Effects of the Utilization of Sawdust During Composting of Poultry Manure on Physico-Chemical Characteristics and Earthworm (계분의 퇴비화 과정중 이화학적 성상 변화가 지렁이 생존에 미치는 영향)

  • This study was conducted to investigate effects of different ratios of sawdust to poultry manure as an earthworm feed on the change of physico-chemical characteristics and earthworm viability during composting, and further to provide the basic information for an efficient vermicomposting using the manure of livestock. Experimental earthworm used in this study was Eisenia foetida and the ratio of sawdust and poultry was determined to 100:0(A), 80:20(B), 70:30(C), 60:40(D) and 50:50(E) on the basis of volume. The results are summarized as follows. Earthworm appeared after 6 wk of aging period, and it was possible for the earthworm to survive in treatments of C. D and E including 30 $^{\sim}$ 50% of sawdust. C/N ratio was significantly higher (P<0.05) in treatments of sawdust mixture (B, C, D, and E) than poultry manure alone (A treatment).Appropriate C/N ratio for earthworm survival was the range of 16.9 to20.7. During period of aging, pH value and electrolyticconductivity (EC) were the highest in the treatment of poultry manure alone. However, as the inclusion of sawdust was increased, pH showed decreasing tendency. Appropriate pH and EC for earthworm survival were 7.69 to 7.79, and 2.77 to 2.87mS/cm, respectively. The results from this study indicated that the mixing of poultry manure with sawdust more improved physico-chemical environment for earthworm growth in comparison with poultry manure alone, and sawdust showed the sufficient possibilityas a bulking agent for an efficient vermicomposting of poultry manure.

Effects of Mixing Ratios of Swine Manure and Rice Straw on Physico-Chemical Characteristics and Earthworm(Eisenia foetida) Survival (돈분에 볏짚 혼합 수준이 퇴비화 과정중 이화학적 성상 변화와 지렁이 생존에 미치는 영향)

  • This study was conducted to investigate the effect of different ratios of rice straw to swine manure as an earthworm feed on the change of physico-chemical characteristics and earthworm viability during composting, and further to provide the basic information for an efficient vermicomposting using the manure of livestock. Experimental earthworm used in this study was Eisenia foetida and the ratio of swine manure and rice straw was determined to 100:0(SM), 90:10(SRS 10), 80:20(SRS20), 70:30(SRS30) and 60:40(SRS40) on the basis of volume. The results are summarized as follows. Earthworm appeared after 8 week of aging period, and it was possible for the earthworm to survive in all the treatments. C/N ratio was significantly higher (P<0.05) in the treatments of rice straw mixture (SRS30 and SRS40) than swine manure alone (SM treatment). Appropriate C/N ratio for earthworm survival was the range of 10.43 to 11.16. During the period of aging, pH value and electrolytic conductivity (EC) were the highest in the treatment of swine manure alone. However, as the inclusion of rice straw was increased, pH value and EC showed a decreasing tendency. Appropriate pH value and EC for earthworm survival were 7.7 to 7.8, and 2.72 to 4.59mS/cm, respectively. The results from this study indicated that mixing swine manure with rice straw more improved more physico-chemical environment for earthworm growth in comparison with swine manure alone, and rice straw showed the sufficient possibility as a bulking agent for an efficient vermicomposting of swine manure.

Studies on the Immobilization of Lipase by Adsorption Method (흡착법에 의한 Lipase의 고정화)

  • To utilize lipase obtained from Candida cylindracea for lipid hydrolysis, methods to immobilize lipase by adsorption and reaction characteristics of the immobilized lipase by adsorption were investigated. Among the tested adsorbents, silica gel was selected as a suitable adsorbent. The optimum condition for adsorption of lipase was when 47.5 units of lipase were adsorbed to 1.6g of silica gel at pH7.0 and $5^{\circ}C$ for 100 min. Optimum pH and temperature for activity of the immobilized lipase were at $37^{\circ}C$ and pH7.0, which were same as the soluble lipase. Optimum enzyme concentration of the immobilized lipase were 30g for milk fat and 80g for olive oil, whereas those of the soluble lipase were 800 units for milk fat and 1200 units for olive oil. The optimum substrate concentrations of the immobilized and soluble lipases were 20% lipid, regardless of lipid types. Rapid hydrolysis of milk fat was observed with the soluble lipase for the initial 4 hours and with the immobilized lipase for the initial 8 hours. The immobilized lipase produced same amount of capric acid as the soluble lipase, but more myristic acid and less butyric acid than the soluble lipase.

Quality Characteristics of Gugija.Heukimja Jook Containing Different Levels of Black Sesame Powder (흑임자 첨가량을 달리한 구기자흑임자죽의 품질특성)

  • This study was to investigated optimum preparation conditions for Gugija Heukimja jook by response surface methodology (RSM). The important materials used for the Gugija Heukimja product were Lycium chinese, Seasamum indicum L., Ziziphus jujuba Mill. and rice. Moisture content, crude protein, crude fat, crude ash and crude fiber content were 15.62, 16.60, 13.62, 4.93 and 11.17% for Lycium chinese, 3.62, 20.94, 43.56, 5.67 and 10.72% for Seasamum indicum L., 24.12, 5.38, 0.73, 2.30 and 1.25% for Ziziphus jujuba Mill., and 12.36, 6.98, 0.46, 0.43 and 0.41% for rice, respectively. Optimal extraction conditions of brix, color, pH and total acidity in the materials for Gugija Heukimja jook were temperature $80^{\circ}C$, water volume 850 mL and extraction time of 63 min. The spreadability of jook with Lycium chinese and Ziziphus jujuba Mill. extracts was the highest when black sesame was 12 g pH was the lowest when black sesame was 9 g. PH showed a significant difference when the added black sesame ratio changed. The 'value L' of chromaticity was lower with an increase of the black sesame ratio, 'value a' was higher with an increase of the black sesame ratio, 'value b' was lower with an increase of the black sesame ratio. Brix was lower when black sesame in jook was added. Total acidity was highest (7.27%) when black sesame was 12 g. The degree of gelatinization tended to be lower with additional black sesame. Higher scores of sensory evaluation were found for the product containing 9 g black sesame with Lycii furctus and Ziziphus jujuba Mill. extracts ompared to the other samples with 3 and 6g. No significant difference in taste occurred for Gugija Heukimja jook prepared with over 9 g black sesame, while a great difference showed in color, pH and total acid. Therefore, the Gugija Heukimja jook prepared under the optimal conditions of 9 g black sesame with Lycium chinese and Ziziphus jujuba Mill. extracts exhibited higher qualities. Gugija Heukimja jook can use the easier for Yacksun jook. It is expected to become functional in the product's development.

  • Thiamine derivatives of thiamine tetrahydrofurfuryl disulfide (TTFD) and thiamine propyl disulfide (TPD) were obtained in a good yield and purity by use of thiothiamine $(SB_1)$ and also described in details for the preparing sodium tetrahydrofurfuryl thiosulphate (Bunte's salt) with and without KI. The optimum reaction conditions for the preparation of TTFD was set in which the Box-Wilson plan was applied. The reaction conditions are as follows; 1. pH value of aqueous solution of thiol type of $B_1{\cdot}HCl;12.09$ 2. Quantity of Bunte's salt (to $B_1{\cdot}HCl\;20G$); 35.01g 3. Reaction temperature; $15.59^{\circ}C$ Studies on ultraviolet absorption were made at various pH and showed that the absorption maxima are shifted with change of pH. The absorption maxima are at $244{\sim}246nm,\;234{\sim}235nm$ in TTFD, and $245{\sim}246nm,\;233{\sim}235nm$ in TPD. The structure was proved by the infrared spectral evidence. Quantitative determination was studied.

Selection and Physico-Chemical Characteristics of Lactic Acid Bacteria which had Cholesterol Lowering Activities (콜레스테롤 저하 유산균의 분리 및 이들 균주의 이화학적 특성)

  • Four strains of lactic acid bacteria which had cholesterol lowering activities were selected from foreign fermented milk. The strains were identified as Lactobacillus(L.) rhamnosus 2084, L. casei 0781, Lactococcus (Lacto.) lactis spp. 204, and Enterococcus(E.) faecium 402. We observed that the L. rhamnosus 2084 was the most tolerant against pH 1.5, L. casei 0781 against pH 2.0, but not significantly different in the tolerance against pH 3.0. The L. rhamnosus 2084 was the most tolerant against bile acid and prominent in the degree of lowering cholesterol level. All four strains were used as starters in producing yogurt, and then investigated physico-chemical characteristics, such as pH, titratable acidity, and viable cell counts of yogurt base. L. casei 0781, L. rhamnosus 2084, Lacto. lactis 204, and E. faecium 402 were incubated for 6 hours at $40^{\circ}C$, 4 hours at $40^{\circ}C$, 6 hours at $37^{\circ}C$, and 12 hours at $37^{\circ}C$ and $40^{\circ}C$, respectively, for the optimum conditions of fermented milk.

Improvement on Storage Stability of Soybean Curd using Cuttle Bone Powder Treated with Acetic Acid (유기산처리 갑오징어갑을 이용한 두부의 저장성 개선)

  • Storage stabilities of soybean curds using cuttle bone powder treated with acetic acid (ATC-soybean curd) and calcium chloride (calcium chloride-soybean curd) as a coagulants were examined. The pH, turbidity of soaking solution, titratable acidity, and viable cell counts of calcium chloride-soybean curd did not change up to 6 days of storage, then decreased rapidly in pH and turbidity and increased rapidly in titratable acidity and viable cell counts, whereas those of ATC-soybean curd did not change up to 9 days of storage, then decreased slowly in pH and turbidity and increased slowly in titratable acidity and viable cell counts. Regardless of coagulants and storage period, white index of soybean curds did not change during cold storage. Hardness and brittleness of calcium chloride-soybean curd decreased rapidly up to 3 days in of storage, remained stable from 3 days to 6 days, then decreased rapidly. Hardness and brittleness of ATC-soybean curd also decreased rapidly up to 3 days in of storage and remained stable from 3 to 9 days, but then decreased slowly. Regardless of coagulants, results of sensory evaluation were similar to those of physicochemical tests. Judging from the above results, shelf-lives were determined as 9 and 6 days for ATC- and calcium chloride-soybean curds, respectively.

  • Effects of hemoglobin (Hgb) concentration and degree of hydrolysis (DH) of Hgb on the separation of heme-iron were examined to produce highly enriched heme-iron from Hgb hydrolysate. Separation efficiency of Hgb hydrolysate with different DH was studied at wide pH range (pH $1.0{\sim}11.0$). Separation efficiency expressed as heme-iron/peptide ratio increased with decreasing Hgb concentration. When 5% Hgb (pH 10.0) was hydrolyzed using commercially available Esperase for 5 h at $50^{\circ}C$, DH was 25%. The precipitation of heme-iron-enriched peptides were remarkably high at pH range $3{\sim}6$. Optimal pH range for heme-iron with high heme-iron/peptide ratio shifted to acidic pH with increasing DHs of Hgb. The enriched heme-iron fraction in the precipitates showed a single band through urea-SDS-PAGE, with a molecular mass of 1 kDa. In the dry heme-iron product produced in a pilot bioreactor, content of heme-iron and heme-iron/peptide ratio were 27.1 and 38.7%, respectively, and production yield was 9.3%.

  • This study was aimed to find out the influence of long-term fertilization for 21 years on soil enzyme activities in the silty clay loam textured normal paddy soil. Total urease activity (TUA) and the microbial urease activity (MUA) were shown to be changed significantly, but the accumulated urease activity (AUA) was similar within trial plots. Especially the MUA of the plots annually applied N.P.K. fertilizers with compost and N.P.K. fertilizers with silicate fertilizer were the highest among plots. The total L-glutaminase activity (TGA) and the accumulated L-glutaminase activity(AGA) were changed significantly among trial plots, but the microbial L-glutaminase activity (MGA) was not. By the simple correlation analysis, it was shown that the TGA and the AGA correlated highly significant to available phosphorus available $SiO_2$ content and pH. Addition of the toluene to the incubation mixture did not markedly affect the activity of phosphatase, but the difference of phosphatase activity among plots was significant. By the simple correlation analysis, it was shown that the phosphatase activity ; correlated highly significant to pH, available $SiO_2$, available phosphorus and exchangeable calcium in soils.

  • This experiment was carried out to examine the MBC adsorption on clay minerals by the slurry method. The adsorption of MBC on Kaolinite nearly reached equilibrium after shaking lot 2hrs. but on Bentonite and Zeolite, 10hrs., respectively. The adsorption amount was in the order of Zeolite>Bentonite>Kaolinite, and Na-saturated clay mineral adsorbed more MBC than Mg-saturated clay mineral did. The more the amount of clay minerals, the more MBC was adsorbed by clay minerals, whereas the amount of MBC adsorbed per unit gram of clay minerals reduced and Kd value cf MBC was greatest on the Na-Zeolite. The lower the pH of the water-clay system at each concentration, the more MBC was adsorbed by clay minerals. The pH dependence of the adsorption of MBC may be due to protonation of MBC in acid solution.
