Candida sp. LSY2 중금속 내성주를 이용한 BOD 분석 (BOD Determination by Heavy Metal Tolerant Candida sp. LSY2)

  • 용존 유기물의 농도를 BOD로 신속히 측정하여 폐수 처리 공정에 feed back할 수 있는 Candida BOD sensor를 이용하여 BOD를 측정하였다. Glucose, acetic acid, aspartic acid 및 glutamic acid에 대한 sensitivity는 lactose, sucrose및 lactic acid의 sensitivity보다 더 높게 나타나서 오염물질의 종류에 따라 BOD sensor의 출력 신호가 다르게 제시되었다.$Zn^{2+}$내성주가 충전된 BOD sensor는 중금속 비내성주의 경우와 유사한 정도의 감응도를 나타내었으나 $Cd^{2+}$ 내성주를 사용하였을 때는 매우 낮은 sensitivity를 보였다. 수용액의 $Cd^{2+}$의 농도가 증가함에 따라 중금속 비내성주와 $Zn^{2+}$ 내성주의 sensor 의 sensitivity는 거의 일정하였으나 $Cd^{2+}$ 내성주의 BOD sensor의 경우에는 sensitivity가 오히려 감소하였다. 반면 수용액 중의 $Zn^{2+}$ 의 농도가 증가함에 따라서는 중금속 비내성주, $Zn^{2+}$ 내성주 및 $Cd^{2+}$ 내 성주 모두 거의 동일한 감응도를 나타내었다.

Selection and Isolation of a Mutant Yeast Strain Tolerant to Multiple Targeted Heavy Metals

  • BACKGROUND: This study was performed for selecting yeast mutants with a high tolerance for targeted metals, and determining whether yeasts strains tolerant to multiple heavy metals could be induced by sequential adaptations. METHODS AND RESULTS: A mutant yeast strain tolerant to the heavy metals cadmium (Cd), copper (Cu), nickel (Ni), and zinc (Zn) was selected by sequential elevated exposures to each metal with intermittent mutant isolation steps. A Cd-tolerant mutant was isolated by growing yeast cells in media containing $CdCl_2$ concentrations that were gradually increased to 1 mM. Then the Cd-tolerant mutant was gradually exposed to increasing levels of $CuCl_2$ in growth media until a concentration of 7 mM was reached, thus generating a strain tolerant to both Cd and Cu. In the subsequent steps, this mutant was exposed to $NiCl_2$ (up to 8 mM), and a resultant isolate was further exposed to $ZnCl_2$ (up to 60 mM), allowing the derivation of a yeast mutant that was simultaneously tolerant to Cd, Cu, Ni, and Zn. CONCLUSION: This method of inducing tolerance to multiple targeted heavy metals in yeast will be useful in the bioremediation of heavy metals.

Sulfate Reduction for Bioremediation of AMD Facilitated by an Indigenous Acid- and Metal-Tolerant Sulfate-Reducer

  • Acid mine drainage (AMD) has been a serious environmental issue that threatens soil and aquatic ecosystems. In this study, an acid-tolerant sulfate-reducing bacterium, strain S4, was isolated from the mud of an AMD storage pond in Vietnam via enrichment in anoxic mineral medium at pH 5. Comparative analyses of sequences of the 16S rRNA gene and dsrB gene involved in sulfate reduction revealed that the isolate belonged to the genus Desulfovibrio, and is most closely related to Desulfovibrio oxamicus (with 99% homology in 16S rDNA sequence and 98% homology in dsrB gene sequence). Denaturing gradient gel electrophoresis (DGGE) analyses of dsrB gene showed that strain S4 represented one of the two most abundant groups developed in the enrichment culture. Notably, strain S4 was capable of reducing sulfate in low pH environments (from 2 and above), and resistance to extremely high concentration of heavy metals (Fe 3,000 mg/l, Zn 100 mg/l, Cu 100 mg/l). In a batch incubation experiment in synthetic AMD with pH 3.5, strain S4 showed strong effects in facilitating growth of a neutrophilic, metal sensitive Desulfovibrio sp. strain SR4H, which was not capable of growing alone in such an environment. Thus, it is postulated that under extreme conditions such as an AMD environment, acid- and metal-tolerant sulfate-reducing bacteria (SRB)-like strain S4 would facilitate the growth of other widely distributed SRB by starting to reduce sulfate at low pH, thus increasing pH and lowering the metal concentration in the environment. Owing to such unique physiological characteristics, strain S4 shows great potential for application in sustainable remediation of AMD.

Selection of Newly Isolated Mushroom Strains for Tolerance and Biosorption of Zinc In Vitro

  • Nine newly isolated mushroom strains were tested to assess both their zinc tolerance and potential for zinc removal from an aqueous solution. Four strains of ectomycorrhizal fungi, namely Clavariadelphus truncatus (T 192), Rhizopogon roseolus (T 21), Lepista nuda (T 373), and Tricholoma equestre (T 174), along with five strains of white rot fungi, Lenzites betulina (S 2), Trametes hirsuta (T 587), Ganoderma spp. (T 99), Polyporus arcularius (T 438), and Ganoderma carnosum (M 88), were investigated using zinc-amended solid and liquid media. Their biosorption properties were also determined. The colony diameter and dry weight were used as tolerance indices for fungal growth. C. truncatus and T. equestre were not strongly inhibited at the highest concentrations of (225 mg/l) zinc in solid media. The most tolerant four strains with solid media, C. truncatus, G carnosum, T. hirsuta, and T. equestre, were then chosen for tolerance tests in liquid media. An ectomycorrhizal strain, C. truncatus, was also detected as the most tolerant strain in liquid media. However, the metal-tolerant strains demonstrated weak activity in the biosorption studies. In contrast, the highest biosorption activity was presented by a more sensitive strain, G. carnosum. In addition, seven different biosorbent types from G. carnosum (M 88) were compared for their Zn (II) biosorption in batch experiments.

Serratia marcescens LSY4 중금속 내성주를 이용한 BOD센서의 감응도 (Sensitivity of BOD Sensor with Heavy Metal Tolerant Serratia marcescens LSY4)

  • S. marcescens LSY4가 충전된 BOD센서를 제작하여 표준오염물질을 포함한 수용액의 BOD를 측정하였다. 배양시간이 9∼16시간일 때에는 배양시간에 따른 BOD센서의 감응도에 큰 차이를 나타내지 않았으며, 균체량도 0.22~0.75mg $cm^{-2}$의 범위에서는 거의 동일한 센서의 감응도를 나타내었다. 수용액의 pH가 4~9사이로 변화할 경우 센서의 감응도가 가역적으로 변화하였으나 수용액이 더 산성이거나 더 염기성이 되면 센서의 감응도가 비가역적으로 저하되었다. 수용액에 중금속이온이 첨가되면 센서의 감응도가 감소하였으며, $Zn^{2+}$$Cd^{2+}$보다 $Cu^{2+}$혹은 $Ag^+$가 첨가되었을 때 센서의 감응도가 더 급격히 감소하였다. 중금속에 대한 내성이 유도된 균체를 충전하였을 때 중금속이온의 첨가에 따른 센서의 감응도 감소가 크게 완화되었으며, 이런 효과는 $Cd^{2+}$내성주보다 $Cu^{2+}$내성주에서 더 현저하였다.

카드뮴 내성균(耐性菌)의 분리(分離), 동정(同定)및 균체내(菌體內) 카드뮴 축적(蓄積) 특성(特性) (Isolation of Cadmium-Tolerant Bacteria and Characterization of Cadmium Accumulation into the Bacteria Cell)

  • 폐수중의 카드뮴을 효과적으로 제거하기 위하여 경남 및 부산지역의 오염된 하천수, 공단폐수처리장의 폐수 및 sludge 그리고 공단주변지역의 토양으로 부터 카드뮴에 내성이 있는 미생물 162균주를 분리하여, 이들 균주중 2,000ppm의 카드뮴이 함유된 한천평판배지에서 생장이 우수한 6균주를 선별한 후 이들 균주의 카드뮴 흡수량을 조사하여, 그 중에서 카드뮴 흡수가 가장 우수한 1균주를 선별하여 동정한 결과 Pseudomonas putida 또는 그 유연균으로 밝혀졌으며, 그 분리균의 최적생장 온도는 $30^{\circ}C$였고, 최적 pH는 pH 7.0이었다. 분리균주의 항생제 내성, 중금속 내성 및 탄화수소 자화능을 조사한 결과, 항생제인 ampicillin(Ap), chloramphenicol(Cm) 및 streptomycin(Sm)과 중금속인 Li, Cu, Pb 및 Zn에 내성을 나타내었다. 그리고 탄화수소인 salicylate, naphthalene 및 xylene을 단일탄소원으로 이용하였다. 분리균의 카드뮴 농도에 따른 생장을 조사한 결과 카드뮴이 첨가되지 않은 배지에서는 배양 1일 후에 최대생장에 도달했고, 카드뮴 100ppm의 농도에서는 2일 후에 최대생장에 도달하였으며 대조구 (카드뮴 무첨가 배지)와 큰 차이가 없었다. 카드뮴의 농도가 높을수록 균의 생장이 심히 저해되었다. 균체내의 카드뮴횹수량은 카드뮴농도가 낮을수록 증가되었다. 배지중 카드뮴농도가 1ppm과 10ppm인 경우에는 배양 1일 후에 최고의 흡수량을 보였고 100ppm에서는 배양 2일 후에 최고의 축적율을 보였다. 균체내 카드뮴 흡수율은 배지중에 카드뮴 농도가 1ppm인 경우에는 최고 78%, 10ppm에서는 60%, 그리고 100ppm에서는 약 40%의 흡수율을 보였다. 균체내에 다량의 카드뮴을 축적시키기 위하여 배양시 계면활성제를 넣어 배양한 결과, 비이온계 계면활성제인 Triton X-100을 0.1% 첨가했을 때 약 37%의 카드뮴축적 증가를 보였다.

A cold-active acidophilic endoglucanase of Paenibacillus sp. Y2 isolated from soil in an alpine region

  • A cellulolytic strain Y2 was isolated from soil obtained in the Canadian Alpine region. The isolate was identified as Paenibacillus sp. Y2 by 16S rRNA sequencing. When grown in LB medium supplemented with carboxymethyl-cellulose (CMC), CMCase production increased to 122.0% of that observed in LB without CMC. Culture supernatant was concentrated by ultrafiltration and 80% ammonium sulfate precipitates were separated by Hi-Trap Q and CHT-II chromatography. The purified enzyme (EG-PY2) showed a homogeneous single band and the molecular mass was estimated to be 38 kDa by SDS-PAGE. Optimum pH and temperature of the enzyme were 4.5 and $30^{\circ}C$, respectively. The half-life of enzyme activity at 50 was 140.7 min, but the enzyme was drastically inactivated within 5 min at $55^{\circ}C$. The enzyme was highly activated to 135.7 and 126.7% by 5.0 mM of $Cu^{2+}$ or $Mg^{2+}$ ions, respectively, and moderately activated by $Ba^{2+}$ and $Ca^{2+}$ ions, whereas it was inhibited to 76.8% by $Fe^{2+}$, and to ${\leq}50%$ by $Mn^{2+}$, $Co^{2+}$, $Zn^{2+}$, and EDTA. The enzyme was activated to 211.5% in the presence of 0.5 M of NaCl and greatly tolerant to 3.15M of NaCl. The enzyme showed 2.98 times higher ${\beta}$-glucanase activity than CMCase activity. Based on these results, it can be concluded that EG-PY2 is an acidophilic, cold-active, and halotolerant endoglucanase. The authors suggest it is considered to be useful for various industrial applications, such as, fruit juice clarification, acidic deinking processes, high-salt food processing, textile and pulp industries, and for biofuel production from seaweeds.
