaneous polyploids, and plant growth characteristics were evaluated in four mandarin hybrids with polyembryony such as 'Amakusa', 'Haruka', 'Hayaka', and 'Seminole' and two with monoembryony such as 'Benibae' and 'Harehime'. The mean number of the developed seeds per fruit was 10.0 and frequency of small seeds was 25.1%. Polyploids were selected from plants germinated in vitro by a flow cytometry and confirmed by chromosome analysis. One triploid was produced from 'Harehime', one tetraploid, 'Amakusa', and one tetrapoid, 'Benibae'. There were little differences in leaf shape, thickness, petiole length, and internode length between diploids and polyploids such as tri- or tetraploid. However, polyploids had larger stomata and lower density of stomata in abaxial epidermis than diploids. SPAS indicating chlorophyll content and photosynthetic rate were significantly affected by ploidy level. The results indicated that spontaneous polyploids might be produced by open pollination in some mandarin hybrids and monoembryony had higher frequency in polyploid occurrence than polyembryony.

Binomial Sampling Plans for the Citrus Red Mite, Panonychus citri(Acari: Tetranychidae) on Satsuma Mandarin Groves in Jeju (온주밀감에서 귤응애의 이항표본조사법 개발)

  • The density of citrus red mite(CRM), Panonychus citri(McGregor), on the commercial satsuma mandarin Citrus unshiu L. groves were determined by counts of the number of CRM per leaf using by leaf sample in Jeju for 2 years. Binomial sampling plans were developed based on the relationship between the mean density per leaf(m) and the proportion of leaf infested with less than T mites per leaf($P_{T}$), according to the empirical model $ln(m)={\alpha}+{\beta}ln(-ln(1-P_{T}))$. T was defined as tally threshold, and set to 1, 3, 5 and 7 mites per leaf in this study. Increasing sample size, regardless of tally threshold, had little effects on the precision of the binomial sampling plan. Increasing sampling size had little effect on the precision of the estimated mean regardless of tally thresholds. T=1 was chosen as the best tally threshold for estimating densities of CRM based on the precision of the model. The binomial model with T=1 provided reliable predictions of mean densities of CRM observed on the commercial satsuma mandarin groves. Binomial sequential sampling procedure were developed for classifying the density of CRM. A binomial sampling program for decision-making CRM population level based on action threshold of 2 mites per leaf was obtained.

Classification of the Edible Plants on the Market in Busan (시장에 유통되는 식용식물의 분류)

  • The edible plants sold at the market in Busan were classified into 8 divisions, 11 classes, 46 orders, 67 families, 156 genera, 183 species and 45 varieties, and a total of 228 kinds of plants were included in them. Among the 228 kinds of edible plants, Angiospermae were the highest number of 202 kinds (88.60%), and next came Fungi of 9 kinds (3.95%), Phaeophyta of 5 kinds (2.19%), Rhodophyta of 4 kinds (1.75%), Pteridophyta of 3 kinds (1.32%) Gymnospermae of 2 kinds (0.88%), Chlorophyta of 2 kinds (0.88%), and Lichenes of 1 kind (0.44%) in order In the taxon of family, Cruciferae, Rosaceae, Leguminoceae, Cucurbitaceae, Umbelliferae, Compositae, Liliaceae, and Gramineae include many species. In the taxon of genus, Brassica(14 species), Prunus(9), Lactuca(9), Allium(7), Citrus(5), Cucumis(4), Cichorium(4) in order contain many species. Among tile edible plants, Chamjuknamu(Cedrela sinensis) is wrongly named Gajuknamu(Ailanthns altissima). And there are others which are called in a wrong way: Endive is wrongly called Chicory, Saeneutaribeoseot, Saesongi and Padeudeknamul(Japanese hornwort), Chamnamul. Gyul and Milgam, Geumgam and Gamgyul, Banana and Pacho, Paprika and Pimang(Pimento) are the names of the same plants. The number of the kinds of edible plants is different in each season. Fall has the most edible plants, and it is followed by spring, summer and winter. The edible parts of plants which are sold at the market are leaves, young sprouts, fruits, grains, roots, stems and petioles.
