The inhibitory effects of (-)-epigallocatechin gallate (EGCG), a major compound of green tea, on the development of locomotor sensitization, conditioned place preference (CPP) and dopamine receptor supersensitivity induced by the repeated administration of morphine were investigated in mice. A single administration of morphine produces hyperlocomotion. The repeated administration of morphine develops sensitization, a progressive enhancement of locomotion, which is used as a model for studying the craving and drug-seeking behaviors characterizing addiction, and CPP, which is used as a model for studying drug reinforcement, respectively. EGCG inhibited morphine-induced hyperlocomotion, sensitization and CPP. In addition, EGCG inhibited the development of postsynaptic dopamine receptors supersensitivity, which may be an underlying common mechanism that mediates the morphine-induced dopaminergic behaviors such as sensitization and CPP. Apomorphine (a dopamine agonist)-induced climbing behaviors also were inhibited by a single direct administration of EGCG These results provide evidence that EGCG has anti-dopaminergic activity, as inhibiting the development of dopamine receptor supersensitivity and apomorphine-induced climbing behaviors. Therefore, it is suggested that green tea may be useful for the prevention and therapy of these adverse actions of morphine.
Biofouling in brackish water reverse osmosis (RO) membranes still needs extensive research to understand cause and mechanism and to obtain methods for reduction of its impact on RO applications. Natural compounds with biofilm formation inhibitory properties are being investigated. Two compounds, vanillin and Epigallocatechin gallate (EGCG), were selected due to their great potential on biofilm formation inhibition. Vanillin shows inhibition on quorum sensing mechanisms of biofilm formation. EGCG has potential to inactivate microbial activity. The two compounds were incorporated in typical polyamide reverse osmosis membranes and evaluated on flux behaviours and biofilm formation potential. The surface properties of membrane coated with vanillin were changed tremendously compared to those with EGCG. As a result, the flux was reduced substantially. The biofilm formation seems hindered with EGCG coated membranes compared to the virgin membranes. More research is needed to optimize coating methods applicable to RO membranes and to enhance biofouling reduction.
천연 녹차추출물인 epigallocatechin gallate (EGCG)의 메추리알 장조림에서 천연식품첨가제로서의 적용 가능성을 탐색하고자 S. Enteritidis에 대한 항미생물 활성을 냉장온도($4^{\circ}C$)와 일반실내온도($25^{\circ}C$)에서 평가하였다. 순수배양액에서의 실험결과 첨가된 EGCG의 농도($200-800{\mu}g/mL$)에 따라 $4^{\circ}C$에서 5일에서 16일 사이에서 그리고 $25^{\circ}C$에서 1일부터 6일 사이에서 S. Enteritidis균이 검출한계 이하로 억제되는 것이 관찰되었다. 이와 대조적으로 간장소스에서의 실험에서는 $25^{\circ}C$에서 첨가된 EGCG의 농도와 관계없이 S. Enteritidis균이 억제되지 않았으며, $4^{\circ}C$에서는 $400{\mu}g/mL$ 이상의 농도일 때 16일에 S. Enteritidis균이 검출한계 이하로 억제되는 것이 관찰되었다. 메추리알 장조림의 경우, $25^{\circ}C$에서 간장소스의 결과와 동일하게 어떠한 EGCG의 항미생물 활성이 나타나지 않았지만, $4^{\circ}C$에서 $400{\mu}g/mL$의 EGCG 농도 첨가 시 16일에 S. Enteritidis 균이 검출한계 이하로 억제되는 것이 확인되었다. 따라서, 메추리알 장조림 완제품에서 EGCG는 $4^{\circ}C$ 이하의 저온유통체계(cold chain system)하에서 S. Enteritidis과 같은 식중독세균을 제어할 수 있는 적합한 천연 식품첨가제로서 활용될 수 있을 것으로 기대된다.
A facile and convenient method was developed for the mass production of epigallocatechin gallate (EGCG) rich green tea extract (Er-GTE). The Er-GTE was successfully obtained from the crude water extract of green tea by the combination of two step purification, i.e., a simple adsorption process on the cation exchange resins (Trilite SCR-B) followed by the chromatography with Diaion HP-20 resins. The green tea extract produced by water extraction under $45^{\circ}C$ was subjected to adsorb on the strongly acidic cation exchange resin, Trilite SCR-B. The eluate passed through the resin was reabsorbed on Diaion HP-20 resin, which was subjected to elute with a mixture of water and alcohol by conventional chromatographical manner. The EGCG content in Er-GTE was estimated above 97% by HP-LC analysis and the newly developed method was regarded as the most suitable and appropriate process for the mass production of epigallocatechin gallate rich green tea extract (Er-GTE).
본 연구에서는 다양한 생리활성이 보고된 폴리페놀 화합물인 EGCG의 화학안정성, H2O2 생성능 및 세포독성에 대하여 다양한 항산화제와의 조합에 의한 변화를 분석하였다. EGCG는 생리적 조건에서 갈색화합물로 산화되면서 불안정화되는데, catalase를 제외한 각종 항산화제제 SOD, ascorbic acid, NAC 및 GSH는 EGCG 갈색산화물의 생성을 유의적으로 저해하였다. EGCG에 의해 생성되는 $H_2O_2$는 catalase에 의해 거의 완벽하게 제거되었으며, SOD와 NAC에 의해서도 유의적으로 감소하였다. 하지만 GSH 및 고농도의 ascorbic acid의 존재 시 오히려 $H_2O_2$ 수준이 증가하는 현상을 나타내었다. EGCG의 HeLa 및 HT-29 세포에 대한 독성은 catalase, SOD 및 NAC 등과 같은 항산화제 존재 하에 유의적으로 감소하였고 NAC에 의한 EGCG 세포독성의 감소는 첨가된 NAC의 농도 증가에 따라 더욱 두드러졌다. 그러나 GSH 존재하에 EGCG의 독성은 GSH와 EGCG농도에 따라 다른 조절 양상을 나타내었으며, ascorbic acid에 의해서 EGCG의 세포독성이 약간 증가하는 현상을 나타내었다. 본 결과는 EGCG와 함께 처리된 다양한 항산화제들이 ROS의 소거 뿐만 아니라 EGCG 화학안정성 등 다른 요인에 영향을 미칠 수 있으며, 항산화제의 존재 하에 변형된 EGCG활성에 대해 ROS관련 기작 외에 다양한 요인들에 대한 고려가 함께 이루어져야 함을 시사한다.
Epigallocatechin 3-gallate (EGCG), one of the major catechins of tea, was isolated from the decaffeinated, crude methanolic extract of Indian green tea (Camellia sinensis L. O. Kuntze) using chromatographic techniques. EGCG was then screened for antidiarrhoeal activity against 30 strains (clinical isolates) of V. cholerae, which is a well known Gram negative bacillus functioning as the pathogen of cholera. V. cholerae strains like V. cholerae 69, 71, 83, 214, 978, 1021, 1315, 1347, 1348, 569B and ATCC 14033 were inhibited by EGCG at a concentration of $25\;{\mu}g/ml$ whereas V. cholerae 10, 522, 976 were even more sensitive, being inhibited at $10\;{\mu}g/ml$ level. However, V. cholerae DN 16, DN 26, 30, 42, 56, 58, 113, 117, 564, 593, 972 and ATCC 14035 were inhibited at $50\;{\mu}g/ml$ level of EGCG. Only four strains were inhibited at $100\;{\mu}g/ml$. In this study the isolated compound was found to be bacteriostatic in its mechanism of action. In the in vivo experiment using the rabbit ileal loop model two different dosages of EGCG ($500\;{\mu}g/ml$ and $1,000\;{\mu}g/ml$) were able to protect the animals when they were challenged with V. cholerae 569B in the ileum.
배경: 혈관내막과다증식은 혈관평활근세포가 내막에서 과다증식하여 나타나며, epigallocatechin-3-gallate(EGCG)는 혈관평활근세포의 증식을 억제하는 효과가 있는 것으로 알려져 있다. 따라서 EGCG는 혈관내막과다증식을 억제할 수 있을 것으로 생각된다. 대상 및 방법: 다른 농도의 EGCG를 포함한 배양 배지에서 인간제대정 맥내피세포(HUVEC)와 쥐대동맥평활근세포(RASMC)를 배양하고, 세포증식과 이동속도를 측정하였다. 20마리의 개의 양측 경동맥에 자가경정맥을 이식하였으며, 실험군(n=10)에 대해서는 정맥도관을 이식 전 30분간 EGCG용액에 보관하였으며, 이식 후 300mM의 EGCG를 혈관주위에 도포하였다. 6주 후에 경정맥이식편의 내막과 중간막의 두께를 측정하였다. 결과: 내피세포와 혈관평활근세포의 증식은 EGCG에 의해 억제되었다. 혈관평활근세포의 이동성은 EGCG에 의해 억제되었으나, 내피세포의 이동성은 영향을 받지 않았다. 동물실험 결과 대조군에 비해 EGCG군에서 내막의 두께가 얇았으며(p<0.05), 중간막의 두께는 두 군간에 차이가 얼었고, 내막/중간막의 두께비는 EGCG군에서 낮게 관찰되었다(p<0.05). 결론: EGCG는 혈관수술 후 혈관내막과다증식을 억제하는 효과가 있으며, 이는 혈관평활근세포의 증식 및 이동을 억제하여 효과를 나타낸다고 생각된다. 따라서 EGCG는 혈관내막과다증식을 예방하는 치료에 효과적으로 사용될 수 있을 것으로 생각된다.
Epigallocatechin-3-gallate(EGCG)는 녹차에 함유된 flavonoid 계열 화합물로, 녹차의 대표적인 유효성분으로 알려져 있다. 본 연구는 3T3-L1 세포에서 EGCG의 항비만 작용 및 항산화 효소 활성을 평가하였다. EGCG는 지방세포 분화를 유의적으로 억제하였으며, 지방세포 분화와 관련된 단백질인 $C/EBP{\alpha}$와 aP2의 발현을 감소시켰다. 동시에 EGCG는 GSH의 활성을 유도하였으며, 항산화 효소인 SOD, CAT, GPx 및 GR의 활성을 증가시켰다. EGCG는 지방세포 분화 동안 활성산소종의 형성을 감소시켰다. 체내 phase II 효소인 HO-1과 GCLC는 EGCG에 의해 발현이 유도되었다. 따라서 EGCG는 3T3-L1 세포에서 항산화 효소의 활성화를 유도하며 동시에 지방세포로의 분화를 억제하는 것으로 나타났다. 또한, EGCG는 활성산소종의 생성을 억제하였으며, 이는 EGCG의 항산화 작용에 의한 지방세포 분화 억제 작용이 관련이 있는 것으로 판단된다. EGCG에 의한 항비만 효과와 항산화 작용을 연구한 본 연구는 녹차를 활용한 체지방 감소 효능을 지닌 식품 개발에 기초 자료로 활용될 수 있을 것으로 생각된다.
목적 : 항균제 polyhexamethylene Biguanide(PHMB), epigallocatechin gallate(EGCG) 및 PHMB/EGCG 혼합물의 각막상피세포(primary human corneal epithelial cells, HCEpiCs)에 대한 급성독성을 평가하고자 하였다. 방법 : 각막상피세포를 0.00001~0.005% PHMB, 0.001~5% EGCG 및 0.00005% PHMB/0.05% EGCG 혼합물이 각각 포함된 배양액에서 30분, 60분, 120분 및 240분 동안 배양하였다. 배양한 각막상피세포를 고정한 다음 Draq 5로 염색하고 공초점현미경과 ImageXpress $Ultra^{TM}$를 이용하여 세포형태를 관찰하여 세포생존율과 세포자살(apoptosis)을 비교하였다. 결과 : 배양된 각막상피세포는 0.00005% 이하의 PHMB 농도 및 0.05% 이하의 EGCG 농도에서는 세포 독성이 나타나지 않았다. 0.00005% PHMB/0.05% EGCG 혼합물이 포함된 배양액에서 급성 세포독성은 관찰되지 않았으나 240분 배양시킨 경우에는 손상된 각막상피세포 수가 증가하고 생존 세포의 수는 감소하였다. 결론 : 항균 시너지 효과를 갖는다고 보고된 0.00005% PHMB/0.05% EGCG 혼합물의 경우 각막상피세포에 대한 급성독성은 없었으나 만성효과에 대해서는 추가적인 연구가 필요할 것으로 생각된다.

EGCG에 의한 포스포리파제 D의 활성화는 포스포리파제 C효소의 특이적인 억제제와 지질분해효소 활성이 억제제 포스포리파제 \gama1$의 변이체를 이용하여 실험한 결과 세포내 칼슘에 의존적인 양상을 보였주었다. 이러한 포스포리파제 D의 활성화는 아마도 칼슘 의존성 단백질 키나아제 II (CaM kinase II)를 통하여 일어나는것으로 여겨진다. 흥미롭게도, EGCG는 포스포리파제 \gama1$를 세포질에서 세포막으로의 이동을 유도하였으며, 포스포리파제 \gama1$와 PLD1의 결합을 유도하였음을 관찰하였다. 이상의 결과들을 종합하여 볼 때, EGCG가 사람의 교세포에서 포스포리파제 \gama1$-칼슘-CaM kinase II의 신호전달 경로를 통하여 포스포리파제 D의 활성을 유도하는 것으로 사료된다.
