한국산과 중국산 고본(藁本)중 정유성분의 정성.정량에 관한 연구 (A study on the Qualitative and Quantitative Analysis of Essential oil in Angelicae tenuissimae Radix or Ligustici rhizoma)

  • Angelicae tenuissimae Radix의 정유성분에 대한 구조규명과 채취장소에 따른 정유성분의 정량실험 결과 및 Ligustici rhizoma와의 정성 정량적 차이점에 관한 실험을 하여 다음과 같은 결론을 얻었다. Ligustici rhizoma와 Angelicae tenuissimae Radix 산지별 네가지 시료의 용출함량을 비교한 결과 경북 석포에서 채집한 시료가 11.32%로 가장 높은 값을 나타내었고, 중국산 Ligustici rhizoma가 3.49%로 가장 낮은 용출함량을 나타냈다. GC/MS로 구조를 분석한 결과 Angelicae tenuissimae Radix의 정유성 분은 ${\alpha}-pinene,\;{\alpha}-phellendrene,\;{\delta}-3-carene,\;{\gamma}-terpinene$, 4-vinylguaiacol, ${\alpha}-farnesene,\;(t)-{\beta}-farnessene,\;{\gamma}-elemene$, sesquiterpene alcohol, butylphthalide, senkyunolide, butylidenephthalide, z-butylidenedihydrophthalide (z-ligustilide) 등을 확인할 수 있었는데, Ligustici rhizoma 정유성분에서는 Angelicae tenuissimae Radix와 다르게 senkyunolide가 38.7%로 주성분이었으며, butylphthalide(17%), cnidilide(8.8%), z-ligustilide(8.8%) 등을 함유하였다. 따라서 Ligustici rhizoma와 Angelicae tenuissimae Radix의 주성분이 서로 상이하며, 그 조성비도 큰 차이가 있음을 확인할 수 있었다. 본 연구 결과로부터 고본의 주성분은 ligustilide (한국산 고본)과 senkyunolide (중국산)로서 앞으로 임상실험을 통하여 이들 성분의 정확한 약리효과를 규정함으로써 고본(藁本)을 규격화 하여야 할 것으로 생각된다.

Comparison of Chemical Compositions and Antimicrobial Activities of Essential Oils from Three Conifer Trees; Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa

  • The chemical compositions, and antibacterial and antifungal effects of essential oils extracted from three coniferous species, Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa, were investigated. Gas chromatography mass analysis of the essential oils revealed that the major components and the percentage of each essential oil were 16.66% $\beta$-phellandrene and 14.85% $\alpha$-pinene in P. densiflora; 31.45% kaur-16-ene and 11.06% sabinene in C. japonica; and 18.75% bicyclo [2,2,1] heptan-2-ol and 17.41% 2-carene in Ch. obtusa. The antimicrobial assay by agar disc diffusion method showed that $2.2{\mu}g$ of Ch. obtusa oil inhibited most effectively the growth of Escherichia coli ATCC 33312 and Klebsiella oxytoca ATCC 10031, whereas the C. japonica oil gave weak antimicrobial activity. The minimal inhibitory concentration(MIC) values for bacterial strains were in the range of 5.45-21.8 mg/ml depending on essential oils, but most Gram-negative bacteria were resistant even at 21.8 mg oil/ml. P. densiflora oil showed the most effective antifungal activity and the MIC values for Cryptococcus neoformans B42419 and Candida glabrata YFCC 062CCM 11658 were as low as 0.545 and 2.18 mg/ml, respectively. Cryp. neoformans B42419 was the most sensitive to all essential oils in the range of 0.545-2.18 mg/ml. Our data clearly showed that the essential oils from the three conifers had effective antimicrobial activity, especially against fungi.

Fragrance Composition in Six Tree Peony Cultivars

  • Tree peony is a traditional famous flower of China, and plays an important role in Chinese traditional culture. But the floral scent of tree peony in vivo is little known. In this study, in order to explore the floral composition of tree peony, floral volatiles of six cultivars, including Paeonia suffruticosa 'Zhaofen' (ZF), P. suffruticosa 'Luoyanghong' (LYH), P. ostii 'Fengdanbai' (FDB), P. ${\times}$ lemonei 'High noon' (HN), P. ${\times}$ lemonei 'Renown' (R), and P. rockii 'Gaoyuanshenghuo' (GYSH) were collected by dynamic headspace and then identified by Automated Thermal Desorption-Gas Chromatography/Mass Spectometry. The results showed that floral fragrances of the six cultivars were qualitatively and quantitatively distinct. A total of 105 volatiles involving ten categories were detected. But not all volatile categories were emitted from these cultivars. The six peony cultivars emitted some shared compounds and peculiar compounds. The total released amounts of volatiles emitted from six cultivars were found significantly different, which was greatest for 'GYSH'. The most abundant volatile compounds detected from 'ZF', 'LYH', 'FDB', 'HN', 'R', and 'GYSH' were respectively ${\alpha}$-pinene, 2,3-dihydroxy propanal, 3-methyl-1-butanol, 2-ethyl-1-hexanol, acetic acid 1-methylethyl ester, and 5-ethyl-2,2,3-trimethyl heptane. This result may contribute to exploring the biosynthesis and emission mechanism of floral scent in tree peony.

  • 소나무의 monoterpene조성(組成)가 솔잎혹파리에 대(對)한 저항성(抵抗性) 관계(關係)를 구명(究明)하기 위(爲)하여 솔잎혹파리 저항성(抵抗性) 소나무 5개(5個) 수종(樹種)가 감수성(感受性)소나무 2개(2個) 수종(樹種), 그리고 감수성(感受性) 해송(海松)가운데 피해목(被害木)과 선발목(選拔木)의 침엽(針葉)을 GLC로 분석(分析)하여 monoterpenes의 조성(組成)을 근거(根據)로 저항성(抵抗性) 또는 감수성(感受性) 수종(樹種)을 구별(區別)할 수는 없었다. 그러나 적송수종내(赤松樹種內)의 피해목(被害木)과 선발목간(選拔木間)에는 limonene, ${\alpha}-pinene$, myrcene 및 ${\beta}-phellandrene$의 농도(濃度)아 조성비(組成比)에 차이(差異)가 인정(認定)되였다. 적송(赤松)의 선발목(選拔木)에서 limonene의 농도(濃度)는 솔잎혹파리 산란이후(産卵以後) 점차(漸次) 증가(增加)하였으나 피해목(被害木)에서는 증가(增加)하지 않았다. 적송(赤松)의 선발목(選拔木)에서 솔잎혹파리에 대(對)한 내충성분은limonene으로 보였다.

돼지풀 잎의 휘발성 물질이 수용체 유식물 생장에 미치는 알레로파시 효과 (Allelopathic Effects of Volatile Compounds from Ambrosia artemisiifolia Leaves on the Selected Species)

  • 돼지풀 잎에서 검정된 monoterpenoids는 약 29종이었고, 그 가운데 octene, $\alpha$-ponene, $\beta$-myrcene, 1.3.6-octatrine, endo-borneol, (-)-bomyl acetate의 함량이 높다. Monoterenoids 총 함량은 초봄부터 점차 증가하여 7월에 가장 높았고 그후 감소하였다. 돼지풀 잎에 함유된 휘발성물질은 수용체 유식물의 뿌리와 줄기 생장에 억제효과를 나타내었으며, 생엽량의 증가에 따라 억제효과도 증가하였다. 이에 따른 억제효과 차이의 유의성은 대부분이 P<0.05 수준이었다. 억제효과는 처리식물에 따라 큰 차이가 있었다. 돼지풀 생엽의 채취시기에 따른 유식물 생장 억제효과의 경향성은 대부분의 처리식물에서 7월>6월$\geq$5월 순 이었다.

Antileishmanial and Cytotoxic Effects of Essential Oil and Methanolic Extract of Myrtus communis L.

  • Plants used for traditional medicine contain a wide range of substances that can be used to treat various diseases such as infectious diseases. The present study was designed to evaluate the antileishmanial effects of the essential oil and methanolic extract of Myrtus communis against Leishmania tropica on an in vitro model. Antileishmanial effects of essential oil and methanolic extract of M. communis on promastigote forms and their cytotoxic activities against J774 cells were evaluated using MTT assay for 72 hr. In addition, their leishmanicidal activity against amastigote forms was determined in a macrophage model, for 72 hr. Findings showed that the main components of essential oil were ${\alpha}$-pinene (24.7%), 1,8-cineole (19.6%), and linalool (12.6%). Findings demonstrated that M. communis, particularly its essential oil, significantly (P<0.05) inhibited the growth rate of promastigote and amastigote forms of L. tropica based on a dose-dependent response. The $IC_{50}$ values for essential oil and methanolic extract was 8.4 and $28.9{\mu}g/ml$ against promastigotes, respectively. These values were 11.6 and $40.8{\mu}g/ml$ against amastigote forms, respectively. Glucantime as control drug also revealed $IC_{50}$ values of 88.3 and $44.6{\mu}g/ml$ for promastigotes and amastigotes of L. tropica, respectively. The in vitro assay demonstrated no significant cytotoxicity in J774 cells. However, essential oil indicated a more cytotoxic effect as compared with the methanolic extract of M. communis. The findings of the present study demonstrated that M. communis might be a natural source for production of a new leishmanicidal agent.

Chemical Composition and Biological Activities of Essential Oils Extracted from Korean Endemic Citrus Species

  • The aim of this study was to analyze the chemical composition of 14 kinds of citrus oils and to test their biological activities. Citrus essential oils were obtained by steam distillation from immature fruits collected from Jeju Island and were analyzed using gas chromatograph (GC)-flame ionization detectors (FID) and GC-MS. Limonene (55.4% to 91.7%), myrcene (2.1% to 32.1%), ${\alpha}$-pinene (0.6% to 1.6%) and linalool (0.4% to 6.9%) were the major components in most citrus species. To evaluate in vitro antibacterial activity, all essential oils were tested against Propionibacterium acnes and Staphylococcus epidermidis. Nine out of fourteen citrus oils exhibited antibacterial activity against P. acnes, but not against S. epidermidis. The effects of the citrus oils on DPPH radical scavenging, superoxide radical anion scavenging, nitric oxide radical, and cytotoxicity were also assessed. Three essential citrus oils, Joadeung, Dongjunggyul, and Bujiwha, exhibited potent inhibitory effects on nitric oxide production. Two essential oils, Dongjunggyul and Joadeung, showed potent free radical scavenging activities in the DPPH assay. For future applications in cosmetic products, we also performed MTT assays in a human dermal fibroblast cell line. The majority of the essential oils showed no cytotoxicity. The results indicate that citrus essential oils can be useful natural agents for cosmetic application.

5-Lipoxygenase Inhibition of the Fructus of Foeniculum vulgare and Its Constituents

  • The fruits of Foeniculum vulgare (Foeniculi Fructus) have been widely used in Chinese medicine as an antiemetic, ameliorating stomach ailments and as an analgesic. In order to establish its potential for antiallergic use, inhibitory actions of the fruit on 5-lipoxgenase (5-LOX) and ${\beta}$-hexosaminidase release were evaluated. The 70% ethanol extract of this plant material (FR) considerably inhibited 5-LOX-catalyzed leukotriene production from A23187-induced rat basophilic leukemia (RBL)-1 cells. The $IC_{50}$ was $3.2{\mu}g/ml$. From this extract, 12 major compounds including sabinene, fenchone, ${\gamma}$-terpinene, ${\alpha}$-pinene, limonene, p-anisylacetone, panisylaldehyde, estragole (4-allylanisole), trans-anethole, scopoletin, bergapten and umbelliferone were isolated. And it was found that several terpene derivatives including ${\gamma}$-terpinene and fenchone as well as phenylpropanoid, trans-anethole, showed considerable inhibitory action of 5-LOX. In particular, the $IC_{50}$ of trans-anethole was $51.6{\mu}M$. In contrast, FR and the isolated compounds did not show considerable inhibitory activity on the degranulation reaction of ${\beta}$-hexosaminidase release from antigen-treated RBL-2H3 cells. Against arachidonic acid-induced ear edema in mice, FR and trans-anethole showed significant inhibition by oral administration at doses of 100-400 mg/kg. In conclusion, FR and several major constituents are 5-LOX inhibitors and they may have potential for treating 5-LOX-related disorders.

GC-MS 기반 대사체학 기술을 응용한 참당귀의 산지비교분석 (Comparative Analysis of Cultivation Region of Angelica gigas Using a GC-MS-Based Metabolomics Approach)

  • Background: A set of logical criteria that can accurately identify and verify the cultivation region of raw materials is a critical tool for the scientific management of traditional herbal medicine. Methods and Results: Volatile compounds were obtained from 19 and 32 samples of Angelica gigas Nakai cultivated in Korea and China, respectively, by using steam distillation extraction. The metabolites were identified using GC/MS by querying against the NIST reference library. Data binning was performed to normalize the number of variables used in statistical analysis. Multivariate statistical analyses, such as Principal Component Analysis (PCA), Partial Least Squares-Discriminant Analysis (PLS-DA), and Orthogonal Partial Least Squares-Discriminant Analysis (OPLS-DA) were performed using the SIMCA-P software. Significant variables with a Variable Importance in the Projection (VIP) score higher than 1.0 as obtained through OPLS-DA and those that resulted in p-values less than 0.05 through one-way ANOVA were selected to verify the marker compounds. Among the 19 variables extracted, styrene, ${\alpha}$-pinene, and ${\beta}$-terpinene were selected as markers to indicate the origin of A. gigas. Conclusions: The statistical model developed was suitable for determination of the geographical origin of A. gigas. The cultivation regions of six Korean and eight Chinese A. gigas. samples were predicted using the established OPLS-DA model and it was confirmed that 13 of the 14 samples were accurately classified.

Volatile Component of Pine Needles from Pinus densiflora S. using Solid Phase Microextraction-Ges Chromatography-Mass Spectrometry

  • The volatile components of Pinus densiflora needles were studied by gas chromatography-mass spectrometry(GC-MS), using seven kinds of solid phase microextraction (SPME) fibers, seven in SPME fibers: 100 ${\mu}m$ PDMS, 65 ${\mu}m$ PDMS/DVB, 65 ${\mu}m$ SF-PDMS/DVB, 85 ${\mu}m$ PA, 75 ${\mu}m$ CAR/PDMS, 65 ${\mu}m$ CW/DVB and 50/30 ${\mu}m$ DVB/CAR/PDMS fibers. A total of 40 components were identified by using the seven different SPME fibers. The identified components were classified, according to their functionalities, as follows: 26 hydro-carbons, 7 alcohols, 4 carbonyl compounds, and 3 esters. The major volatile components of Pinus densiflora needles identified by these SPME fibers were $\alpha$-pinene ($1.7\~21.7\;{\mu}g/g$), $\beta$-myrcene ($2.0\~20.1\;{\mu}g/g$), $\beta$-phel-landrene ($4.6\~22.8\;{\mu}g/g$), $\beta$-caryophyllene ($6.7\~26.0\;{\mu}g/g$) germacrene D ($1.1\~11.9\;{\mu}g/g$). In the comparison of the seven SPME fibers, PDMS appeared to be the most suitable fiber for the analysis of hydrocarbon compounds and CAR/DPMS, PDMS/DVB, CW/VB and DVB/CAR/PDMS are shown to be optimal for analysis of the alcohols and carbonyl compounds.
