The Characteristics of Growth of Brochothrix thermosphacta on Packaged Beef (포장우육에서 Brochothrix thermosphacta의 생육특성에 관한 연구)

  • This work was carried out to get the basic data about packaged meat by investigating the characteristics of growth of B. thermosphacta. For this purpose. B. thermosphacta and Lactic acid bacteria were isolated from the vacuum-packaged beef and the effect of various conditions such as pH of beef, packaging method, and Lactobacilli on the growth of B. thermosphacta was investigated. In aerobic packaged beef, the pH did not affected the growth of the B. thermosphacta, while it did affected critically in vacuum packaged beef. The lowest pH for the growth of B. thermosphacta in aerobic-packaged beef was 5.3-5.4 and 5.4-5.5 in vacuum-packaged beef. The growth of B. thermosphacta was completely stopped when the beef with pH 5.7-6.0 was packaged in 100% $CO_2$, but it was not affected significantly when the beef was packaged in a mixture of $CO_2$(20%) and $N_2$(80% ). The inhibitory effect of lactic acid bacteria on the growth of B. thermosphacta was observed faintly in aerobic-packaged beef, but in vacuum-packaged beef, the lower the pH , the more significant its inhibitory effect. The growth of B. thermosphacta was almost stopped in a beef with pH 5.5-5.6.

Enhanced Anthocyanin Accumulation by UV-B and JA Treatment in Cell Suspension Culture System of Grope (Vitis vinifera L.)

  • Effects of jasmonic acid treatment, UV-B and white light treatment on the anthocyanin biosynthesis and cell growth were investigated using the cell suspension culture system of grape (Vitis vinifera L.). Cell growth was not affected by white light irradiation, while it was remarkably suppressed by UV-B irradiation from 8 to 32 h. Anthocyanin accumulation dramatically increased after 16 h from irradiation of UV-B. Simultaneous treatment of jasmonic acid and UV-B increased anthocyanin accumulation by 10-fold. The cell division was restored when anthocyanin was abundantly accumulated after 32 h from UV-B irradiation. Optimum concentration of jasmonic acid was found to be 5 uM for maximum accumulation of anthocyanin. Application of jasmonic acid to grape suspension cells rapidly induced the expression of CHS gene after 2 h from treatment and showed maximum level at 32 h. Simultaneous treatment of jasmonic acid and light also induced CHS gene expression after 2 h, but the maximum level of CHS transcript was observed at 16 h with white light and 8 h with UV-B exposure. The synergistical effects could be explained by the defense mechanism that UV irradiation is mediated in part by alterations in JA and its signaling pathway.

Improvement of the Optimum pH of Aspergillus niger Xylanase towards an Alkaline pH by Site-Directed Mutagenesis

  • In an attempt to shift the optimal pH of the xylanase B (XynB) from Aspergillus niger towards alkalinity, target mutation sites were selected by alignment between Aspergillus niger xylanase B and other xylanases that have alkalophilic pH optima that highlight charged residues in the eight-residues-longer loop in the alkalophilic xylanase. Multiple engineered XynB mutants were created by site-directed mutagenesis with substitutions Q164K and Q164K+D117N. The variant XynB-117 had the highest optimum pH (at 5.5), which corresponded to a basic 0.5 pH unit shift when compared with the wild-type enzyme. However, the optimal pH of the XynB-164 mutation was not changed, similar to the wild type. These results suggest that the residues at positions 164 and 117 in the eight-residues-longer loop and the cleft's edge are important in determining the pH optima of XynB from Aspergillus niger.

  • The reaction of 6-chloro-2-hydrazinoquinoxaline 4-oxide (11) with ethyl chloroglyoxylate provided ethyl 8-chloro-4H-1,3,4-oxadiazino[5,6-b]quinoxaline-2-carboxylate (12) through the intramolecular cyclization. The reaction of 12 with hydrazine hydrate afforded the $C_2$-hydrazinocarbonyl derivative 13, whose reactions with substituted benzaldehydes or heteroaryl aldehydes provided 8-chloro-2-(substituted benzylidenehydrazinocarbonyl)-4H-1,3,4-oxadiazino[5,6-b]quinoxalines (14) or 8-chloro-2-[(2-substituted methylidene)hydrazinocarbonyl]-4H-1,3,4-oxadiazino[5,6-b]quinoxalines(15), respectively.

  • It is shown that every almost linear mapping $h:{\mathcal{A}}{\rightarrow}{\mathcal{B}}$ of a unital Poisson Banach algebra ${\mathcal{A}}$ to a unital Poisson Banach algebra ${\mathcal{B}}$ is a Poisson algebra homomorphism when h(xy) = h(x)h(y) holds for all $x,y{\in}\;{\mathcal{A}}$, and that every almost linear almost multiplicative mapping $h:{\mathcal{A}}{\rightarrow}{\mathcal{B}}$ is a Poisson algebra homomorphism when h(qx) = qh(x) for all $x\;{\in}\;{\mathcal{A}}$. Here the number q is in the functional equation given in the almost linear almost multiplicative mapping. We prove that every almost Poisson bracket $B:{\mathcal{A}}\;{\times}\;{\mathcal{A}}\;{\rightarrow}\;{\mathcal{A}}$ on a Banach algebra ${\mathcal{A}}$ is a Poisson bracket when B(qx, z) = B(x, qz) = qB(x, z) for all $x,z{\in}\;{\mathcal{A}}$. 광-펜톤 반응을 이용한 Rhodamine B의 색 제거

  • 수용성 안료인 RhB를 대상으로 광-펜톤 공정의 최전 운전조건을 구하고, 광-펜톤 공정을 구성하는 개별 공정을 비교한 결과 다음의 결론을 얻었다. 광-펜톤 공정의 최적 $Fe^{2+}$$H_2O_2$ 투입량은 각각 0.0031 mmol과 0.625 mmol이었으며, 최적 pH는 3으로 나타났으나, 7이하의 pH 범위에서는 RhB 색 감소에 큰 영향을 미치지 않는 것으로 나타났다. 초기 반응 속도에 가장 큰 영향을 주는 인자는 UV 광 전력 > $H_2O_2$> 철염의 순으로 나타났다. 80분간의 반응시간 경과 후 최종 RhB 농도를 고찰한 결과 UV 광 전력이 낮을 경우 색도가 다 제거되지 않기 때문에 광-펜톤 공정에서 UV 광 전력이 색 제거에 대한 가장 큰 인자라고 사료되었다. 광-펜톤 공정의 개별 공정인 UV, $H_2O_2$, 펜톤을 이용하여 RhB의 농도감소를 고찰한 결과 초기 반응속도상수는 펜톤 공정의 빠른 초기 반응로 인해 펜톤 > UV >$H_2O_2$로 나타났으나, 최종 RhB 농도를 고려할 경우 UV> 펜톤 > $H_2O_2$로 나타났다.

Exploratory Study to Develop Customers' Experience Measurement Scale of H&B Store

  • Purpose: Recently, Korean cosmetics distribution market has been reorganized with the H&B store. In the domestic cosmetics distribution market, existing brand road shops are decreasing, and multi-shops are leading the H & B stores, which have greatly improved their experience and content. In these environmental changes, the offline distribution channels are turning into the multi-editing shops that have introduced products of various brands and greatly enhanced experiences and contents. Nevertheless, most studies of factors and measurement items for measuring customer experience in the H&B store use Schmitt (1999)'s Strategic Experience Modules (SEMs). Therefore, the purpose of this study is to propose a measure that is practicable through consideration of the in-store customer experience components of the H&B store. Research design, data and methodology: Based on Schmitt's Strategic Experience Modules (SEMs), which are widely used in customer experience marketing, the metric pool was constructed through customer and literature research on H & B store managers. Since then, 101 preliminary surveys and 211 main surveys have been conducted in order to propose a dimension of customer experience and refine the metrics. Results: As a result of the research, H&B store's customer experience was derived from a measurement model consisting of 19 measurement items in total of five dimensions: environmental experience, intellectual experience, behavioral experience, tech experience, and relationship experience. This study analyzed that compared to the existing Schmitt's Strategic Experience Modules (SEMs), (1) emotional experience expanded to environmental experience, (2) Cognitive and relationship experiences are maintained (3) behavioral experience was subdivided into physical and technical experiences. In particular, the environmental experience has been proposed as a major component is an important point because the H&B store recently opened a large flagship store and is competitive in constructing a differentiated space. Conclusions: Related experience was seen as an important component of customer experience in the offline store, but in the process of refining the scale, interaction items with employees of the H&B store were removed, and rather, participation in the APP or SNS channel of the company, event Participation, interaction with other customers, etc. appear to be important, while suggesting the practical implications.

B-H Loop Measurement of a High Tensile Steel Plate (사각판재형 강재의 자기특성측정)

  • Minor B-H loop measurement for a rectangular high tensile steel was obtained by using Labview. A ferrite cores of high permeance with primary and secondary windings on the steel plate were used to form a closed loop of magnetic flux. To compensate errors due to an extremely small gap between a pair of ferrite core, and between the ferrite core and the rectangular high tensile steel, quadratic function of least square method was used. Also a 3D FEM magnetic analysis tool was used to measure H and B of the steel. B-H loop of the high tensile steel plate can be measured up to 520 A/m of a magnetic field and 0.15 T of a magnetic flux density.

  • Recently, pre-engineering building (P.E.B.) systematic frames are increasingly being used in steel factory buildings, but almost of the related techniques are dependent on the engineering program (e.g, MBS, LTI), which is usually imported from other countries. These are designed under the AISC-ASD because at present there is no Korean design code for P.E.B. frames. Also, there are few studies onbehaviour and we need to develop the element techniques by using H-shaped components.In particular, there is a tendency towards overestimated design because column-rafter connections have been designed with extended end plate type joint, which is treated asrigid joint,so structural examinations are needed. Therefore, this study represents a basic step in ascertaining the application of P.E.B. systematic frames by using H-shaped column-rafter connectionwith flush type end plate. Its structural performance is compared with that of existing extended type joint using a structural performance test. The structural behaviour of specimen was understood qualitatively and the possibility of application (e.g, design aid charts) of semi-connection (flush type) with H-shaped column-rafter was determined.
