The Preventive Effects of Paeoniae Radix Extract against LPS-induced Acute Hepatotoxicity (LPS로 유도된 급성 간독성에 대한 백작약 추출물의 보호 효과)

  • The purpose of this study was to investigate the preventive effects of Paeoniae Radix Extract(PRE) against the acute hepatotoxicity-inducing lipopolysaccharide(LPS) in the liver. PRE of 100 mg/kg concentration was intraperitoneally administered into rats at dose of 1.5 ml/kg for 20 days. On day 21, 5 mg/kg of LPS dissolved in saline was injected 4 hours before anesthetization. We examined the levels of glutamate oxaloacetate transaminase(GOT), glutamate pyruvate transaminase(GPT), lactate dehydrogenase(LDH) in serum of rats, superoxide dismutase(SOD) in mitochondrial fractions, and malondialdehyde(MDA), catalase(CAT), glutathione peroxidase(GPx) in liver homogenates. LPS-treatment markedly increased the levels of GOT, GPT, LDH and MDA, and significantly decreased those of SOD, CAT and GPx. But PRE-pretreatment decreased the levels of GOT, GPT, LDH and MDA, by 59.7%, 43.6%, 59.6% and 63.5%, respectively and increased those of SOD, CAT and GPx, by 85.5%, 57.8% and 62.9%, respectively. These results showed that the PRE had the preventive effects against the acute hepatotoxicity-inducing LPS in the liver.

Comparative Hepatotoxicity Assessment of Cadmium and Nickel with Isolated Perfused Rat Liver(IPRL) (적출간 관류법을 이용한 카드뮴과 니켈의 간독성 비교)

  • Objectives : It is the objective of this study to compare hepatotoxicity of nickel chloride and cadmium chloride with each other through IPRL(Isolated Perfused Rat Liver) method. Methods : Biochemical indicator of hepatic function such as AST(aspartate aminotransferase), ALT(alanine aminotransferase), LDH(lactate dehydrogenase) and perfusion flow rate were used as the indicator of hepatotoxicity. Oxygen consumption rate were used as vability indicator. $300({\pm}50)g$ - weighted rats were allocated randomly to each group($0{\mu}M,\;50{\mu}M,\;200{\mu}M\;NiCl_2\;and\;CdCl_2$ exposure) by 5, totally 25. After Krebs-Ringer bicarbonate butler solution flowed into the penal vein and passed the liver cell, it flowed out of vena cava. Liver was administered with each $NiCl_2\;and\;CdCl_2$ of each concentration and observed with buffer solution sampling time. Butler which got out of liver was sampled and then biochemical indicator of hepatotoxicity was measured. Results : AST, ALT, and LDH in buffer increased with sampling time much more in $CdCl_2$ exposure group than $NiCl_2$ exposure group in both 50 and $200{\mu}M$ and statistical significance w3s verified with 2-way repeated ANOVA. Viability was decreased more and more in all substances during passed time. Conclusions : It is inferred that $CdCl_2$ has stronger hepatotoxicity than $NiCl_2$. IPRL method would be used widely for acute hepatotoxicity when considerating the benefit of it.

Hepatoprotective Activity of Fermented Curcuma longa L. on Galactosamine-Intoxicated Rats (발효울금의 갈락토사민 투여에 의한 흰쥐의 간독성에 대한 보효 효과)

  • In this study, the general components and minerals of fermented Curcuma longa L. (FC) by Aspergillus oryzae were examined as well as the hepatoprotective effects of FC on acute hepatotoxicity induced by a single dose of galactosamine (GalN, 650 mg/kg body weight (b.w.)). The FC was found to consist of 0.15% moisture, 4.68% crude fat, 4.35% crude protein, 6.92% crude fiber, and 6.83% crude ash. The P, Ca, and Mg levels in FC were also quantitatively analyzed. Male Sprague-Dawley rats were divided into six groups; nontreated control, GalN, 150 mg/kg b.w. of silymarin plus GalN, 30 mg/kg b.w. of FC plus GalN, 100 mg/kg b.w. of FC plus GalN, and 300 mg/kg b.w. of FC plus GalN. Pretreatment 300 mg/kg b.w. of FC during 14 days significantly decreased the increased in aspartate aminotransferase, alanine amino transferase, and triglyceride (TG) induced by GalN. Severe liver damage, hepatocellular necrosis, infiltration of inflammatory cells, and councilman body necrosis on histopathological liver tissues were observed in GalN treated rats. Administration of 300 mg/kg b.w. of FC significantly decreased the degree of live damage. These results suggest that FC displays hepatoprotective activity and FC was able to lower the TG levels in serum; thus, FC may serve as a useful material for health food and clinical conditions associated with liver disease.

  • 천연물 성분의 약효와 간독성에 관여하는 기전연구의 일환으로 사염화탄소로 유발된 흰쥐에서 혈청 중의 효소 활성도, 지질함량 및 과산화지질을 측정하여 홍화자 추출물과 분획물이 사염화탄소로 유발되는 간독성에 미치는 효과를 규명하고자 하였다. 홍화자는 홍화(Carthamus tinctorius L.)의 씨로서 성미는 신, 온하며 활혈거제 및 소 지통의 효능을 가진 구어혈제로서 자궁에 대한 흥분작용과 관상동맥을 확장시키는 작용이 있으며 혈압을 하강시키는 작용이 있다. 또한 고cholesterol 혈증을 낮추고 혈전폐색성 맥간염을 치료하는 효과도 있다고 한다.

  • 우리나라 사람들에게는 유달리 높은 간염 발병률이 나타나고 있고 더우기 바이러스성 간염의 경우 풍토병 내지 전염병이라고 할 정도의 양상을 띠고 있다. 현재까지는 이러한 간질환에 적절한 치료약이 없는 점을 감안하여 저자들은 생약으로 부터 간질환에 유효한 성분을 분리하여 치료약으로 사용할 수 있도록 하기위하여 집중적인 연구를 지난 5년간 계속하여 왔다. 연구의 내용은 동서양의 고전및 연구보문을 조사하여 간보호 및 치료학으로 사용한 생약에 관한 문헌적 조사, 이들 생약중 채집 및 구입이 가능한 식물을 구하여 생약엑기스 제조, 이들 엑기스를 간염 동물 모델의 하나인 $CCl_{4}$로 유발시킨 간독성에 대한 보호작용의 검색, 보호작용을 나타내는 생약중에서 자원적 측면을 고려하여 국내에서 많이 생산되는 차전자(Plantago asiatica seeds)를 선택하였고, 차전자로 부터 간보호 작용을 나타내는 유효성분으로 iridoid 계열물질인 aucubin을 분리하였다. 그러나 aucubin이 차전자에 소량 밖에 없으므로 aucubin이 다량 함유된 식나무(Aucuba japonica)로 aucubin 추출자료생약을 바꾼후 aucubin의 간보호 작용을 간염 동물모델인, $CCl_{4}$ D-galactosamine 및 $\alpha$-amanitin등으로 유발시킨 간독성에 대한 보호작용을 보여 주었기 때문에, 이러한 간보호작용의 기전을 규명하는 연구를 진행하였고, 다음은 aucubin 자체의 급성 독성 및 기타 독성 유발여부를 밝히는 연구 등으로 요약 될 수 있다.

In vitro hepatocyte inflammation by Ephedra sinica extracts (마황 추출물의 in vitro 간세포 염증반응 유도)

  • In this study, the in vitro hepatotoxic mechanism of Ephedra sinica (ma-huang) was investigated by measuring the degree of cell death, secretion of cytokine, and fat accumulation by treating HepG2 cells with 70% ethanolic extracts of ma-huang. Cell death was observed at concentrations of around $5-100{\mu}g/mL$ by treatment with ma-huang extracts (p<0.05). The secretion of interleukin 8 (IL-8) and macrophage colony-stimulating factor (M-CSF), which are inflammatory cytokines, were significantly promoted at concentrations of around 0.05-100 and $0.5-100{\mu}g/mL$, respectively (p<0.05). In this experiment, it was shown that the extracts of ma-huang stimulate the secretion of inflammatory cytokines, such as IL-8 and M-CSF, and lead to fat accumulation in the hepatocytes, thereby causing inflammation of the hepatocytes. Hepatotoxicity was observed at around 10-500 times lower concentration than the concentration required to cause serious toxicity, such as cell death, suggesting that hepatic toxicity (hepatitis) may be induced at a low dose.

Protective Effect of Lithospermum erythrorhizon on Galactosamine Induced Liver Injury (자초(Lithospermum erythrorhizon)추출물의 투여가 Galactosamine으로 유도된 간손상에 미치는 보호 효과)

  • The present study was carried out to investigate the hepatoprotective effect of water extract of Lithospermum erythrorhizon on acute hepatotoxicity induced in Sprague-Dawley (SD) rats by a single dose of galactosamine (400 mg/kg, i.p). The animals were divided into four groups. The animals in the Con group were fed basal diet. GalN group were administered with galactosamine. LE200 and LE500 groups treated with water extract of Lithospermum erythrorhizon (such as 200 and 500 mg/kg/day, p.o) for 7 days before galactosamine injection. In the change of AST, ALT, ALP, GGT and LDH contents, as compared with GalN group, LE200 group were significantly decreased. According to the electron microscopical observation, liver cells were increased the lipid droplet, change of mitochondria in the GalN compared with LE200. These results suggest that administration of water extract of Lithospermum erythrorhizon suppress or retard galactosamine induced acute liver injury.

The preventive effect of Daeshiho-tang on liver damage induced by acetaminophen in the rats (대시호탕이 acetaminophen으로 유도된 간독성 흰쥐에 미치는 영향)

  • Acetaminophen은 세계적으로 널리 사용되는 해열 진통제이지만, 또한 과용 및 남용, 알코올 중독과 같은 여러 원인에 의해 간독성을 유발할 수 있는 약물이다. 이러한 acetaminophen의 간독성은 CYP에 의해 생성되는 대사산물인 NAPQI와 활성산소에 의해 유발되는 것으로 알려져 있다. 본 연구에서 5주된 수컷 백서에 acetaminophen (500 mg/kg)을 투여하기 전에 대시호탕 (500 mg/kg)를 일주일간 투여하였다. 이후 GOT, GPT, GST 그리고, 조직사진으로 대시호탕의 간보호작용을 측정하였다. 또한 대시호탕의 간보호작용 기전을 항산화작용과 CYP 2E1 발현조절을 통한 NAPQI 생성억제의 두 가지 면에서 측정하였다. GOT, GPT 그리고 조직사진에서 나타난 결과들은 대시호탕이 고용량의 acetaminophen에 대한 간보호작용이 있음을 증명할 수 있었다. 또한 LPO와 catalase, 그리고 GSH 실험에서 나타난 결과들을 통해 대시호탕이 항산화작용이 있음을 알 수 있었다. 그리고 GSH, GST, RT-PCR, western blot 실험에서 대시호탕이 CYP 2E1의 발현을 조절하여 NAPQI 생성을 억제한다는 것도 알 수 있었다. 이상의 결과들을 바탕으로 대시호탕은 항산화작용에 의한 활성산소 제거력과 CYP 2E1의 발현조절을 통한 NAPQI 생성억제로 고용량 acetaminophen에 의해 유도된 간손상에 대해 유의성 있는 보호작용을 한다는 것을 알 수 있었다.
