Optimization of Analytical Methods for Octacosanol in Related Health-functional Foods with GC-MS (GC-MS를 이용한 건강기능식품 중 옥타코사놀 분석법 개발 연구)

  • The Ministry of Food and Drug Safety (MFDS) is amending its test methods for health-functional foods (dietary food supplements) to establish regulatory standards and specifications in Korea. In this regard, we continue our research on developing analytical methods for the items. Octacosanol is the major component of polycosanol and is a high-molecular-mass primary fatty alcohol, obtained from sugar cane wax. Previous researchers have shown that octacosanol can lower cholesterol and has antiaggregatory properties, cytoprotective uses, and ergogenic properties for human health. Recently, octacosanol products have been actively introduced into the domestic market because of their functional biological activity. We have developed a sensitive and selective test method for octacosanol that the TMS derivatives by means of gas-chromatographic-tandem mass spectrometry (GC-MS). The trimethylsilyl ether derivative of the target analyte showed excellent chromatographic properties. The procedure was validated in the range of $12.5{\sim}200{\mu}g/L$. Standard calibration curves presented linearity with the correlation coefficient ($r^2$) > 0.999, and the limits of detection (LOD) and limits of quantitation (LOQ) were $4.5{\mu}g/L$ and $13.8{\mu}g/L$, respectively. The high recoveries (92.5 to 108.8%) and precision (1.8 to 2.4%) obtained are in accordance with the established validation criteria. Our research can provide scientific evidence to amend the octacosanol test method for the Health-Functional Food Code.

Distribution of Biomass and Production of Robinia pseudoacacia Plantation in Korea (아까시나무조림지(造林地)의 물질생산량(物質生産量)에 관(關)한 연구(研究))

  • To study the comparison of the aboveground biomass of Robinia pseudoacacia L. of 8-, 13- and 20-year-old plantations, the experimental plot of $100m^2$ in size located in Youngin-gun were selected. Seven sample trees at different stand ages selected taking account of DBH distribution were felled and the diagram of oven-dry weight distribution of stem, branch and leaf for each 1m segment was constructed. The dry weight of each part of plots was estimated by the method of basal area ratio. If the estimations are extended to a hectare area stand, it contains 36.72, 69.28 and 118.67 tons of the aboveground standing crops in 8-, 13- and 20- year-old stand respectively. The net production was estimated to be as much as 12.56, 13.23 and 16.78 tons per hectare per year and the net assimilation ratio 3.66, 4.13 and 2.50 kg/kg/yr in 8-, 13- and 20-year-old stand respectively. And the efficiency of leaves to produce stem was 1.69, 2.10 and 0.96 kg/kg/yr in same order.

A Study on the Effect of Environmental Pollution on the Biomass Productivity of Paulownia coreana (환경오염(環境汚染)이 오동나무인공림(人工林)의 물질생산(物質生産)에 미치는 영향(影響)에 관한 연구(硏究))

  • To study the comparison of aboveground biomass of paulownia coreana Uyeki of 6-year-old, located in Seongju of non-attacked forest and Ulsan of damaged forest by the air pollution were selected. Ten sample trees in Seongju district and seven trees in Ulsan selected taking account of DBH were measured for 16 trees in total within a $10{\times}10m$ experimental plot. The diagram of oven-dry weight distribution of stem, branch and needle for each 1m segment was constructed. The logarithmic regression equations between dry weight of each component and the two-variables, DBH and tree height, combined term were presented. IF the estimations are extended to a hectare area stand, it contains 47.49 tons of aboveground biomass in Seongju district and 19.05 tons of it in Ulsan. The annual net productions were 11.64 tons of above 2.29 kg/kg/yr in Ulsan and the efficiency of leaf to produce stem was 2.99kg/kg/yr in Seongju and 0.83kg/kg/yr in Ulsan.

A Study on the Structure of Biomass Production in Thrifty-Mature Quercus mongolica Stand (신갈나무 장령임분(壯齡林分)의 물질생산(物質生産) 구조(構造)에 관(關)한 연구(硏究))

  • This study was conducted to explain the characters of the structure of biomass production in the thrifty-mature Quercus mongolica stands and investigate the relationships between the leaf weight or leaf area and the sapwood area in the bole. Also we intended to identify the allocation ratio of stem, branches, and leaves or heartwood, sapwood and bark in trees and the characters of productive structure of stem and leaf biomass by the tree height. The results obtained were as follows : 1. The allocation ratio of biomass based on dry weight was 70-84% in stem, 11-25% in branches, and 3-6% in leaves. 2. In the bole, the ratios of composition of heartwood, sapwood, and bark were showed 37-43%, 38-46%, and 16-19%, respectively. 3. The volume of sapmood was exceeded more than that of heartwood in dominant and intermediate trees, while it was reversely appeared in suppressed trees. 4. The weight and area of leases significantly correlated with the sectional area of sapwood in bole (r>0.9. 1% significant level). 5. The ratio of leaf area($m^2$) to sapwood areal($cm^2$), k varied 0.35 to 2.05. 6. The basal diameter and the cross sectional area of a branch significantly correlated with the leaf weight r>0.9. 1% significant level. 7. The leaf weight in a tree is showed a normal distribution curve and the accumulative volume of bole is showed a tapering type.

Studies on the Ray Parenchyma of Salicaceae in Korea (한국산(韓國産) 버드나무류(類)의 방사조직(放射組織)에 관한 연구(硏究))

  • These studies were carried out to identify anatomical characteristics of ray parenchyma of xylem tissue of trunk in 6 species of Populus and 5 species of Salix which were grown in Korea. The results of these experiments were summarized as follows: In the type of ray parenchyma, Populus had uniseriate homogeneous ray tissue and Sahx had uniseriate heterogeneous ray tissue. Upright ray cells among uniseriate heterogeneous ray tissue in Salix were subdivided into rectangular type and square type. The minimum and maximum length of procumbent ray cells of Populus ranged $26.84-212.28{\mu}$ and those of Salix were $46.36-170.80{\mu}$. However rectangular type of upright ray cell n Salix were $26.84-70.76{\mu}$ and square type were $17.08-43.92{\mu}$. The minimum and maximum width of procumbent ray cells of Populus ranged $12.20-24.40{\mu}$ but those of Salix were $12.20-26.84{\mu}$. However, rectangular type of upright ray cell in Salix were $9.76-41.48{\mu}$ and square type were $19.52-46.36{\mu}$. The height of ray parenchyma of Populus in tangential section ranged $65.88-414.80{\mu}$ but Salix were $65.88-439.20{\mu}$. Ray parenchyma width of Populus ranged $4.88-24.40{\mu}$ but those of Salix were $7.32-21.96{\mu}$. The number of ray parenchyma cells of Populus in tangential section were 3-26 cell, but Salix were 2-21 cells.

  • The starting point of the research is the relation between capital restructuring and urban restructuring. The economic restructuring, which has been caused by the economic crisis in the early 1970s in the United States has brought a spatial restructuring at different geographic scales. The degree of the success of urban restructuring is contingent to the local economic and political environments. The local contingent factor such as local politics should not be neglected for investigating the restructuring process. Through the case study of Chicago, the research provides two inconsistencies in applying the structural approach to the local level: first, the lack of the theoretical link between crisis and restructuring; and second, the crucial importance of local politics in shaping urban development.

Alteration of MicroRNAs Targeted Integrins by PD-MSCs Transplantation Is Involved in Hepatic Regeneration in a Rat Model with BDL (담관결찰 쥐 모델에서 태반유래중간엽줄기세포 이식에 의한 miRNA 표적 인테그린 변화의 간재생 효과)

  • Placenta-derived mesenchymal stem cells (PD-MSCs) are promising candidates for cell-based therapy in regenerative medicine. The migration and homing potential of PD-MSCs to injured sites is a critical property of MSC engraftment. MicroRNAs (miRNAs) have recently been shown to regulate the critical functions of MSCs, such as proliferation, survival, and migration. The objective of the present study was to identify the miRNA and target genes involved in PD-MSCs homing in a bile duct ligation (BDL) rat model. We selected candidate miRNAs targeting genes for PD-MSCs homing based on microarray analysis. PD-MSC engraftment in BDL-injured rat liver was identified by immunofluorescence assay and human-specific Alu gene expression by quantitative real-time polymerase chain reaction (qRT-PCR) one week after transplantation. Compared with migrated naïve PD-MSCs under hypoxic and normoxic conditions (Hyp/Nor), the transplanted group with PD-MSCs (Tx) showed distinct differences in miRNA expressions in BDL-injured rat liver. We also validated the miRNAs and their target genes for PD-MSCs homing. The expressions of integrin α4 (ITGA4) and integrin α5 (ITGA5) target genes for miR-199a-5p and miR-148a-3p were significantly upregulated in the Tx group (p<0.05). In addition, integrin β1 (ITGB1) and integrin β8 (ITGB8) were upregulated by suppressing miR-183-5p and miR-145-5p, respectively. These results demonstrated that PD-MSCs regulate miRNA expression related to the integrin family for their homing effects on the BDL-injured rat liver. The findings further suggest that miRNA-mediated regulation of the integrin family contributes to the therapeutic efficacy of PD-MSCs in the rat hepatic fibrosis model by BDL.

Estimation of Productivity for Quercus variabilis Stand by Forest Environmental Factors (삼림환경인자(森林環境因子)에 의한 굴참나무임분(林分)의 생산력추정(生産力推定))

  • This study was initiated to estimate productivity of Quercus variabilis stand. However the practical objective of this study was to provide some information to establish the basis of selecting the suitable site for Quercus variabilis. The productivity measured in terms of DBH, height, basal area and stem volume was hypothesized, respectively, to be a function of a group of factors. This study considered 32 factors, 20 of which were related to the forest environmental factors such as tree age, latitude, percent slope, etc. and the rest of which were related to soil factors such as soil moisture, total nitrogen, available $P_2O_5$, etc. The data on 4 productivity measurements of Quercus variabilis growth and related factors cited were collected from 99 sample plots in Kyeongbook and chungbook provinces. Some factors considered were, in nature, discrete variables and the others continuous variables. Each kind of factor was classified into 3 or 4 categories and total numbers of such categories were eventually amounted to 110. Then each category was treated as an independent variable. This is amounted to saying that individual variable was treated a dummy variable and assigned a value 1 or 0. However the first category of each factor was deleted from the normal equation for statistical consideration. First of all, each of 4 productivity measurements of Quercus variabilis growth was regressed and, at the same time, those 110 categories. Secondly, the partial correlation coefficients were measured between each pair of 4 productivity measurements and 32 individual foctors. Finally, the relative scores were estimated in order to derive the category ranges. The result of these statistical analyses could be summarized as follows: 1) Growth measurement in terms of height seems to be a more significant criterion for estimation of productivity of Quercus variabilis. 2) Productivity of forest on stocked land may better be estimated in terms of forest environmental factors, on the other hand, that of unstocked land may be estimated in terms of physio-chemical factors of soil. 3) The factors that a strongly positive relation to all growth factors of tree are age group, effective soil, soil moisture, etc. This implies that these factors might effectively be used for criteria for selecting the suitable site for Quercus variabilis. 4) Parent rock, latitude, total nitrogen, age group, effective soil depth, soil moisture, organic matter, etc., had more significant category range for tree growth. Therefore, the suitable site for Quercus variabilis may be selected, based on this information. In conclusion, the above results obtained by the multivariable analysis can be not only the important criteria for estimating the growth of Quercus variabilis but also the useful guidance for selecting the suitable sites and performing the rational of Quercus variabilis forest.
