  • To investigate hepatoprotective activities of the root extract of Rosa davurica, the activities of hepatic enzymes, aminopyrine N-demethylase, aniline hydroxylase, glutathione S-transferase and epoxide hydrolase in rats intoxicated with bromobenzene were studied. Pretreatment with the methanol extract from the roots of Rosa davurica did not show any significant effects on the increases of the activities of aminopyrine N-demethylase and aniline hydroxylase, enzymes forming toxic epoxide by bromobenzene. There was no change in glutathione S-transferase activity by Rosa davurica. However, the activity of epoxide hydrolase, and epoxide-removing enzyme, was increased 33% by the administration of 500 mg/kg of the methanol extract. From the results, the protection of Rosa davurica against bromobenzene-induced hepatotoxicity is thought to be via enhancing the activity of epoxide hydrolase, an enzyme removing toxic epoxide rather than through epoxide-producing system.

  • The purpose of present study was to investigate the protective effect of silkworm excrement powder (SEP) on alcohol-induced hepatotoxicity in rats. Semisynthetic diet supplemented with SEP (3%, w/w) given to alcohol-feeding rats for 30 days, then blood and tissues were collected, processed and used for alcohol concentration mensuration, various biochemical estimations and histopathological examination. Chronic alcohol administration resulted in significantly increase in the activities of the clinically important liver marker enzymes, alanine aminotransferase (ALT), aspartate aminotransferase (AST), $\gamma$-glutamyl transpeptidase ($\gamma$-GTP) and lactate dehydrogenase (LDH). Also, a highly significant increase in the blood alcohol level by alcohol treatment was observed. But alcohol-induced elevation of ALT and LDH levels markedly prevented and the level of blood alcohol decreased in SEP treated rats as compared to alcohol-administered control rats. SEP supplementation showed highly decreased the concentrations of total lipid, triglyceride and cholesterol in serum, as compared with alcohol treated control rats. Alcohol treatment induced the marked accumulation of large lipid droplets, hepatocytes necrosis and inflammation in the liver, but SEP administration attenuated to alcohol-induced accumulation of lipid droplets and hepatocyte necrosis. The results indicated that SEP may exert a protective effect against alcoholic hepatotoxicity through decreasing the activity of hepatic marker enzymes.

  • Testosterone deficiency syndrome (TDS) is normally observed in elderly men and is a clinical and biochemical syndrome, characterized by a decline in plasmic testosterone levels resulting in a significant decrease in quality of life. The aim of this study was to evaluate the effect of a mixed extract of fenugreek seeds and Lespedeza cuneata (YHM) on TDS. Rats were divided into three groups: the negative control, YHM-40 (40 mg/kg), and YHM-80 (80 mg/kg) groups. After 4 weeks of YMH administration, an increase was observed in the plasmic testosterone levels, vastus lateralis muscular strength, forced swimming time, total sperm counts, and motile sperm counts in YHM-40 and YHM-80 groups compared to the negative control group. Moreover, sex hormone binding globulin, the epididymal fat pad, total plasmic cholesterol, and triglyceride levels were significantly decreased in the YHM-fed groups. However, prostate specific antigen, aspartate aminotransferase, and alanine aminotransferase levels did not differ among the groups. These results suggest that YHM may enhance testosterone levels in elderly men and alleviate TDS without common side effects.

  • This study was carried out to investigate the effects of Rhynchosia molubilis saponin on carbon tetrachloride ($CCl_4$)-induced hepatotoxicity. Sprague-Dawley rats were intraperitoneally administered the Rhynchosia molubilis saponin at 100 mg/kg every day for two weeks, then $CCl_4$ (3.3 ml/kg) was injected into rats. 12 hours later, they were anesthesized with ether and dissected. Rhynchosia molubilis saponin-administered group showed 59.92% and 62.28% of inhibitory effects on aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities compared to $CCl_4$-treated group (p<0.05). Malonedialdehyde (MDA) levels of Rhynchosia molubilis Saponin-administered and $CCl_4$-treated (RSC) group in liver homogenate and mitochondria were significantly inhibited to 61.83%, 81.11 %, respectively, compared to $CCl_4$-treated group (p<0.05). Superoxide dismutase (SOD) activities of RSC group in liver homogenate and mitochondria were significantly inhibited to 66.53%, 31.04%, respectively, compared to $CCl_4$-treated group (p<0.05). GPx activities of RSC group in liver homogenate and mitochondria were significantly inhibited to 72.74%, 72.68%, respectively, compared to $CCl_4$-treated group (p<0.05). The histological examinations showed that the liver cell necrosis and centrilobular congestion aggregation induced by $CCl_4$ were dearly eliminated by the administration of Rhynchosia molubilis saponin. These results suggest that Rhynchosia molubilis saponin could have the protective effects against hepatotoxicity.

  • The hepatoprotective effects of water extracts composed of Adenophora triphylla var japonica Hara (ATJH) on acetaminophen (APAP)-induced hepatotoxicity were investigated in vivo and in vivo. The effects of ATJH on liver toxicity induced by APAP were assessed by blood biochemical and histopathological analyses. APAP treatment (350 mg/kg) caused severe liver injury in mice as indicated by their significantly elevated plasma aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels. Pretreatment with ATJH for 3 or 7 days attenuated the increases in ALT and AST when challenged with APAP. The reductions in viability caused by high dose of APAP (450 mg/kg) in vivo were reversed by pretreatment with ATJH. These protective effects of ATJH against APAP-induced toxicity were consistent with the results from the histopathological examinations. We next examined the effects of ATJH on the gene expression of glutathione S-transferases (GSTs) that detoxify the metabolic intermediates of APAP in H4IIE cells. The hepatic GST protein levels [$\alpha$ class (GSTA2, GSTA3/5)] were significantly elevated in a dose-dependent manner by ATJH treatment. In summary, ATJH is effective at protecting against APAP-induced hepatotoxicity by GST induction, implying that ATJH should be considered a potential chemopreventive agent.
