당귀로부터 항암활성을 화합물을 silicagel column chromatography를 이용하여 분리 및 정제한 후, $^1H$와 $^{13}C-NMR$ MS, HMQC 및 COSY spectrum(500 MHz $CDCl_3$) 분석을 통하여 분자량 328의 decursin임을 구조 동정하였다. 정제된 decursin을 MCF-7에 24시간 처리한 결과 $20{\mu}g/mL$ 이상에서 농도 의존적으로 암세포주의 성장을 억제하였으며, SW480, HepG2, MCF-7및 293과 같은 인체 암세포주에 대한 decurcin의 $IC_{50}$값은 각각 31.04, 27.24, 20.45 및 $33.60{\mu}g/mL$로 나타나, decursin은 인체 정상세포주인 293세포를 포함한 다른 세포에 비하여 MCF-7에 대하여 가장 높은 성장억제 활성을 보여주었다. MCF-7세포에 decursin을 처리한 결과 대조군에서 균일한 핵의 형태가 관찰된 것에 반해 처리군의 경우는 핵의 응축과 apoptotic body를 보였고, DNA 절편이 관찰되었다. 이 결과는 당귀에서 분리한 decursin은 MCF-7세포에서 apoptosis를 통하여 세포의 성장을 억제하는 것을 암시한다.
참당귀 뿌리로부터 2종의 pyranocoumarin 성분을 분리하고 그들의 구조는 NMR 분석에 의해 각각 decursinol angelate(1), decursin(2)로 결정하였다. 이들 화합물에 대하여 hACAT 저해활성을 검정해본 결과, decursinol angelate(1)가 약한 저해활성을 나타낸 반면 decursin(2)은 hACAT1와 hACAT2에 대하여 $IC_{50}$ 값이 각각 137, $168\;{\mu}M$로서 우수한 저해활성을 나타내었다. 한편, 참당귀의 뿌리를 비롯한 꽃, 종자, 잎 등 여러부위에서 decursinol angelate(1), decursin(2)를 정량분석하였는데, 이때 LC/MS/MS(ESI, positive ion mode, MRM mode) 분석을 이용하였다. Decursinol angelate의 함량은 뿌리>잎>종자>꽃 순으로 높았으며, decursin의 함량은 뿌리>종자>꽃>잎 순으로 높게 측정되었다. 이상의 결과들은, 참당귀의 decursin이 동맥경화와 같은 심혈관 질환의 개선과 치료를 위해 유용하게 이용될 수 있음을 시사하였다. 또한 뿌리 이외에 그 동안 이용되지 않고 있던 참당귀 꽃, 잎과 같은 비상용부위에도 기능성물질인 decursin, decursinol angelate이 상당량 함유되어 있음이 구명되어 이러한 부위도 새로운 천연 기능성 소재로서 활용될 수 있음을 기대할 수 있었다.
A reversed-phase liquid chromatographic method for decursin and decursinol angelate of Angelicae gigantis Radix, an important crude drug in Korean traditional medicine, was developed and validated. Decursin and decursinol angelate, the structure isomer (pyranocoumarin) each other, are the main organic constituents in Angelicae gigantis Radix. This method was developed using a RP-18 column, UV detector at 280 nm and 50% acetonitrile solution containing 0.01 M sodium dodecyl sulfate and 25 mM sodium dihydrogen phosphate (pH 5.0) as the mobile phase. Various validation parameters were included and evaluated satisfactorily. Linearity was established in range 2-75 mg/ml of decursin and decursinol angelate (correlation coefficient = 0.9997 and 0.9995, respectively). This analytical method showed good accuracy (98.1% and 99.5%, respectively). Precision (repeatability) revealed a relative standard deviation value of 1.71% (decursin) and 3.19% (decursinol angelate). For intermediate precision measure the considered variables were equipment and days. A robustness test showing the influence of deferent counter-ion concentration in mobile phase was also performed.
The (S)-(+)-decursin and its analogues are reported as potent inhibitors of melanin production in B16 murine melanoma cells. In order to understand the factors responsible for potency as well as inhibition of potency of (S)-(+)-decursin and its analogues, three-dimensional quantitative structure-activity relationship (3D-QSAR) studies were performed. Since receptor structures are not available, a pharmacophore model was constructed. Using PHASE, we generated 3 different models and selected the seven-site model, which returned excellent statistical values ($r^2$ = 0.9127, $Q^2$ = 0.6878, Pearson-R = 0.9014). Using the generated pharmacophore model, we screened a natural products library and obtained 4'-epi-decursin as the most related compound. 4'-epidecursin is similar to (S)-(+)-decursin, but shows additional interaction possibilities with tyrosinase. The study thus sheds some light on possibility of developing more potent tyrosinase inhibitors.
Decursin, decursinol and nodakenin which were isolated from the root of Angelica gigas $N_{AKAI}$ (Umbelliferae) that is used as a botanical drug Dang-Gui (當歸) in Korea show following general pharmacologic activities. Decursin and decursinol increased the motility of the excised duodenum of the rabbit, but nodakenin did not. All of these three compounds depressed the heart contraction of the frog and blood pressure of the carotid artery and respiration of the rabbit. Decursin increased uterus contraction of the rabbit, but decursinol depressed it. And the tonus of the muscle of the earth worm was increased by decursinol, but was decreased by decursin.

Traditionally, Cham dang-gui (Angelica gigas Nakai) is one of the most popular herbal medicines in Asian countries including Korea. A. gigas has been used as a functional food product for treatment anemia, women's health care, a sedative, an anodyne or a tonic agent. Decursin and decursinol angelate isolated from the roots of A. gigas are pyranocoumarin compounds. Recently, as the global herbal medication market is increasing, investigations about pharmacological effects of decursin and decursinol angelate are rapidly increasing. We summarized previous studies about pharmacological effects of decursin and decursinol angelate, and reviewed relation with pharmacological effects of decursin and decursinol angelate on human disorder, focused on the approach for new drug development. Pharmacological effects of decursin and decursinol angelate were classified as anti-tumor activity, anti-bacterial activity, improvements of the circulating system, inhibition of cytochrome P-450 activity, anti-inflammation activity, anti-oxidant activity and cognitive-enhancing activites. The activity of A. gigas with improvement of the circulating system may have wide therapeutic potential for circulatory diseases, including diabetes, hyperlipidemia and atherosclerosis. Also, anti-inflammation activity A. gigas may be beneficial for the treatment and prevention of asthma, atopic dermatitis and rheumatism arthritis. This relation could potentially lead to the development of herbal new drugs. In order to development a new drug containing decursin and decursinol angelate, it is also necessary to consider the safety profile, and the information in this review would contribute to development a new drug from herbal medicine.
In oriental medicine, it is well known that Angelica gigantis tonifies and harmonizes the blood, regulates the menses, and moistens the intestines. This herb has been used for deficient blood pattern, menstural problem, abdominal pain, and traumatic injuries in oriental medicine. A major component of Angelica gigantic consist of a various coumarines, decursin, schooltime, umbelliferone, safrole, 3-butylphalide sistesterol. In this study, we aimed to measure the contents of decursin, one of major coumarine derivative, to determine the standardization of Angelica gigantis.
This study was performed to elucidate the influence on the plasma decursinol concentration after administration of combined decursin and Cnidii Rhizoma or decurisin and Bupleuri Radix extracts in comparison with single decursin administration. The identification of decursinol isolated from Angelica gigas was carried out by GC/MS and the concentration of decursinol in the plasma after oral administration was determined by HPLC. The value of area under the plasma concentration (AUC) and the maximal concentration (Cmax) of decursinol after administration of methanol and ether Angelicae Gigantis Radix extract was higher than those of single decursin-treated group. It was also found that the AUC and the Cmax of decursinol after combined administration with decursin and Cnidii Rhizoma were higher than those of decursin administration, whereas those of combined with decursin and Bupleuri Radix was lower. Moreover, the studies were performed with decursinol only or with combined decursinol and Cnidii Rhizoma/Bupleuri Radix, both of combined treatment increased the plasma decursinol concentrations compared with decursinol-treated group in rats. On the basis of these results, it is concluded that co-existing substances or co-administration with other drugs may influence the plasma levels of decurisinol after oral administration.
