For the purpose of establishing the most appropriate method of bonding between glass ionomer liners and composite resin and comparing the materials for sandwich technique, an experiment was performed to measure the shear bond strengths between the two with the variables in the surface treatment of liners and elapsed time till composite buildup. Materials used were Vitrebond and Fuji II LC, each as the restorative and liner respectively, and each group was subdivided by surface treatment (acid etching and sandblasting) and time elapsed from GIC filling to composite buildup (immediately, 1 day, 7 days), consisting 12 groups as a whole. Each subgroup was composed of 10 specimens and the shear bond strength between GIC liners and composite resin was measured under UTM and analyzed. The result were as follows: 1. The shear bond strength between two materials was highest when initially filled Fuji II LC was sandblasted after 1 days and composite built-up (Group FS1). And the lowest value was found when GIC was acid-etched after 7 days and composite built-up (Group FE7). Significant difference was found between the two groups. (P<0.01) 2. In regard of surface treatment of GI liners, acid-etched group (VE) showed higher bond strength than sandblasted group (VS) for Vitrebond. But, the reverse was true for Fuji II LC. (P<0.05) 3. In regard to the time elapsed from GI filling to composite buildup, the group of 1 day elapse showed relatively higher strength for Vitrebond. On the contrary, immediate buildup group (FE0) was stronger for acid-etched group and 1 day elapse group(FS1) was higher for sand-blasted group in Fuji II LC. (P<0.05)
The purpose of this study was to compare the bond strength of the glass ionomer luting cements after the functional loads were applied to the crowns respectively 10 min, 30 min, 1 hr, 24 hrs later after mixing of cements. The conventional (GC Fuji I; GC Int. Co., Japan), water hardening type (Ketac Cem; ESPE, W. Germany) glass ionomer luting cement and C-dent ZPC (Confi-Dental Products Co., U.S.A.) as a control were used. The results of this study were obtained as follows: 1. In the bond strength of GC Fuji I, 24 hr-group was highest and there were no statistically significant differences among 10 min-, 30 min- and 1 hr- groups. 2. l3 the bond strength of Ketac Cem and C-dent ZPC, there was no statistically significant differences among 10 min-, 30 min-, 1 hr- and 24 hr-groups respectively. 3. Comparing the bond strength among cements in 10 min-, 30 min-groups respectively, GC Fuji I was lowest and there was no statistically significant difference between Ketac Cem and C-dent ZPC. 4. The bond strengths between GC Fuji I and Ketac Cem were not significantly different in 1 hr-, 24 hr-group. 5. The bond strengths among 3 types of cements were not significantly different in 24 hr-groups.
Transformation of Fuji apple (Malus domestica 'Fuji') was performed using Agrobacterium tumefaciens harboring a coat protein (CP) gene of Cucumber mosaic virus (CMV). A plasmid DNA containing the virus CP and NPT II genes was introduced into the loaves of apple by th e Agrobacterium - mediated transformation procedure. Regenerated transformants of the apple were obtained by kanamycin resistance conferred by the introduced NPT II gene. PCR analysis showed that 3 out of 20 putatively selected R0 plant lines contain the CMV-CP gene. Nine putative transgenic lines out of 20 lines were investigated with the PCR analysis; 5 regenerants produced a 450 bp DNA band and 3 regenerants showed a 671 bp DNA band for the NPT II and CMV-CP genes, respectively. Southern hybyidization results demonstrate the successful integration of the CMV-CP gene into the genome of the apple. This is the first report on the generation of useful vius resistance source of transgenic apple for molecular breeding program.
Bond strength of luting cements to dentin is a critical consideration for success of complete cast crowns. This study was performed to evaluate the relationship between surface characteristics of teeth prepared for complete cast crowns and retention of cemented restorations. Eighty artificial crowns were cast for standardized complete crown tooth preparations accomplished with the use of a special device on recently extracted human teeth. Coarse diamond(#102R, Shofu) and superfine finishing diamond(#SF102R, Shofu) burs of similar shape were used. Crowns in each group were randomly subdivided into few subgroups of 10 for luting cements selected for this study: zinc phosphate cement (FLECK' S), polycarboxylate cement (Poly-F), rein-forced glass ionomer cement (Fuji PLUS). and adhesive resin cement (Panavia 21). Retention was evaluated by measuring the tensile load required to dislodge the artificial crown from tooth preparations with an Instron testing machine, and analysed by one-way ANOVA and Student's t-test. The obtained results were as follows ; 1. When tooth preparation was done with coarse diamond bur, retentive force was diminished in order of Panavia 21 Fuji PLUS, FLECK'S, and Poly-F. Retentive forces showed the significant difference between Fuji PLUS group and FLECK'S group(p<0.001). 2. When tooth preparation was done with superfine diamond bur, retentive force was diminished in order of Fuji PLUS, Panavia 21, FLECK'S, and Poly-F. Retentive forces showed the significant difference between Panavia 21 group and FLECK'S group(p<0.001). 3. Retentive force in coarse tooth surfaces was significantly higher than that in superfine tooth surface with all luting cements(p<0.001), and cement residues were almost retained with-in the cast crown in all groups.
The purpose of this study was to investigate the effect of cavity configurations on the marginal leakage of class 5 glass ionomer cement and composite resin restorations. Four types of cavities such as saucer shape. notch shape. combined shape(notch shape occlusally and saucer shape gingivally). and U shape were prepared on the buccal and lingual surfaces of 80 extracted premolars(40 cavities for each shape). Occlusal cavity margins were placed at enamel and cervical margins were placed at dentin. 10 cavities of each shape were restored with Ketac Fil as a conventional glass ionomer cement. Fuji II LC improved as a resin modified glass ionomer cement, Z 100 as a hybrid composite resin. and Tetric Flow as a flowable composite resin (40 cavities for each material). After thermocycling, teeth were immersed in 5% basic fuchsin solution for 6 hours and sectioned longitudinally in a buccolingual direction through the center of the restoration. The dye penetrations at the tooth restoration interface were examined by stereomicroscope. The Result were as follows 1. In saucer shape, notch shape and combined shape, composite resin restorations showed lesser leakage than glass ionomer restorations(p<0.05) and in U shape. Tetric Flow showed the least marginal leakage and others were decreased as Z 100. Fuji II LC improved, Ketac Fil in that order. There were statistically significant difference between Tetric Flow and Fuji II LC improved. Ketac Fil and between Z 100 and Ketac Fil(p<0.05). 2. In Ketac Fil restoration group, saucer shape showed the highest marginal leakage and U shape showed the least marginal leakage and others were decreased as notch shape, combined shape in that order. There were statistically significant difference between saucer shape and combined shape, U shape and between notch shape and U shape(p<0.05). 3. In Fuji II LC improved restoration group, U shape showed the least marginal leakage. There were statistically significant difference between U shape and other three shapes(p<0.05). 4. The cavity configuration had no significant effect on marginal leakage of composite resin restorations(p>0.05).
Objectives: The purpose of this study was to evaluate the microleakage of 4 temporary materials in teeth with Class II-type endodontic access preparations by using a glucose penetration model. Materials and Methods: Glucose reaction test was performed to rule out the presence of any reaction between glucose and temporary material. Class II-type endodontic access preparations were made in extracted human premolars with a single root (n = 10). Each experimental group was restored with Caviton (GC), Spacer (Vericom), IRM (Dentsply-Caulk), or Fuji II(GC). Microleakage of four materials used as temporary restorative materials was evaluated by using a glucose penetration model. Data were analyzed by the one-way analysis of variance followed by a multiplecomparison Tukey test. The interface between materials and tooth were examined under a scanning electron microscope (SEM). Results: There was no significant reaction between glucose and temporary materials used in this study. Microleakage was significantly lower for Caviton and Spacer than for Fuji II and IRM. SEM observation showed more intimate adaptation of tooth-restoration interfaces in Caviton and Spacer than in IRM and Fuji II. Conclusions: Compared to IRM and Fuji II, Caviton and Spacer can be considered better temporary sealing materials in Class II-type endodontic access cavities.
The purpose of this study was to determine the fluroide release levels of new fluoride-containing liner/base cements and the fluoride uptake by dentin surfaces. Ten specimens of each brand (Fuji ionomer Type III, Fuji Lining LC, Timeline, Vitrebond and XR ionomer) were made, polymerized and placed in fluoride-free distilled water at $37^{\circ}C$, 100% relative humidity for 24 hours. The extracting solution of specimen was exchanged and fluoride release was measured daily for the 30 days. For fluoride uptake study, twenty-five extracted human lower molars were sectioned longitudinally in the mesiodistal direction with a diamond disc. Five teeth were filled with each material and then stored at $37^{\circ}C$, 100% humidity for 4 weeks. Fluoride uptake by dentin from the test materials was evaluated using electron probe micro X-ray analyzer. The following results were obtained : 1. The amounts of fluoride release showed no significant difference between Fuji ionomer Type III and Fuji Lining LC, but showed significant difference between other groups. XR ionomer released significantly greater fluoride than any other group(P<.001). 2. All the materials have a burst effect which more fluoride released in then first 3 day and showed significant decrease over the test period (P<0.001). 3. XR ionomer group showed fluoride penetration to approximately $50{\mu}m$ deep in dentin. But other material groups showed very little fluoride uptake by dentin.
