The Role of Mitochondrial ATP-sensitive Potassium Channel on Intestinal Pacemaking Activity

  • Interstitial cells of Cajal (ICCs) are the pacemaker cells that generate slow waves in the gastrointestinal (GI) tract. In the present study, we investigated the effect of mitochondrial ATP-sensitive potassium (mitoKATP) channel on pacemaking activity in cultured ICCs from murine small intestine by using whole-cell patch clamp techniques. Under current clamp mode, at 10μM glibenclamide, there was no change in pacemaking activity of ICCs. At $30{\mu}M$ glibenclamide, an inhibitor of the ATP sensitive $K^+$ channels, we could find two examples. If pacemaking activity of ICCs was irregulating, pacemaking activity of ICCs was changed into regulating and if in normal conditions, membrane potential amplitude was increased. At $50{\mu}M$ glibenclamide, the resting membrane potential was depolarized. At 3mM 5-HDA, an inhibitor of the mitoKATP channels, inhibited the pacemaking activity of ICCs. Both the amplitude and the frequency were decreased. At 5 mM 5-HDA, both the amplitude and the frequency were completely abolished. Diazoxide, an opener of the mitoKATP channels, was applied to examine its effect on pacemaking activity of ICCs. At $50{\mu}M$ concentration, the pacemaking activity of ICCs was inhibited. Both the amplitude and the frequency were decreased. At 1 mM concentration, both the amplitude and the frequency were completely abolished and the resting membrane potential was shaked.These results indicate that mitoKATP channel has an important role in pacemaking activity of ICCs.

The Protective Effect of Chunghyul-dan(Qingxuedan) Against 6-hydroxydopamine Induced Neurotoxicity. (청혈단(淸血丹)의 6-hydroxydopamine에 의해 유발된 독성에 대한 신경세포보호효과)

  • Objectives : This Study was performed to assess the antioxidant and neuroprotective effect of Chunghyul-dan(Qingxuedan) in PC12 cells and primary rat mesencephalic dopaminergic neurons. Methods : The anioxidant effect was investigated using the DPPH radical and ABTS cation scavenging assays and total polyphenol amout of Chunghyul-dan(Qingxuedan). The neuroprotective effect of Chunghyul-dan(Qingxuedan) in PC12 cells was evaluated using MTT assay. The scavenging activity of Chunghyul-dan(Qingxuedan) on ROS production induced by 6-OHDA(6-hydroxydopamine) in PC12 cells was evaluated, as well as the attenuating effect on GSH reduction. Finally, we examined the neuroprotective effect of Chunghyul-dan(Qingxuedan) against 6-0HDA-induced toxicity in the primary culture of rat mesencephalic doperminergic neurons. Results : Chunghyul-dan(Qingxuedan) showed concentration-dependent scavenging activities in DPPH radical and ABTS cation scavenging assays and it was not cytotoxic to PC12 cells. In postand co-treatment, Chunghyul-dan(Qingxuedan) protected PC12 cells from the 6-OHDA induced toxicity at 50 and 100 ${\mu}$g/mL significantly. And Chunghyu!-dan(Qingxuedan) decreased the 6-OHDA induced ROS production at a dose dependent manner, while increaing the 6-OHDA induced GSH reduction at 50 and 100 ${\mu}$g/mL significantly. Finally, Chunghyul-dan(Qingxuedan) showed signicant protection of rat mescencephalic dopaminergic neurons from 6-OHDA at 1 ${\mu}$g/mL. Conclusions : These results demonstrate that Chunghyul-dan(Qingxuedan) has the antioxidant and neuroprotective effect against 6-0HDA induced cytotoxicity through decreasing ROS production and increasing GSH reduction.

  • Sensor nodes used in Geosensor network are resource limited and power constrained. So it is necessary to research on routing protocols to gather data by using energy efficiently. Wireless sensor networks collect data gathered from sensor nodes by transfering it to the sink using multihop. However, it has two problems. First, the existing works require unnecessary data transmission for choosing a proper parent node to transfer data. Secondly, they have a large number of data transmission because each sensor node has a different path. To solves the problems, we, in this paper, propose a designated path based data aggregation scheme for efficient energy management in WSNs. The proposed scheme can reduce unnecessary data transmission by pre-determining a set of paths and can enable all the sensor nodes to participate in gathering data by running them in round-robin fashion. We show from performance analysis that the proposed scheme is more energy efficient than the existing directed diffusion(DD) and the hierarchical data aggregation(HDA).

Antioxidant Activity of Solvent Extract Isolated from Barley Leaves (보리잎에서 분리된 용매 추출물의 항산화 작용)

  • The antioxidant activity of solvent extracts isolated from barley leaves was investigated by measuring peroxide value. The fractions of methanol extract obtained from preparative TLC was also studies, with UV-Visible spectrum, total phenol contents and hydrogen donating ability(HDA) The antioxidant activity of various solvent extracts was, in decreasing order, methanol> ethyl ether> methylene chloride $\geq$ ethyl acetate $\geq$acetone> hexane. The antioxidant activity of the fractions of methanol extract was, in decreasing order, fraction 2> fraction 3> fraction 1 and their activity was all superior to that of tocopherol at 500 ppm level. All fraction(1, 2 and 3) exhibited a strong UV absorption at 280 m which would be specifically produced by phenolic compound. UV absorption at 280 m of fraction 2 was greater than those of fraction 1 and 3. In the visible spectrum of these fractions, the maximum .absorption wavelengths of fraction 1, 2 and 3 were 660, 460 and 460 m, respectively. Antioxidant activity of barley leaves seemed to be due to the flavonoids containing phenolic group by UV spectrum and total phenol content.

Enzymatic Formation of 13,26-Dihexyl-1,14-dioxacyclohexacosane-2,15-dione via Oligomerization of 12-Hydroxystearic Acid

  • The enzymatic polymerization of 12-hydroxystearic acid (12-HSA) was carried out with Lipase $CA^{(R)}$ in benzene to produce poly(12-hydroxystearate) (PHS) with a low molecular weight. When this polymerization was continued for a long reaction time, the PHS once formed was depolymerized into a cyclic diester, 13,16-dihexyl-1,14-dioxacyclohexacosane-2,15-dione (12-HSAD). Similar polymerization and depolymerization were observed when 12-hydroxyoleic acid (12-HOA) was treated with Lipase $CA^{(R)}$, whereas only polymerization occurred when 12-hydroxydodecanoic acid (12-HDA) was treated in a similar manner. The preferential formation of cyclic diesters for 12-HSA was attributed to the structural requirements due to the bulky n-hexyl side groups stemming from the ring systems.

Performance Improvement of Korean Connected Digit Recognition Using Various Discriminant Analyses (다양한 변별분석을 통한 한국어 연결숫자 인식 성능향상에 관한 연구)

  • In Korean, each digit is monosyllable and some pairs are known to have high confusability, causing performance degradation of connected digit recognition systems. To improve the performance, in this paper, we employ various discriminant analyses (DA) including Linear DA (LDA), Weighted Pairwise Scatter LDA WPS-LDA), Heteroscedastic Discriminant Analysis (HDA), and Maximum Likelihood Linear Transformation (MLLT). We also examine several combinations of various DA for additional performance improvement. Experimental results show that applying any DA mentioned above improves the string accuracy, but the amount of improvement of each DA method varies according to the model complexity or number of mixtures per state. Especially, more than 20% of string error reduction is achieved by applying MLLT after WPS-LDA, compared with the baseline system, when class level of DA is defined as a tied state and 1 mixture per state is used.

Atom Transfer Radical Polymerization of Hexadecyl Acrylate Using CuSCN as the Catalyst

  • The atom transfer radical polymerization (ATRP) of hexadecyl acrylate (HDA) was carried out in Ν,Ν-dimethylformamide (DMF) in the presence of CuSCN/Ν,Ν,Ν′,Ν"Ν"-pentamethyldiethylenetriamine (PMDETA). The results indicate that the polymerization is well-controlled: a linear increase of molecular weights occurs with respect to conversion and the polydispersities are relatively low. In particular, the use of CuSCN as the catalyst resulted in faster polymerization rates for hexadecyl acrylate than did those using either CuBr or CuCl; the polydis-persity, however, was larger than those obtained in the cases when CuBr and CuCl were used. In addition, we report the thermodynamic data and activation parameters for the solution ATRP of hexadecyl acrylate.

Antioxidative Activity of Extract from Bangah Herb (방아 추출물의 항산화 효과)

  • Bangah, one of the herbs grown in Korea, was investigated for its antioxidant activity. The ether extracts of bangah herb was separated into neutral, phenolic, acidic and basic fractions and further separated into subfractions. Antioxidative activities were measured by hydrogen donating activity (HDA), peroxide value (POV), thiobarbituric acid (TBA) value and inhibition activity against lipid peroxidation of rat liver microsomes, The subfraction components were identified by GC/MS and NMR. Phenolic, though being very small in quantity, showed higher antioxidant activity at all assay system by hydrogen donating activity. POV, TBA value and inhibition activity against lipid peroxidation of rat liver microsomes. Five subfractions(P-1, P-2, P-3, P-4 and P-5) were fractionated from phenolic fraction of bangah herbs, and subfraction P-2 among them showed strong antioxidant activity on a level with BHT or gallic acid at each assay system. Four compounds (peak I, peak II, peak III and peak IV) were isolated by gas chromatogram of TMS derivatives of subfraction P-2 and thes compounds were confirmed to be phenolic substance having -OH and COOH group. There subfractions (N-1, N-2 and N-3) were fractionated from neutral fraction of bangah herbs, and subfraction N-2 among them showed highest antioxidant activity and inhibition activity against lipid peroxidation of rat liver microsomes. Subfraction N-2 was indentified to be estragole by H-NMR spectroscopy.

Behavioral Pharmacological Studies of Nootropic Candidates on the Central Dopaminergic Activity Rats (중추도파민신경활성에 미치는 뇌기능개선 후보약물의 행동약리학적 연구)

  • The present study was undertaken to elucidate the behavioral characteristics of nootropic candidates, entrophenoxine, N-methyl-D-glucamine, piracetam and red ginseng saponin components on stereotyped sniffing behavior induced by apomorphine in rats. Apomorphine, a direct dopaminergic receptor agonist, induced stereotyped behaviors including sniffing licking growing and biting in a dosedependent manner, and that behaviors were completely inhibited when measured at 1 week after 6-ydroxydopamine(6-HDA) treatment. Centrophenoxine, N-methyl-D-glucamine, red ginseng total saponin(TS), panaxatriol (PT), and Rg1 enhanced but panaxadiol (PD) inhibited, whereas piracetam and Rb1 were not effective of the sterotyped sniffing behavior induced by apomorphine(1mg/kg). The enhanced stereotyped behavior by centrophenoxine, N-methyl-D-glucamine, red ginseng total saponin, panaxatriol(PT), and Rg1 seems to have a similarity to entrophenoxine, N-methyl-D-glucamate in modulating of dopaminergic neuroal activity and also my be useful for the nootropic candidates.

Construction of a linkage Map in Capsicum annuum L. Using RAPD Markers and Identification of Two QTLs.

  • A linkage map of Capsicum annuum L. was constructed by random amplified polymorphic DNA (RAPD) markers followed in a backcross population of an intraspecific cross between cultivars HDA210 and Yatsufusa. A total of 420 random primers were tested and 311 polymorphic bands were generated by 158 random primers. Among them, 86 Yatsufusa specific bands generated by 52 primers were examined for mapping. Most bands except three segregated in Mendelian fashion fitting the expected 1:1 ratio. The total length of the map was 533 cM distributed in 15 linkage groups. The map distance between adjacent markers ranged 0 to 32.8 cM, with an average distance of 9.1 cM (63 markers). Some markers were clustered and this may be due to the amplification of a repetitive sequence by the RAPDs. Primer pairs for a sequence characterized amplified region (SCAR) were developed and the segregation scores by the SCAR primers were in accordance with the RAPD data. Two QTL markers for number of axillary shoots and for early flowering were developed. One QTL for early flowering located in the linkage group 3 and explained 61 "io of the phenotypic variation. The other QTL for the number of axillary shoots located in the linkage group 4 explained 55 % of the phenotypic variation.tion.
