Effects of Catechin on Lipid Composition and Adipose Tissue in Obese Rats Fed High Fat Diet (고지방 식이로 유도된 비만쥐에서 녹차 Catechin이 체지방 조성 및 지방조직에 미치는 영향)

  • The current study examined the effects of catechin on lipid composition of serum and liver and adipocyte of epididymal fat pads in obese rats fed high fat diet. Sprague-Dawley male rats weighing $100{\pm}10g$ were randomly divided into eight groups, four normal diet groups and four high fat diet groups according to the level of dietary catechin supplement. The rats were fed ad libitum experimental diets for 4 weeks and then they were sacrificed. Body weight in HF group was heavier than that of NC group, but HFCM and HFCH groups were significantly reduced compared to HF group. Relative body weight to abdominal weight and relative body weight to epididymal weight in HF group were increased to 103% and 106%, respectively, compared to NC group, but HFCM and HFCH groups were significantly reduced as compared to HF group. The levels of serum triglyceride, total cholesterol, LDL-cholesterol and atherogenic index in HFCH groups were significantly lower than those of HF group, whereas HDL-cholesterol levels were increased. Total lipid contents of liver in HF group was significantly higher than that of NC group, but HFCH group maintained the NC level. There were no significant difference in hepatic triglyceride contents of high fat diet groups. Contents of hepatic cholesterol in HF group was 29% higher than that of NC group, but HFCM and HFCH groups were significantly reduced as compared to HF group. Cell number and cell size of epididymal fat pads in HFCM and HFCH groups were significantly reduced, respectively, compared to HF group. Improved lipid metabolism observed in rats fed catechin may be caused by an alteration of number and size in epididymal fat pad and lipid composition.

Biofunctional Activities of Sanguisorbae officinalis L. Leaves Ethanol Extract (오이풀잎 에탄올 추출물에 대한 기능성 연구)

  • This study was conducted to investigate the antioxidant activity and the bioactive compounds found in 70% ethanol extracts taken from Sanguisorbae officinalis L. leaves(SO) cultivated in Korea. The extracts were tested for their total phenolic contents (TPC), total flavonoid contents (TFC), phenolic compounds, and antioxidative activities using various in vitro assay such as DPPH, ABTS radical scavenging activity, FRAP activity, and reducing power. The TPC and TFC were found to be $119.3{\pm}1.54mg$ gallic acid and $59.6{\pm}1.43mg$ rutin at mg of 70% ethanol extracts, respectively. Catechin was the major material among the phenolic compounds in SO extracts. The DPPH radical scavenging activity, ABTS radical scavenging activity, FRAP activity, and reducing power of SO extracts were increased in a dose-dependent manner. These results suggest that SO extracts could be considered as a good source of natural antioxidants and functional food ingredient.

The Serum Cholesterol Level Lowering Effect from the Underground Parts of Rosa rugosa (해당화 지하부의 혈청 Cholesterol치 저하효과)

  • The present paper describes the serum cholesterol level lowering effect of the underground parts of Rosa rugosa(Rosaceae) which have been used in a folkloric medicine for treating diabetics. The methanol extract was shown to lower the serum lipid level against the normal and 1% cholesterol and 0.5% cholic acid diet feeding to rats, respectively. When various fractions prepared from the above methanol extract was administered, the ethyl acetate-soluble fractions lowered the serum cholesterol level most remarkably. And when (+)-catechin from the ethyl acetate-soluble fractions was only administered to them, the serum cholesterol level was significantly lowered to 260$\pm$9.3mg/dl, the decrease of 17% for the control group. Therefore, it is suggested that (+)-catechin is one of the active principles of this plant.

  • Phannacokinetics of epigallocatechin gallate(EGCG) was studied following i.v. bolus and oral administration in rats. The values of systemic clearance(CL) were $67.9{\pm}5.2$ and $26.5{\pm}1.4\;ml/min/kg$ following i.v. bolus administration of 1 mg and 5 mg EGCG, respectively. The values of volume of distribution at steady state (Vss) were $380{\pm}56$ and $835{\pm}84\;ml/kg$ after i.v. bolus administration of 1 mg and 5 mg EGCG, respectively. The decrease in the value of CL and the increase in the value of $V_{ss}$ as a function of EGCG dose (1 mg to 5 mg) suggest saturable mechanism(s) responsible for the distribution and elimination of EGCG. The fraction absorbed of EGCG after oral and intraduodenal administration of GTC were 13% and 22% of the dose, respectively. This result suggests a considerable degradation or elimination of EGCG in the gastrointestinal absorption after oral administration in rats.

  • This study was performed to investigate the catechins which separated from mulberry leaf. The Epicatechin contents in mulberry leves was 1.21${\pm}$0.05 $\mu\textrm{g}$/mg ethylacetate fraction. epigalocatechingallte contents WAS 3.56 $\mu\textrm{g}$/mg ethylacetate fraction. The cathechins (epicatechin and epigalocate-chingallate) separated from mulberry leaf was shown a strong activity to growth inhibition by the concentration of 5 mg/disc on the Clostridium perfringens.

  • The purpose of this study was to evaluated the antioxidative constituents and their activities of the 80% methanolic extracts from fruit of Sorbaria sorbifolia var. stellipila MAX. The isolation of active compound was performed in three steps: solvent partition, open column chromatography, and high-performance liquid chromatography (HPLC). The solvent fractions were tested for their antioxidant activities by oxygen radical absorbance capacity (ORAC). The antioxidant activity of 80% methanolic extracts by various solvent partitions was in the order of 80% MeOH (1.68 ${\pm}$ 0.027), n-hexane (1.02 ${\pm}$ 0.036), $CH_2Cl_2$ (0.95 ${\pm}$ 0.025), EtOAc (1.98 ${\pm}$ 0.065), n-BuOH (1.94 ${\pm}$ 0.054) and Water (1.28 ${\pm}$ 0.032). Therefore, the results indicated that the potential antioxidant activities and functional values were observed significantly at EtOAc fraction from fruit of S. sorbifolia, flavonoid compound isolated.

Fusion-Complex Activity of Camellia extract (동백 열수추출물의 융복합 활성)

  • To determine fusion-complex activity of Camellia extract extracted hot water, this study was conducted. Special catechin was identified at this extract by analysis of catechin. In analysis of antioxidant activity, the extract was contained antioxidant material and flower among the other parts showed high antioxidant activity. flavonoid showed most high content of Camellia. Flavonoid content was found to be $223.29{\pm}0.005mg/g$ to measured most high to stem, a control group was measured Green tea new leaves $126.14{\pm}0.005mg/g$. Total polyphenol content exhibited most high content at new leaves and flavonoid showed most high content at stem. ${\gamma}$-Amino butyric acid (GAVA) among the free amino acid showed highly at all parts.

A Comparative Study on Anthocyanin and Polyphenol Contents in Colored Agricultural Products (유색 농산물 중 안토시아닌과 폴리페놀 함량 비교 연구)

  • In this study, 17 kinds of polyphenols and 5 kinds of anthocyanins were analyzed to compare the contents of polyphenols and anthocyanins in 76 colored agricultural products. A total of 17 polyphenols were analyzed simultaneously by 9 phenolic acids (gallic acid, protocatechuic acid, chlorogenic acid, vanillic acid, caffeic acid, syringic acid, p-coumaric acid, t-ferulic acid, t-cinnamic acid) and 8 flavonoids ((+)catechin, syringic aldehyde, rutin, epicatechin gallate, naringin, luteolin, naringenin, kaempferol) and 5 anthocyanins (delphinidin-3-glucoside, delphinidin-3-rutinoside, cyanidin-3-galactoside, cyanidin-3-glucoside, cyanidin-3-arabinoside) were simultaneously analyzed. The total content of 17 polyphenols was determined as seoritae $255.1{\pm}7.5{\mu}g/g$, seomoktae $275.8{\pm}5.3{\mu}g/g$, black rice $78.5{\pm}4.6{\mu}g/g$, black sesame $75.8{\pm}3.2{\mu}g/g$, blueberry $143.3{\pm}5.5{\mu}g/g$, aronia $195.2{\pm}4.9{\mu}g/g$ and blackcurrent $131.6{\pm}3.2{\mu}g/g$, the highest content was found in the order of seomoktae > seoritae > aronia > blueberry > blackcurrant > black rice > black sesame. The total content of 5 anthocyanins was determined as seoritae $82.4{\pm}17.2{\mu}g/g$, seomoktae $95.2{\pm}6.1{\mu}g/g$, black rice $74.1{\pm}9.7{\mu}g/g$, black sesame were not detected, blueberry $110.8{\pm}1.9{\mu}g/g$, aronia $218.9{\pm}6.1{\mu}g/g$ and blackcurrent $209.7{\pm}4.0{\mu}g/g$, the highest content was found in the order of aronia > blackcurrant > blueberry > seomoktae > seoritae > black rice. These results indicated that seomoktae and aronia possessed the high level of functional components and further study will be needed to develop high value-added foods based on the colored agricultural products.

Manufacturing of the Enhances Antioxidative Wine Using a Ripe Daebong Persimmon (Dispyros kaki L) (대봉감 연시를 이용한 항산화 활성이 강화된 와인 제조)

  • In this study, the characteristics of alcohol fermentation using ripe Daebong persimmon juice were studied in static fermentation condition by Saccharomycess cerevisiae CS02 in an effort to develop new types of functional wine. Attempts were made to modify the ripe Daebong persimmon juice in order to find suitable conditions for alcohol fermentation. The modified ripe Daebong persimmon juice that was most suitable for alcohol fermentation contained $24^{\circ}brix$ of sugar supplemented with sucrose as a carbon source and 0.5 g/L of $(NH_4)_2HPO_4$ as a nitrogen source. After 9 days of fermentation at $25^{\circ}C$, $12.2{\pm}0.02%$ of alcohol was produced from the modified juice and its pH markedly decreased to $3.97{\pm}0.02$. The wine contained free sugar such as fructose ($0.12{\pm}0.02$ g/L), some organic acids such as malic acid ($35.92{\pm}0.24$ g/L), succinic acid ($8.12{\pm}0.03$ g/L), oxalic acid ($22.14{\pm}0.11$ g/L), and citric acid ($13.63{\pm}0.08$ g/L), as well as some flavanols and phenolic acids such as catechin gallate ($38.99{\pm}0.32$ mg/L), epicatechin gallate ($110.21{\pm}0.16$ mg/L), gallic acid ($163.88{\pm}1.11$ mg/L), epigallocatechin ($15.97{\pm}0.18$ mg/L), and tannic acid ($13.36{\pm}0.02$ mg/L). In addition, the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical (84.25%) and $ABTS^{\cdot+}$ radical (99.65%) scavenging activities were increased significantly with a corresponding increased in the organic acid and phenolic acid contents, but decreased in the flavonoids.
