전남지역 숲길의 모노테르펜류 농도와 공기질 특성 (The Characteristics of Monoterpene and Air Quality in Ambient Air at Forested Road in Jeollanam-do)

  • 본 연구는 숲속 환경대기에서 모노테르펜류(${\alpha}$-Pinene, camphene, sabinene, ${\beta}$-pinene, myrcene, terpinolene, limonene, 1,8-cineol, linalool, camphor)의 농도분포, 숲속 대기의 청정도 및 공기음이온 분포를 파악하기 위해 전남도 내 5개 유명 숲을 대상으로 3 계절(봄, 여름, 가을) 동안 수행하였다. 4 시간 평균농도 기준으로 대나무가 우점하고 있는 죽녹원지점에서 ${\beta}$-pinene+myrcene이 958 pptv, 우드랜드와 순천만에서 ${\alpha}$-pinene(524 pptv)과, limonene(445 pptv)이 각각 높은 농도로 나타났다. 계절별로는 우드랜드와 선암사는 봄, 죽녹원과 순천만은 여름에 가장 최고치를 보였다. 대기환경기준물질 농도는 환경정책기본법의 환경기준보다 매우 낮은 농도로 나타났다. 공기음이온은 지점별로 공기 1 mL당 최고 400~3,000개로 조사되었다. 모노테르펜과 공기음이온사이에 유의한 상관은 나타나지 않았다.

곰취와 한대리곰취의 휘발성 향기성분 분석 (A Comparison the Volatile Aroma Compounds between Ligularia fischeri and Ligularia fischeri var. spiciformis Leaves)

  • The volatile aroma of fresh leaves is one of main factor in taste of all the edible green plants. The volatile aroma in almost edible green leaves are suggested as essential oil compounds. Ligularia fischeri, Synurus deltoides, Ligularia fischeri var. spiciformis and Aster scaber are one of the favourable edible green plants in Korea. In this study, volatile aroma compounds from Ligularia fischeri and Ligularia fischeri var. spiciformis species were analyzed by the SPME/GC/MSD method. Ligularia fischeri had 78 volatile aroma compounds such as D-limonene(20.28%), ${\alpha}$-pinene(dextro, 14.15%), L-${\beta}$-pinene(12.85%), 3-carene, ${\beta}$-cubebene(10.39%), etc. Ligularia fischeri var. spiciformis had 83 volatile aroma compounds such as D-limonene(36.97%), ${\beta}$-cubebene(13.95%), L-${\beta}$-pinene(13.38%), ${\alpha}$-pinene(dextro, 4.76%), caryophylle-ne(3.33%) etc. Conclusively, the commom volatile aroma compounds in Ligularia fischeri and Ligularia fischeri var. spiciformis leaves were D-limonene, ${\alpha}$-pinene, L-${\beta}$-pinene, ${\beta}$-cubebene, Caryophyllene, ${\alpha}$-farnesene, terpinolen. However, the composition and amount of volatile aroma compounds were very different between the two species.

북방수염하늘소의 Ipsenol, Ipsdienol에 대한 유인효과 (Attractive Efficacy of Ipsenol and Ipsdienol against Monochamus saltuarius Gebler (Coleoptera: Cerambycidae))

  • 국내 전역에 분포되어 있는 잣나무(Pinus koraiensis)에 소나무재선충(Bursaphelenchus xylophilus)을 옮기는 새로운 매개충으로 북방수염하늘소(Monochamus saltuarius)에 의한 발병이 보고되면서 소나무재선충의 빠른 확산이 우려되고 있다. 잣나무의 주요 성분이며 Monochamus속에 유인성분으로 보고된 $\alpha$-pinene과 에탄올, 나무종류의 유인성분으로 알려진 Ips 속 페로몬(ipsenol, ipsdienol)에 대하여 혼합 유인효과를 검토한 결과, ipsenol + ipsdienol에서 67.9%, 그리고 ipsenol과 ipsdienol을 $\alpha$-pinene + 에탄올과 혼합 처리 시 각각 67.7%, 79.3%의 유인력을 보였다. 특히 ipsenol에 비해 ipsdienol의 $\alpha$-pinene + 에탄올에 대한 혼합효과가 높았으며, 북방수염하늘소의 수컷에 비해 암컷이 높은 유인력을 나타냈다.

소나무 잎과 가지의 정유와 그 성분들의 항산화 및 항노화 작용 (Anti-oxidant and Anti-aging Activities of Essential Oils of Pinus densiflora Needles and Twigs)

  • To investigate effects of pine essential oil isolated by steam distillation from Pinus densiflora needles (PN) and twigs (PT) on anti-oxidant and anti-aging activities, the experiments were carried out to determine anti-oxidant and antiaging activities on DPPH radical scavenging activity, NBT/xanthine oxidase-superoxide scavenging activity, silica-induced intracellular $H_2O_2$ and hydroxyl radical generation in RAW264.7 cells, hyaluronidase and elastase activities in vitro. Essential oil of PN and PT were contained 0.225 and 0.176 (w/v) %, respectively. PN was contained with 30 kinds of essential oil and its major constituent is $\alpha$-pinene (21.5%). Further PT was contained with 40 kinds of essential oil and its major constituent is $\beta$-pinene (22.4%) in GC/MS assay. Other essential oils of PN were $\beta$-pinene, camphene, myrcene, limonene, terpinolene, $\alpha$-terpineol, bornyl acetate, $\alpha$-copaene, caryophellene and humulene and PT were $\alpha$-pinene, camphene, phellandrene, limonene, terpinolene, $\alpha$-terpineol, bornyl acetate, $\alpha$-copaene, caryophellene and humulene. The essential oil of PT have more active than that of PN in anti-oxidant activity which has significant DPPH radical and superoxide scavenging activity, and significant inhibitory activities on silica-induced intracellular $H_2O_2$ and hydroxyl radical generation, as well as, significantly inhibited elastase and hyaluronidase activities. Further, phellandrene, myrcene and $\alpha$-pinene have DPPH radical and superoxide scavenging activities, $\beta$-pinene, terpinolene, myrcene and phellandrene inhibited silica-induced intracellular $H_2O_2$ and hydroxyl radical generations. And also phellandrene and $\beta$-pinene inhibited hyaluronidase and elastase activities. In conclusion, the essential oils isolated from PN and PT have anti-oxidant and anti-aging activities.

  • In continuation of our studies on essential oils of Angelica genus(Umbelliferae), We report on the components of essential oils obtained from the root of Angelica tenuissima Nakai(藁子). Oils were obtained from the dried roots by steam distillation and fractionated by column chromatography. Each isolate or fraction was identified by GC, GC-MS and spectral analysis. Essential oils of the root of A. tenuissima(Gaoben) were found to contain $\alpha-pinene,\;camphene,\;\beta-pinene,\;myrcene,\;\alpha-phellandrene,\;\Delta-3-carene,\;p-cymene,\;limonene,\;\gamma-terpinene,\;terpinolene,\;4-vinylguauacol,\;\gamma-elemene$, one aromatic compound, three unidentified sesquiterpene alcohols, butylidenephthalide, senkyunolide and Z-ligustilide which was the most abundant compound comprising 75% of the whole oil. Also butylphthalide and hydroxybutylidenephalide were tentatively identified.

계절에 따른 울산지역 산책로의 피톤치드 특성연구 (Study on Seasonal Variation Characteristics of Forest Phytoncide in Ulsan Metropolitan Trails)

  • This study was conducted to investigate the seasonal variation characteristics of phytoncide in trail of Ulsan Metropolitan. Air samples were collected from May to December 2011. They were collected using Tenax Ta tube and phytoncides were detected and quantified using a Gas Chromatograph Mass Spectrometer (GC/MSD). This study are summarized as follows; The seasonal concentrations of phytoncide are Munsu Mt. 272.3 ${\mu}g/m^3$, Samho Mt. 192.4 ${\mu}g/m^3$, Shinbul Mt. 50.9 ${\mu}g/m^3$, Sibli Bamboo forest 22.4 ${\mu}g/m^3$ and Joongbu Fire Station 24.4 ${\mu}g/m^3$. In Munsu Mt., Samho Mt. and Shinbul Mt. major component ratio is ${\alpha}$-Pinene > ${\beta}$-Pinene+Myrcene >Limonene >Camphene > ${\gamma}$-Terpinene > ${\alpha}$-Terpinene. Bamboo forest and Joongbu fire station major component ratio is ${\alpha}$-Pinene >Limonene > ${\beta}$-Pinene+Myrcene >Camphene > ${\alpha}$-Terpinene. The variation of seasonal concentration is summer >spring >fall >winter. The phytoncide concentration of coniferous forest(Munsu Mt., Samho Mt.) is higher than broadleaf forest(Shinbul Mt.).

침엽수 4종 목부 정유의 항염증 효과 평가 (Anti-Inflammatory Effect of Essential Oils Extracted from Wood of Four Coniferous Tree Species)

  • 본 연구는 국내산 편백, 소나무, 잣나무, 일본잎갈나무 목부 정유의 항염증 효과를 평가 하였다. 실험에 사용된 정유는 각 수종의 목부에서 증류법(hydrodistillation)으로 추출되었다. 추출한 정유들을 GC-MS를 통해 성분 분석한 결과, 편백의 경우 ${\alpha}$-cadinol (19.25%), ${\tau}$-muurolol (14.20%) 및 ${\alpha}$-pinene (13.74%)이, 소나무는 ${\alpha}$-pinene (47.16%), longifolene (14.31%), ${\beta}$-phellandrene (11.78%) 및 ${\beta}$-pinene (11.02%)이 주요성분이었으며, 잣나무의 경우 ${\alpha}$-pinene (13.49%)와 longifolene(10.79%), 낙엽송의 경우 geranyl linalool (23.58%) 및 ${\alpha}$-pinene (18.57%)이 주요성분으로 확인되었다. 침엽수 목부 정유들의 항염증 효과는 LPS(Lipopolysaccharide)로 염증반응이 유도된 RBL-2H3 비만세포주에 정유를 처리한 후 IL-4(interleukin-4), IL-13(interleukin-13), ${\beta}$-hexosaminidase의 발현 변화를 조사함으로써 평가하였다. MTT assay를 통한 세포 독성 평가 결과, 침엽수 4종 정유 모두 $10^{-7}{\sim}10^{-5}%$ 농도에서 세포 독성을 보이지 않았다. 염증반응이 유도된 RBL-2H3 세포에 침엽수 4종의 목부 정유를 $10^{-7}%$ 농도로 처리하였을 때, LPS 처리군 대비 침엽수 4종의 목부 처리군에서 IL-4와 IL-13의 상대적 발현량이 큰 폭으로 감소하였으며, ${\beta}$-hexosaminidase 분비가 유의하게 억제되었다. 침엽수 정유 4종의 IL-4 상대적 발현량은 LPS 처리군 대비 소나무 69.6%, 잣나무 63.2%, 편백 55.1% 그리고 낙엽송 45.8% 감소하였다. IL-13의 상대적 발현량은 소나무 57.8%, 잣나무 57.1%, 편백 51.1% 그리고 낙엽송 34.5% 감소하였다. ${\beta}$-Hexosaminidase 분비의 경우 LPS 처리군 대비 편백 38.1%, 소나무 33.0%, 잣나무 27.4% 그리고 낙엽송 9.1% 억제하였다. Il-4, IL-13 및 ${\beta}$-hexosaminidase 분비를 비교한 결과, 침엽수 목부 정유 4종 중 소나무 정유의 활성이 가장 우수하였다. 이러한 결과는 국내 침엽수 4종의 목부 정유는 염증반응을 유발하는 싸이토카인의 발현 억제를 통하여 항염증 효과를 나타냄을 확인할 수 있었다.

초임계 이산화탄소로 추출한 유자껍질의 향기성분 (Volatile Compounds of Citron (Citrus Junos) Peel extracted by Supercritical Carbon Dioxide)

  • 초임계 이산화탄소를 이용하여 유자(Citrus junos) 껍질의 향기성분을 추출하기 위해 동결건조한 유자과피분말을 4,000psi와 4$0^{\circ}C$에서 운전하면서 이산화탄소의 소비량에 따라 4단계로 분획하여 분획별 향기성분의 특성을 조사하였다. 초임계 이산화탄소에 대한 유자의 향기성분의 수율은 0.011g/CO2(L)였으며, 최대추출 수율은 8.812g/kg이었다. 초임계 이산화탄소로 추출한 유지과피의 주요향기성분은 dl-limonene, Υ-terpinene, linalool, sabinene, $\beta$-myrcene, $\alpha$-pinene, $\beta$-farnesene, $\alpha$-terpineol과 terpinolene 등 9종이었다. 분획별 향기성분은 휘발성이 높은 $\alpha$-pinene, $\beta$-myrcene과 dl-limonene은 추출시간이 길어짐에 따라 함량이 감소하였으며, 이와 반대로 휘발성이 낮은 $\alpha$-terpineol과 $\beta$-farnesene은 추출시간이 길어짐에 따라 함량이 증가하였다. 따라서 초임계 이산화탄소로 유자의 향기성분을 추출할 경우 추출시간에 따라 얻어지는 분획의 향기성분 조성이 서로 다른 것으로 나타났다.

Antifungal Activities of Essential Oils from Glehnia littoralis Alone and in Combination with Ketoconazole

  • The antifungal activities of essential oils from the leaves of Glehnia littoralis, which is cultivated in Korea, were evaluated against pathogenic Trichophyton species by the broth dilution method and the disk diffusion test. Additionally, the effects of the oils together with ketoconazole were tested by the checkerboard titer test. The essential oil fraction and its main components showed significant inhibition of the tested Trichophyton fungi, with minimal inhibitor concentrations (MICs) in the range of 16-32 mg/ml. The results suggest that activities of this oil are based mainly on the contents of ${\alpha}-pinene$ (22.17%), the next prominent component of the oil fraction, while the first main components ${\beta}-pinene$ (57.83%) have relatively mild activity. The MICs of ${\alpha}-pinene$ and ${\beta}-pinene$ were 1-4 mg/ml and 4-32 mg/ml, respectively. Additionally the Glehnia oil fraction and its main components as well, exhibited significant synergism with ketoconazole against Trichophyton rubrum.

구릿대 뿌리 정유의 항진균 효과 (Antifungal Activities of Essential Oil from the Roots of Angelica dahurica Bentham et Hooker f.)

  • The essential oil was extracted from the roots of Angelica dahurica Bentham et Hooker f. by steam distillation and its composition was analyzed by GC-MS. The antifungal activities were evaluated by micro-dilution method against five Aspergillus and three Trichophyton species. The most abundant component was ${\alpha}$-pinene (17.21%) among 40 compounds identified in this oil. The essential oil fraction of A. dahurica and ${\alpha}$-pinene exhibited marked inhibiting activities against the tested Aspergillus and Trichophyton species with MICs (minimum inhibitory concentrations) between 0.12 mg/ml and 8 mg/ml.
