Activity and application of 20α-hydroxysteroid dehydrogenase in rat 1.   • Progesterone catabolizing enzyme, the enzyme $20{\alpha}$-hydroxysteroid dehydrgenase($20{\alpha}$-HSD) is pivotal in the regulation of ovarian function in rodents, which catabolizes progesterone into biologically inactive $20{\alpha}$-hydroxypregn-4-en-3-one($20{\alpha}$-OHP). In this study was carried out the influence of $20{\alpha}$-HSD activity of ovarian function, we investigated changes in ovarian cytosol $20{\alpha}$-HSD activity and serume progesterone concentration during the estrous cycles and pregnancy in rat. During the estrous cycles, the $20{\alpha}$-HSD activities were highest on the progestrous, but serum progesterone concentration was lowest on this phase. During the pregnancy, the $20{\alpha}$-HSD activities were relatively higher early pregnancy(day-1-3 gestation) and late pregnancy(day 21 to parturition), serum progesterone concentration was maintained significantly high to day 19 of gestation. The $20{\alpha}$-HSD activities were lower during the middle pregnancy. From these results, ovarian $20{\alpha}$-HSD activities may possibly act as physiologically very important in the control and maintenance of estrous cycles in rat.

Comparative Analysis of $\alpha$-glucosidase Activity in Bombyx mori and Antheraea yamamai

  • [ $\alpha$ ]Glucosidase (EC 3.2.1.20) is a glycosidase that hydrolyzes disaccharides, oligosaccharides, and polysaccharides resulting in the release of α-D-glucose. In this study, $\alpha$-glucosidase activity in the hemolymph and midgut of the mulberry silkworm Bombyx mori and Japanese oak silkmoth Antheraea yamamai was measured using maltose, sucrose, trehalose, and p-nitrophenyl $\alpha$-D-glucopyranoside as substrates. In general, hemolymph $\alpha$-glucosidase activity was higher in B. mori than in A. yamamai. In contrast, midgut $\alpha$-glucosidase activity was higher in A. yamamai than in B. mori for all of the substrates tested. $\alpha$-Glucosidase activity in the midgut of both B. mori and A. yamamai showed similar responses to changes in pH and temperature for all of the substrates tested. Native (7.5%) PAGE of hemolymph and midgut proteins from B. mori and A. yamamai followed by staining with 4-methylumbelliferyl $\alpha$-D-glucoside (MUG) indicated that the $\alpha$-glucosidases of these related lepidopterans are functionally similar but structurally different. In comparison to $\alpha$-glucosidase activity from A. yamamai, $\alpha$-glucosidase activity from B. mori was generally less sensitive to the $\alpha$-glucosidase inhibitors, 1-deoxynojirimycin (DNJ), acarbose, and voglibose when the activity was determined using maltose, sucrose, and trehalose.

  • Four thiazolidines and following five new compounds were prepared by the addition reaction of cysteine and thiophenol to ${\alpha}$,N-diphenylnitrones, respectively ; ${\alpha}$,thiophenoxy-benzylidene aniline ;${\alpha}$,thiophenoxy-p-hydroxybenzylidene aniline ; ${\alpha}$,thiophenoxy-p-chlorobenzylidene aniline ;${\alpha}$,thiophenoxy-p-methoxybenzylidene aniline ; ${\alpha}$,thiophenoxy-p-nitrobenzylidene aniline. The structure of these compounds were confirmed by the elemental analysis, UV-, IR-and NMR-spectra.

  • In order to establish a biochemical genetic system in Drosophila populations in Korea, the $\\alpha$-glycerophosphate dehydrogenase alleles of eleven natural populations of D. melanogaster in Korea were examined by means of agarose gel electrophoresis. The results obtained are presented below: 1. $\\alpha$-Glycerophosphate dehydrogenase ($\\alpha$-GPDH) allele is scored for eleven natural populations of D. melanogaster in Korea, resulting that $\\alpha$-GPDH is found to be widely polymorphic for two electrophoretic variants. 2. The heterozygosity of $\\alpha$-GPDH is calculated as $40\\sim50%$. 3. The frequency of the FF genotype of $\\alpha$-GPDH is found to be roughly same as the SS genotype, but less than the FS genotype. 4. The F gene of $\\alpha$-GPDH is distributed almost frequently as the S gene.

The Sensory Evaluation of Bread with Added $\alpha$-Azuki bean Powder for Manufacturing Small Red Bean Bread by Balanced Incomplete Block Design ($\alpha$-팥 분말 첨가 식빵 제조를 위한 균형된 불완비 블록법에 의한 관능 평가)

  • This study was prepared to optimize $\alpha$-azuki bean powder content by sensory evaluation method when manufacturing bread with added $\alpha$-azuki bean powder. These sensory characteristics were designed to investigate sensory evaluation about appearance, color, texture, taste and overall acceptability of small red bean bread by balanced incomplete block design. According to evaluated mean of adjusted treatments, appearance was represented high value in bread with added 3% and 6% $\alpha$-azuki bean powder than bread without $\alpha$-azuki bean powder. Bread without $\alpha$-azuki bean powder was revealed maximum sensory score value of color among whole treatments and vread with added 6% $\alpha$-azuki bean powder was revealed second highest sensory value. As $\alpha$-azuki bean powder content increased, sensory score of texture, taste and overall acceptability increased. and bread added 12% $\alpha$-azuki bean powder revealed maximum sensory score. On the results of this research about sensory characteristics for manufacturing bread with added $\alpha$-azuki bean powder, bread with added $\alpha$-azuki bean powder was considered optimum when12% $\alpha$-azuki bean powder was added wheat flour because of highest score of texture, taste and overall acceptability in spite of lower score of appearance and color.

TNF-α in the Pleural Fluid for the Differential Diagnosis of Tuberculous and Malignant Effusion (결핵성 및 악성흉수의 감별에 있어 흉수 내 TNF-α의 유용성)

  • Background : Determining the cause of an exudative pleural effusion is sometimes quite difficult, especially between malignant and tuberculous effusions. Twenty percent of effusions remain undiagnosed even after a complete diagnostic evaluation, including pleural biopsy. The activity of tumor necrosis factor-alpha (TNF-${\alpha}$), which is the one of proinflammatory cytokines, is increased in both infectious and malignant effusions. The aim of this study was to investigate the diagnostic efficiency of TNF-${\alpha}$ activity in distinguishing tuberculous from malignant effusions. Methods : 46 patients (13 with malignant pleural effusion, 33 with tuberculous pleural effusion) with exudative pleurisy were included. TNF-${\alpha}$ concentrations were measured in the pleural fluid and serum samples using an enzyme-linked immunosorbent assay (ELISA). In addition, TNF-${\alpha}$ ratio (pleural fluid TNF-${\alpha}$ : serum TNF-${\alpha}$) was calculated. Results : TNF-${\alpha}$ concentration and TNF-${\alpha}$ ratio in the pleural fluid were significantly higher in the tuberculous effusions than in the malignant effusions (p<0.05). However, the serum levels of TNF-${\alpha}$ in the malignant and tuberculous pleural effusions were similar (p>0.05). The cut off points for the pleural fluid TNF-${\alpha}$ level and TNF-${\alpha}$ ratio were found to be 136.4 pg/mL and 6.4, respectively. The sensitivity, specificity and area under the curve were 81%, 80% and 0.82 for the pleural fluid TNF-${\alpha}$ level (p<0.005) and 76%, 70% and 0.72 for the TNF-${\alpha}$ ratio (p<0.05). Conclusion : We conclude that pleural fluid TNF-${\alpha}$ level and TNF-${\alpha}$ ratio can distinguish a malignant pleural effusion from a tuberculous effusion, and can be additional markers in a differential diagnosis of tuberculous and malignant pleural effusion. The level of TNF-${\alpha}$ in the pleural fluid could be a more efficient marker than the TNF-${\alpha}$ ratio.

Study on Progesterone $6{\alpha}-Steroid$ Hydroxylase from New-born Rat Liver (신생 쥐 간의 Progesterone $6{\alpha}-Steroid$ Hydroxylase에 대한 연구)

  • Five subcellular fractions were obtained by successive centrifugation from the liver of rats within 6 hours of life and characterized by comparing marker compound or marker enzyms. After incubating $3{\beta}$-hydroxy-$5{\alpha}$-pregnan-20-one with the each fraction, the steroids were analyzed by TLC, GLC and GC-MS. A $6{\alpha}$-hydroxylase which hydroxylizes the tetra-hydrogenated compound of progesterone, $3{\beta}$-hydroxy-$5{\alpha}$-pregnan-20-one, was localized in the crude plasma membrane fraction, but not in the microsome fraction. The maximum 6α-hydroxylation was observed at pH 7.0. While this 6α-steroid hydroxylase was not able to hydroxlyze the progesterone, the $3{\alpha}$-isomer was hydroxylized at the $6{\alpha}$-position.

  • E. M. Silvia introduced the class $S^\lambda_\alpha$ of $\alpha$-spirallike functions f(z) satisfying the condition $$ (A) Re[(e^{i\lambda} - \alpha) \frac{zf'(z)}{f(z)} + \alpha \frac{(zf'(z))'}{f'(z)}] > 0, $$ where $\alpha \geq 0, $\mid$\lambda$\mid$ < \frac{\pi}{2}$ and $$\mid$z$\mid$ < 1$. We will generalize Silvia class of functions by formally replacing f(z) in the denominator of (A) by a spirallike function g(z). We denote the new class of functions by $Y(\alpha,\lambda)$. In this note we obtain some results for the class $Y(\alpha,\lambda)$ including integral representation formula, relations between our class $Y(\alpha,\lambda)$ and Ziegler class $Z_\lambda$, the radius of convexity problem, a few coefficient estimates and a covering theorem for the class $Y(\alpha,\lambda)$.

Polyclonal Antibody to a 37-kDa Recombinant Protein Derived from Bovine $20{\alpha}$-Hydroxysteroid Dehydrogenase

  • We prepared the polyclonal antibody anti-$20{\alpha}$-hydroxysteroid dehydrogenase (anti-$20{\alpha}$-HSD) against the recombinant full-length protein bovine $20{\alpha}$-HSD in Escherichia coli. The specificity of anti-$20{\alpha}$-HSD was demonstrated using Chinese hamster ovary (CHO) cells transfected with recombinant bovine $20{\alpha}$-HSD and bovine placental tissues. According to western blot analysis, anti-$20{\alpha}$-HSD specifically recognizes the 37-kDa protein bovine $20{\alpha}$-HSD. The protein is not present in untransfected CHO cells. Anti-$20{\alpha}$-HSD also recognizes a specific protein in the ovaries and placenta of other animals. Immunostaining was used to detect expression of bovine $20{\alpha}$-HSD protein in the cultured luteal cells during the estrous cycle later.

An additive mapping $d:R{\rightarrow}R$ is called an $({\alpha},{\beta})^*$-derivation of R if $d(xy)=d(x){\alpha}(y^*)+{\beta}(x)d(y)$ is fulfilled for any $x,y{\in}R$, and an additive mapping $F:R{\rightarrow}R$ is called a generalized $({\alpha},{\beta})^*$-derivation of R associated with an $({\alpha},{\beta})^*$-derivation d if $F(xy)=F(x){\alpha}(y^*)+{\beta}(x)d(y)$ is fulfilled for all $x,y{\in}R$. In this note, we intend to generalize a theorem of Vukman [12], and a theorem of Daif and El-Sayiad [6], moreover, we generalize a theorem of Ali et al. [4] and a theorem of Huang and Koc [9] related to generalized Jordan triple $({\alpha},{\beta})^*$-derivations.
