The Effects of Enzyme Complex on Performance, Intestinal Health and Nutrient Digestibility of Weaned Pigs

  • Two experiments were conducted to evaluate the effect of supplementing a corn-soybean meal-based diet with an enzyme complex containing amylase, protease and xylanase on the performance, intestinal health, apparent ileal digestibility of amino acids and nutrient digestibility of weaned pigs. In Exp. 1, 108 piglets weaned at 28 d of age were fed one of three diets containing 0 (control), 100, or 150 ppm enzyme complex for 4 wks, based on a two-phase feeding program namely 1 to 7 d (phase 1) and 8 to 28 d (phase 2). At the end of the experiment, six pigs from the control group and the group supplemented with 150 ppm enzyme complex were chosen to collect digesta samples from intestine to measure viscosity and pH in the stomach, ileum, and cecum, as well as volatile fatty acid concentrations and composition of the microflora in the cecum and colon. There were linear increases (p<0.01) in weight gain, gain: feed ratio and digestibility of gross energy with the increasing dose rate of enzyme supplementation during the whole experiment. Supplementation with enzyme complex increased the digesta viscosity in the stomach (p<0.05) and significantly increased (p<0.01) the concentrations of acetic, propionic and butyric acid in the cecum and colon. Enzyme supplementation also significantly increased the population of Lactobacilli (p<0.01) in the cecum and decreased the population of E. coli (p<0.05) in the colon. In Exp. 2, six crossbred barrows (initial body weight: $18.26{\pm}1.21$ kg), fitted with a simple T-cannula at the distal ileum, were assigned to three dietary treatments according to a replicated $3{\times}3$ Latin Square design. The experimental diets were the same as the diets used in phase 2 in Exp. 1. Apparent ileal digestibility of isoleucine (p<0.01), valine (p<0.05) and aspartic acid (p<0.05) linearly increased with the increasing dose rate of enzyme supplementation. In conclusion, supplementation of the diet with an enzyme complex containing amylase, protease and xylanase improved piglet performance. This is likely a result of improvement in nutrient digestibility, volatile fatty acid concentrations and bacteria ratio in the large intestine.

Volatile Fatty Acid Production from Saccharina japonica Extracts by Anaerobic Fermentation: Evaluation of Various Environmental Parameters for VFAs Productivity (혐기성 발효에 의한 다시마 추출물로부터 휘발성 유기산 제조: 휘발성 유기산 생산성에 대한 환경적 영향인자 평가)

  • Volatile fatty acids (VFAs) production from marine brown algae, Saccharina japonica, was investigated in anaerobic dark fermentation. In order to evaluate the VFAs productivity, various experimental parameters (i.e., physicochemical pre-treatment, microorganism inoculation ratio, substrate concentration, and pH) were evaluated. According to the physicochemical pre-treatment methods, the maximum concentrations of VFAs were obtained in the order of sulfuric acid, subcritical water and subcritical water with lipid-extraction. Also, we investigated the operating parameters such as microorganism inoculation ratio (MV/M = 10 to 30), the substrate concentration (18.0 to 72.0 g/L) and pH (6.0 to 7.0) in sulfuric acid pre-treatment method. When the substrate concentrations were 18.0, 36.0, 54.0 and 72.0 g/L at $35^{\circ}C$, microorganism inoculation ratio 15, pH 7.0 for 372 hours, the maximum concentrations of VFAs were respectively 9.8, 13.9, 18.6 and 22.3 g/L. The change in VFAs concentrations was detected that acetic- and propionic acids increased according to increasing pH, while the butyric acid increased with decreasing pH. The VFAs obtained from concentration and separation process may be used as basic chemistry materials and bio-fuel, and they will expect to produce alternative energy of fossil fuel.

The Nutritive Values and Manufacture of Total Mixed Fermentation Feeds using Green Forage Crops and RiCE-straw (청예 사료작물과 볏짚을 이용한 완전배합발효사료의 제조와 영양적가치)

  • Adequate forage amounts in ruminant animal are necessary for proper ruminal function in dairy cow. This study was carried out to determine the effect of total mixed fermentation feeds made by different green forage crops and grain processings on chemical compositions, RFV (relative feed value) and ruminal characteristics in sheep. The experiment was arranged in a split plot design with 4 replications. The main plot consisted of 6 kinds of green forages (corn, grass, rye, rape, alfalfa and oat) and the sub plots three different grain processings such as non-milling, half milling (7mm mesh over), and regular milling (7mm mesh below). And the different TMFFs (total mixed fermentation feeds) were analyzed for chemical composition and fed to 8 ruminally fistulated sheep for ruminal charactics and palatability. RFV, daily feed intake, acetate/propionate ratio of the rape-TMFFs were higher compared with the other treatment. Ruminal content of VFA (volatile fatty acid) of corn-TMFF was highest as 90.19 mmol% and pH of the feed was lowest as 3.82. But, acetic acid, propionic acid and butyric acid were no difference among treatments. In conclusion, the effect of grain proceeding was not appeared but if consider of only RFV, palatability and dry matter disappearance, grade of TMFF was improved in order of rape-, corn-, alfalfa-, grass-, oat- and rye-TMFF.

Effects of Dietary Lactobacillus brevis Supplementation on Growth Performance, Dry Matter and Nitrogen Digestibilities, Blood Cell Counts and Fecal Odor Emission Compounds in Growing Pigs (육성돈사료에 Lactobacillus brevis의 첨가가 성산성, 건물과 질소 소화율, 혈구수 및 분 내 악취 발생 물질에 미치는 영향)

  • This study was conducted to investigate the effects of dietary Lactobacillus brevis (3.4×108 CFU/g) supplementation on growth performance, DM and N digestibilities, blood cell counts and fecal odor emission compounds in growing pigs. Ninety six crossbred [(Landrace×Yorkshire)×Duroc] pigs with an initial BW of 24.60±1.28kg were used for 42-d feeding trial according to a completely randomized design. Three corn- soybean meal based dietary treatments included: 1) CON (basal diet); 2) LB1 (basal diet + Lactobacillus brevis 0.2%) and 3) LB2 (basal diet+Lactobacillus brevis 0.4%). There were three dietary treatments with eight replicate pens per treatment and four pigs per pen. Through the entire experimental period, ADG, ADFI and gain/feed had no significant differences among treatments(P>0.05). Nitrogen digestibility was increased in LB1 and LB2 treatments compared to CON treatment (linear effect, P<0.05), however, DM digestibility had no significant difference among all the treatments (P>0.05). The WBC, RBC and lymphocyte concentrations in whole blood were not affected by treatments (P>0.05). Fecal NH3N and H2S concentrations were significant decreased in LB2 treatment compared to CON treatment (linear effect, P<0.05). Fecal VFA (acetic acid and propionic acid) concentration was also reduced in LB2 treatment compared to CON treatment (linear effect, P<0.05). In conclusion, Lactobacillus brevis (3.4×108 CFU/g) supplementation at the level of 0.4% can improve nitrogen digestibility and decrease the concentrations of fecal odor emission compounds in growing pigs.

Effect of Microbial Additives on Metabolic Characteristics in Sheep and Milking Performance of Lactating Dairy Cows (미생물제제의 첨가가 면양의 반추대사 및 젖소의 유생산성에 미치는 영향)

  • Two experiments were conducted to observe the effects of direct fed microbials on metabolic characteristics in sheep and milking performance in dairy cows. A metabolic trial with four ruminally cannulated sheep(60±6kg) was conducted in a 4×4 Latin square design to investigate the supplementation effects of Saccharomyces cerevisiae, Clostridium butyricum or mixed microbes of S. cerevisiae and C. butyricum on ruminal fermentation characteristics and whole tract digestibility. Sheep were fed 1.25 kg of total mixed ration(TMR, DM basis) supplemented with S. cerevisiae (2.5g/day), C. butyricum (1.0g/day) or its mixture(S. cerevisiae 1.25g/day+C. butyricum 1g/day), twice daily in an equal volume. But control sheep were fed only TMR. A feeding trial with 28 lactating Holstein cattle was also conducted for 12 weeks to investigate the effects of the same microbial supplements as for the metabolic trial on milking performance. The cows were fed the TMR(control), and fed S. cerevisiae(50g/day), C. butyricum(15g/day) or its mixture (S. cerevisiae 25g/day + C. butyricum 7.5g/day) with upper layer dressing method. Total VFA concentration and the digestibility of whole digestive tract in the sheep increased by supplementation of S. cerevisiae, C. butyricum or their combined microbials compare to control group. The proportion of propionic acid at 1h(P<0.039) and 3h(P<0.022) decreased by supplementation of S. cerevisiae while tended to increase acetic acid proportion at the same times. Daily dry matter intake(DMI) was not influenced by the microbial treatments, but milk yield(P<0.031) and feed efficiency(milk yield/DMI, P<0.043) were higher for the cow received C. butyricum than those for other treatments. The milk fat content was higher (P<0.085) when cows fed S. cerevisiae(4.11%) than that fed the control (4.08%), the diets with C. butyricum (3.85%) and the microbial mixture. Based on the results obtained from the current experiments, supplementation of C. butyricum or mixture with S. cerevisiae might be increased milk fat content and milk productivity of lactating daily cows. Studies on the Characteristics of Volatile Fatty Acid Evolution from Fresh Animal Feces (축분의 휘발성 지방산 발현 양상 연구)

  • This work was carried out to measure volatile fatty acids emissions from different manure (poultry, swine, cattle) incubated at $10^{\circ}C$, $25^{\circ}C$, and $37^{\circ}C$ for 6 days under anaerobic condition. Following are summary of these tests results. 1. Amounts of Acetic acid generated were 1,128.05mg/kg, 628.21mg/kg and 592.50mg/kg for swine, poultry, and cattle manure, respectively, during the period of incubation. In the case of swine and cattle manure, 83.87%(946.10mg/kg) and 57.49%(340.63mg/kg) from all the temperature treatments were produced in the $25^{\circ}C$, respectively. 83.57% in swine and 78.79% in cattle manure were intensively emerged from 3 day, 4 day and 5 day of the $25^{\circ}C$ treatment. In the case of poultry manure, 45.36%(284.93mg/kg) and 45.36%(284.93mg/kg) in the $25^{\circ}C$ and in the $37^{\circ}C$, respectively, were produced. Accordingly, acetic acid generated from poultry manure was characteristic of being mainly produced in more than $25^{\circ}C$. 2. Amounts of propionic acid generated were 238.56mg/kg, 162.14mg/kg and 155.49mg/kg for swine, poultry, and cattle manure, respectively, during the period of incubation. In the case of swine manure, 78.52%(187.32mg/kg) of propionate emitted from all the temperature treatments was produced in the $25^{\circ}C$ and 79.1% of them was intensively emerged from 3day, 4day and 5day of the $25^{\circ}C$ treatment. In the case of poultry manure, 35.12%(56.95mg/kg) and 45.89%(74.40mg/kg) of the propionate amounts were produced in the $25^{\circ}C$ and in the $37^{\circ}C$, respectively. In the case of cattle manure, 28.21% (43.86mg/kg) and 49.30% (76.66mg/kg) of the propionate amounts were produced in the $10^{\circ}C$ and in the $25^{\circ}C$, respectively. Accordingly, propionate evolved from poultry manure was characteristic of being mainly produced in more than $25^{\circ}C$ and from cattle manure, in less than $25^{\circ}C$, respectively. 3. Amount of butyric acid generated were 1,463.87mg/kg, 96.72mg/kg and 129.18mg/kg for swine, poultry, and cattle manure, respectively, during the period of incubation. The time intensively emerged from the period of incubation was differently generated from the incubation temperature and animal feces. 4. Amounts of iso-valeric acid generated were 6,885.99mg/kg, 399.28mg/kg and 307.47mg/kg for swine, cattle and poultry manure, respectively, during the period of incubation. In the case of swine and cattle manure, 28.22%(1,943.52mg/kg) and 48.56%(193.90mg/kg) in the $25^{\circ}C$, 68.76%(4,734.90mg/kg) and 46.93%(187.40mg/kg) in the $37^{\circ}C$, respectively, were occupied. Accordingly, iso-valeric acid evolved from swine and cattle manure was characteristic of being mainly produced in more than $25^{\circ}C$. In the case of poultry manure, 59.89%(184.13mg/kg) of iso-valeric acid generated from all the temperature treatments was produced in the $37^{\circ}C$ and 100% of them was intensively emerged from 2 day and 3 day of the $37^{\circ}C$ treatment.

Effects of Molasses Addition on the Feeding Value of Socheongryongtang(Herbal Medicine) Meal Silage (당밀 첨가가 소청룡탕박 사일리지의 품질에 미치는 영향)

  • Two experiments were conducted to investigate effects of molasses addition to silage materials in ensiling Socheongryongtang meal on the nutritive quality of silage, palatability and various ruminal parameters in Korean native goats. In Experiment 1, Socheongryongtang meal silage was produced by the addition of 0, 0.5, or 1.0% molasses and stored for 40 days at room temperature. There were three replicates per treatment. Lactic acid contents of Socheongryongtang meal silage containing molasses(0.5 and 1.0%) were significantly(p<0.05) higher than that of the control(0%). However, the pH and butyric acid contents of Socheongryongtang meal silage containing molasses(0.5 and 1.0%) were lower than those of the control(0%). In addition, molasses(0.5 and 1.0%) increased the number of lactobacillus, but decreased the number of fungi in Socheongryongtang meal silage. In vitro dry matter disappearance tended to increase by molasses addition. In Experiment 2, three 1.5-yr-old Korean native female goats were employed in cross-over design to measure the palatability and various ruminal parameters of Socheongryongtang meal silage. Feed intake of Socheongryongtang meal silage containing molasses(0.5 and 1.0%) for 30 min significantly higher than that of control(0%). Silmilar trend was found when the feed intake was measured for 6 hr. In various ruminal parameters, molasses addition to silage materials increased propionic acid(P) and total volatile fatty acid contents, but decreased pH, acetic acid (A), and butyric acid. The A/P ratio was the lowest in 1.0% molasses treatment group. It is concluded that molasses addition in ensiling Socheongryongtang meal could enhance its nutritional quality and lengthen storage period. Also the palatability of silage was improved by the addition of molasses.

Effects of Volatile Fatty Acids on IGF-I, IGFBP-3, GH, Insulin and Glucagon in Plasma, and IGF-I and IGFBP-3 in Different Tissues of Growing Sheep Nourished by Total Intragastric Infusions

  • Twelve Suffolk${\times}$Small-tail-Han male sheep (body weight 21-26 kg), aged four months, were used to study the effects of volatile fatty acids (VFA) on IGF-I (insulin-like growth factor-I), IGFBP-3 (insulin-like growth factor binding protein-3), GH (growth hormone), insulin and glucagon in plasma, and IGF-I and IGFBP-3 in different tissues. The sheep were randomly divided into four groups with 3 sheep in each group. The sheep were sustained by total intragastric infusions and four levels of mixed VFA (the molar proportion of acetic acid, propionic acid and butyric acid was 65:25:10), which supplied 333, 378, 423 and 468 KJ energy/kg $W^{0.75}$/d, were infused into the rumen as experimental Treatments I, II, III and IV, respectively. The experiment lasted 12 days, of which the first 8 days were for pretreatment and the last 4 days for collection of samples. At the end of the experiment, blood samples were taken and then the sheep were slaughtered and tissue samples from the rumen ventral sac, rumen dorsal sac, liver, duodenum and Longissimus dorsi muscle were obtained. IGF-I, IGFBP-3, GH, insulin and glucagon in plasma and IGF-I and IGFBP-3 in different tissues were analysed. Results showed that the concentration of IGF-I, IGFBP-3, GH, insulin or glucagon in plasma and the content of IGF-I and IGFBP-3 in the rumen dorsal sac, rumen ventral sac, liver or Longissimus dorsi muscle were increased with VFA infusion level (p<0.05). No significant differences were found in duodenum IGF-I between Treatments I and II and in rumen dorsal sac IGFBP-3 between Treatments II and III (p>0.05). It was concluded that IGF-I, IGFBP-3, GH, insulin and glucagon in plasma and IGF-I and IGFBP-3 in rumen dorsal sac, rumen ventral sac, liver and Longissimus dorsi muscle were increased significantly with increasing level of ruminal infusion of mixed VFA.

Effect of Season on Volatile Organic Compounds and Volatile Fatty Acids Concentration in finishing Pig Slurry to Grassland (초지환원용 비육돈 슬러리의 계절에 따른 휘발성유기물과 휘발성지방산 농도 분석연구)

  • The objective of this study is to investigate the changes in levels of odorous compounds in pig slurry during different seasons. Slurry from pens of finishing pigs was sampled every 4-wk and concentration of odorous compounds was analyzed. There was no difference in the range of phenols level (123 to 156 ppm) during spring (April to May), summer (July to August) and fall (October to November). The concentration of indoles was higher (P<0.05) during spring (14.3 ppm) than summer and fall (5.4~7.6 ppm). Level of BCFA ranging from 727 to 1,194 ppm was not different at any season. Among SCFA, there was no difference in propionic acid during any season but levels of acetic acid and butyric acid were highest (P<0.05) during spring season. Concentration of odorous compounds in pig slurry was highest during spring season. Interestingly, it tended to be lower during summer season compare to fall. This result might be due to relatively lower ventilation rate in order to maintain a constant temperature during spring season. Further study will be necessary to determine the relationship between the concentration of odorous compounds and ventilation system.

Effect of different feeding times using a diet containing betaine on production, blood profile and a short chain fatty acid in meat ducks exposed to a scorching heat wave (베타인 사료의 급여시기가 폭염에 노출된 오리의 짧은 사슬지방산, 혈액 프로파일 및 생산성에 미치는 효과)

  • The effects of different feeding times, using a diet containing 800 ppm betaine, on production, blood profile, and the short-chain fatty acid (SCFA), was investigated using 240 Cherry Valley (Anas platyrhynchos) meat ducks exposed to a scorching heat wave. The animals were randomly assigned to four groups, each of which was replicated three times with twenty ducks per replicate. The experimental period was 42 days for each group. Four groups were assigned into C (heat wave control group without betaine), T1 (ad libitum group fed a diet containing 800 ppm betaine), T2 (diet-restricted group fed twice daily between 05:00-10:00 and 17:00-20:00, using a diet containing 800 ppm betaine), and T3 (night-restricted group, fed from 17:00-10:00, with a diet containing 800 ppm betaine). At 42 days, body weight increased in order of T2, T1, T3 compared to the heat wave control group although. However, these differences were not found between the groups of T1 and T3. The heat wave control group, provided an ad libitum diet without betaine (C), showed an elevated feed conversion ratio compared to the groups fed a diet containing betaine. However, these differences were not found between the groups of T1, T2 and T3. RBC and platelet profiles except for PLT and MPV showed statistically significant differences between study groups fed a diet containing betaine. T2 presented significantly higher blood electrolytes $Na^+$ and $Cl^-$ than the other groups. T2 also showed a blood gas level that was generally higher than the other groups. Total SCFA, acetic acid and propionic acid concentration has been the increasing trend in T2, but butyric acid, isobutyric acid and valeric acid concentration has been the decreasing trend in T2 compared to the other groups. It is concluded that the feeding-restricted group, fed two times daily between 05:00-10:00 and 17:00-20:00, with a diet containing 800 ppm betaine may improve growth performance in meat ducks exposed to a scorching heat wave.
