Critical Thermal Maximum (CTM) of Cultured Black Rockfish, Sebastes schlegeli

  • The critical thermal maximum (CTM) of black rockfish, Sebastes schlegeli, was evaluated. Black rockfish were acclimated at $24^{\circ}C$, and then exposed to temperatures from 24 to $33^{\circ}C$. Black rockfish were kept in constant darkness and subjected to a gradual temperature increase $(1 ^{\circ}C\;12^{-1})$. The oxygen consumption rate (OCR) was measured using an automatic intermittent­flow-respirometer (AIFR) during the exposure period (from 119.3 to 143.5 h). The OCR increased from 94.5 to 214.2mL $O_2 kg^{-1}\;ww\;h^{-1}$ as the temperature rose from 24 to $29.4-30.9^{\circ}C$. Subsequently, the OCR increased abruptly, reaching 245.8-412.7mL $O_2 kg^{-1}\;ww\;h^{-1}$ at $32^{\circ}C$. This study suggests that the CTM for black rockfish is $29.4-30.9^{\circ}C$ when temperature is increased at $1^{\circ}C\;12h^{-1}$ following acclimation at $24^{\circ}C$.

  • An efficient synthetic route for preparing novel $4'{\alpha}$-C phenyl branched carbocyclic nucleoside is described. The installation of phenyl group at the $4'$-position of carbocyclic nucleoside was successfully accomplished via a sequential [3,3]-sigmatropic rearrangement and ring-closing metathesis (RCM) beginning from simple ketone such as 2-hydroxy acetophenone.

The Storage Temperature Effect on Vibrio parahaemolyticus in Fish Homogenates (생선회에 오염원 장염비브리오균에 미치는 저장온도의 영향)

  • The change of cell counts of Vibrio parahaemolyticus in fish muscle by the storage time and temperature was examined to get basic informations for precautionary steps against food poisoning of slices of raw fish (sashimi). There fore, we inoculated fish homogenate of oceanic bonito (Katsuwonus pelamis), yellow tail (Seriola quinqueradiata) with Kanagawa positive Vibrio parahaemolyticus and stored it at $30^{\circ}C,\;18^{\circ}C,\;4^{\circ}C\;and\;-20^{\circ}C$ for 24 hours. The number of the Vibrio parahaemolyticus upon fish homogenate stored at $30^{\circ}C\;and\;18^{\circ}C$ decreased for the first two hours and increased thereafter. When the fish homogenates inoculated with Vibrio parahaemolyticus at about $10^3$ per gram were stored at $18^{\circ}C\;and\;30^{\circ}C$ for 10 hours, the cell numbers increased about 10 times and 1,000 times initial cell numbers, respectively. The survival rate of Vibrio parahaemolyticus was about $20\%$, when the inoculated fish homogenates were stored at $-20^{\circ}C$ for 24 hours. Vibrio parahaemolyticus inoculated in fish homogenates was decreased by about $10\%$ of initial cell numbers by the storage at $4^{\circ}C$ for 4 hours and it was decreased by about $50\%$ after 24 hours storage of the samples at the same temperature. The decreasing rate of inoculated Vibrio parahaemolyticus in fresh fish muscle homogenate was higher than that in frozen fish muscle homogenate during the storage time at a refrigerator.

Physicochemical Properties of Phosphatidylcholine (PC) Monolayers with Different Alkyl Chains, at the Air/Water Interface

  • Physicochemical properties of a series of PC monolayers with different alkyl chains (C24, C20, C16, and C8), at the air/water interface were investigated. The surface pressure is influenced mainly by the hydrophobicity of the PCs, which is confirmed by the curve shape and the on-set value of π-A isotherms at the air/water interface by increasing the number of alkyl chain. The on-set values of surface pressure were 125 Ų/molecule for DOPC(C8), 87 Ų/molecule for DPPC(C16), 75 Ų/molecule for DAPC(C20), and 55 Ų/molecule for DLPC(C24), respectively. The orientations of alkyl chains at the air/water interface are closely connected with the rigidity of the monolayers, and it was confirmed by the tendency of monolayer thickness in ellipsometry data. The temperature dependence of a series of PCs shows that the surface pressure decreases by increasing temperature, because the longer the alkyl chain length, the larger the hydrophobic interaction in surface pressure. The temperature effects and the conformational changes of unsaturated and saturated PCs were confirmed by the computer simulation study of the cis-trans transition with POPC and DPPC(C16). The cistrans conformational energy difference of POPC is 62.06 kcal/mol and that of DPPC(C16) is 6.75 kcal/mol. Due to the high conformational energy barrier of POPC, phase transition of POPC is limited in comparison with DPPC(C16).

  • In order to study the optimum environmental condition for larvae culture of the freshwater crab, Eriocheir japonicus, larvae from different growth stages and young crab were cultured under the 16 different conditions of $4\times4$ temperature-salinity combinations (4 different temperatures at 22, 24, 26, $28^{\circ}C$ with 10.5, 17.5, 24.5 and $31.5\%$ of salinity). The duration of metamorphosis, metamorphosis rate, the interval of moulting period, and survival rate were measured from each experimental group of larvae and young crab under the different conditions. The results indicated that the optimum conditions may be a $24.5\%o$ of salinity at water temperature at 22, 24, and $26^{\circ}C$. At $28^{\circ}C$ with $24.5\%o$), the duration of metamorphosis reduced somewhat, nevertheless metamorphosis and survival rate decreased a lot. And the lower the salinity showed the lower the metamorphosis and survival rates at $28^{\circ}C$.

Studies on the Utilization of Straw for Production of Glucose Isomerase (볏짚을 이용한 Glucose Isomerase 생산에 관한 연구)

  • The hemicellulose fraction of ryegrass straw was extracted with NaOH and used for production of glucose isomerase by Streptomyces flavogriseus. Up to 25% crude hemicellulose (20% as pentosan) could he obtained by treating straw with 1 to 24% NaOH. The yield of hemicellulose was increased proportionately with increasing NaOH concentration up to 4%, but the rate of increase was slowed thereafter. The optimum condition for hemicellulose extraction from ryegrass straw was to treat straw with 4% NaOH for 3hrs at 9$0^{\circ}C$ or 24hrs at 3$0^{\circ}C$. Highest level of glucose isomerase activity (3.04 units/ml culture) was obtained when the organism was grown for 2 days at 3$0^{\circ}C$ on 2% straw hemicellulose. The organism also produced a good quantity of glucose isomerase on xylan, xylose or H$_2$SO$_4$-hydrolysate of straw. The hemicellulose-extracted straw residue could be used as animal feed, because the residue had 75% higher digestibility and 20% better feed efficiency for weanling meadow voles than the untreated straw.

  • The superconducting properties of high temperature superconducting (HTS) GdBCO coated conductor (CC) tape (Ag/GdBCO/Buffer-layers/Stainless Steel) were investigated, specifically a series of samples prepared by vacuum heat treatment (200℃ to 600℃), using a Quantum Design PPMS-14. The critical current density Jc value was obtained by applying the modified Bean model to the irreversible magnetization ∆Mirr(H) data which was estimated from the magnetization M(H) loop. The reduction rates of lnJc and Tc values according to the increase of the vacuum annealing temperature Tan were d(lnJc)/dTan = - 0.016 A/(cm2∙℃) and dTc/dTan = - 0.24, respectively. We examined the effect of recovery temperature Tre (475℃ to 700℃) and recovery duration time t (0.5 h to 24 h) on the restoration of previously completely lost superconductivity in samples that subsequently received heat treatment in an O2 gas flow space. All samples were fully restored to superconductivity by heat treatment in an O2 gas flow space. The recovery temperatures Tre (475℃ to 700℃) and recovery duration times t (0.5 h to 24 h) were both independent of the superconductivity recovery characteristics.

  • Distribution of the marine cladoceran Evadne spinifera in waters adjacent to Korea was studied on the bases of samples collected during the Cooperative Study of the Kuroshio and Adjacent Regions, 1965-1974, and during the Exploitation Research of Marine Resources on the yellow Sea, September October 1992. This species was mainly occurred in August and September in the sea of Japan Distribution in the Yellow Sea was sparse. temperature and salinity ranged $23.42-28.24^{\circ}C$ and $32.11-24.20\textperthousand$, respectively. Maximum abundance of $11{\;}indiv./\textrm{m}^3$ was recorded in coastal waters of Honshu, Japan with $26.80^{\circ}C$ and $32.81\textperthousand$ water. E. spinifera was considered to be a stenothermal animal preferring high temperature.

  • Rhodotorula glutinis K-24 was found to produce internal, cell wall bound, and external invertase. Internal invertase was purified by column chromatographies on DEAE-Sephadex A-50, Sp-sephadex C-50, gel filtration on Sephadex G-200 and isoelectric focusing. Cell wall bound invertase was partially purified by the following procedures; column chromatography on DEAE-Sephadex A-50 and gel filtration on Sephadex G-100. Optimum pH and temperature for enzymatic activities of internal and cell wall bound invertase were pH 3.0 and 6$0^{\circ}C$, respectively. Both enzymes were inhibited by HgC1$_2$, AgNO$_3$, MnSO$_4$, and sodium dodecylsulfate. The molecular weights of internal and cell wall bound invertases were estimated to be 310,000 and 61,000, respectively. Other physicochemical properties of the both enzymes were similar.
