The comparative of Naringin and Chitosan using Natural preservation agents by LM and TEM (천연보존제 나린진과 키토산의 비교....LM & TEM적 소견)

  • This study investigated the effects of naringin, and chitosan in rabbits' corneas. Naringin, a glycone of naringenin, is a widely distributed bioflavonoid in the grapefruit and citrus peel, and it has already been reported as an antioxidant, antimicrobial, and anticancer agent. It has been used as a food preservatives and cosmetics. One of the natural preservatives, chitosan has also used in food preservatives, health drinks, and teas. Chitosan is distributed in the epithelium of crustacea, insects, and fungi. Naringin and chitosan have no harmful effects of cytotoxicity in the human body and they are recognized as an antibacterial for various forms of bacteria. The purpose of this study is to search for the ideal percentage of natural products to substitute the chemical preservatives occuring within the cornea and conjunctiva cytotoxicity and inflammations as wearing on soft contact lens. The present study compared the morphology of corneal epithelium and endothelium observed by light microscopy (LM) and transmission electron microscopy (TEM). In vivo methods, We investigated the effects of natural preservatives on soft contact lens. We inserted 3-4 drops of the naringin and chitosan, directly on rabbits' corneas 4 times per a day during one week. After enucleation of cornea, morphorgical damages of the epithelium and endothelium were observed by LM and TEM. In view of ultrastructure, chitosan caused siginficant damage on the epithelium and endothelium of cornea. The damage of cells was higher in chitosan treated cornea than 0.01, 0.1, and 1% of naringin. The 1% of naringin also expressed cell damage seriously. The results suggest that the most important thing is to use the reasonable percentage of preservatives for contact lens solutions.

  • Calcification in patients with endstage renal disease on renal replacement therapy can occur in extraskeletal area such as conjunctiva and cornea. Conjunctival and corneal calcification(CCC) has mostly has been reported in adults with endstage renal disease on hemodialysis. CCC seems to be associated with the duration of renal replacement therapy, and high Ca$\times$P value. We report a 10-year-old girl who was on peritoneal dialysis for 31 months and presented with CCC on both eyes. Her corneal calcification was resolved after the epithelial debridement and ethylenediaminetetraacetic acid(EDTA) soaking therapy.

  • The corneal tissue consists of three layers : epithelium, stroma, and endothelium. Central cornea is a highly differentiated tissue whereas the limbus contains the epithelial stem cell. In the present study. we report the engineering of the three-dimensional reconstructed cornea derived from rabbit limbal epithelial and stromal cells. The differentiation degree of corneal stem cells were assessed in serum concentration and inoculation density of stromal cells. Optimal condition differentiation of corneal stem cells is achieved when 5% FBS was supplemented to culture medium and $1-2{\times}10^5$ cells/ml inoculation density of stromal cells.

  • The present study was conducted to investigate the structural changes in rat cornes. Sixty eyes from one-day-old uneyed rats, fourty eyes from 10-weeks-old adult rats were used. With the increase of age, the epithelial layer was thickened by the addition of new successive cellular layers. Then, the new-born rat's epithelial cells formed a pentagonal shape, and the quality of decidual cells showed a high electron-density, although the boundary between cells was distinctive. The newly produced cells showed a low electron-density so that there was the distinctive difference between light and darkness. In Bowman's layer, collagen fibrils demonstrate a regularly arranged structure along with the age. In stroma's layer, the density of keratocytes was decreased and thereby progressively flattened during the development. The collagenous layer of the adult rats was more distinctive than that of the new-born rats in a form of vertical and horizontal parallel alignment running vertically and horizontally. Descemet's membrane changed its structure significantly along with the age. It changed the shape from "banded-layer" to "non-banded layer" gradually. The thickness was also increased along with the age. Regardless of developmental stages, the endothelium is usually monolayer. During the developmental process, endothelial cells disappeared, so the density of endothelium was also decreased. The empty spaces were replaced by the expansion of adjacent cells.

Exploration of Beneficial Herbal Medicines to Attenuate Particulate Matter-induced Cellular Injury in Human Corneal Epithelial Cells (인간 각막상피세포에서 미세먼지로 인한 세포 손상을 완화할 수 있는 유익한 한약재의 탐색)

  • Particulate matter (PM) is known to be involved in the onset and progression of various diseases by promoting oxidative and inflammatory reactions as air pollutants containing various small particles that are harmful. In this study, the protective efficacy of herbal medicines was evaluated in human corneal epithelial cells (hCECs) to select natural products that can protect the eye, the primary organ directly exposed to external pollutants from PM. As a result, five candid ate herbal medicines [Cheonmundong, Asparagus Rhizome; Seokchangpo, Aciru Gramineri Rhizoma; Hwangryeon, Coptidis Rhizoma; Gamgug, Chrysanthemi Indici Flos; and Geumjanhwa (Marigold flower petals)] which showed inhibitory efficacy on PM2.5-induced cytotoxicity, were selected from among 12 candidate herbal medicines. To evaluate the antioxidant activity of these candidate substances, the reactive oxygen species (ROS) scavenging ability was investigated, and it was found that the extracts of Seokchangpo, Cheonmundong and Hwangryeon showed a significant inhibitory effect on PM2.5-induced ROS production, which was correlated with the preservation of mitochondrial activity. In addition, it was confirmed that they could block DNA damage caused by PM2.5 through analysis of 8-hydroxy-2'-deoxyguanosine generation and phosphorylated-H2A histone family member X (γ- H2AX) expression. Furthermore, the increase in inflammasome activity and inflammatory response in PM2.5-treated hCECs was also canceled in the presence of these extracts. Although additional studies are needed, the results of this study will be used as primary data to find novel natural compounds that protect hCECs from PM.

  • Purpose: We report two cases of corneal edema in patients who presented with bilateral blurry vision due to vaporized amines while working in a polyurethane processing plant. Case summary: A 28-year-old male presented with bilateral blurred vision. His work involved solidifying polyurethane liquid and he often found himself exposed to polyurethane heat and gas. The central corneal thickness (CCT) was also increased in both eyes, measuring $698{\mu}m$ (right) and $672{\mu}m$ (left). After prescribing 0.5% moxifloxacin and, 1% fluorometholone eye drops for 3 days in both eyes, the UCVA recovered to 20/40 (right) and 20/20 (left). The CCT decreased to $644{\mu}m$ (right) and $651{\mu}m$ (left), and the microbullae improved significantly in the left eye. The second patient was a 34-year-old female who presented with bilateral decreased visual acuity while at work. She worked in a factory that produced car seat filling. Her UCVA was 20/25 (right) and 20/20 (left). The CCT by specular microscopy was $537{\mu}m$ (right) and $541{\mu}m$ (left). On slit lamp examination, both eyes demonstrated bilateral central subepithelial edema. The patient did not attend any follow-up outpatient appointments after the initial presentation. Conclusions: Exposure to vaporized amines such as polyurethane may causereversible corneal toxicityeven without direct contact.   • 동양달팽이속(Nesiohelix)에 속하는 종으로 한국에서는 유일하게 서식하고 있는 종으로알려진 동양달팽이(Nesiohelix samarangae)성체의 눈에 대하여 해부학적 및 미세구조관찰을 실시한 결과 다음과 같은 결론을 얻을 수 있었다. 동양달팽이의 눈은 각막, 무세포성의 수정체, 망막및 신경망 등으로 구성되어 있는 바 가장 복잡한 구조를 보이는 부분은 망막으로서 이의상피는 2종의 원주세포로 구성되어있다. 그 하나는 광수용세포로서 세포의 상부세포질이 원추형 또는 피라미드형의 돌기를 이루었고 그 유리표면에는 긴 감간(microvilli)이 무수히 많이 존재하고 있으며, 돌기를 이룬 세포질 내에는 용해소체의 활성이 높게 나타난다. 또한 광수용세포들의 상부세포질에는 이웃해 있는 색소세포들의 세포질 돌기들이 여러개 침입해 들어와 있다. 그리고 핵 주변의 세포질에는 수많은 photic vesicle들이 군집을 이루어 존재한다. 다른 한 종류의 세포는 색소세포들로서 광수용세포들과는 달리 세포의 상부에 원추형의 세포질 돌기가 없고 다만 그 유리표면을 따라 광수용세포의 것보다는 굵고 짧은 많은 감간이 존재하는데 두 종류의 세포들에서 뻗어나온 감간들은 서로 만나 때로 파상을 이루고 있다. 망막 상피세포층의 기저막 아래에는 신경망이 컵모양의 망막을 따라 존재한다.

  • 본 연구에서는 압전체의 횡 진동모드를 이용한 압전 초음파 진동자를 설계 분석하였다. 이전의 란쥬반 진동자는 진동을 얻기 위하여, 복수의 원판 또는 사각판 형태의 압전 세라믹을 서로 반대 방향으로 분극하여 마주 보도록 설치한 후 전기적으로 병렬로 연결하고 상단 및 하단에 금속부을 부착하여 전체를 볼트로 조인 복잡한 구조와 큰 출력 파워를 갚는 반면, 본 연구에서는 판상형의 압전소자와 일체형 금속진동체를 이용하여 기계적 출력 파워 조절이 용이한 구조의 압전 진동자를 고안하여, 압전진동자의 횡 진동 모드를 이용함으로써 신뢰성과 정확도가 높고 진동효율이 최대가 되도록 설계하였다. 설계 개발된 압전 진동자는 진동의 크기 조절이 용이하여 일반 진동자 뿐 만 아니라, 외과 및 안과 수술에 있어서 인체조직이나 각막상피의 활성화를 유지한 상태에서 안전하게 절개 및 분리 시술용 진동자로 사용 할 수 있는 이점이있다. 압전 진동자는 유한요소법 시뮬레이션 프로그램 (ATILA 5.2.4)을 이용하여 설계를 하였으며 압전소자의 두께는 각각 0.2 mm, 0.5 mm로 제작하여 시뮬레이션 결과와 제작된 샘플의 특성을 비교하였고, 변위측정은 칼날을 결합 한 상태에서 공진 주파수대역 부근 주파수별로 측정 비교하였다.

Effect of Environmental Factors on the Growth of Rabbit Oral Keratinocytes (토끼 구강점막 상피세포 성장에 미치는 환경인자의 영향)

  • Isolation and primary culture technique of rabbit oral keratinocytes, and the study for effect of environmental factors on the cell growth were carried out in T75-flask. $1.92{\pm}0.59{\times}10^6$ viable cells were isolated by trypsin enzymatic digestion method from $0.25cm^2$ biopsy of rabbit oral mucosa. Primary culture with 10 mL of K-SFM containing 50 mg/L BPE, $5.0{\mu}g/L$ EGF and 0.15 mM $Ca^{2+}$ showed confluence after 8 days and doubling time was 2.54 days. Effect of medium types, medium volume and supplement types on the cell growth was investigated after the cultured keratinocytes had been harvested from primary confluence. Serum addition showed adverse effect and the increase of serum concentration didn't have an effect on the cell growth. The increase of medium volume decreased the cell growth. The increase of calcium concentration increased the cell growth and 2.0 mM was optimum value. In conclusion, when rabbit oral keratinocytes was cultured in T75-flask, the most effective conditions was to use 10 mL of K-SFM containing 50 mg/L BPE, $5.0{\mu}g/L$ EGF and 2.0 mM $Ca^{2+}$, and doubling time was 1.32 days. This study can provide the useful informations to develop a process and design a bioreactor for the culture of keratinocytes in human body like skin and cornea, as well as mucosa.
