Morphological Traits of Trisomic Plant in Durum Wheat (듀럼밀 3염색체 식물의 형태적 특성)

  • The morphological traits of different types of primary trisomics(2n=28+1) in durum wheat, Triticum durum var. hordeiforme(2n=28 AABB) were compared with disomics (2n=28) through the examination of reciprocal gene action on the extra chromosomes. However it was not easy to distinguish morphologically the trisomies containing A genome from those containing B genome. These results suggested that the chromosomal location of the major genes for some morphological traits exists on homoeologous chromosome. It is important that these results revealed the homoeology and linkage groups of both A and B genomes in durum wheat. These primary trisomies will be valuable materials for the trisomic analysis and genetic mapping on the chromosome of both A and B genomes in durum wheat. Furthermore, it must be useful for the evolutionary study of Triticum durum(AABB) and Triticum squarrosa(DD) by way of the ancester species of Triticum aesitivum(AABBDD).

Development and Prey Consumption of Phytoseiid Mites, Amblyseius womersleyi, A. fallacis, and Typhlodromus occidentalis under controlled Environments (점박이응애 천적인 3종 이리응애의 발육 및 포식량 비교)

  • Comparative studies on some ecological characteristics of three phytoseiid mitespecies (one native; Arnblyseius womersleyi Schica, and two introduced species; A. fallacis Garmanand Typhlodromus occidentalis Nesbit) were carried out. The two-spotted spider mite (Tetranychusurticae Koch) was supplied as prey. Under four constant temperatures of 20, 23, 25 and 30f OS$^{\circ}$C,duration of growth from egg to adult for A. womer.vleyi was 11.5, 7.7, 6.7 and 5.6 days. While twoother species needed slightly shorter time but not significantly different. Critical temperature andeffective degree-days (DD) of A. womerslevi females were 83$^{\circ}$C and 1 1 1.6 DD, whereas those of A..fallacis were 10.7"C and 86.0 DD, and those of 7: occidentalis were 10.7"C and 94.1 DD. Also, thoseof males were similar to their females. Average longevity of females of A. womersleyi, A. fallucisand 7: occidentalis were 18.2 k 8.67, 19.6 3~7.18 and 13.0f5.66 days, total fecundity were 34.3 $-11.93, 39.8k 12.64 and 23.6k8.86, respectively. Under four constant temperatures of 20, 23, 25 and30-t0.S$^{\circ}$C, A. womersleyi consumed 9.1 f2.49, 9.7 k2.00, 9.7 f 2.61, and 10.3 k2.33 eggs of 7:urticae throughout their development. A. ,fizlluc~i.sc onsumed 10.2 k 2.52, 9.7 f2.29, 10.7 f 2.37 and10.1 k2.62 eggs, while, 7: occidentalis consumed 1 1.9 k3.43, 14.2 f4.50, 14.8 k 3.2 1 and 12.7 f2.95 eggs, respectively. Gravid females of A. womersleyi, A. f~zllacis and 7: occidentalis consumed11.4f1.59, 12.5k1.43 and 11.7k3.07 eggs, or consumed 11.9f 2.63, 12.4k3.82, and 12.5f 3.73protonymphs of 7: urticae in a day at 25-30$^{\circ}$C.e in a day at 25-30$^{\circ}$C.

Association Study between Porcine LEPR-derived Microsatellite Polymorphisms and Economic Traits (돼지 leptin receptor내 초위성체 다형성과 경제형질과의 연관성 구명)

  • The leptin receptor gene(LEPR) produces a high affinity receptor that mediates the regulation of the leptin gene. Leptin secreted from adipose tissue plays an important role in regulating feed intake and energy balance. In this study, a microsatellite marker within LEPR was selected and genotyped for the F2 population composed of 354 individuals from an intercross between Korean Native boars and Landrace sows. Totally, six alleles (255, 259, 261, 263, 265 and 267bp) and nineteen genotypes were detected in the population, of which the CE (261/265), CC (261/261) and EE (265/265) types were observed by 20.0%, 10.1% and 9.6%, respectively. Relationships between their genotypes and economic traits were analyzed. We found specific genotypes associated with economic traits such as body weight at 12 weeks of age/body fat including abdominal and trimmed fat/shear force (P〈0.001), body weight of 30 weeks of age (P〈0.01) and body weight of 3 weeks of age/back fat thickness (P〈0.05). The DD (263/263) and DF (263/267) types were associated with body weight at 3, 5, 12 and 30 weeks of age. The DF (263/267) type showed a highly significant effect on back fat thickness and body fat including abdominal and trimmed fat. The DF (263/267) type showed positive effect on shear force, whereas the BB (259/259) and DD (263/263) types negatively affected on tenderness.

Effect of Massages on The Skeletal Muscles of Sciatic Nerve Neurectomized ddY Mice (마사지가 좌골신경 절단 마우스의 골격근에 미치는 영향)

  • To demonstrate the effect of massages on the skeletal muscles of immobilization ddY mice models induced by right side sciatic nerve neurectomy, the cross sectional histological profiles of the muscularis (M) gastrocnemius, M. tibialis cranialis and M. tibialis caudalis were observed after 28 days of treatment of massages with the changes of body weight, thickness of hind limb and individual muscle weights. In addition, changes of serum aspartate amino- transferase (AST) and creatine phosphokinase (CK) levels were also demonstrated with diameter of individual muscle fiber and muscle fasciculata, and number of muscle fiber in each of three types of muscles located in the calf. The experimental groups were divided into five groups, 1) Sham-operated group (Sham), 2) Neurectomized but not massage control group (Control), 3) Neurectomized and massage at knee joint regions (T1), 4) Neurectomized and massage at calf regions (T2), and 5) Neurectomized and massages at achilles tendon regions (T3). The experimental animals were used 5 per groups. The results are as follow: 1. No significant changes of body weight were demonstrated in this groups during whole experimental periods compared to that of Sham group. In addition, no significant changes of body weight gains during experimental periods (Day 0 - Day 27: 28 days of treatment) were also observed. 2. The thickness of intact left side hind limb was not changes in all tested groups compared to that of Sham group. However, the thickness of right side hind limb were remarkedly decreased in Control, T1, T2 and T3 groups compared to that of Sham group. In addition, the differences of thickness of hind limb between right and left sides were also significantly increased in Control and massage groups (T1, T2 and T3) compared to that of Sham group. However, the thickness of right side of hind limb were significantly increased and the differences of thickness between both sides of hind limb were dramatically decreased in massage groups compared to that of Control group in followed order: T1 > T3 > T2. 3. Absolute and relative weight of M. gastrocnemius in the intact left side of hind limb was not changes in all tested groups compared to that of Sham group. However, the weight of right side M. gastrocnemius were remarkedly decreased in Control, T1, T2 and T3 groups compared to that of Sham group. In addition, the differences of weight of M. gastrocnemius between right and left sides were also significantly increased in Control and massage groups compared to that of Sham group. However, the weight of right side M. gastrocnemius were significantly increased and the differences of weight between both sides of M. gastrocnemius were dramatically decreased in massage groups compared to that of Control group in followed order: T3 > T1 >. 4. Absolute and relative weight of M. tibialis cranialis in the intact left side was not changes in all tested groups compared to that of Sham group. In addition, the differences of weight of M. tibialis caudalis between right and left sides were also significantly increased in Control and massage groups compared to that of Sham group. However, the weight of right side M. tibialis caudalis were significantly increased and the differences of weight between both sides of M. tibialis caudalis were dramatically decreased in massage groups compared to that of Control group in followed order: T3 > T1 > T2. 6. A significant increase of serum AST levels were demonstrated in Control group. However, serum AST levels of massage groups were significantly decreased compared to that of Control group in followed order: T3 > T1 > T2. In case of 73 group, the values decreased into similar levels compared to that of Sham group. 7. A significant increase of serum CK levels were demonstrated in Control group. However, serum CK levels of massage groups were significantly decreased compared to that of Control group in followed order: T3 > T1 > T2.

Possible involvement of temperature-entrainable timing system in arrhythmic mutant flies in Drosophila melanogaster

  • In Drosophila melanogaster, it is known that the circadian clock consists of an autoregulatory feedback loop, which includes so-called clock genes, such as per, tim, dClk and cyc and produces periodical expression of per. It is recently suggested, however, that the circadian oscillation without the rhythmical expression of per is involved in the regulation of circadian locomotor rhythms. In the present study, we examined the existence and the property of the possible per-less oscillation using arrhythmic clock mutant flies carrying per$^{01}$, tim$^{01}$, dClk$^{Jrk}$ or cyc$^{01}$. When temperature cycles consisting of 25$^{\circ}$C and 30$^{\circ}$C with varying periods (T = 8~32 hr) were given, they showed rhythms synchronizing with the given cycle under constant darkness (DD). per$^{01}$ and tim$^{01}$ flies always showed a peak around 7 hr after the onset of thermophase irrespective of Ts of temperature cycles, while dClk$^{Jrk}$ and cyc$^{01}$ flies did not. In addition, several days were necessary to establish a clear temperature entrainment in per$^{01}$ and tim$^{01}$ flies, when they were transferred from a constant temperature to a temperature cycle under DD. These results suggest that per$^{01}$ and tim$^{01}$ flies have a temperature-entrainable weak oscillatory mechanism. The fact that dClk$^{Jrk}$ and cyc$^{01}$ flies did not show any sign of the endogenous oscillation suggests that the per-less oscillatory mechanism may require CLK and CYC.

Association between Single Nucleotide Polymorphisms of Fatty Acid Synthase and Fat Deposition in the Liver of the Overfed Goose

  • Goose fatty liver is one of the most delicious and popular foods in the world, but there is no reliable genetic marker for the early selection and breeding of geese with good liver-producing potential. In our study, one hundred and twenty-four 78-day-old Landes geese bred in Shunda Landes goose breeding farm, Jiutai, Jilin, China were selected randomly. The fatty livers were sampled each week after overfeeding during a three week period. Polymerase chain reaction-single strand conformation polymorphism and DNA sequencing were used to identify single nucleotide polymorphisms (SNPs) of fatty acid synthase (FAS), which is an important enzyme involved in the synthesis of fat under both physiological and pathological conditions. Least-squares correlation was established between these SNPs and fatty liver weight, abdominal fat weight, and intestinal fat weight of the overfed Landes geese, respectively. The results showed that fatty liver weight of geese with EF and FF genotypes (amplified by primer P1) was significantly higher than that of the EE genotype (p<0.05), and liver weight of CD and DD genotypes (amplified by primer P2) was significantly higher than that of the CC genotype (p<0.05). Different genotype combinations showed different liver weights, and from highest to lowest were ABDD, DDEF, DDFF, DDEE, ABEF, ABFF, AADD, and CDEF. Further analysis of DNA sequencing showed that there were two SNPs within the 5' promoter region the FAS gene. The geese of EF and FF genotypes carried a change of T to C, and the geese of CD and DD genotypes carried a change of A to G. The changes of the bases could potentially influence the binding of some transcription factors to this region as to regulate FAS gene. To our knowledge, this is the first report of SNPs found within the 5' promoter region of the Landes goose FAS gene, and our data will provide an insight for early selection of geese for liver production.

A 2.5-V, 1-Mb Ferroelectric Memory Design Based on PMOS-Gating Cell Structure (PMOS 게이팅 셀 기반 2.5-V, 1-Mb 강유전체 메모리 설계)

  • In this paper, a FRAM design style based on PMOS-gating cell structure is described. The memory cell consists of a PMOS access transistor and a ferroelectric capacitor. Its plate is grounded. The proposed scheme employs three novel operating methods: 1) $V_{DD}$ precharged bitline, 2) negative-voltage wordline technique and 3) negative-pulse restore, Because this configuration doesn`t need the on-pitch plate control circuitry, it is effective in realizing cost-effective chip sizes. Implementation for a 2.5-V, 1-Mb FRAM prototype design in a $0.25-{\mu}m$, triple-well technology shows a chip size of $3.22\;mm^{2}$, an access time of 48 ns and an active current of 11 mA. The cell efficiency is 62.52 $\%$. It has gained approximately $20\;\%$ improvement in the cell array efficiency over the conventional plate-driven FRAM scheme.

Efficacy of Dose Dense Doxorubicin and Cyclophosphamide Followed by Paclitaxel versus Conventional Dose Doxorubicin, Cyclophosphamide Followed by Paclitaxel or Docetaxel in Patients with Node-Positive Breast Cancer

  • Background: Adding taxanes to adjuvant antracycline and cyclophosphamide (AC) in combination may provide significant improvement in node-positive and high risk node-negative breast cancer (BC) patients. However, the optimal dose and the role of dose-dense (DD) chemotherapy have yet to be determined. The aim of this study was to compare the efficacy of a DD paclitaxel (P)-AC combination with conventional weekly P-AC or docetaxel D-AC combinations in patients with node-positive breast cancer. Materials and Methods: Newly diagnosed 280 node-positive BC patients diagnosed from 1998 to 2013 in three clinics were retrospectively analyzed. Demographic and medical data were collected from the medical charts. Patients were categorized to 3 groups according to treatment arms: arm A, ddAC-P; arm B, weekly P and AC combination; and arm C; T and AC combination. Adjuvant trastuzumab was added for HER2-positive patients. Kaplan-Meier survival analysis was carried out for disease free survival (DFS) and overall survival (OS). The log-rank test was used to examine the statistical significance of the differences observed between the groups. Two-sided P values <0.05 were considered statistically significant. Results: Of the total of 280 patients, 101 were in arm A, 114 in arm B and 65 in arm C.The median ages were 49, 50 and 46, respectively (p=0.11). Median follow-up was 39 (3-193) months. Stage, lymphovascular and perineural invasion, receptor patern, and menopausal status were similar in the 3 treatment arms, but HER2 positivity was significantly lower in arm A, compared to arms B and C (25.7%, 53.1%, 41.5% in arms A, B and C, respectively; p<0.001). Also grade 3 tumors were significantly less frequent in treatment arm A compared to arm B and C (27.3%, 56.8% and 49.2%, respectively, p=0.01). Afterunivariate and multivariate analysis were performed, 3-year DFS rates were 89%, 81%, and 75%, respectively (p=0.12) and three year OS rates were 96.6%, 89%, and 75% (p=0.62). Conclusions: In this study, no significant difference was found between adjuvant dose dense and conventional taxane treatment regimens.

Characteristics of Individual Growth Curve by Porcine LEPR-derived Microsatellite Polymorphisms (돼지의 Leptin receptor 유전자내 초위성체 다형성에 따른 개체별 성장곡선 특성)

  • This study was conducted to estimate the growth curve parameters of 253 heads of F2 population produced by inter-crossing F1 from Korean Native boars and Landrace sows, and to estimate the effects of Leptin receptor gene(LEPR) on their growth characteristics. Growth curve parameters were estimated from nonlinear regression using Gompertz model individually. Average mature weight and average maturing rate estimated were 179.69${\pm}$4.40kg and 0.3103${\pm}$0.0043, respectively. The effect of sex was insignificant for all the parameters estimated from Gempertz model(p〉.05), and the effect of calving group was significant for mature weight and maximum growth rate at inflection point (p〈.05). The effect of LEPR genotype were significant for all the growth curve parameters(p〈.05). According from the results of the least squares means of growth curve parameters by LEPR genotypes, mature weight and point of inflection were highest in genotype AA in which the maturing rate was the lowest, and were lowest in genotype DD in which maturing rate was the highest, reversely.

Comparison of Bite Forces between Pre- and Post-Treatment in Patients with Temporomandibular Disorders (측두하악장애 환자의 치료 전후 교합력 비교)

  • The purpose of this study was to compare the maximum bite forces between pre- and post-treatment related to specific diagnostic groups of TMD including masticatory muscle disorder (MMD), disc derangement (DD), joint inflammation (JI) and osteoarthritis (OA). Bite force between pre- and post-treatment was compared in 36 patients with unilateral TMD, successfully-managed in the Department of Oral Medicine, Dankook University Dental Hospital, for this study. The ratio of men to women was 7:29 and their mean age of $28.1{\pm}13.7$ years. The patients were categorized, through clinical and radiographic examination, into aforementioned 4 groups; MMD (N=18), DD (N=6), JI (N=5) and OA (N=7). The maximum bite force measurements were done at the antagonizing canines and 1st molars using a bite force recorder. Paired t-test, ANOVA, Multiple Comparison t-tests were used for statistical analysis. The results of this study showed that the maximum bite force before treatment increased after TMD treatment, which was noticeable at the canines (p=0.001 and p=0.000 for the affected and unaffected sides, respectively). In comparison related to the diagnostic groups of TMD, patients with osteoarthritis of TMJ exhibited the lowest strength while those with inflammatory disorder of TMJ had the highest strength on the affected sides. Increase of bite force after treatment was also found in each group. Significant difference between pre- and post-treatment was found at canines on the affected sides in MMD (p=0.045) and DD groups (p=0.009) while on the unaffected sides in OA group (p=0.003). Conclusively, the reduced bite force due to TMD could be recovered by conservative TMD treatment and that the difference of bite forces between pre- and post-treatment was noticeable at the canines.
