Chemical Components and Biological Activities of Red Onion Powder (붉은 양파 분말의 화학성분 및 생리활성)

  • We investigated the chemical components of red onion powder dried using the low temperature vacuum method and the inhibitory effects of solvent extracts of the dried red onion powder on the growth of HT-1080 human fibrosarcoma and HT-29 human colon cancer cells and $H_2O_2$-induced oxidative stress. The moisture content of the dried red onion powder was 17.95%, while the vitamin C content was 96 mg/100 g and the total phenols content was 39.1 mg/mL. The inhibitory effects of acetone with methylene chloride (A+M) and methanol (MeOH) extracts of the red onion powder on the growth of HT-1080 and HT-29 cancer cells increased in a dose dependent manner (p<0.05). The inhibitory effect was greater on the growth of HT-29 cells, while the A+M extracts had a higher inhibitory effect than the MeOH extracts. Treatment with the hexane, 85% aq. methanol, butanol and water fractions of the extract led to significant inhibition of the growth of both cancer cell lines (p<0.05). Among the fractions, the hexane and 85% aq. methanol fractions showed a greater inhibitory effect. To determine the protective effect on $H_2O_2$-induced oxidative stress, a DCFH-DA (dichlorodihydrofluorescin diacetate) assay was conducted. All fractions, including the crude extracts of dried red onion, appeared to lead to a significant reduction in the levels of intracellular reactive oxygen species (ROS), and these reductions occurred in a dose dependent fashion (p<0.05). Among the fractions, the 85% methanol fraction showed the greatest protective effect on the production of lipid peroxides.

Screening of ${\alpha}$-Glucosidase Inhibitory Activity of Vietnamese Medicinal Plants : Isolation of Active Principles from Oroxylum indicum

  • Among 38 Vietnamese medicinal plant extracts investigated for their ${\alpha}$-glucosidase inhibitory activity, 35 extracts showed $IC_{50}$ values below $250{\mu}g/mL$. The MeOH extracts of the heartwood of Oroxylum indicum, the seeds of Caesalpinia sappan, and the fruits of Xanthium strumarium exhibited strong ${\alpha}$-glucosidase inhibitory activity with $IC_{50}$ values less than $50{\mu}g/mL$. Fractionation of the MeOH extract of the heartwood of O. indicum led to the isolation of oroxylin A (1), oroxyloside (2), hispidulin (3), apigenin (4), ficusal (5), balanophonin (6), 2- (1-hydroxymethylethyl)-4H,9H-naphtho[2,3-b]furan-4,9-dione (7), salicylic acid (8), p-hydroxybenzoic acid (9), protocatechuic acid (10), isovanillin (11), and ${\beta}$-hydroxypropiovanillon (12). Compounds 1 - 3, 5, 6, 8, 10, and 12 showed more potent activities, with $IC_{50}$ values ranging from 2.13 to $133.51{\mu}M$, than a positive control acabose ($IC_{50}$, $241.85{\mu}M$). The kinetic study indicated that oroxyloside (2) displayed mixed-type inhibition with inhibition constant (Ki) was $3.56{\mu}M$.

  • A crude alkaloidal fraction from white ginseng (Panax.gilfsertg C.A. Meyer) showed over thirteen Dragendorff positive spots by TLC using eluent of $CHCl_3$/MeOH(10 : 1). TLC pattern of white, red and Sanchii ginseng (P. notoginseng) was mostly not different from each other, but, in American ginseng (P. quinqgtefolium),two spots having Rf 0.71 and 0.68 were not detected. An alkaloid component was isolated from white ginseng and identified as $N_9$-formyl-1-methyl-$\beta$-carboline (mp $174^{\circ}C$, $C_13H_10N_2O$).

In vitro Peroxynitrite Scavenging Activity of Arctic Seaweed Extracts

  • In this study, antioxidant activities of acetone/dichloromethane and methanol extracts of Arctic seaweeds were investigated. The antioxidant properties of both extracts of arctic seaweed were evaluated using two different peroxynitrite tests, including scavenging power on authentic peroxynitrite and inhibitory activities on peroxynitrite generation from 3-morpholinosydnonimine (SIN-1) producing superoxide anion and nitric oxide simultaneously in vitro. At concentration of $10\;{\mu}g/ml$, the acetone/dichloromethane and methanol extracts of Odonthalia dentata exhibited 54.6 and 64.2% inhibition against peroxynitrite generation from SIN-1 while they exhibited 24.3 and 23.0% scavenging activities on authentic peroxynitrite, respectively. On the other hand, the acetone/dichloromethane extract of Polysiphonia stricta exhibited 61.8% inhibition only against the induced peroxynitrite from SIN-1. Furthermore, the crude extracts of Odonthalia dentata and Polysiphonia stricta were fractionated into n-hexane, 85% aq. MeOH, n-BuOH, and $H_2O$ fractions, successively, and only 85% aq. MeOH fraction exhibited the best inhibition.

  • A unique brick series based on Vietnamese clay was manufactured at 114.22 MPa pressure rate for γ-ray attenuation purposes, consisting of (x) metallic waste & (90%-x) red clay mineral & 10% (hardener mixed with epoxy resin), where (x) is equal to the values 0%, 20%, 40%, 50%, and 70%. The impacts of industrial metal waste ratio in the structure and radiation protective characteristics were evaluated experimentally. The increase in metallic waste doping concentrations from 0% to 70% was associated with an increase in the manufactured brick's density (ρ) from 2.103 to 2.256 g/cm3 while the fabricated samples' porosity (Φ) decreased from 11.7 to 1.0%, respectively. Together with a rise in fabricated brick's density and a decrease in their porosities, the manufactured bricks' γ-ray attenuation capacities improved. The measured linear attenuation coefficient (μ, cm-1) was improved by 30.8%, 22.1%, 21.6%, and 19.7%, at Eγ equal to the values respectively 0.662, 1.173, 1.252, and 1.332 MeV, when the metallic waste concentration increased from 0% to 70%, respectively. The study demonstrates that manufactured bricks exhibit superior radiation shielding properties, with radiation protection efficiencies of 88.4%, 90.0%, 91.7%, 92.1%, and 92.4% for bricks with industrial metal waste contents of 0%, 20%, 40%, 50%, and 70%, respectively, at γ-ray energy (Eγ) of 1.332 MeV.

Immuno-Regulatory Activities of an Isoflavone Glycoside, 4', $6-Dimethoxylsoflavone-7-O-{\beta}-D-Glucopyranoside$ and the Crude Extract Isolated from Amorpha fruticosa LINNE

  • The methanolic (MeOH) extract of A. fruticosa bark, which showed immune-regulatory activities, was separated to purify an active compared by means of a multi-stage column chromatography. This resulted in the isolation and characterization of an isoflavone glycoside named 4', $6-Dimethoxyisoflavone-7-O-{\beta}-D-glucopyranoside$. Immuno-regulatory activities of the crude extract of Amorpha fruticosa LINNE bark were compared with that of an isoflavone glycoside (4', $6-dimethoxyisoflavone-7-O-{\beta}-D-glucopyranoside$). The crude methanolic extract of A. fruticosa and purified single compound showed 16% of relatively low cytotoxicity at a maximum concentration of 1.0 g/L in cultivated normal human lung cell line (HEL299). Cell growth of human T cells was increased up to 15%, 0.5 g/L of the crude extract added group. This was higher than a single compound added one. On the other hand, specific production rates of IL-6 and $TNF-{\alpha}$ from T cell were higher in the purified compound treat group ($0.82{\times}10^{-4}\;pg/cell$ and $1.08{\times}10^{-4}\;pg/cell$, respectively), compared to 0.5 g/L of the crude extract added group ($0.65{\times}10^{-4}\;pg/cell$ and $0.84{\times}10^{-4}\;pg/cell$, respectively). In addition, the growth of NK-92MI cells incubated with the crude extract was higher up to 56% over the cells grown with a single compound (0.5 g/L). In overall, the crude extract showed relatively higher immuno-regulatory activities compared with a single compound, probably due to the synergic effect given by other substances existed in the crude extract. Even though the siolated compound stimulated higher secretion of cytokines from human T cells.

Isolation and Identification of Inhibitory Compounds from Morus alba cv. Kuksang on α-amylase and α-glucosidase (국상(Kuksang) 뽕잎(Morus alba L.)으로부터 α-amylase와 α-glucosidase 저해 물질 분리 및 동정)

  • The objective of this research was to evaluate the inhibitory activities of phenolic compounds isolated from mulberry (Morus alba) leaves of 109 types against α-amylase and α-glucosidase. The inhibitory activity of the water extracts from Morus alba cv. Kuksang against α-amylase and α-glucosidase were determined as 93.8% and 48.7% respectively. The total phenolic content of extracts from Morus alba cv. Kuksang was 9.7±0.2 mg/g soluble in water and 14.3±0.2 mg/g soluble in ethanol. The inhibitory activity of the water extracts from Morus alba cv. Kuksang at 200 μg/ml phenolics concentration against α-amylase and α-glucosidase were determined as 100% and 82.6% respectively. The purification of inhibitory compounds was carried out by Sephadex LH-20 and MCI-gel CHP-20 column chromatography using a gradient elution procedure by nomal phase type (EtOH→distilled water) and reverse phase type (distilled water→MeOH). The quercetin was confirmed to be the chemical structure of the inhibitory compound against α-amylase and α-glucosidase by spectroscopic analysis of FAB-MS, NMR and IR spectrum.

Analysis of Higenamine Contents in Plants with HPLC and GC/MSD (HPLC 및 GC/MSD를 이용한 식물 중의 Higenamine 함량 분석)

  • Higenamine is known as a cardiotonic principle of Aconiti tuber. The analytical procedures were established for the detection of higenamine in plants. The amounts of higenamine in several Aconiti tubers and the embryo of Nelumbo nucifera, another plant species known to contain higenamine, were determined. The $H_2O$ soluble fraction prepared from MeOH extract was first treated with AD-2 resin and then applied to either HPLC or GC/MSD systems. With HPLC, $6.4{\sim}19.2\;{\mu}g/g$ of higenamine were detected from various Aconiti tubers and $182.3\;{\mu}g/g$ of higenamine from the embryo of Nelumbo nucifera. The results obtained with GC/MSD also provided comparable data with those obtained with HPLC.

CMP Properties of ZnO thin film deposited by RF magnetron sputtering (RF-sputtering에 의해 제작된 ZnO박막의 연마특성)

  • ZnO는 육방정계(wurtzite) 결정구조를 지니며 상온에서 3.37eV의 wide band gap을 갖는다. ZnO의 엑시톤 결합 에너지는 GaN에 비해 2.5배 높은 60meV로서 고효율의 광소자 적용 가능성이 높다. 또한 고품위의 박막합성이 가능하다. 이러한 특성 때문에 display소자의 투명전극, 광전소자, 바리스터, 압전소자, 가스센서 등에 폭 넓게 응용되고 있다. ZnO박막의 제조는 스퍼터링, CVD, 진공증착법, 열분해법 등이 있다. 본 논문에서는 RF 마그네트론 스퍼터에 의해 제작된 ZnO 박막에 CMP공정을 수행하여 연마율과 비균일도 특성 및 광투과 특성을 연구하였다. ZnO박막은 $2{\times}2Cm$의 Corning glass위에 증착되었다. 로터리 펌프와 유확산 펌프를 이용하여 초기진공을 $2{\times}10^{-6}$ Torr까지 도달시킨 후 Ar과 $O_2$를 주입하였다. 증착은 상온에서 이루어졌으며 공정압력은 $6{\times}10^{-2}$Torr이였다. 초기의 불안정한 상태의 풀라즈마를 안정시키기 위해 셔터를 이용하여 pre-sputtering을 하였다. CMP 공정조건은 플레이튼 속도, 슬러리 유속, 압력은 칵각 60rpm, 90ml/min, $300g/cm^2$으로 일정하게 유지하였으며 헤드속도는 20rpm에서 100rpm까지 증가시키면서 연마특성을 조사하였다. 실리카슬러리의 적합성을 알아보기 위해 DIW와 병행하여 CMP공정을 수행하고 비교 분석하였다. CMP공정 결과 광투과도는 굉탄화된 표면의 확보로 인해 향상된 특성을 보였다. 실리카 슬러리를 사용하여 CMP를 할 경우는 헤드속도는 저속으로 하여야 양호한 연마특성을 얻을 수 있었다.

Evaluation of True Metabolizable Energy and the Effect of Corn Distillers Dried Grains with Solubles in the Diets on Broiler Performance and Nutrient Availability (육계에 있어서 옥수수 주정박의 진정 대사 에너지가 측정 및 사료내 첨가가 생산성과 영양소 이용률에 미치는 영향)

  • This study was conducted to investigate the effect of corn distillers dried grains with solubles (CDDGS) in broiler diet on ileal amino acid digestibility and broiler performance. In experiment 1, TMEn of CGGDS ranged from 2,863 to 2,976 kcal/kg and averaged 2,904 kcal/kg on an 86% DM basis. In experiment 2, 192 male Ross broilers (one-day-old) were assigned randomly to 4 dietary treatment groups (12 birds/pen, 4 pens/treatment) as CDDGS levels (0, 6, 12 and 18%) and fed experimental diets for 6 weeks (starter ($0{\sim}2wk$); grower ($2{\sim}4wk$); finisher ($4{\sim}6wk$)). There was no difference in BWG of the chicks. However, feed intake and feed efficiency of chicks fed with non CDDGS groups were better than those of chicks fed with CDDGS fed groups (P<0.05). The amino acid digestibilities of diets containing CDDGS were high for phosphorus but low for lysine, threonine, aspartic acid, cystine and glycine (P<0.05) in the treatments compared to those of the control diet. This study showed that CDDGS has a high ME value and the treatments were low for the ileal amino acid digestibility, but there was no effect on the broiler performances.
