Mitochondrial calcium uniporter inhibition attenuates mouse bone marrow-derived mast cell degranulation induced by beta-1,3-glucan

  • Mast cells are primary mediators of allergic inflammation. Beta-1,3-glucan (BG) protects against infection and shock by activating immune cells. Activation of the BG receptor induces an increase in intracellular $Ca^{2+}$, which may induce exocytosis. However, little is known about the precise mechanisms underlying BG activation of immune cells and the possible role of mitochondria in this process. The present study examined whether BG induced mast cell degranulation, and evaluated the role of calcium transients during mast cell activation. Our investigation focused on the role of the mitochondrial calcium uniporter (MCU) in BG-induced degranulation. Black mouse (C57) bone marrow-derived mast cells were stimulated with $0.5{\mu}g/ml$ BG, $100{\mu}g/ml$ peptidoglycan (PGN), or $10{\mu}M$ A23187 (calcium ionophore), and dynamic changes in cytosolic and mitochondrial calcium and membrane potential were monitored. BG-induced mast cell degranulation occurred in a time-dependent manner, and was significantly reduced under calcium-free conditions. Ruthenium red, a mitochondrial $Ca^{2+}$ uniporter blocker, significantly reduced mast cell degranulation induced by BG, PGN, and A23187. These results suggest that the mitochondrial $Ca^{2+}$ uniporter has an important regulatory role in BG-induced mast cell degranulation.

보존처리 후 철제유물에 생성된 부식물 특성 연구 (Study on Characteristics of Corrosion Products Generated in Iron Artifacts after Conservation Treatments)

  • 철제유물은 출토되는 순간 급격한 환경변화로 인해 빠른 속도로 부식이 진행되므로 보존처리 과정을 거쳐 부식을 억제한다. 그러나 보존처리가 완료된 철제유물도 재부식이 발생하는 경우가 다수이며, 재부식된 유물의 보존처리는 1차 보존처리 시보다 그 처리가 어렵고 처리기간 또한 길어진다. 본 연구는 보존처리가 완료된 이후 보관과정에서 발생하는 부식생성물을 과학적으로 분석하여 재부식의 요인을 찾고자 하였다. 경주지역의 세 유적에서 출토된 철제유물을 동일한 약품과 방법으로 2002~2009년 동안 보존처리를 완료하였으나, 포장 보관 상태의 일부 철제유물에서 재부식 징후가 관찰되었다. 이중 재부식의 징후가 확인된 단조 철제유물 9점을 선별하여 질량측정, 육안관찰, 현미경을 통한 물리적 변화를 관찰하였고, SEM-EDS, XRD, IC, ICP분석을 통해 화학적 변화를 분석하였다. 그 결과, 유물에서 탈락된 부분의 접면에 형성된 황갈색 부식생성물은 군집한 형상만 다를 뿐 결정상은 공통적으로 침상형이 확인되었으며, 적색에서 황갈색으로 갈수록 침상의 형태가 뚜렷하였다. 보존처리가 완료된 시점의 경과에 따라 부식생성물이 증가할수록 유물의 질량이 증가하였고 염화이온의 농도가 상대적으로 높아지는 경향이 나타났다. 채집된 모든 시료의 부식생성물에 대한 XRD분석에서는 ${\beta}$-FeOOH(akaganeite)이 확인되었고, ICP분석 결과 $Na^+$, $Ca^{2+}$성분을 확인할 수 있었다.

톳 분획물이 3T3-L1 지방전구세포의 분화 및 지방생성의 억제에 미치는 영향 (Effects of Hizikia fusiforme Extracts on Adipocyte Differentiation and Adipogenesis in 3T3-L1 Preadipocytes)

  • 본 연구에서는 톳 분획물의 항비만 효과 및 그에 따른 생화학적 기전의 해석을 위하여 톳 분획물이 비만유도인자에 의하여 인위적으로 유발된 adipogenesis 과정에 있어서 어떠한 영향을 미치는 지를 조사하였고, 이때 $PPAR{\gamma}$, C/$EBP{\alpha}$ 및 C/$EBP{\beta}$ 등과 같은 adipogenic transcription factor들의 발현에 어떠한 변화가 유발되었는지를 조사하였다. 각각의 톳 분획물들이 성숙한 지방세포에서 나타나는 lipid droplet 및 TG 생성에 어떠한 영향을 미치는 지를 확인한 결과, 모든 분획물에서 lipid droplet 및 TG 생성억제가 나타났지만 특히 WFHF 처리군에서 이러한 현상이 가장 강하게 나타났다. 또한 lipid droplet 및 TG 생성에 중요한 역할을 하는 것으로 알려진 adipogenic transcription factor들의발현에각각의분획물들이 어떠한영향을미치는지를확인한결과,WFHF 처리군에서 $PPAR{\gamma}$, C/$EBP{\alpha}$ 및 C/$EBP{\beta}$의 발현이 현저하게 감소하였음을 확인하였다. 이상의 결과를 종합해 보면 다섯 종류의 톳 분획물 모두 비만억제 효과가 있는 것으로 나타났고 특히 WFHF의 비만억제 효과가 강하게 나타났음을 알 수 있었다. 본 연구 결과는 톳의 비만억제 가능성을 제시하는 것으로서 항비만 기전에 대한 생화학적 해석 및 이를 활용한 향후 지속적인 연구를 위한 자료로서 그 가치가 매우 높을 것으로 생각된다.

Studies on the Mode of Uptake of Plasma Glucose, Acetate, β- hydroxybutyrate Triglyceride Fatty Acids and Glycerol by the Mammary Gland of Crossbred Holstein Cattle Feeding on Different Types of Roughage

  • The present experiment was carried out to study the utilization of substrates in the mammary gland of crossbred Holstein Friesian during feeding on different types of roughage. Sixteen pregnant crossbred Holstein heifers consisted of two breed types of eight animals each; Holstein Friesian${\times}$Red Sindhi (50:50=50%HF) and Holstein Friesian${\times}$Red Sindhi (87.5:12.5=87.5%HF). Animals were divided into four groups of the same breed type in each group which were fed with either rice straw treated with 5% urea or pangola hay (Digitaria decumbens) as the source of roughage throughout the experiments. Four consecutive experimental periods were carried out in late pregnancy (20-23 days before parturition), early lactation (30 days postpartum), mid-lactation (120 days postpartum) and late lactation (210 days postpartum). Measurement of mammary blood flow in combining with measurement of AV difference was performed for the mammary uptake of substrates. In the period of lactation, udder blood flow was nearly three times higher than that of late pregnant period (p<0.05) in both 50%HF and 87.5%HF feeding on either hay or urea treated rice straw. During mid- and late lactation of both groups of 87.5%HF animals, mammary blood flow and milk yield showed decrease when compared to those during the early lactating period while the trends for persistency were apparent in both groups of 50%HF animals throughout experimental periods. The mean arterial plasma concentrations of glucose, acetate, $\beta$-hydroxybutyrate and free glycerol in each group remained constant throughout experimental periods. During late pregnancy in all groups, the AV difference and extraction ratio of glucose, $\beta$-hydroxybutyrate and triacylglycerol across the mammary gland markedly lowered (p<0.05), which coincided with a lower net uptake by the mammary gland in comparison to the early lactating period. The mean arterial plasma concentration, AV difference and extraction ratio for acetate showed no significant differences between late pregnancy and the early lactating period. The AV difference of free glycerol showed apparent release from mammary tissue during late pregnancy in all groups. In mid- and late lactation, the mammary uptake for glucose, acetate and $\beta$-hydroxybutyrate in both groups of 87.5%HF animals showed apparent decrease as compared to that in the early lactating period, whereas no appearances were observed in 50%HF animals feeding either hay or urea treated rice straw. The mean arterial plasma concentrations for free fatty acid (FFA) and triacylglycerol (C16 to C18) were higher in late pregnancy than in early lactation in both types of crossbred animals. The values of AV difference and the net uptake by the mammary gland for FFA were variable during late pregnancy and lactating periods in all groups. There were no significant differences for AV difference, extraction ratio and net uptake of triacylglycerol during lactation advance in both groups of 50%HF and 87.5%HF animals feeding either hay or urea treated rice straw. These results suggest that the adaptations to either hay or urea treated rice straw by the mammary gland of crossbred HF animals allow for an adequate nutrient supply during pregnancy and lactation. There is no difference in the mode of mammary uptake of substrates in the same crossbred animals in response to feeding hay or urea treated rice straw. The differences in utilizing nutrients by the mammary gland for milk production between 87.5%HF and 50%HF animals would be dependent on changes in both intra-mammary factors and extra-mammary factors.

cDNA microarray에 의한 치주인대세포의 광물화 결절형성에 관여하는 유전자들의 분석 (Identification of Matrix Mineralization-Related Genes in Human Periodontal Ligament Cells Using cDNA Microarray)

  • 치주인대세포는 시험관적 실험에서 광물화 결절형성을 유도할 수 있으므로 광물화 결절형성에 관여하는 유전자들을 특이하게 발현할 것으로 여겨진다. 이에 본 실험은 cDNA microarray를 이용한 동시 유전자분석을 시행하여 치주인대세포의 분화에 의한 광물화 결절형성시 나타나는 유전자의 특징적 발현 양상을 알아보고자 하였다. 교정치료를 목적으로 경북대학교병원에 내원한 환자의 제일소구치를 발치하여 통상적 방법으로 치주인대세포를 분리, 배양하였고, 3세대의 치주인대세포를 사용하여 실험을 시행하였다. 치주인대세포를 100mm 배양접시에 넣고 배양하여 매 2일 마다 배지를 교환해 주고, 10% FBS만을 투여한 대조군으로, ascorbic acid $(50\;{\mu}g/ml)$, ${\beta}-glycerophosphate$ (10 mM) 및 100 nM dexamethasone을 투여한 군을 실험군으로 하였다. 배양된 치주인대세포에 ascorbic acid, ${\beta}-glycerophosphate$, 그리고 dexamethasone을 투여한 실험군에서 21일째 광물화된 결정을 관찰할 수 있었으나 대조군에서는 관찰할 수 없었다. 3063개의 유전자를 분석한 결과 35개 유전자가 대조군에 비해 2배이상 발현이 증가하였고, 38개 유전자는 2배이상 발현이 감소하였다. 형태학적 검사에서 보여준 바와 같이 광물화 형성과정시 관여하는 JGF-2과 IGFBP2와 같은 유전자가 실험군에서 증가하였으며, 세포골격과 세포외기질 형성에 관여하는 proteogycan 1, fibulin-5, keratin 5, ${\beta}-actin$, ${\alpha}-smooth$ muscle actin, capping protein 등도 발현이 실험군에서 증가하였다. 한편 periostin and S100 calcium-binding protein A4는 대조군에서 오히려 높게 나타나므로 이는 배양된 치주인대세포가 그 자체의 표현형을 유지하고 있음을 보여 주고 있다. 그 외 apoptosis를 유발시키는데 관여하는 Dkk-1와 Nip3는 실험군에서 높게 발현되었고, apoptosis를 억제시키는데 관여하는 Btf와 TAX1BP1는 오히려 낮게 발현됨을 알 수 있으므로 이는 실험군에서 치주인대세포가 골아세포로의 분화되었음을 나타낸다.

Comparative study on the cellular activities of osteoblast-like cells and new bone formation of anorganic bone mineral coated with tetra-cell adhesion molecules and synthetic cell binding peptide

  • Purpose: We have previously reported that tetra-cell adhesion molecule (T-CAM) markedly enhanced the differentiation of osteoblast-like cells grown on anorganic bone mineral (ABM). T-CAM comprises recombinant peptides containing the Arg- Gly-Asp (RGD) sequence in the tenth type III domain, Pro-His-Ser-Arg-Asn (PHSRN) sequence in the ninth type III domain of fibronectin (FN), and the Glu-Pro-Asp-Ilu-Met (EPDIM) and Tyr-His (YH) sequence in the fourth fas-1 domain of ${\beta}$ig-h3. Therefore, the purpose of this study was to evaluate the cellular activity of osteoblast-like cells and the new bone formation on ABM coated with T-CAM, while comparing the results with those of synthetic cell binding peptide (PepGen P-15). Methods: To analyze the cell viability, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay was performed, andto analyze gene expression, northernblot was performed. Mineral nodule formations were evaluated using alizarin red stain. The new bone formations of each group were evaluated using histologic observation and histomorphometrc analysis. Results: Expression of alkaline phosphatase mRNA was similar in all groups on days 10 and 20. The highest expression of osteopontin mRNA was observed in the group cultured with ABM/P-15, followed by those with ABM/T-CAM and ABM on days 20 and 30. Little difference was seen in the level of expression of collagen type I mRNA on the ABM, ABM/T-CAM, and ABM/P-15 cultured on day 20. There were similar growth and proliferation patterns for the ABM/T-CAM and ABM/P-15. The halo of red stain consistent with $Ca^{2+}$ deposition was wider and denser around ABM/T-CAM and ABM/P-15 particles than around the ABM particles. The ABM/T-CAM group seemed to have bone forming bioactivity similar to that of ABM/P-15. A complete bony bridge was seen in two thirds of the defects in the ABM/T-CAM and ABM/P-15 groups. Conclusions: ABM/T-CAM, which seemed to have bone forming bioactivity similar to ABM/P-15, was considered to serve as effective tissue-engineered bone graft material.

고추장 제조시 발효가 항돌연변이 및 in vitro 항암효과에 미치는 영향 (Fermentation process increased antimutagenic and in vitro anticancer effects during Kochujang manufacturing)

  • 본 연구에서 고추장 제조시 발효가 항돌연변이 효과와 in vitro 항암효과에 미치는 영향을 확인하기 위하여 고추장 제조 재료인 원료 밀, 1차발효밀, 2차발효밀, 최종발효밀, 고춧가루와 최종발효밀로 제조된 고추장의 Sal. Typhimurium TA100을 이용한 Ames test를 실시하여 MNNG에 대한 항돌연변이 효과를 살펴보았으며, HT-29 인체 대장암세포와 AGS 인체 위암세포의 성장 억제효과를 살펴보았다. 실험한 결과 원료 밀보다 1차발효밀, 2차발효밀보다 최종발효밀이 발효진행에 따라 점차 증진된 항돌연변이 효과와 in vitro 항암효과가 나타났으며 최종발효밀과 고춧가루로 제조된 고추장이 가장 높았다. 15, 30일 발효시킨 고추장이 발효되지 않은 고추장보다 더 높은 항암효과가 나타났다. 따라서 발효과정(기간)이 고추장의 항돌연변이 효과와 항암효과에 중요하게 관련되어 있다고 할 수 있으며 고추장의 주재료인 고춧가루의 capsaicin, 비타민C와 ${\beta}$-carotene 등의 작용만이 아닌 밀에서 온 발효 숙성 중에 많이 증식된 미생물 그리고 발효에 의한 생성된 다른 활성물질이 항돌연변이와 항암 효과에 관련되어 있다고 하겠다.

부자(附子)와 오가피(五加皮) 물 추출물의 골수유래 지방세포와 파골세포 분화 억제 효과 (Inhibitory Effect of Water Extracts of Aconiti Lateralis Preparata Radix and Acanthopanacis Cortex on Differentiation of Bone Marrow-Derived Adipocytes and Osteoclasts)

  • Objectives : The aim of this study was to evaluate the efficacy of Aconiti Lateralis Preparata Radix (AP) and Acanthopanacis Cortex (AT) extracts in bone-derived adipocyte OP9 cell, osteoclast and osteoblast-like MG63 cells. Methods : MTT assay was used to evaluate the cytotoxicity of AP and AT extracts on OP9, osteoclast and MG63 cells. OP9 cells were treated with AP and AT, and the alterations in fat storage in the cells were determined by the Oil red O. To explain effects of RANKL-induced osteoclast differentiation in bone marrow macrophages, we performed the TRAP staining. The protein level of CAAAT/enhancer binding protein alpha ($C/EBP{\alpha}$) and peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$) as a adipocyte differentiation marker, and adiponectin was examined using western blot in differentiated OP9 cells. Effects of related genes were confirmed by luciferase assay using reporter assay. Results : AP and AT was not toxic on OP9 and MG63 cells, but AT was a little cytotoxic to osteoclast at the dose of $100{\mu}g/m{\ell}$. They could inhibit differentiation of OP9 cells and osteoclast with results of oil red O staining and TRAP staining. By western blot, AP and AT decreased the expression of $PPAR{\gamma}$ and $C/EBP{\alpha}$ which is the key transcription factor in adipogenesis and adiponectin secretion. AT also inhibited the BMP-4 activity in luciferase assay. AP also inhibited BMP-4 and Wnt3a activity, stimulated ER-${\beta}$ activity but inhibited androgen receptor activity. Conclusions : These results show AP and AT can be useful in osteoporosis and obesity via inhibition of osteoclast and adipocyte differentiation.

2-Bromopropane의 수컷 랫트를 이용한 고환 독성시험 (Testicular Toxicity of 2-Bromopropane in Male Sprague-Dawley Rats)

  • 택트 스위치 제조공정 침지액의 주성분인 2-bromopropane독성에 대한 연구로 최단기의 폭로로 농도를 달리하여 3주간 반복 투여 시험을 시행하여, 흰쥐의 혈액 및 세정관의 변화를 관찰하기 위해 투여기간 동안의 체중의 변화, 고환, 간, 신장 등의 장기무게, 혈액화학과 혈액학적 분석 및 고환의 병리조직의 변화 등을 관찰하여 2-bromopropane의 급성투여 조건에서의 중독현상을 비교.분석하였다. 농도를 달리하여 투여에 따른 체중변동은 통계적으로 유의한(P<0.05)체중의 감소를 나타내었다. 1.000 mg/kg 투여군에서 백혈구수, 적혈구수, 혈구용적과 혈색소 농도에서 유의한 변화(p<0.05)를 보였다. 조직병리학적 소견으로 정 소에서는 세정관의 정조세포와 정모세포의 괴사를 볼 수 있었고, 기저막의 비후, 세정관의 Sertoli세포는 광범위하게 세포질성 공포현상을 보여주고 있다. 또한 간질조직에서는 Leydig 세포의 증식을 볼 수 있었다. 2-bromopropane의 손상부위는 조혈과 생식계가 표적으로 생각되며, 고농도 투여가 저농도 투여에 비해 독성이 심하며 독성물질의 양-반응 관계를 보여주고 있다.

Plasma Levels of Hormones and Metabolites as Affected by the Forages Type in Two Different Types of Crossbred Holstein Cattle

  • An experiment was carried out to study plasma levels of hormones and metabolites of crossbred Holstein cattle during late pregnancy (28 days pre partum), early lactation (30 days post partum), mid-lactation (120 days post partum) and late lactation (210 days post partum). Two breed types of Holstein $Friesian{\times}Red$ Sindhi (50:50 = 50%HF) and Holstein $Friesian{\times}Red$ Sindhi (87.5:12.5 = 87.5%HF) were divided into four groups of four animals each. Two groups of each breed were fed with either rice straw treated with 5% urea or pangola hay (Digitaria decumbens) as the source of roughage throughout the experiments. There were a substantial increases in the mean levels of total triiodothyronine ($T_3$), insulin and glucagon at the onset of lactation, and maintained in a high levels during lactation advance for all groups of experiments. The mean levels of prolactin and thyroxine ($T_4$) were not significantly different among groups of animals, but the plasma cortisol concentration was slightly higher in both groups of 50%HF in comparison with those of 87.5%HF animals. The mean levels of plasma growth hormone (GH) of both groups of 87.5%HF animals feeding on either hay or urea treated rice straw markedly rose in the early period of lactation and markedly reduced in mid- and late lactation. These changes were accompanied with changes of milk yield. In contrast to 50%HF animals, plasma GH levels were considerably higher in the late pregnant period than in the early period of lactation and it remained constant as its value at the early lactation throughout the experimental period. The high levels of both plasma progesterone and estradiol concentration significantly declined after parturition and remained low through lactating period. The plasma glucose level in the 50%HF animals feeding on either hay or urea treated rice straw was higher than the 87.5%HF animals in all periods of experiments. Changes in plasma FFA levels of both types of crossbred animals were depended on the endocrine status during late pregnancy and lactation. The levels of plasma FFA of 50%HF animals were significantly higher (p<0.05) than those of 87.5%HF animals during late pregnancy. Both plasma ${\beta}$-hydroxybutyrate and lactate concentrations were not affected by feeding on either hay or urea treated rice straw during late pregnancy and lactation. These data demonstrate that there were no differences in the physiological performances in the same crossbred animals fed either hay or urea treated rice straw. The 87.5%HF animal has the genetic potential for a high milk yield and homeorhetic adaptation for mammary function differed from 50%HF animals during periods of lactation. Altering lactation persistency in 87.5%HF is regulated mainly by chronically acting growth hormones through the period of lactation.
