Histone deacetylase family in balloon flower (Platycodon grandiflorus): Genome-wide identification and expression analysis under waterlogging stress

  • Histone deacetylases (HDACs) play a pivotal role in epigenetic regulation, affecting the structure of chromatin and gene expression across different stages of plant development and in response to environmental stresses. Although the role of HDACs in Arabidopsis and rice has been focused on in extensive research, the role of the HDAC gene family in various medicinal plants remains unclear. In the genome of the balloon flower (Platycodon grandiflorus), we identified 10 putative P. grandiflorus HDAC (PlgHDAC) proteins, which were classified into the three families (RPD3/HDA1, SIR2, and HD2 HDAC families) based on their domain compositions. These HDACs were predicted to be localized in various cellular compartments, indicating that they have diverse functions. In addition, the tissue-specific expression profiles of PlgHDACs differed across different plant tissues, indicating that they are involved in various developmental processes. Furthermore, the expression levels of all PlgHDACs were upregulated in leaves after waterlogging treatment, implying their potential role in coping with waterlogging-induced stress. Overall, our findings provide a comprehensive foundation for further research into the epigenetic regulation of PlgHDACs, and particularly, on their functions in response to environmental stresses such as waterlogging. Understanding the roles of these HDACs in the development and stress responses of balloon flower could have significant implications for improving crop yield and the quality of this important medicinal plant.

Effects of Coffee and Aflatoxin B1 on the Pancreatic Exocrine Function and Structure (Coffee와 Aflatoxin B1이 췌장의 외분비 기능 및 조직에 미치는 영향)

  • Coffee is known to increase pancreatic secretion of digestive enzymes. The mutagen, aflatoxin B1(AFB1) is contained in fermented foods and known to increase the specific activities of pancreatic chymotrypsin, trypsi, amylase, and lipase. Nowadays, coffee intake is increased among Koreans who have consumed relatively high amount of traditional fermented foods. Therefore, this study was performed to examine the effect of coffee and AFB1 on pancreatic exocrine function and structure. Rats were divided into 10 experimental groups. The first five groups were W(control group), LD(0.2g decaffeinated coffee/Kg B.W), HD(3g decaffeinated coffee/Kg B.W), LC(0.2g coffee/Kg B.W), and HC(3g coffee/Kg B.W). The second five groups were WA, LDA, HDA, LCA, HCA, same as first five groups in caffieine level but treated with AFB1. The result of this experiment showed that the caffeine intake did not influence significantly on the growth and feed efficiency. But water intake was increased by caffeine intake and AFB1 treatment. The weights of pancreas and liver were increased as the caffeine intake was increased. Trypsin activities were tend to increase in concentrated coffee groups(HD, HC). AFB1 treated groups showed the higher trypsin level than the AFB1 untreated groups. Amylase activities were tend to increase in concentrated coffee groups(HD, HC) of AFB1 untreated animals. AFB1 treated did not show the additional effect on the stimulated amylase secretion by coffee. Lipase activities were tend to decrease in concentrated coffee groups(HD, HC) of AFB1 untreated animals. Lipase activities were increased in the order named WA group, coffee groups, decaffeinated coffee groups in AFB1 treated animals. AFB1 treated groups showed the higher lipase level than AFB1 untreated groups. In the histologic observation of pancreas HCA group showed more dense compound tubuloalveolar glands and proliferation of nuclei than normal. The result suggested a development of a atypia which is ongoing phase to a cancer.

A Nutritional Survey on the Actual Condition of Group Feeding in Seoul City and Kyongki Province (서울, 경기 일부지역(一部地域)의 단체급식소(團體給食所)에 대(對)한 영양실태조사(營養實態調査))

  • The purpose of this nutritional survey on the actual conditions of group feeding is to be of value for promoting the health of the blue collar workers through consice understanding of food intake trends in businesses as well as realizing our desirable target. The results of the survey are summarized as follow. 1, Status of food economics 1) The average feeding cost amounts to 453 won per day per person. 2) The average school years of food buyers were 12.3 years, cookers were 12.0 years and dieticians were 14.5 years. 3) Kerosene(57.6%), gas(24.2%), electric(12. 1%) and cool briguette(6.1%) were used for fuel. 2. Status of food intake The average food intake per person per day in surveyed businesses was 1193.4g. The total intake of food was consisted of 42% grains, 40.4% vegetables, 4.7% legumes, 4.5% fish and shellfish, 4.0% potatoes, 1.6% meats, 1.0% seasonings. 0.8% eggs, 0.5% sea weeds, 0.3% oil and fats, 0.1% sugar. These findings led us to the conclusion that workers in the surveyed businesses retied heavily on plant foods. 3. Status of nutrient intakes 1) The average intake of calorie was 2752.6 kcal per day, which was slightly higher than 2,700 kcal of RDA. Though the quantity was above the level of RDA, it was largely from plant foods. 2) The average intake of protein was 84.7g per day, which was slightly higher than 80g of RDA. 3) The average intake of fat was 26g per day, which was much lower than 36g of RDA. 4) The averse intake of calcium was 532.3mg per day, which was lower than 600mg of RDA. 5) The average iron intake was 9.8mg per day, which was much lower than 10mg of RDA and 18mg of HDA for women. 6) Intakes of vitamin group were high. Especially, intake of vitamin A was 2-3 times higher than RDA and intake of vitamin C and niacin were 2 times higher than RDA.

Antioxidant Activity of Royal Jelly Hydrolysates Obtained by Enzymatic Treatment

  • Recently, research on the processing of raw functional materials with the aim of improving various physiological activities has been conducted. In this study, we investigated the antioxidant activity of royal jelly (RJ) hydrolysates obtained from three commercial proteases. Enzyme-treated royal jelly (ERJ), in which the RJ hydrolysates were converted into easy-to-absorb shorter chain monomers through the removal of two known allergen proteins, showed no difference in the content of (E)-10-hydroxydec-2-enoicacid (10-HDA) or the freshness parameter and showed a significant increase in total free amino acid content. The antioxidant activity of ERJ was determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and chemical assays. The ERJ showed about 80% DPPH-radical scavenging activity at same concentration of ascorbic acid. The antioxidant effect of ERJ was confirmed to be due to reduction of intracellular reactive oxidative species (ROS) and nitric oxide (NO) production in LPS-treated macrophages. Moreover, ERJ significantly increased the activity of the antioxidant enzyme superoxide dismutase (SOD) and the level of the antioxidant glutathione (GSH) in a dose-dependent manner. Interestingly, these antioxidant activities of ERJ were stronger than those of non-treated RJ. These findings indicate that ERJ has high potential as an antioxidant agent for use in human and animal diets.

A Study on the Test Evaluation Method of AEB (V2P) Considering the Road Environment in Korea and Euro NCAP Test Protocol v3.0.1 (국내 도로환경과 Euro NCAP VRU Test Protocol v3.0.1을 고려한 AEB(V2P) 시험평가 방법에 관한 연구)

  • In the world, traffic accidents and environmental pollution caused by the increase of vehicles are becoming a serious social problem. According to the 2016 data published by the Korea Highway Traffic Authority, Korea owns 49.9 vehicles per 100 people. This is the 28th largest number among the 35 OECD member countries. In addition, the number of deaths from traffic accidents in Korea totaled 4,292, of which 1,714 were caused by traffic accidents involving vehicles and pedestrians. To reduce these human casualties, the automotive industry is constantly working on the development and commercialization of Adaptive Driver Assist System (ADAS). ADAS is the system providing convenience and safeness for drivers. In general, ADAS consists of Autonomous Emergency Braking (AEB), Highway Driving Assist (HDA), Adaptive Cruise Control (ACC), Lane Keeping Assist System (LKAS). Among them, the AEB detects the possibility of collision by the vehicle itself and plays a role of avoiding the collision or reducing the damage through active braking. For such AEB, Euro NCAP has been developing test-evaluation methods for the vulnerable since 2017. Therefore, In this paper analyzes the scenario of Euro NCAP VRU Test Protocol v3.0.1, which will be established in 2020, and proposes test conditions according to the Korean road traffic law. In addition, the reliability of the proposed scenario and test conditions was verified by comparing and analyzing the proposed theoretical evaluation formulas and actual test results.

Performance Analysis of Landing Point Designation Technique Based on Relative Distance to Hazard for Lunar Lander (달 착륙선의 위험 상대거리 기반 착륙지 선정기법 성능 분석)

  • Lidar-based hazard avoidance landing system for lunar lander calculates hazard cost with respect to the desired local landing area in order to identify hazard and designate safe landing point where the cost is minimum basically using slope and roughness of the landing area. In this case, if the parameters are only considered, chosen landing target can be designated near hazard threatening the lander. In order to solve this problem and select optimal safe landing point, hazard cost based on relative distance to hazard should not be considered as well as cost based on terrain parameters. In this paper, the effect of hazard cost based on relative distance to hazard on safe landing performance was analyzed and it was confirmed that landing site designation with two relative distances to hazard results in the best safe landing performance by an experiment using three-dimensional depth camera.

Quantitation of Plasma Apolipoprotein A-I with a Sandwich Type Enzyme-Linked Immunosorbent Assay Using Monoclonal Antibodies

  • A sandwich-type enzyme-linked immunosorbent assay (ELISA) for the quantification of human apolipoprotein A-I (apoA-I) was developed using monoclonal antibodies. For this assay, we used three monoclonal antibodies to trap and detect apo A-I. HDAI16 and HDA15 monoclonal antibodies were used for trapping apoA-I and HDAI8 monoclonal antibody was for detecting apoA-I. These three monoclonal antibodies were produced by immunizing mice with high density lipoprotein (HDL) isolated from human plasma. By immunoblot analysis, these three monoclonal antibodies were specific to apoA-I and showed no cross-reactivities with other plasma proteins. The results of competition assays for epitope cross-reactivity test also verified that these monoclonal antibodies identified separate and distinct epitopes on HDL and apoA-I. Affinity constants of monoclonal antibodies were measured by ELISA. Their association constants ranged from $10^7$ to $10^8$ $M^{-1}$. For this assay, pure apoA-I was isolated by affinity chromatography using monoclonal antibodies. In this sandwich assay, the amount of HRP-labeled HDAI8 bound to apoA-I trapped by HDAI16 and HDAI5 was proportional to apoA-I concentration in the range of 0 to 500ng/ml. ApoA-I concentration in plasma was calculated from the linear regression equation of standard curve. The precision and reliability of the assays are reflected in the low intra-and interassay coefficients of variation that averaged 3.25% and 4.30%, respectively. This assay is sensitive, simple, reproducible, convenient in incubation interval, and does not use radioisotope: thus it can be widely applied in clinical laboratories.

  • Objectives : A self-made "Tomotherapy couch device" capable of correcting the Yaw direction was fabricated and evaluated for its usefulness. Materials and Methods : "Tomotherapy couch device" capable of correcting the Yaw direction is made of rigid fibreboard with a flexural strength of $200kg/cm^2$. CBCT Image from Novalis Tx and Iso-Align Phantom from MED-TEC were used to evaluate the physical accuracy. The treatment plan was designed using Accuray $Precision^{TM}$ and In House Head and Phantom. Accuray $PrecisionART^{TM}$ and $Precision^{TM}$ was used to evaluate dose. Results : Evaluation results, the self-fabricated device accurately corrected the setup error, Target dose was within 95 %~107 % of all. In order to directly evaluate the OAR dose according to the Yaw change, the absolute dose was measured. As a result, when the error in the Yaw direction was $3^{\circ}$, the specific OAR showed a maximum difference of 18.4 %. Conclusion : "Tomotherapy couch device" capable of correcting the Yaw direction can be manufactured at a lower cost compared to the effect, and it can prevent the patient's MVCT image dose for re-imaging. Accurate radiation therapy without errors can be performed.
