Lethal toxin is a critical virulence factor of anthrax. It is composed two protein: protective antigen (PA) and lethal factor (LF). PA binds to specific cell surface receptors and, forms a membrane channel that mediates entry of LF into the cell. LF is a zinc-dependent metalloprotease, which cleaves MKKs [MAPK (mitogen-activated protein kinase) kinases] at peptide bonds very close to their N-termini. In this study, we suggest application of cell-based assays in the early phase of drug discovery, with a particular focus on the use of yeast cells. We constructed MEK1 expression system in yeast to determine LF activity and approached cell-based assay system to screen inhibitors, in which the results covering the construction of LF-substrate in yeast expression vector, expression, and LF-mediated proteolysis of substrate were described. These results could provided the basic steps in design of cell-based assay system with the high efficiency, rapidly and easy way to screening of inhibitors.
Purpose: This study was conducted to identify the effects of home exercise programs and manual therapy on shoulder function and quality of life in patients with adhesive capsulitis. Methods: A total of 31 patients were enrolled in the study. All subjects were randomly assigned to a home exercise group (n=15) or a manual therapy group (n=16). Both groups performed each intervention program three times a week for four weeks. After four weeks, both groups performed a home exercise program continuously until week 12. Shoulder function was evaluated using the Korean Shoulder Scoring (KSS) system, while quality of life was evaluated using the WHO Quality of Life-BREF (WHOQOL-BREF) before and 4, 8, and 12 weeks after the intervention. Data were analyzed using two-way repeated measures analysis of variance (ANOVA). Results: There were significant differences in KSS before intervention and 4, 8, and 12 weeks after intervention for both the home exercise group and manual therapy group (p<0.001). There was also a significant difference in KSS at 4, 8, and 12 weeks when compared to the home exercise group and manual therapy group (p<0.001). Moreover, there was a significant difference in WHOQOL-BREF before and 4, 8, and 12 weeks after intervention in the home exercise group and manual therapy group (p<0.001). Conclusion: Home exercise and manual therapy improved shoulder function, but manual therapy led to a greater improvement in shoulder function better than home exercise. Home exercise and manual therapy improved quality of life, but there was no significant difference between groups.
Corrugated steel plate shear wall (CSPSW) as an innovative lateral load resisting system provides various advantages in comparison with the flat steel plate shear wall, including remarkable in-plane and out-of-plane stiffnesses and stability, greater elastic shear buckling stress, increasing the amount of cumulative dissipated energy and maintaining efficiency even in large story drifts. Employment of low yield point (LYP) steel web plate in steel shear walls can dramatically improve their structural performance and prevent early stage instability of the panels. This paper presents a comprehensive structural performance assessment of corrugated low yield point steel plate shear walls having circular openings located in different positions. Accordingly, following experimental verification of CSPSW finite element models, several trapezoidally horizontal CSPSW (H-CSPSW) models having LYP steel web plates as well as circular openings (for ducts) perforated in various locations have been developed to explore their hysteresis behavior, cumulative dissipated energy, lateral stiffness, and ultimate strength under cyclic loading. Obtained results reveal that the rehabilitation of damaged steel shear walls using corrugated LYP steel web plate can enhance their structural performance. Furthermore, choosing a suitable location for the circular opening regarding the design purpose paves the way for the achievement of the shear wall's optimal performance.
Objective: The purpose of this study was to measure the temperature of the plugger tip of 3 cordless heat carriers set at $200^{\circ}C$. Materials and Methods: Pluggers of the same taper (0.06, 0.08, 0.10) and similar tip sizes (sizes of 50 and 55) from 3 cordless heat carriers, namely SuperEndo-${\alpha}^2$(B & L Biotech), Friendo (DXM), and Dia-Pen (Diadent), were used and an electric heat carrier, System B (SybronEndo), was used as the control. The plugger tips were covered with customized copper sleeves, heated for 10 seconds, and the temperature was recorded with a computerized measurement system attached to a K-type thermometer at room temperature (n = 10). The data were analyzed with 2-way analysis of variance at a 5% level of significance. Results: The peak temperature of the plugger tips was significantly affected by the plugger taper and by the heat carrier brand (p < 0.05). The peak temperature of the plugger tips was between $177^{\circ}C$ and $325^{\circ}C$. The temperature peaked at $207^{\circ}C-231^{\circ}C$ for the 0.06 taper pluggers, $195^{\circ}C-313^{\circ}C$ for the 0.08 taper pluggers, and $177^{\circ}C-325^{\circ}C$ for the 0.10 taper pluggers. Only 5 of the 12 plugger tips showed a temperature of $200^{\circ}C{\pm}10^{\circ}C$. The time required to reach the highest temperature or $200^{\circ}C{\pm}10^{\circ}C$ was at least 4 seconds. Conclusion: When using cordless heat carriers, clinicians should pay attention to the temperature setting and to the activation time needed to reach the intended temperature of the pluggers.

Objectives: The purpose of this study was to compare the maximum screw-in forces generated during the movement of various Nickel-Titanium (NiTi) file systems. Materials and Methods: Forty simulated canals in resin blocks were randomly divided into 4 groups for the following instruments: Mtwo size 25/0.07 (MTW, VDW GmbH), Reciproc R25 (RPR, VDW GmbH), ProTaper Universal F2 (PTU, Dentsply Maillefer), and ProTaper Next X2 (PTN, Dentsply Maillefer, n = 10). All the artificial canals were prepared to obtain a standardized lumen by using ProTaper Universal F1. Screw-in forces were measured using a custom-made experimental device (AEndoS-k, DMJ system) during instrumentation with each NiTi file system using the designated movement. The rotation speed was set at 350 rpm with an automatic 4 mm pecking motion at a speed of 1 mm/sec. The pecking depth was increased by 1 mm for each pecking motion until the file reach the working length. Forces were recorded during file movement, and the maximum force was extracted from the data. Maximum screw-in forces were analyzed by one-way ANOVA and Tukey's post hoc comparison at a significance level of 95%. Results: Reciproc and ProTaper Universal files generated the highest maximum screw-in forces among all the instruments while M-two and ProTaper Next showed the lowest (p < 0.05). Conclusions: Geometrical differences rather than shaping motion and alloys may affect the screw-in force during canal instrumentation. To reduce screw-in forces, the use of NiTi files with smaller cross-sectional area for higher flexibility is recommended.
This paper aims at providing a critical view over the cybernetics theory especially of first generation on which the artificial intelligence heavily depends nowadays. There has been a commonly accepted thought that the conception of artificial intelligence could not has been possible without being influenced by N. Wiener's cybernetic feedback based information system. Despite the founder of contemporary cybernetics' ethical concerns in order to avoid an increasing entropy phenomena(social violence, economic misery, wars) produced through a negative dynamics of the western modernity regarded as the most advanced form of humanism. In this civilizationally changing atmosphere, the newly born cybernetic technology was thus firmly believed as an antidote to these vices deeply rooted in humanism itself. But cybernetics has been turned out to be a self-organizing, self-controlling mechanical system that entails the possibility of telegraphing human brain (which are transformed into patterns) through the uploading of human brain neurons digitalized by the artificial intelligence embedded into computing technology. On this background emerges posthuman (or posthumanism) movement of which concepts have been theorized mainly by its ardent apostles like N. K. Hayles, Neil Bedington, Laurent Alexandre, Donna J. Haraway. The converging of NBIC Technologies leading to the opening of a much more digitalizing society has served as a catalyst to promote the posthuman representations and different narratives especially in the contemporary visual arts as well as in the study of humanities including philosophy and fictional literature. Once Bruno Latour wrote "Modernity is often defined in terms of humanism, either as a way of saluting the birth of 'man' or as a way of announcing his death. But this habit is itself modern, because it remains asymmetrical. It overlooks the simultaneous birth of 'nonhumaniy' - things, or objects, or beasts, - and the equally strange beginning of a crossed-out God, relegated to the sidelines."4) These highly suggestive ideas enable us to better understand what kind of human beings would emerge following the dazzlingly accelerating advancement of artificial intelligence technology. We wonder whether or not this newly born humankind would become essentially Homo Artificialis as a neuronal man stripping off his biological apparatus. However due to this unprecedented situation humans should deal with enormous challenges involving ethical, metaphysical, existential implications on their life.
The purpose of this study was to compare the effect of curing methods of adhesive resins and resin cements in the root canal. Crown portions of 32 single-rooted mandibular premolars were removed. Routine endodontic treatment was done, and 9 mm deep post spaces were prepared within root canals. No.3 FRC Postec posts (Ivoclar-Vivadent AG, Liechtensteih) were cemented in the post spaces by self-(SC) or light-curing (LC) using two dual-cured adhesives (Adper Scotchbond multi-purpose plus and Exite DSC )and resin cements (RelyX ARC and Variolink II). They were assigned to 4 groups (n=8): R-SC, R-LC, V-SC, V-LC group. After stored in distilled water for 24 hours, each root was transversally sectioned with 1.5 mm thick and made three slices. The specimens were subjected to push-out test in a universal testing machine (EZ Test, Shimadzu Co., Japan) with a crosshead speed of 1 mm/min. The data were analyzed with repeated ANOVA and one-way ANOVA. Also the interface of post-resin cement and resin cement-canal wall of each group was observed under FE-SEM. When fiber posts were cemented into the root canal using total-etch adhesives, the bond strength and adaptation between post and root canal dentin was affected by curing method. Self-cure of adhesives and resin cements showed higher bond strength and closer adaptation than light-cure of them.
Generally, the rainfall and the influent of wastewater treatment plants (WWTPs) have strong relationship at the case of combined sewers. With the fact that the influent variations in terms of quantity and sewage quality is the most common and significant disturbance, the impact factor to the characteristics of sewage should be searched for. In this paper, the relationship between weather conditions such as humidity, temperature and rainfall and influent flowrate and contaminant concentration was analysed using factor analysis. Additionally, 3 influent types were deduced using cluster analysis and the distributions of operational variables were compared to the each groups by one-way ANOVA. The applied dataset were clustered to three groups that have the similar weather and influent conditions. These different conditions can cause the different operating conditions at WWTPs. That is, the Group 1 is for the condition with high humidity and rainfall, so DO concentration in the reactor was very high but MLSS concentration was very low because of too large flowrate. However, the Group 3 is classified to the case having low humidity, temperature, and rainfall, therefore, the SRT was the longest and the SVI was the highest due to the worst settleability in the winter for a year.

Nervous system development takes place after positional information has been established along the dorsal-ventral (D/V) axis. The initial subdivision provided by a gradient of nuclear dorsal protein is maintained by the zygotic genes expressed along the D/V axis. In this study, an investigation was conducted to determine the range of Dpp function in repressing the expression of eagle (eg) that is present in intermediate neuroblasts defective (ind) and muscle specific homeobox (msh) gene domain. eg is expressed in neuroblast (NB) 2-4, 3-3 and 6-4 of the msh domain, and NB7-3 of the ind domain at the embryonic stage 11. In decapentaplegic (dpp) loss-of-function mutant embryos, eg was ectopically expressed in the dorsal region, while in dpp gain-of-function mutants produced by sog or sca-GAL4/UAS-dpp, eg was repressed by Dpp. It is worthy of note that Dpp produced from sim;;dpp embryos showed that Dpp could function at long range. However, Dpp produced from en-GAL4/UAS-dpp or wg-GAL4/UAS-dpp primarily acted at short-range. This result demonstrated that this discrepancy seems to be due to the repression of Dpp to EGFR signaling in sim;;dpp embryos. Taken together, these results suggest that Dpp signaling works at short-range, but can function indirectly at long-range by way of repression of EGFR signaling during embryonic neurogenesis.
The aim of this study was to examine the epidemiological features of Eimeria in calves with acute diarrhea. Samples were collected from between 15 days and 90 days old calves (n=83) in Gimje area from March 2006 to March 2007. Feces of bloody diarrhea were examined for the presence of Eimeria oocysts using a sucrose flotation method. Out of 83 calves, 62 (74.6%) had Eimeria oocysts. In the results of monthly analysis, the highest prevalence (12.0%) of Eimeria oocysts was found on June. In the seasonal infection rate, spring was the highest prevalence (30.1%), followed by summer (24.0%). Furthermore, the highest prevalence (44.5%) was found in calves from between 31 - 60 days old in the analysis by ages. However, there was no significant differences between female and male sex even though the prevalence was slightly bigger in female than in male. The prevalence of the present study to detect Eimria oocysts for infection may have been affected by weather-conditions in the spring. Young calves should be separated to minimize the infection from cattle as much as possible. Additional studies are necessary to find other factors for infection and combining molecular methods with a highly sensitive system for Eimeria detection could be a reliable and economic way of Eimeria eradication.
