레틴알 안정화를 위한 사이클로덱스트린-리포좀에 관한 연구 (Study on Stabilization of Retinaldehyde using Drug-in-Cyclodextrinin-Liposome (DCL) for Skin Wrinkle Improvement)

  • 레틴알(RA)은 레티놀과 레티노익애씨드의 중간체로 비타민A 유도체이며 주름개선 효과가 우수하다. 본 연구에서는 drug-in-cyclodextrin-in-liposome (DCL)을 이용하여 레틴알의 안정성을 높였다. 레틴알과 hydroxypropyl-β-cyclodextrin (HP-β-CD) 복합체를 동결건조 방식으로 제조하였고, UV-Vis 분광법, FT-IR 및 SEM 이미지로 레틴알의 포접 여부를 확인하였다. 레틴알과 HP-β-CD의 비율이 1 : 15 (w/w)일 때 약 95.6% 포집되었다. 레틴알-HP-β-CD 복합체는 호모믹서 및 마이크로플루다이저로 리포좀에 담지시켰으며, 평균 입자 크기는 215.3 ± 4.2 nm, 제타포텐셜 -33.2 ± 1.5 mv로 나타났다. 레틴알의 분해 안정도 평가에서, 물에서 레틴알-HP-β-CD-리포좀의 레틴알 감소율은 1.8%로 레틴알-리포좀(5.8%), 레틴알-HP-β-CD복합체(9.7%), 레틴알 단독(37.6%)보다 높게 나타났다. 레틴알-HP-β-CD-리포좀이 함유된 크림(0.05% RA 함유)을 제조하여, 미간, 이마, 목, 눈가, 입가, 팔자 주름개선 효능 및 피부 치밀도를 2 ~ 4 주간 평가하였다. 그 결과 레틴알크림은 피부 자극 없이 유의한 주름 개선 효과를 보였다. 결론적으로, DCL시스템을 이용한 이중 안정화 기술은 레틴알의 안정화를 높여 피부 주름 개선 효과에 기여함을 확인하였다.

8주 케톤체 투여가 마우스 지구성 운동수행능력과 골격근의 자가포식에 미치는 영향 (The Effects of 8-week Ketone Body Supplementation on Endurance Exercise Performance and Autophagy in the Skeletal Muscle of Mice)

  • 마우스 모델을 사용하여, 8주 케톤체(베타-하이드록시뷰티레이트, β-HB)가 지구성 운동 수행능력과 골격근의 단백질 합성 및 분해에 미치는 영향을 조사하였다. 48마리의 수컷 ICR 마우스(8주령)를 무작위로 4개 그룹으로 나누었다: 비운동 통제군(Sed+Con), 비운동+베타-하이드록시뷰티레이트(Sed+β-HB), 운동 통제군(Exe+Con), 운동+베타-하이드록시뷰티레이트(Exe+β-.HB). β-HB 투여를 위해 β-HB를 PBS (150 mg/mL)에 용해시켜 8주 동안 매일 피하 주사(250 mg/kg)하였다. 운동 훈련을 위해 마우스는 8주 동안 20분 트레드밀 달리기 운동 훈련을 주 5일 수행하였다. 훈련은 10° 경사도에서 10 m/min 속도에서 5 min 동안 실시하고 나서, 매 1 min 마다 1 m/min의 속도를 나머지 15 min 동안 증가시켰다. 8주간의 처치 후, 내장 지방량과 골격근량, 혈액 매개변수, 자가포식 및 단백질 합성 마커가 분석되었다. 데이터는 SPSS 21 프로그램을 사용하여 ANOVA (p<0.05)로 분석되었다. Exe+β-HB 그룹의 혈중 젖산 수치는 모든 3개 그룹(Sed+Con, Sed+β-HB 및 Exe+β-HB)보다 유의하게 낮았다(p<0.05). Sed+Con 및 Exe+Con 그룹에 비해, 8주 Exe+β-HB 처치는 최대 달리기 수행 시간(탈진 시간)을 유의하게 증가시켰다(p<0.05). 8주 β-HB 투여는 마우스의 골격근에서 자가포식 유동과 자가포식 관련 단백질을 유의하게 감소시켰다(p<0.05). 반대로, β-HB와 지구력 운동 훈련의 조합된 처치는 단백질 합성(mTOR 신호 및 번역 속도)을 증가시켰다(p<0.05). 8주간의 β-HB 처치와 지구력 운동 훈련은 마우스 골격근에서 지구력 수행능력 증가, 단백질 합성 증가, 단백질 분해 감소 효과를 보여주었다.

자소엽(Perillae Folium) 열수추출물의 식물화학성분 연구 (Phytochemical Study of Hot-Water Extract of Perillae Folium)

  • In this study, 15 compounds were elucidated from the hot-water extract of Perillae Folium. Fifteen isolates were determined to be protocatechuic acid (1), caffeic acid (2), (R)-rosmarinic acid (3), (S)-shisoflavanone A (4), luteolin-7-O-β-D-glucuronopyranoside (5), scutellarein-7-O-β-D-glucuronopyranoside (6), apigenin-7-O-β-D-glucuronopyranosyl(1→2)-O-β-D-glucuronopyranoside (7), luteolin-7-O-β-D-glucuronopyranosyl(1→2)-O-β-D-glucuronopyranoside (8), kelampayoside A (9), trans-N-feruloyloctopamine (10), 3-(4-hydroxy-3-methoxyphenyl)-N-[2-(4-hydroxyphenyl)-2-methoxyethyl]acrylamide (11), perilloside C (12), perilloside A (13), (6S,9R)-9-hydroxy-megastigma-4,7-dien-3-one-9-O-β-D-glucopyranoside (14) and (6S,9R)-roseoside (15) through spectroscopic evidences. The HPLC analysis revealed that hot-water extract of Perillae Folium contained caffeic acid, rosmarinic acid and glycosides of apigenin, luteolin and scutellarein as main constituents.

  • The adsorption of CO on W(111) surface in the range of adsorption temperature between 300 K and 1000 K has been studied using AES, LEED, and TDS in an UHV system. After CO saturation at 300 K, four desorption peaks are observed at temperatures (K) of about 400, 850, 1000, and 1100 in thermal desorption spectra, called as α, β1, β2, and β3 state, respectively. The state was attributed to molecular species of CO, which is well known. Because the CO in βstates (especially the β3 state) is still debated as to whether it is dissociative or non-dissociative, the β3 state is mainly discussed. By using the variation method of heating rate in the thermal desorption spectrometry, the desorption energy and pre-exponential factor for the β3 state are evaluated to be 280 kJ/mol and 1.5×10 12 s-1 , respectively. A lateral interaction energy of 5.7 kJ/mol can also be estimated by Bragg-Williams approximation. To interpret the thermal desorption spectra for the β3 state, moreover, those for the model of a first order and a second order desorption are simulated using quasi-chemical approximation. In this study, a model of lying-down CO species is proposed for the β3 state of CO adsorption.

기혼남성의 아버지 역할행동에 관한 연구 (A Study on Paternal Role-Behavior of Married Men)

  • The purpose of this study was to identify the paternal role-behavior. Data were collected from questionnares with 270 fathers at the thirties-forties residing in Seoul. The data were analyzed with the SPSS WIN 7.0 package. The major findings were as follows: (1) The general trends of paternal role-behavior showed relatively high(mean=3.60) (2) Paternal role-behavior differed significantly according to their age education level occupation among the demograpic variables (3) Paternal role-behavior differed significantly according to age of oldest child number of sons number of daughters among the family-environmental variables. (4) Paternal role-behavior was correlated with the perceptions on this own father's ways of rearing marital satisfaction job satisfaction experience of bringing up child (5) The result of the regression analysis for the paternal role-behavior the seven variables most affected were as follows; father's role perception(β=3.6) age of oldest child (β= -1.9) experience of bringing up child(β=.19) perception on his own father's nurturance(β=.16) number of child(β=-.16) number of sons(β=-.13) level of income(β=.12)

Transforming growth factor-beta and liver injury in an arginine vasopressin-induced pregnant rat model

  • Objective: Approximately 30% of preeclamptic pregnancies exhibit abnormal liver function tests. We assessed liver injury-associated enzyme levels and circulating transforming growth factor beta (TGF-β) levels in an arginine vasopressin (AVP)-induced pregnant Sprague-Dawley rat model. Methods: Pregnant and non-pregnant Sprague-Dawley rats (n=24) received AVP (150 ng/hr) subcutaneously via mini-osmotic pumps for 18 days. Blood pressure was measured, urine samples were collected, and all animals were euthanized via isoflurane. Blood was collected to measure circulating levels of TGF-β1-3 isomers and liver injury enzymes in pregnant AVP (PAVP), pregnant saline (PS), non-pregnant AVP (NAVP), and non-pregnant saline (NS) rats. Results: The PAVP group showed significantly higher systolic and diastolic blood pressure than both saline-treated groups. The weight per pup was significantly lower in the AVP-treated group than in the saline group (p<0.05). Circulating TGF-β1-3 isomer levels were significantly higher in the PAVP rats than in the NS rats. However, similar TGF-β1 and TGF-β3 levels were noted in the PS and PAVP rats, while TGF-β2 levels were significantly higher in the PAVP rats. Circulating liver-type arginase-1 and 5'-nucleotidase levels were higher in the PAVP rats than in the saline group. Conclusion: This is the first study to demonstrate higher levels of TGF-β2, arginase, and 5'-nucleotidase activity in PAVP than in PS rats. AVP may cause vasoconstriction and increase peripheral resistance and blood pressure, thereby elevating TGF-β and inducing the preeclampsia-associated inflammatory response. Future studies should explore the mechanisms through which AVP dysregulates liver injury enzymes and TGF-β in pregnant rats.

Enhancement of paclitaxel-induced breast cancer cell death via the glycogen synthase kinase-3β-mediated B-cell lymphoma 2 regulation

  • Glycogen synthase kinase-3β (GSK-3β) is a serine/threonine protein kinase that is known to mediate cancer cell death. Here, we show that B-cell lymphoma 2 (Bcl-2), an anti-apoptotic protein, is regulated by GSK-3β and that GSK-3β-mediated regulation of Bcl-2 is crucial for mitochondrial-dependent cell death in paclitaxel-stimulated cells. We demonstrate that MCF7 GSK-3β siRNA cells are more sensitive to cell death than MCF7 GFP control cells and that in the absence of GSK-3β, Bcl-2 levels are reduced, a result enhanced by paclitaxel. Paclitaxel-induced JNK (c-Jun N-terminal kinase) activation is critical for Bcl-2 modulation. In the absence of GSK-3β, Bcl-2 was unstable in an ubiquitination-dependent manner in both basal- and paclitaxel-treated cells. Furthermore, we demonstrate that GSK-3β-mediated regulation of Bcl-2 influences cytochrome C release and mitochondrial membrane potential. Taken together, our data suggest that GSK-3β-dependent regulation of Bcl-2 is crucial for mitochondria-dependent cell death in paclitaxel-mediated breast cancer therapy. Decrease of glycogen synthase kinase 3β phosphorylation in the rat nucleus accumbens shell is necessary for amphetamineinduced conditioned locomotor activity

  • Phosphorylation levels of glycogen synthase kinase 3β (GSK3β) negatively correlated with psychomotor stimulant-induced locomotor activity. Locomotor sensitization induced by psychomotor stimulants was previously shown to selectively accompany the decrease of GSK3β phosphorylation in the nucleus accumbens (NAcc) core, suggesting that intact GSK3β activity in this region is necessary for psychomotor stimulants to produce locomotor sensitization. Similarly, GSK3β in the NAcc was also implicated in mediating the conditioned effects formed by the associations of psychomotor stimulants. However, it remains undetermined whether GSK3β plays a differential role in the two sub-regions (core and shell) of the NAcc in the expression of drug-conditioned behaviors. In the present study, we found that GSK3β phosphorylation was significantly lower in the NAcc shell obtained from rats expressing amphetamine (AMPH)-induced conditioned locomotor activity. Further, we demonstrated that these effects were normalized by treatment with lithium chloride, a GSK3β inhibitor. These results suggest that the behavior produced by AMPH itself and a conditioned behavior formed by associations with AMPH are differentially mediated by the two sub-regions of the NAcc.

Characteristics of high β-glucan resources quality of Barley

  • β-glucan, a nonstarch polysaccharide, is one of the main functional component in the cell wall of barley. This study was quality characteristics to use a korean variety with a high β-glucan as an original material for developing functional food. The high β-glucan 'Jeonju528, and 'Betaone' were compared with 'Hyeyang', 'Dahyang', 'Hwanggeumchal' and 'Glacier AC38' total 6 varieties. Seed section dyed to classify of Waxy/non-waxy type, starch granule was tested and moisture, protein, amylose, and β-glucan of whole grains and pearl barleys were experiment. Whole grains were the average protein of 13.2% and were the average starch 50.1%. β-glucan of whole grains were 5.3-10.0%, and amylose were 3.0-23.4%. Pearl barleys were the average protein of 11.7% and were the average starch of 65.0%. β-glucan of pearl barleys were 6.5-12.3%, and amylose were 3.6-31.1%. As a results of the correlation analysis were recognized significance among varieties for protein, starch and β-glucan but there was no difference in other traits. It was concluded that amylose showed a positive correlation with starch and β-glucan showed a negative correlation with amylose.

  • Diabetes Mellitus (DM) is a major health issue increasing worldwide. Currently, nearby half a billion people have diabetes. Two major types of DM that type 1 and type 2-DM have different etiologies but feature a crucial common pathological transition into dysfunction of pancreatic β-cells and consequently leading hyperglycemia and finally go into DM. Therefore, maintaining of β-cells such as preventing β-cells degeneration, and promoting β-cells regeneration and proliferation will be essential approaches in prevention and/or treatment of DM. There are many reports that various types of seaweed control metabolic diseases such as obesity, high blood pressure, and blood sugar control. However, no new drug candidates have been developed yet. Additionally, although seaweed has excellent blood sugar control effects, there is no evidence that it directly proliferates or regenerates beta cells. Therefore, we studied on the promotion of β-cell regeneration by a seaweed, Polysiponia morrowii extract (PME) which preserves β-cells and maintains its function. As a result, it was confirmed that PME directly promotes the proliferation of pancreatic islet β-cells with insulin secretion function in in vivo. Therefore, PME shows potential as a candidate for β-cell regeneration that may play a fundamental role in the treatment of diabetes.
