Effect of Application of Rice Bran Extract on Quality of Agaricus bisporus during Storage (쌀겨추출물을 적용한 양송이의 저장 중 품질 변화)

  • Postharvest browning of mushroom (Agaricus bisporus) reduces the shelf life of harvested mushrooms. Here, mushrooms were dipped in various solutions (distilled water; DW, 0.25% rice bran extract; RB, 0.1% ascorbic acid; AA, RB + AA) for 3 min. After air-drying at room temperature, the dipped mushrooms were packaged in a polypropylene (PP) films and stored at 4 or $15^{\circ}C$. The quality changes of mushrooms were measured in terms of color, gas composition, firmness, and sensory evaluation during storage. Rice bran extract was measured for total polyphenol content, total flavonoid content, DPPH, ABTS radical scavenging, chelating activity and PPO inhibition activity. No difference in firmness were found in the mushroom samples regardless of dipping solution or storage temperature. At both 4 and $15^{\circ}C$ storage temperatures, RB + AA solution-dipped samples showed the highest L value and lowest delta E value. During the storage period, sensory evaluation showed that overall acceptability of mushrooms treated with RB and RB + AA solution was higher than that of the untreated mushrooms. Total polyphenol and flavonoid contents of 0.25% rice bran extract were $36.42mg\;GAE{\cdot}g^{-1}$ and $4.85mg\;QE{\cdot}g^{-1}$, respectively. The DPPH and ABTS radical scavenging activity of 0.1% ascorbic acid was higher than that of 0.25% rice bran extract. The highest copper ($Cu^{2+}$) chelating activity was found in the 0.25% rice bran extract. The PPO inhibition activity of 0.1% ascorbic acid was higher than that of 0.25% rice bran extract. Our results suggest that 0.25% rice bran extract with 0.1% ascorbic acid is effective anti-browning agent for maintaining quality of Agaricus bisporus during storage.

The Cross-talk Mechanisms of Constitutive Androstane Receptor (CAR) in the Regulation of its Activity, Energy Metabolism, Cellular Proliferation and Apoptosis (Constitutive Androstane Receptor (CAR)의 활성, 에너지 대사 및 세포의 증식과 사멸의 조절에 대한 CAR의 cross-talk 기전)

  • The activity of CAR can be regulated not only by ligand binding but also by phosphorylation of regulatory factors involved in extracellular signaling pathways, cross-talk interactions with transcription factors, and the recruitment, degradation, and expression of coactivators and corepressors. This regulation of CAR activity can in turn have effects on the control of diverse physiological homeostasis, including xenobiotic and energy metabolism, cellular proliferation, and apoptosis. CAR is phosphorylated by the ERK1/2 signaling pathway, which causes formation of a complex with Hsp-90 and CCRP, leading to its cytoplasmic retention, whereas phenobarbital inhibits ERK1/2, which causes dephosphorylation of the downstream signaling molecules, leading to the recruitment to CAR of the activated RACK-1/PP2A components for the dephosphorylation, nuclear translocation, and the transcriptional activation of CAR. Activated CAR cross-talks with FoxO1 to induce inhibition of its transcriptional activity and with PGC-1α to induce protein degradation by ubiquitination, resulting in the transcriptional suppression of PEPCK and G6Pase involved in gluconeogenesis. Regulation by CAR of lipid synthesis and oxidation is achieved by its functional cross-talks, respectively, with PPARγ through the degradation of PGC-1α to inhibit expression of the lipogenic genes and with PPARα through either the suppression of CPT-1 expression or the interaction with PGC-1α each to induce tissue-specific inhibition or stimulation of β-oxidation. Whereas CAR stimulates cellular proliferation by suppressing p21 expression through the inhibition of FoxO1 transcriptional activity and inducing cyclin D1 expression, it suppresses apoptosis by inhibiting the activities of MKK7 and JNK-1 through the expression of GADD45B. In conclusion, CAR is involved in the maintenance of homeostasis by regulating not only xenobiotic metabolism but also energy metabolism, cellular proliferation, and apoptosis through diverse cross-talk interactions with extracellular signaling pathways and intracellular regulatory factors.

Quality Characteristics of Cabbage Kimchi by Different Packaging materials (포장재에 따른 양배추 김치의 품질특성)

  • In this study, cabbage ($Brassica$ $oleracea$ var. $capitata$) Kimchi was made packed into four kinds of packaging materials, PET vessel, PP tray, OPP/AL/PE film and Nylon/PE/LLDP film, and the effects of these packaging materials on Kimchi quality characteristics, such as lactic acid bacteria counts, salinity, sugar contents, pH, total acidity, electron donating ability were examine as well as their effects on the sensory qualities after storage at a temperature of $4^{\circ}C$. The pH change ranged from pH 6.24 to 6.43 shortly after manufacture, and did not significantly change until 7 days of storage. However, it began to decrease rapidly after 14 days. On the 35th day of storage, the acidity was 0.79% in the PET vessel and 0.83% in OPP/AL/PE. Therefore, the PET vessel and OPP/AL/PE were considered appropriate packaging materials for Kimchi storage. The salinity did not change significantly during the storage period, and the sugar content generally increased in the four kinds of packaging materials, but decreasing after the 7th day of storage. After 14 days of storage, the Kimchi stored in the OPP/AL/PE film showed the highest lactic acid bacteria counts. Although the electron donating ability was the highest after proper time for fermentation, it decreased in all the packaging materials after the proper time for fermentation. However, the OPP/AL/PE film had an antioxidant potential of up to 93.18%. In the sensory evaluation, fermented Kimchi was found to be superior unfermented Kimchi. In addition, the Kimchi stored in the OPP/AL/PE film for 14 days showed the high score of 6.70 and 6.60 in overall preference. Therefore, the results of this study provide basic knowledge on the fermentation level and packaging material's condition for commercialization of small packed cabbage Kimchi. Henceforth, industrialization must include a variety of studies under these conditions to increase the merchantability.

Influence of Vetiver Grass (Vetiveria zizanioides) on Rhizosphere Chemistry in Long-term Contaminated Soils (중금속으로 오염된 토양에서 근권부의 화학적 특성에 미치는 vetiver grass (Vetiveria zizianioides)의 영향)

  • A detailed understanding and appreciation of the important mechanisms operating at the soil:root interface, commonly identified as the rhizosphere, is critical for evaluating the potential for particular plant species to be successfully used as part of a phytoremediation technique. For specific plants, mechanisms may exist to overcome the inherit limitation of the phytoremediation technique when poorly mobile soil metals are of interest. In the present study, the influence of root exudates on the rhizosphere chemistry of soil and consequential metal uptake were investigated following culture of vetiver grass (Vetiveria zizanioides), recognized as a promising plant for land stabilization, in three different long-term contaminated soils and one non-contaminated control soil. The soil solution pH increased (0.3-1.1 units) following vetiver grass culture and dissolved organic carbon (DOC) also significantly increased in all soils with the highest increase in PP02 (23 to $173mg\;L^{-1}$). Chemical changes are contributed to root exudation by vetiver grass when exposed to high concentration of heavy metals. Chemical changes, consequently, influenced metal (Cd, Cu, Pb, and Zn) solubility and speciation in the rhizosphere. The highest solubility was observed for soil Ko01 (eg. 2091 and $318{\mu}g\;L^{-1}$ for Cd and Pb, respectively). Initial heavy metal solubility in soils varied with soil and either increased or decreased following vetiver grass culture depending on the soil type. An increase in pH following plant culture generally resulted in a decrease in metal solubility, while elevated DOC due to root exudation resulted in an increase in metal solubility via the formation of metal-DOC complexes. Donnan speciation demonstrated a significant decrease in free Cd and Zn in the rhizosphere and the concentration of Cd, Pb, and Zn in vetiver grass shoot was highly correlated with soluble concentration rather than total soil metal concentration.

The Risk Factors of the Pre-hypertension and Hypertension of Rural Inhabitants in Chungnam-do (충남 농촌 지역 주민의 고혈압 전단계와 고혈압의 위험요인)

  • The purpose of this study is to investigate risk factors of pre-hypertension and hypertension in rural residents. Nine hundred and ninety four subjects aged 40-70 yrs in Chungnam-do participated in this study. The subjects (n = 824) were classified into three groups of hypertensive, pre-hypertensive, and normotensive according to the Joint National Committee (JNC)-7 criteria. The weight, body mass index (BMI), waist-hip ratio (WHR), and serum total protein, albumin, BUN, and triglyceride (TG) were positively correlated with SBP and DBP. After adjusted by age, sex and BMI, the total protein, albumin and TG were significantly correlated with SBP and DBP (p < 0.01). There was no significant difference in eating habits according to the level of blood pressure. The serum albumin, creatinine, Glu-FBS, Glu-PP l20, and triglyceride were higher in both prehypertensive and hypertensive group than in the normotensive group. However, mean serum cholesterol was not different among three blood pressure groups. In this study, the common risk factors of pre-hypertension and hyper-tension were male, age of fifties, lower education level, ex-smoking, higher drinking frequency, higher BMI, body fat %, waist circumference, WHR, serum albumin and diabetes, even though the degree of risks in these variables were higher in the hypertensive group. The higher BUN was a risk factor of prehypertension, while the family history, prediabetes, serum total protein, Glu-PP l20 and higher alcohol drinking amount were the risk factors of hypertension. This result suggests that maintaining good health habit and normal range of blood parameters as well as controlling body weight have to be paid attention in order to prevent hypertention, and further reseasch on the relationship of blood pressure and BUN are needed.

  • Purpose : Cytomegalovirus(CMV) infection still remains as a major cause of morbidity and mortality after stem cell transplantation. In this study, we analyzed the results of antigenemia-guided preemptive therapy among children with allogeneic hematopoietic stem cell transplantation to determine the incidence and risk factors associated with CMV antigenemia, and evaluated the efficacy of the CMV antigenemia based preemptive therapy. Methods : We enrolled 213 pediatric patients following allogeneic hematopoietic stem cell transplantation(HSCT), at the Catholic HSCT center between October 1998 and December 2003. Pre-emptive ganciclovir was started when more than 5 CMV Ag-positive cells were detected in matched sibling HSCT, and when any Ag-positive cells were seen in unrelated allogenic HSCT. Results : CMV antigenemia was observed in 88(41.3 percent) of 213 patients on median day 28(day 11-99). In univariated analysis, use of unrelated donors(other than siblings), age of recipient(more than 5 years at transplant) at transplantation, the presence of recipient CMV-IgG before transplantation, TBI-based conditioning regimen and the presence of acute GvHD(grade ${\geq}II$) were the risk factors for positive CMV antigenemia. In multivariate analysis, unrelated bone marrow transplantation, positive recipient CMV serology and acute GvHD(grade ${\geq}II$) were the independent risk factors for positive CMV antigenemia. Conclusion : Risk factors of CMV infection in children were CMV serostatus of the recipient, the source of stem cells, and acute graft-versus-host disease. The pre-emptive therapy based on CMV antigenemia was effective in the prevention of CMV disease.

Differential Factors of Obstructive Sleep Apnea in Subjects whose Main Sleep Complaint was Insomnia (불면증을 호소하는 환자에서의 폐쇄성 수면 무호흡증 감별 인자)

  • Objective: The purpose of this study is to investigate the prevalence rate of OSA in subjects whose main sleep complaint is insomnia and to find differential factors of OSA in these insomniac subjects. Method: We reviewed the medical records and polysomnographic findings of patients referred to the Sleep Laboratory at Seoul National University Hospital from January 1996 to December 2002. Four-hundred and seventy subjects complained of insomnia as their main sleep problem (235 males and 235 females, mean age $53.6{\pm}12.4\;years$). First, we investigated the prevalence rate of OSA in these insomniac patients. Second, we compared the clinical and demographic characteristics of the OSA-associated group with those of the non-associated group. Third, we examined whether the degree or presence of differential factors within the OSA group correlate with severity of OSA, as determined by the respiratory disturbance index (RDI). Results: Among 470 insomniac subjects, 125 subjects (26.6%) were diagnosed as OSA by nocturnal polysomnography. OSA-associated subjects were significantly older ($58.4{\pm}12.3\;years$ vs. $51.8{\pm}11.2\;years$, p<0.01), and had significantly higher body mass index (BMI) ($23.4{\pm}3.3\;kg/m^2$ vs. $22.5{\pm}3.1\;kg/m^2$, p=0.44) than non-associated subjects. The OSA-associated group had more subjects with male gender (64.0% vs. 44.9%, p<0.01), hypertension (20.0% vs. 9.3%, p<0.01) or snoring (96.0% vs. 63.5%, p<0.01). Within the OSA-associated group, age had a significant positive correlation with RDI (p=0.01). Conclusion: We found that a considerable portion of patients complaining of insomnia as their main sleep problem were diagnosed as OSA. Snoring, old age, male gender, obesity, and comorbid hypertension were found to be differential factors of OSA in insomniac patients. We suggest that diagnostic efforts including nocturnal polysomnography are needed for insomniac patients with any of the above risk factors of OSA.

A Clinical Study on Binaural Hearing Aid (양이 보청효과에 관한 연구)

  • Monaural and binaural hearing aid performance under quiet and noisy conditions were compared in regard to (1) the degree of hearing impairment, (2) the symmetry of pure tone audiogram, (3) the automatic gain control of the hearing aid. (4) hearing impairement with recruitment and, word discrimination ability. Performance using binaural hearing aids was consistently superior to that using monaural hearing aids. The results were as follows. 1. Speech detection thresholds were enhanced by a mean of 4.25dB when tested with danavox 747 PP stereo type hearing aid and by a mean of 4.12 dB when tested hearing aids connected seperately to the right and left ears. 2. Binaurally tested speech reception thresholds were superior to monaurally tested thresholds by a mean of 3.56dB when tested in quiet and by a mean of 5.56dB when tested in noise. 3. Binaurally tested word discrimination scores were also superior by a mean of 17.09% in quiet and by a mean 19.63% in noise. 4. Both SRT and word discrimination scores were performed best by subjects with moderately-severe impairement. The performance by one mildly impaired subject was the poorest of all performances. The levels of performance order were; moderately-severe loss, severe loss. moderate loss and mild loss. 5. The data obtained using AGC aids when compaired with that of linear amplification show that when AGC aids were worn in both ears. the results were very poor but when one AGC aid was worn in one ear and linear amplification in the other. the results were good. 6. The advantages of binaural hearing aids were obvious even in cases 1) with great diferences in hearing thresholds between right and left ears, 2) when the subject was unable to discriminate words without vision and. 3) when the subject had extreme recruitme t phenomenon.

Habitat Classification and Distribution Characteristic of Aquatic Insect Functional Feeding Groups in the Geum River, Korea (금강 수계 서식지 유형분류 및 수서곤충 섭식기능군 분포특성)

  • This study was performed to classify habitat types depending on environmental factors and to find out distribution characteristics of functional feeding groups of aquatic insects which were collected at that habitat types. Field survey was conducted twice in a year for every spring and fall from 2007 to 2008 for 38 sites in the Geum River. During the field survey 15 environmental factors were measured at each 38 sites and analyzed by similarity analysis method to classify habitat types. The result of similarity analysis showed that the 38 sites could be grouped into 7 classes like as C1 and C3 class belong to Head water(HD), C2 and C4 and C5 class belong to Middle stream(MS), C6 and C7 class belong to Large River(LR) based on euclidean distances 4. And also, we could extract the main environmental factors affecting the classification of habitat types such as Stream Width and Elevation of physical environmental factors, Water Temperature, Conductivity and DO of chemical environmental factors, percentages of Sand, Silt and Gravel of substrate factors. Total 142 species of aquatic insects in 46 families, 9 orders were collected during the field surveys and the occurrence number of species and individuals showed high correlation with the Velocity factor and the percentage of Sand factor of each habitat types. In addition, correlation analysis between functional feeding groups and environmental factors represented that (1) Filtering-collectors(FC) affected by Velocity, Stream Width and Silt, (2) Gathering-collector(GC) affected by Velocity, (3) Predator(P) affected by Elevation, Velocity, Boulder, Conductivity and Sand, (4) Plant-piecer(PP) affected by Water Width and Silt, (5) Scraper(SC) affected by Elevation and Conductivity, (6) Shredder(SH) affected by Elevation, Boulder, DO, pH, Conductivity and Water Temperature respectively. As a result of this study, Elevation, Stream Width, Velocity, Conductivity, Water Temperature and percentage of Sand factors which were deduced by stepwise multiple regression analysis had correlations($r{\geqq}0.600$, p<0.01) with biota community inhabitation. Therefore these six environmental factors were regarded as major environmental factors that might affect highly the distribution of functional feeding groups in stream ecosystem of the Geum River.

EFFECTS OF SUBINHIBITORY CONCENTRATIONS OF ANTIMICROBIAL AGENTS ON CELL SURFACE PROPERTIES AND VIRULENCE FACTORS OF MUTANS STREPTOCOCCI (아저해농도(亞沮害濃度)의 항균물질이 mutans streptococci의 세포표면성질과 독력인자에 미치는 영향)

  • Subinhibitory concentrations (sub-MICs) refer to concentrations below minimum inhibitory concentrations (MICs). The antimicrobial agents may be present at relatively high concentration, at least higher than bacterial MIC and thereafter be deserted off a surface and function at sub-MICs, perhaps by interfering with bacterial metabolism. Consequently, the aim of this study was to determine the effects of growth, in the presence of sub-MICs of antimicrobial agents, on the cell surface properties and virulence factors of mutans streptococci and to investigate the efficacy of a chemical approach in vitro. Streptococcus mutans Ingbritt and Streptococcus sobrinus 6715-7 were used. Eight antimicrobial agents (Sanguinaria extract;SG, Chlorhexidine digluconate;CHX, Fluoride;F, Propolis;PP, Hydrogen peroxide;HP, Triclosan;TC, Sodium dodecyl sulfate;SDS Cetylpyridinium chloride; CC) were diluted serially in broth to determine MICs and to compare the growth rate, acid production, hydrophobicity, adhesion activity to saliva coated hydroxyapatite, glucan synthesis and cellular aggregation of experiment groups (in the presence of sub-MICs) with those of control (in the absence of antimicrobial agents). Sub-MICs of antimicrobial agents affected the growth of cells, hydrophobicity, and adhesion of bacteria to saliva coated hydroxyapatite and glucan synthesis. They also resulted in a significant reduction in pH after 12 hours (p<0.05). By cells pretreated with proteinase K, either the aggregation induced by antimicrobial agents was completely inhibited or the aggregation titers were markedly increased. According to the results of the present study, each antimicrobial agent at sub-MICs could affect similar as its known action mechanism and could continually inhibit cariogenic bacteria at such concentrations. Thus, the use of these antimicrobial agents would be one of the effective methods to prevent dental caries.
