대식세포에서 LPS로 유도된 염증에 대한 회향 열매의 항염 효과 (Anti-inflammatory Effects of the Fruits of Foeniculum vulgare in Lipopolysaccharide-stimulated Macrophages)

  • 회향(Foeniculum vulgare) 열매의 메탄올 추출물과 분획물(hexane, methylene chloride, ethylacetate, n-butanol)을 cell culture model system을 이용하여 항염증 활성을 평가하였다. 이를 위해, LPS처리로 염증이 유도된 RAW 264.7 macrophage에서 시료가 세포독성을 나타나지 않는 $100{\mu}g/ml$의 농도를 처리한 후 염증매개물질인 NO, PGE2, 염증성 사이토카인($IL-1{\beta}$, IL-6 및 TNF-${\alpha}$) 생성 및 mRNA 발현을 측정하였다. 그 결과 hexane, methylene chloride 및 ethylacetate 분획물은 대조군에 비해 NO, PGE2 생성을 억제하였으며, 이는 iNOS, COX-2 mRNA 발현 저해에서 기인함을 확인하였다. Methylene chloride와 ethylacetate 분획물은 $100{\mu}g/ml$의 농도에서 $IL-1{\beta}$, IL-6 생성 및 mRNA 발현을 효과적으로 억제하였으며, 특히 ethyl acetate 분획물은 TNF-${\alpha}$ 의 생성과 mRNA발현을 유의적으로 저해하였다. 본 연구 결과를 통해 회향 및 회향 분획물의 항염증 활성을 확인하였으며, 이는 염증 매개물질인 NO, PGE2 그리고 염증성 사이토카인 mRNA 활성 및 분비 억제에 의한 것으로 나타났다.

  • 목 적: 본 연구를 통해 $TGF-{\beta}1$에 의해 유도된 인간자궁내막의 탈락막화 과정에서 ERK와 $PPAR{\gamma}$의 역할을 규명하고자 하였다. 연구방법: 자궁내막 기질세포는 DMEM/F12 (10% FBS, 1 nM E2 and 100 nM P4) 조건에서 배양하였다. 연구 목적에 따라 $TGF-{\beta}1$ (5 ng/ml), Rosiglitazone (50 nM)와 PD98059 ($20{\mu}M$)를 배양액에 첨가하였다. Trypan-Blue와 hematocytometer를 이용하여 현미경하에서 세포의 개수를 측정하였다. Enzyme-linked immunosorbent assay (ELISA)와 western blotting 방법을 사용하여 단백질의 발현 정도를 관찰하였다. 결과 및 결론: 배양액에 $TGF-{\beta}1$을 첨가하여 세포의 증식 정도를 측정한 결과 $TGF-{\beta}1$이 세포의 증식을 억제하는 것을 알 수 있었다. 또한 배양된 세포로부터 PGE2 및 prolactin의 발현을 유도하는 것을 알 수 있었다. 이러한 $TGF-{\beta}1$의 작용은 Smad 및 ERK의 활성화를 통하여 일어남을 알 수 있었다. $PPAR{\gamma}$의 기질인 rosiglitazone을 배양액에 첨가한 결과 $TGF-{\beta}1$에 의한 세포 증식의 억제가 역전되는 것을 알 수 있었다. 뿐만 아니라, 세포 내 ERK의 활성 역시 억제 시켰으며 이 결과 PGE2와 prolactin의 발현이 억제 되는 것을 관찰할 수 있었다. 따라서 본 연구를 통해 $TGF-{\beta}1$에 의한 자궁내막 기질세포의 탈락막화는 Smad와 ERK의 활성화를 통하여 이루어지며 이러한 과정은 $PPAR{\gamma}$에 의해 억제됨을 알 수 있었다.

Analgesic and anti-inflammatory effects of galangin: a potential pathway to inhibit transient receptor potential vanilloid 1 receptor activation

  • Background: Galangin, commonly employed in traditional Chinese medicine for its diverse medicinal properties, exhibits potential in treating inflammatory pain. Nevertheless, its mechanism of action remains unclear. Methods: Mice were randomly divided into 4 groups for 7 days: a normal control group, a galangin-treated (25 and 50 mg/kg), and a positive control celecoxib (20 mg/kg). Analgesic and anti-inflammatory effects were evaluated using a hot plate test, acetic acid-induced writhing test, acetic acid-induced vascular permeability test, formalin-induced paw licking test, and carrageenan-induced paw swelling test. The interplay between galangin, transient receptor potential vanilloid 1 (TRPV1), NF-κB, COX-2, and TNF-α proteins was evaluated via molecular docking. COX-2, PGE2, IL-1β, IL-6, and TNF-α levels in serum were measured using ELISA after capsaicin administration (200 nmol/L). TRPV1 expression in the dorsal root ganglion was analyzed by Western blot. The quantities of substance P (SP) and calcitonin gene-related peptide (CGRP) were assessed using qPCR. Results: Galangin reduced hot plate-induced licking latency, acetic acid-induced contortions, carrageenan-triggered foot inflammation, and capillary permeability in mice. It exhibited favorable affinity towards TRPV1, NF-κB, COX-2, and TNF-α, resulting in decreased levels of COX-2, PGE2, IL-1β, IL-6, and TNF-α in serum following capsaicin stimulation. Galangin effectively suppressed the upregulation of TRPV1 protein and associated receptor neuropeptides CGRP and SP mRNA, while concurrently inhibiting the expression of NF-κB, TNF-α, COX-2, and PGE2 mRNA. Conclusions: Galangin exerts its anti-inflammatory pain effects by inhibiting TRPV1 activation and regulating COX-2, NF-κB/TNF-α expression, providing evidence for the use of galangin in the management of inflammatory pain.

파슬리추출물의 피부 노화 방지와 자극 완화에 대한 효과 (Effects of Parsley Extract on Skin Anti-aging and Anti-irritation)

  • 파슬리추출물이 피부에 미치는 개선효과를 조사하기 위하여, 배양 인체 섬유아세포에서 total Collagen, type I procollagen을 각질형성세포주인 HaCaT 세포에서 prostaglandin E$_2$(PGE$_2$), interleukin l$\alpha$ (IL-l$\alpha$)와 tumor necrosis factor $\alpha$ (TNF$\alpha$)를 무모생쥐(Female albino hairless mice, Skh:hr-1)에서는 진피의 두께와 밀도를 측정하였다. 그 결과, 1 $\mu\textrm{g}$/mL 농도의 파슬리 추출물은 total collagen은 23%, type I procollagen은 18% 증가시켰고, 자외선 B에 의한 PGE$_2$의 생합성은 약 60% 정도 감소시켰다. 10uM RA, 100 $\mu\textrm{g}$/mL SLS와 자외선 B 30 mJ/$\textrm{cm}^2$로 조사했을 때, IL-1$\alpha$ 및 TNF $\alpha$의 생합성 역시 1 $\mu\textrm{g}$/mL 파슬리추출물 처리 시 감소되었다. 4일 동안 1% 파슬리추출물로 폐쇄첩포한 무모생쥐의 진피 두께는 대조군에 비해 약 1.5배 정도 두꺼워지고, 밀도도 훨씬 촘촘해졌다. 본 연구의 결과는 파슬리추출물이 피부에서 노화방지 효과 및 자극완화 효과가 있음을 시사하고 있다.

우슬(牛膝)의 콜라겐 유도 관절염 생쥐에 대한 개선 효과 (The therapeutic effect of Achyranthis Radix on the collagen-induced arthritis in mice)

  • Objectives : The present study was undertaken to determine whether the water extract of Achyranthis Radix, which is the roots of Achyranthes japonica (Achyranthis Radix, AR), is efficacious against collagen-induced arthritis (CIA) in DBA/1J mice. Methods : Mice were immunized with bovine type II collagen and orally treated with AR-W (50 and 100 mg/kg/bw) from days 21 to 42 after immunization. Arthritis was evaluated by arthritic score, histological examination of knee joint and serological markers such as TNF-${\alpha}$, $PGE_2$ and anti-type II collagen (C II)IgG. Results : The results showed that comparing with untreated CIA mice, treated with AR-W significantly suppressed the clinical score and joint tissue pathological damages, reduced the serum levels of TNF-${\alpha}$, $PGE_2$ and anti-C II IgG in CIA-mice. These results suggest that AR-W can effectively alleviate inflammatory response on CIA, and anti-inflammatoy of AR-W can be attributed, at least partially, to the inhibition of inflamamtory mediators, $PGE_2$ and TNF-${\alpha}$, in CIA. Conclusions : This study suggests that AR-W has a therapeutic potential in inflammatory joint diseases such as rheumatoid arthritis.

식이지방과 Dexamethasone 주입이 소장절제후 소장 점막의 증식에 미치는 영향 (Effects of Dietary Lipids and Dexamethasone on Mucosal Hyperplasia after 70% Jejunoileal Resection in Rats)

  • Dietary mehaden oil enhances mucosal hyperplasia that normally occurs after massive small bowel resection. In contrast, dexamethasone and aspirin inhibit the adaptation response. In order to gain insight on the mechanism of these effects, male Sprague-Dawley rats weighing approximately 150gram were randomly divided into two groups and fed diet containing either 15% safflower oil or 14% menhaden oil and 1% safflower oil. Ten days later they were subjected to 70% jejunoilear resection. Immediately after surgery each group was further divided into two groups and receive either vehicle or 125ug/kg/day dexamethasone subcutaneously. All animals were sacrificed seven days after the surgery, and the remaining intestine was removed and divided at the anastomotic site. Dexamethasone, which decreased gut hyperplasia in both dietary groups, decreased both serum IGF-I levels and ileral PGE2 synthesis. Menhaden oil enhanced gut hyperplasia, but did not increase IGF-I or IGF-II levels in serum. PGE2 synthesis was lower in the ileum of menhaden oil-fed rats compared to that of safflower oil-fed rats. The effects of menhaden oil on adaptation did not apper to be mediated either through IGFs or PGE2 synthesis. Other factors could have played a role in enhancing adaptation following menhaden oil feeding.

Inhibitory Effect of the Leaves of Rumex crispus L. on LPS-induced Nitric Oxide Production and the Expression of iNOS and COX-2 in Macrophages

  • In this study, the anti-inflammatory effects of ethanol extract of Rumex crispus L. and its fractions were investigated in RAW 264.7 macrophages. To evaluate the anti-inflammatory effects of extract, we studied nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), and tumor necrosis factor-alpha ($TNF-{\alpha}$) levels in RAW 264.7 cells. The ethanol extract of R. crispus L. significantly decreased NO production and the levels of other inflammatory factors, such as PGE2 and $TNF-{\alpha}$, in lipopolysaccharide (LPS)-stimulated macrophages in a dose-dependent manner. We also assessed the inducible nitric oxide synthase (iNOS) and the cyclooxygenase 2 (COX-2) protein expression by western blot. Ethyl acetate fraction of R. crispus L. had the strongest anti-inflammatory effect. These results suggest that ethyl acetate extract of R. crispus L. might be beneficial in the treatment of chronic inflammatory diseases.

Inhibitory Effects of Jojoongikgi-tang on the Secretion of PGE2 and NO and Cytokines in LPS-stimulated BV2 Microglial Cells

  • Objectives This experiment is about Jojoongikgi-tang(JIT) exerts anti-inflammatory effects in BV2 microglial cells, and the effect of JIT on Nitric oxide(NO) production in lipopolysaccharide(LPS)-stimulated BV2 microglial cells were also demonstrated. Methods To investigate the anti-inflammatory effects of JIT, NO production, expression level of iNOS mRNA, PGE2 synthesis, expression COX-2 mRNA, cell viability, $TNF-{\alpha}$ mRNA expression were examined. Results The expression level of inducible nitric oxide synthase(iNOS) was decreased by JIT, and the production of Prostaglandin E2(PGE2) and the expression of Cox-2 mRNA also were inhibited by JIT. Proinflammatory mediators, such as $TNF-{\alpha}$, $IL-1{\beta}$, IL-12, were inhibited by JIT in a dose-dependent manner. Conclusions JIT have anti-inflammatory effects in BV2 microglial cells and could be used in inflammatory disease.

에폭시 수지의 경화 거동에 미치는 반응성 희석제의 영향 (Effects of Reactive Diluents on the Curing Behavior of Epoxy Resin)

  • 에폭시 수지의 성형 가공성을 조절하기 위해 반응성 희석제를 사용했을 때의 경화특성 및 경화 후 유리 전이온도의 변화를 조사하였다. 반응성 희석제로 사용한 Butyl gilcidyl ether(BGE) 및 Phenyl glycidyl ether(PGE)함량의 증가에 따라 경화에 따른 발열량과 경화 후 유리 전이온도는 낮아지는 특성이 관찰되었다. 에폭시 수지는 BGE를 사용한 경우 PGE를 사용한 경우보다 더 낮은 유리 전이온도를 보였으며, 희석제의 첨가에 따른 유리 전이온도 변화는 가교점 사이의 분자량 증가 때문으로 해석하였다. 에폭시 수지의 경화 거동을 자촉매 반응에 대한 속도론식으로 해석한 결과 속도상수 $k_1$, $k_2$는 희석제의 양에 따라 감소하였다.

Antioxidant and Suppressive Effects of Ethanolic Extract Fractions from Safflower (Carthamus tinctorius L.) Flower on the Biosynthesis of Inflammatory Mediators from LPS-stimulated RAW 264.7 Cells

  • The aim of this study was to elucidate the anti-inflammatory activity of safflower (Carthamus tinctorius L.) ethanolic extract fractions (CFEFs). Butanol fraction had the strongest antioxidant activity, and all CFEFs, except for chloroform fraction, partly inhibited lipopolysaccharide (LPS)-induced nitrite production in RAW 264.7 cells. In the cell-free system, hexane and butanol fractions chemically quenched nitric oxide (NO). In addition, the iNOS mRNA transcription was suppressed by ethanol extract and hexane fraction in LPS-stimulated RAW 264.7 cells. Taken together, the inhibitory effect of CFEFs on NO production from LPS-stimulated RAW 264.7 cells, might be due to both the chemical NO quenching activity and the suppression of iNOS mRNA transcription partially. The synthesis of prostaglandin $E_2$ ($PGE_2$) was potently inhibited by ethanol extract to below basal label, and the transcription of cyclooxygenase-2 (COX-2), an enzyme involving in $PGE_2$ synthesis, was partially suppressed by ethanol extract and hexane fraction. Based on these results, CFEFs may be useful as an alternative medicine for the relief and retardation of immunological inflammatory responses through the reduction of inflammatory mediators, including NO and $PGE_2$ production.
