핵심 잡지의 결정 요인에 관한 연구

  • The purpose of this study is to verify the factors determining core journals in a field based upon the results of the citation analysis of the journals in the field of Korean History(KH). In order to verify the determinant factors, dividing the articles of the KH journals into their novelty and author's seniority, the following hypotheses were established. Hypothesis 1: When a KH journal publishes more articles on prehistoric and ancient eras than others, the journal will have higher chance to be included in the core journals. Hypothesis 2: When a KH journal publishes more articles by senior researchers than others, the journal will have higher chance to be included in the core journals. To achieve the purpose, the cited articles found in the two sample journals, namely, Backsan Hakbo and Hanguksa Yongu were analysed during the past 20 years from 1962 to 1981. Applying the Hirst's Discipline Impact Factor method to the the study, Historical Science Impact Factor(HSIF) on the samples itself and the historical era dealt with in the articles and the seniority of the authors during every five-year period. Major ginsenosides from Panax ginseng promote aerobic cellular respiration and SIRT1-mediated mitochondrial biosynthesis in cardiomyocytes and neurons

  • Background: Aerobic cellular respiration provides chemical energy, adenosine triphosphate (ATP), to maintain multiple cellular functions. Sirtuin 1 (SIRT1) can deacetylate peroxisome proliferator-activated receptor gamma coactivator 1 alpha (PGC-1α) to promote mitochondrial biosynthesis. Targeting energy metabolism is a potential strategy for the prevention and treatment of various diseases, such as cardiac and neurological disorders. Ginsenosides, one of the major bioactive constituents of Panax ginseng, have been extensively used due to their diverse beneficial effects on healthy subjects and patients with different diseases. However, the underlying molecular mechanisms of total ginsenosides (GS) on energy metabolism remain unclear. Methods: In this study, oxygen consumption rate, ATP production, mitochondrial biosynthesis, glucose metabolism, and SIRT1-PGC-1α pathways in untreated and GS-treated different cells, fly, and mouse models were investigated. Results: GS pretreatment enhanced mitochondrial respiration capacity and ATP production in aerobic respiration-dominated cardiomyocytes and neurons, and promoted tricarboxylic acid metabolism in cardiomyocytes. Moreover, GS clearly enhanced NAD+-dependent SIRT1 activation to increase mitochondrial biosynthesis in cardiomyocytes and neurons, which was completely abrogated by nicotinamide. Importantly, ginsenoside monomers, such as Rg1, Re, Rf, Rb1, Rc, Rh1, Rb2, and Rb3, were found to activate SIRT1 and promote energy metabolism. Conclusion: This study may provide new insights into the extensive application of ginseng for cardiac and neurological protection in healthy subjects and patients.

Differential effects of ginsenoside metabolites on slowly activating delayed rectifier K+ and KCNQ1 K+ channel currents

  • Channels formed by the co-assembly of the KCNQ1 subunit and the mink (KCNE1) subunit underline the slowly activating delayed rectifier $K^+$ channels ($I_{Ks}$) in the heart. This $K^+$ channel is one of the main pharmacological targets for the development of drugs against cardiovascular disease. Panax ginseng has been shown to exhibit beneficial cardiovascular effects. In a previous study, we showed that ginsenoside Rg3 activates human KCNQ1 $K^+$ channel currents through interactions with the K318 and V319 residues. However, little is known about the effects of ginsenoside metabolites on KCNQ1 $K^+$ alone or the KCNQ1 + KCNE1 $K^+$ ($I_{Ks}$) channels. In the present study, we examined the effect of protopanaxatriol (PPT) and compound K (CK) on KCNQ1 $K^+$ and $I_{Ks}$ channel activity expressed in Xenopus oocytes. PPT more strongly inhibited the $I_{Ks}$ channel currents than the currents of KCNQ1 $K^+$ alone in concentration- and voltage-dependent manners. The $IC_{50}$ values on $I_{Ks}$ and KCNQ1 alone currents for PPT were $5.18{\pm}0.13$ and $10.04{\pm}0.17{\mu}M$, respectively. PPT caused a leftward shift in the activation curve of $I_{Ks}$ channel activity, but minimally affected KCNQ1 alone. CK exhibited slight inhibition on $I_{Ks}$ and KCNQ1 alone $K^+$ channel currents. These results indicate that ginsenoside metabolites show limited effects on $I_{Ks}$ channel activity, depending on the structure of the ginsenoside metabolites.

Generation of a High-Growth Influenza Vaccine Strain in MDCK Cells for Vaccine Preparedness

  • As shown during the 2009 pandemic H1N1 (A(H1N1)pdm09) outbreak, egg-based influenza vaccine production technology is insufficient to meet global demands during an influenza pandemic. Therefore, there is a need to adapt cell culture-derived vaccine technology using suspended cell lines for more rapid and larger-scale vaccine production. In this study, we attempted to generate a high-growth influenza vaccine strain in MDCK cells using an A/Puerto/8/1934 (H1N1) vaccine seed strain. Following 48 serial passages with four rounds of virus plaque purification in MDCK cells, we were able to select several MDCK-adapted plaques that could grow over $10^8PFU/ml$. Genetic characterization revealed that these viruses mainly had amino acid substitutions in internal genes and exhibited higher polymerase activities. By using a series of Rg viruses, we demonstrated the essential residues of each gene and identified a set of high-growth strains in MDCK cells ($PB1_{D153N}$, $M1_{A137T}$, and $NS1_{N176S}$). In addition, we confirmed that in the context of the high-growth A/PR/8/34 backbone, A/California/7/2009 (H1N1), A/Perth/16/2009 (H3N2), and A/environment/Korea/deltaW150/2006 (H5N1) also showed significantly enhanced growth properties (more than $10^7PFU/ml$) in both attached- and suspended-MDCK cells compared with each representative virus and the original PR8 vaccine strain. Taken together, this study demonstrates the feasibility of a cell culture-derived approach to produce seed viruses for influenza vaccines that are cheap and can be grown promptly and vigorously as a substitute for egg-based vaccines. Thus, our results suggest that MDCK cell-based vaccine production is a feasible option for producing large-scale vaccines in case of pandemic outbreaks.

  • Objectives: Osteoporosis is characterized by bone loss and morbidity with osteoporotic fracture. In this study, the author aimed to evaluate the effect of dried roots of Rehmannia glutinosa extract (RGE) on osteoblast proliferation in murine calvarial cells. Methods: The osteoblast separated from murine calvariae was cultivated for 6 days and evaluated the cell function. After the addition of RGE on the culture medium, we determined the effect of RGE on the cell viability, cell proliferation, protein synthesis, alkaline phosphatase activity, collagen synthesis and calcified nodule formation of the cultivated osteoblast. Results: The results were summarized as follows. 1. RGE did not change the survival rate of rat calvarial osteoblast. 2. RGE increased the proliferation of rat calvarial osteoblast. 3. RGE increased ALP activity of rat calvarial osteoblast., 4. RGE slightly affected protein synthesis of rat calvarial osteoblast. 5. RGE increased collagen synthesis of rat calvarial osteoblast. 6. RGE slightly affected calcified nodule formation of rat calvarial osteoblast. Conclusions: From these results, it is concluded that RG might improve the osteoporosis resulted from augmentation of osteoblast proliferation.

Rhodotorula glutinis 유래의 고효율 재조합 Epoxide Hydrolase를 이용한 라세믹 Styrene Oxide의 비대칭 광학분할 (Asymmetric resolution of racemic styrene oxide using recombinant Escherichia coli harboring epoxide hydrolase of Rhodotorula glutinis)

  • Rhdotorula glutinis epoxide hydrolase 유전자를 pColdI 벡터 와 pET-21b(+) 벡터에 재조합하여 제작한 E. coli를 생촉매로 사용하여 라세믹 styrene oxide에 대하여 회분식 가수분해 반응을 실시하였다. pET-21b(+)/RgEH 재조합 플라스미드 DNA를 가진 E. coli를 $15^{\circ}C$에서 저온 배양할 때 수용성 단백질 형태로 가장 많이 발현되었고, 입체선택적 가수분해 활성과 촉매 안정성이 가장 좋았다. 라세믹 styrene oxide 20 mM에 대하여 반응온도 $30^{\circ}C$에서는 반응시간 20분 동안에 수율 24.0%로 (S)-styrene oxide를 얻은 반면에, 반응온도를 $10^{\circ}C$로 낮추고 0.5% (w/v) Tween 20을 첨가하고 반응시키면 광학순도 99.0% ee 이상의 (S)-styrene oxide을 46.0%의 수율로 얻을 수 있었다. 최적조건에서 E 값은 6.68이었으며, 100 mM의 라세믹 styrene oxide에 대해서는 반응시간 50분에 이론 수율 50% 대비 40%의 높은 수율로 (S)-styrene oxide를 얻을 수 있었다.

  • 엑세스 기술의 발달로 사용자는 점점 더 다양한 엑세스 네트워크를 선택할 수 있게 되고 있다. 엑세스 기술들은 서로 다른 특징을 가지고 있으므로 사용자들의 상황에 따른 적시 핸드오버에 대한 요구는 점점 커지고 있다. 이러한 사용자의 요구에 따라 고속 및 이종망간 핸드오버를 위한 연구가 활발히 진행되고 있으며, 기존의 1, 2 계층과 3, 4 계층을 분리하여 핸드오버를 진행할 때의 한계를 극복하기 위하여 IEEE 802.21 WG 이나 IRTF의 MobOpts (IP Mobility Optimizations) 연구그룹에서는 여러 계층이 통신하고 협력하여 핸드오버를 할 수 있도록 이벤트 기반 통신을 통한 핸드오버에 대한 연구가 진행되고 있다. 본 논문에서는 고속의 핸드오버를 위하여 요구되는 L2 계층의 메시지의 종류와 속성에 대해서 제안한다.

직파 4 ~ 6년생 인삼의 연근 및 직경에 따른 Ginsenoside 함량 비교 (Comparison of Ginsenoside Content According to Age and Diameter in Panax ginseng C. A. Meyer Cultivated by Direct Seeding)

  • This study was carried out to investigate ginsenoside content in different root parts and the correlation between root diameter and ginsenoside composition of Panax ginseng C. A. Meyer cultivated by direct seeding. The unit contents of ginsenoside were 29.65, 28.76, 26.34 mg/g, respectively in 4, 5, 6 years old. However, the total contents of ginsenoside were 431.97, 606.56, 657.80 mg/root, respectively. Total ginsenoside content of fine root was higher than that of main root and lateral root. These tendencies were related to decrease by the increase of root diameter. When diameter of main root and lateral root were the same in different ages, the total ginsenoside content was higher in the order of 4 > 5 > 6 years old roots. Except for ginsenoside-Rg1, other ginsenosides components (PD/PT and total ginsenosides) had highly negative correlation with the root diameter within whole root, main root, lateral root and fine root, which indicated that ginsenoside content is correlated to root diameter. As results, it is suggested that ginsenoside content can be predicted.

Sprague-Dawley 랫드를 이용한 Up & Down 법 (UDP)에 의한 LMK02의 단회 경구투여 독성시험 (Up-and-Down Procedure(UDP) Determinations of Acute Oral Toxicity of LMK02-Jangwonhwan in SD Rats)

  • Objectives : The oriental medicine Jangwonhwan originally described in the Korean medical text, DonguiBogam(amnesia chapter). Recently, a modified formula of Jangwonhwan(LMK02-Jangwonhwan), was shown to reduce $\beta$-amyloid deposition in the brain of Tg-APPswe/PS1dE9 mouse model for Alzheimer's disease. This experiment aimed to investigate the acute oral toxicity of LMK02 in SD rats by up-and-down procedure determinations. Methods : Quality control of tablet form of LMK02 was established by estimating indicative components, Ginsenoside Rg3 of Red Ginseng and Decursin of Angelicagigas Nakai. The toxicity of LMK02 was investigated in 6 week old, specific pathogen free(SPF), Sprageu-Dawley rats. 3 female rats received 5,000 mg/10 ml/kg of test substance and their death rate, clinical sings, weight changes and autopsy findings had been observed for 2 weeks. Results : Any specific symptoms or death were resulted in this experiment. No significant changes in rats' weight. No significant differences in atopsy. Conclusions : The minimum lethal dose(MLD) of LMK02 for female Sprauge-Dawley rats were more than 5,000mg/kg in this experiment.

Simultaneous quantification of six nonpolar ginsenosides in white ginseng by reverse-phase high-performance liquid chromatography coupled with integrated pulsed amperometric detection

  • Background: White ginseng consists of the roots and rhizomes of the Panax species, and red ginseng is made by steaming and drying white ginseng. While red ginseng has both polar and nonpolar ginsenosides, previous studies showed white ginseng to have only polar ginsenosides. Because nonpolar ginsenosides are formed through the manufacture of red ginseng from white ginseng, researchers have generally thought that nonpolar ginsenosides do not exist in white ginseng. Methods: We developed a simultaneous quantitative method for six nonpolar ginsenosides in white ginseng using reverse-phase high-performance liquid chromatography coupled with integrated pulsed amperometric detection. The nonpolar ginsenosides of white ginseng were extracted for 4 h under reflux with 50% methanol. Results: Using the gradient elution system, all target components were completely separated within 50 min. Nonpolar ginsenosides were determined in the rhizome head (RH), main root (MR), lateral root, and hairy root (HR) of 6-year-old white ginseng samples obtained from several regions (Geumsan, Punggi, and Kanghwa). The total content in the HR of white ginseng was 37.8-56.8% of that in the HR of red ginseng. The total content in the MR of white ginseng was 5.9-24.3% of that in the MR of red ginseng. In addition, the total content in the RH of white ginseng was 28.5-35.8% of that in the HR of red ginseng Conclusion: It was confirmed that nonpolar ginsenosides known to be specific components of red ginseng were present at substantial concentrations in the HR or RH of white ginseng.
