친수성 L-ascorbic acid와 소수성 α-tocopherol을 모두 포집하는 ethosome의 특성 (Coencapsulation of L-Ascorbic Acid and α-Tocopherol in Ethosomes and Their Properties)

  • 친수성 아스코르빈산(L-ascorbic acid)과 소수성 토코페롤(${\alpha}$-tocopherol)을 모두 함유하는 ethosome 베시클을 제조하면서 이들의 특성을 살펴보았다. Ethosome에 포집되는 아스코르빈산 수용액 농도가 증가할수록 베시클 입자 크기는 작아졌다. 토코페롤은 HPC에 혼합되어 ethosome 베시클 액정막의 구성 성분이 되는데 토코페롤 혼합에 의해 베시클 입자크기와 아스코르빈산 수용액의 포집효율이 다소 증가하였다. 그러나 혼합비율이 25 wt% 이상이 되면 베시클 막이 불완전한 액정구조로 변하면서 입자크기는 크게 증가하고 포집효율은 크게 감소하였다. 아스코르빈산과 토코페롤이 함께 ethosome에 포집되어 있을 때 항산화 효능이 고온($40^{\circ}C$)에서도 5주간 안정하게 유지됨을 확인하였다. UV 조사 실험에서도 아스코르빈산은 수용액 상태로 있을 때에 비해 ethosome에 포집되어 있을 때 안정성이 향상되었다.

쥐(rats)의 full-thickness skin grafts에 대한 hyperbaric oxygen과 α-tocopherol의 효과 (Effects of hyperbaric oxygen and α-tocopherol on full-thickness skin grafts in rats)

  • To document that effects of hyperbaric oxygen(HBO) and ${\alpha}-tocopherol$ on full-thickness skin grafts in rat, we performed full-thickness skin grafts bilaterally on each rats. The HBO-treated rats were received HBO twice daily for 90 minutes at 2 ATA. Surgical control rats were not treated with HBO. ${\alpha}-tocopherol$ treated rats were received the agent via oral gastric tube daily for 3 days preoperative and a fourth dose 1 to 2 hours postoperative. HBO plus ${\alpha}-tocopherol$ treated rats were received HBO and ${\alpha}-tocopherol$ as mentioned above. Biopsy specimens were taken from each rat at the time of grafting and on days 2, 4, 7, 10, 14, 21, and 28, then were processed for tissue-concentration of total glutathione(GSHt), oxidized/reduced glutathione level, and thiobarbituric acid-reactive substance(TBARS) levels. The percentage of viable graft on day 10 ranged from 67 to 93%, and was not significantly different among the each other groups. The percentage of viable graft were, however, higher in HBO plus ${\alpha}-tocopherol$ treated rats(78.6%) than in HBO alone treated rats(59.1%), ${\alpha}-tocopherol$ alone treated rats(66.7%) and surgical control rats(58.2%). TBARS concentration had a significant increase from preoperative concentration at day 2, and peak concentration at day 4(p<0.01). Concentration then decreased to preoperative concentration at day 28. GSHt concentration of free skin graft had a similar patteren of change in four groups and decreased significantly from preoperative concentration at day 2, returning to preoperative concentration by day 7(surgical control, HBO-treated, and ${\alpha}-tocopherol-treated$, alone) and 28(HBO plus ${\alpha}-tocopherol-treated$). Percentage of the concentration of reduced glutathione decreased in surgical control, HBO-treated and, ${\alpha}-tocopherol-treated$(p<0.05), and HBO plus ${\alpha}-tocopherol-treared$(p<0.01) on day 7 after surgery, whereas the concentration of oxidized increased significantly in HBO-treated(p<0.05), ${\alpha}-tocopherol-treated$(p<0.05), and HBO plus ${\alpha}-tocopherol-treated$(p<0.01).

  • 지용성 토코페롤(${\alpha}$-tocopherol)의 산화 안정성을 높이기 위하여 nanostructured lipid carrier (NLC)에 봉입을 시도하였다. 먼저 구성 성분과 혼합 비율을 달리한 여러 NLC를 만들고 각각의 특성을 살펴보았다. 고체 지질로 세틸 팔미테이트 또는 글리세릴 모노스테아레이트를 사용했을 때는 안정한 상태의 NLC 입자가 만들어지지만 스테아린 산으로 만든 NLC에서는 상분리가 일어났다. NLC 입자는 수백 나노미터 크기로 만들어지는데 지질대비 용매의 비율이 높을수록 입자크기가 작게 나타났다. 고급 지방알콜인 옥틸도데칸올을 고체 지질에 첨가하면 결정화도가 감소하면서 지질 매트릭스의 배열규칙성 이 약해짐을 DSC (Differential Scanning Calorimetry) 열차트와 anisotropy 측정을 통해서 확인하였다. 고온($45^{\circ}C$) 또는 UV 빛이 조사되는 조건에서 토코페롤이 NLC에 봉입되어 있으면 용액이나 에멀젼 상태로 있는 것에 비해 산화 안정성이 크게 향상됨을 DPPH 테스트와 과산화물가 측정으로 확인하였다.

Utilization of Dried Garlic Powder and α-Tocopherol to Improve the Shelf-life of Emulsion-type Sausage during Refrigerated Storage

  • This study investigated the shelf life of emulsion-type sausages containing garlic powder and/or ${\alpha}$-tocopherol during storage at $4^{\circ}C$ for 0, 10, 20, and 30 d. Six groups of emulsion-type sausages were included: control (no additives), GP1 (1% garlic powder), GP3 (3% garlic powder), AT100 (100 IU of ${\alpha}$-tocopherol/kg of sausage), AT200 (200 IU of ${\alpha}$-tocopherol/kg of sausage), and GP1+AT100 (1% garlic powder+100 IU of ${\alpha}$-tocopherol/kg of sausage). During storage, the pH, thiobarbituric acid reactive substances, and residual nitrite content were reduced by the addition of garlic powder and/or ${\alpha}$-tocopherol relative to the control (p<0.05). In addition, emulsion-type sausages supplemented with garlic powder and/or ${\alpha}$-tocopherol improved color stability (p<0.05). The results suggest that a higher amount of garlic powder and their different combinations could improve the shelf life of emulsion-type sausages and protect against lipid oxidation.

A Comparison of Natural (D-α-tocopherol) and Synthetic (DL-α-tocopherol Acetate) Vitamin E Supplementation on the Growth Performance, Meat Quality and Oxidative Status of Broilers

  • The present study was conducted to compare the supplementation of natural (D-${\alpha}$-tocopherol) and synthetic (DL-${\alpha}$-tocopherol acetate) vitamin E on the growth performance, meat quality, muscular antioxidant capacity and genes expression related to oxidative status of broilers. A total of 144 1 day-old Arbor Acres broiler chicks were randomly allocated into 3 groups with 6 replicates of 8 birds each. Birds were given a basal diet (control group), and basal diet supplemented with either 20 IU D-${\alpha}$-tocopherol or DL-${\alpha}$-tocopherol acetate for 42 days, respectively. The results indicated that treatments did not alter growth performance of broilers (p>0.05). Compared with the control group, concentration of ${\alpha}$-tocopherol in the breast muscle was increased by the supplementation of vitamin E (p<0.05). In the thigh, ${\alpha}$-tocopherol content was also enhanced by vitamin E inclusion, and this effect was more pronounced in the natural vitamin E group (p<0.05). Vitamin E supplementation increased the redness of breast (p<0.05). In the contrast, the inclusion of synthetic vitamin E decreased lightness of thigh (p<0.05). Dietary vitamin E inclusion reduced drip loss at 24 h of thigh muscle (p<0.05), and this effect was maintained for drip loss at 48 h in the natural vitamin E group (p<0.05). Broilers given diet supplemented with vitamin E showed decreased malondialdehyde (MDA) content in the breast (p<0.05). Additionally, natural rather than synthetic vitamin E reduced MDA accumulation in the thigh (p<0.05). Neither natural nor synthetic vitamin E supplementation altered muscular mRNA abundance of genes related to oxidative stress (p>0.05). It was concluded that vitamin E supplementation, especially the natural vitamin E, can enhance the retention of muscular ${\alpha}$-tocopherol, improve meat quality and muscular antioxidant capacity of broilers.

국가표준식품성분표 개정을 위한 국내 다소비 해조류 및 채소류의 비타민 E 및 K 분석 (Analysis of vitamin E and K contents in sea algae and vegetables frequently consumed in Korea for National Standard Food Composition Database)

  • 본 연구에서는 국내에서 소비되고 있는 해조류와 채소류의 비타민 E와 K 함량을 분석하고, 사용된 분석법의 유효성과 분석품질관리를 통해 분석 데이터의 신뢰성을 확보하여 식품 영양성분 데이터베이스 구축을 위한 자료를 제공하고자 하였다. 해조류의 비타민 E는 α-tocopherol만 검출되었으며 이들의 활성은 0.15-1.14 mg α-TE/100 g으로 해조류의 종류와 조리 방법에 따라 다르게 나타났다. 해조류에서 비타민 E는 건조된 매생이(dried)가 1.14 mg α-TE/100 g으로 가장 높은 활성을 나타내었으며, 비타민 K는 건조 미역이 1,629.50 ㎍/100 g으로 가장 높은 함량을 나타내었다. 채소류의 경우, 비타민 E 활성은 0.01-2.48 mg α-TE/100 g으로 나타났으며, 가열처리 방법에 따라 비타민 E 함량이 변화하였다. 채소류의 비타민 K1 함량은 16.15-979.60 ㎍/100 g의 범위를 나타내었다. 비타민 K1 은 초록 잎을 가진 시료에서 주로 검출되었으며, 뿌리 채소에서는 검출되지 않았다. 본 연구에서 사용된 비타민 E와 K의 분석법에 대한 검증은 정확성, 정밀성, 직선성, 검출한계, 정량한계를 평가하였으며, 분석품질관리를 통하여 분석 기간 동안 모든 시료에 대한 분석이 신뢰성 있게 진행되었음을 확인하였다. 따라서 본 연구에서 분석된 해조류와 채소류의 비타민 E와 K 데이터는 국가표준식품성분표 개정을 위한 기초자료로 이용될 수 있을 것으로 판단된다.

항산화 성분을 함유한 PE 필름이 반건오징어 저장 중 지질산화에 미치는 영향 (Preparation of Antioxidative Polyethylene Film and Its Effects on the Lipid Oxidation of Semi-dried Squid during Storage)

  • Antioxidative polyethylene films for food packaging were experimentally prepared by fortification of TBHQ(tertiary butylhydroquinone) and dl-a-tocopherol at the weight ratio of 0.05, 0.2 and 0.5%, respectively and laminated with nylon. TBHQ and tocopherol contents in the PE/nylon film were analyzed and the antioxidative effects of film were investigated on the lipid oxidation of semi-dried squid during storage at 5℃ and l5℃. TBHQ contents of TAP 1(TBHQ 0.05%), TAP 2(TBHQ 0.2%) and TAP 3(0.5%) were 38, 146 and 365 mg/100g, respectively. Tocopherol contents of DAP l(tocopherol 0.05%), DAP 2(tocopherol 0.2%) and DAP 3(tocopherol 0.5%) were 33, 139 and 356 mg/l00g, respectively. TBA value and POV during storage of semi-drid squid were affected both by storage temperatures and packaging films. Lipid oxidation during storage was retarded by anti-oxidative films, as TBA value of TAP 3 and DAP 3 revealed about 50% of control after storage at 5℃ for 20 days and similer effects in POV were also observed.

Relationships between Muscle α-Tocopherol Concentrations and Metmyoglobin Percentages during Display of Six Muscles of Japanese Black Steers

  • Relationships between muscle $\alpha$-tocopherol oncentrations and metmyoglobin percentages during display of six muscles, m. serratus ventralis (SV), m. psoas major (PM), m. gluteus medius (GM), m. semimembranosus (SM), m. semitendinosus (ST) and m. longissimus lumborum (LL), of Japanese Black steers slaughtered at 28 months of age were studied. Steers were supplemented with 0, 2,000 and 4,000 mg $\alpha$-tocopheryl acetate/head/day for 28 days prior to slaughter in the VE 0, the VE 2,000 and the VE 4,000 groups, respectively. $\alpha$-Tocopherol concentrations in PM, GM, SM, ST and LL of the VE 2,000 and the VE 4,000 groups were significantly (p<0.05) higher than those of the VE 0 group. There were no significant (p>0.05) differences in $\alpha$-tocopherol concentrations in all muscles between the VE 2,000 group and the VE 4,000 group. The muscle $\alpha$-tocopherol concentrations ($\ell$/g meat) which can retard metmyoglobin formation in muscles were estimated to be 5.3 for SV, 4.5 for PM, 4.2 for GM, 4.0 for SM, 3.6 for ST and 3.5 for LL. The equation to predict color-shelf-life of each muscle from the $\alpha$-tocopherol concentration in each muscle could be obtained.

방사선 조사에 의한 감귤의 β-Carotene, 멸치의 비타민 D3 및 쇠고기의 α-Tocopherol의 함량변화 (Effects of Gamma Irradiation on the Content of β-Carotene in Jeju Orange, Vitamin D3 in Anchovy and α-Tocopherol in Beef)

  • 방사선 조사식품의 지용성 비타민에 대한 안정성을 확인하기 위하여 지용성 비타민 중 $\beta-carotene$, 비타민 $D_3$, $\alpha-tocopherol$을 택하여 한국인의 상용 식품이면서 각각의 급원식품인 귤, 멸치, 쇠고기에 대해 방사선조사 전후의 해당 비타민 함량을 분석하였다. 귤의 $\beta-carotene$의 함량은 조사구가 비조사구에 비해 약간 증가하는 경향을 보였으나 유의적이지는 않았다. 멸치의 비타민 $D_3$ 함량은 조사선량에 비례하여 5kGy 조사 시 $109.30\%$, 10kGy 조사 시 $125.25\%$로 증가하였다. (p<0.05). 소고기의 $\alpha-tocopherol$ 함량은 고지방 부위인 등심육의 경우 조사선량에 따라 큰 폭으로 감소되어 비조사구에 비해 10kGy 조사구에서 $84\%$의 감소율을 보였으나, 지방함량이 비교적 적은 사태육의 경우 조사구 모두에서 비조사구 대비 약 $13\%$의 감소율을 보였다. 순수 용액의 비타민 A, 비타민 $D_3,\;\alpha-tocopherol$은 조사선량에 비례하여 함량이 감소하는 것으로 나타났으며 특히, 비타민 $D_3$는 지용성 비타민 중 조사에 가장 안정한 비타민으로 나타났고, $\alpha-tocopherol$의 경우는 10kGy 처리구에서 전량 감소를 보여 지용성 비타민 중 조사에 가장 취약한 비타민으로 나타났다. 마이크로웨이브 처리구나 가열 처리구에서는 이들 지용성 비타민들의 손실이 거의 없었다. 이사의 결과로부터 10kGy이내의 방사선 조사 시, 귤의 $\beta-carotene$ 함량에는 영향을 미치는 않는 것으로 보이며, 비타민 $D_3$의 경우는 적어도 10kGy까지는 매우 안정함을 보여주었고 지방함량이 높은 등심육의 경우 $\alpha-tocopherol$의 상당한 분해가 일어남을 알 수 있었다.

Free radical에 의한 지질과산화와 단백질산화에 대한 α-tocopherol의 항산화효과 (The antioxidative effects of α-tocopherol on the lipid peroxidation and protein oxidation by free radicals)

  • This study was carried out to investigate the inhibitory effects of vitamin E on the oxidative damage of cellular lipids and proteins in free radical reaction induced by $FeCl_3$, and ascorbic acid. In this experiment, a vitamin E treated rat group was administered with 100mg/kg body weight of $dl-{\alpha}-tocophery$ 1 acetate and an untreated rat group was administered with the same volume of corn oil. And then assays of malondialdehyde and carbonyl group in total homogenate, mitochondrial and microsomal fraction of rat liver were carried out at the scheduled time. The results obtained from this study were summarized as follows; 1. Lipid peroxidation levels in vitamin E administered rat liver cells were significantly (p<0.05) decreased at the intervals between 1 hour and 4 hours in liver homogenate, at all times except for 1 hour point in mitochondrial fraction, and also at the intervals between 0.5 hour and 3 hours in microsomal fraction compared with those of the control rat liver cell. 2. Protein oxidation levels in vitamin E administered rat liver cell were also significantly (p<0.05) decreased at the intervals between 1.5 hours and 4 hours in liver homogenate, at over 4 hours in liver mitochondrial fraction, and at the intervals between 0.5 hour and 3 hours in liver microsomal fraction compared with those of the control rat liver cells.
