Polyhydroxyalkanoic Acid Production by Alcaligenes sp. GB-77 (Alcaligenes sp. GB-77 에 의한 Polyhydroxyalkanoic Acid의 생산)

  • For polyhydroxyalkanoic acid (PHA) production, several microorganisms were isolated from sewage sludge. One of them, GB-77 strain, was chosen from its PHB/HV copolymer production on only fructose without cosubstrate. The isolated strain GB-77 was identified as the genus Alcaligenes. Optimal temperature and pH for cell growth were 36C and 6.8. Optimal medium composition was 10 g/l of fructose and 5 g/l of polypeptone, 1 $\times$ 10$^{-2}$M Na$^{2}$HP0$^{4}$, 1.3 $\times$ 10$^{-2}$M KH$^{2}$PO$^{4}$. To investigate the optimal condition for polyhydroxyalkanoic acid production two-stage culture technique was used; first stage for cell growth and second stage for PHA production on unbalanced growth conditions. Optimal conditions for high PHA production were C/N ratio 50, temperature 36$\circ$C and pH 6.8. To overcome fructose inhibition on cell growth, intermittent feeding fed-batch culture technique was used. Total cell concentration was 17.4 g/l with 9.1 g/l of PHA. The purified PHA was identified PHB/HV copolymer by NMR analysis.

Chemical Properties of Soil Solution under Larix leptolepis, Pinus koraiensis, and Quercus mongolica Stands (일본잎갈나무, 잣나무, 신갈나무 임분 토양의 층위별 토양수의 화학적 특성)

  • Soil solutions were collected by zero-tension lysimeters at Kwangju, Kyunggi Province to estimate differences in ion concentration among species and horizons. Zero-tension lysimeters were installed in O, A, and B horizons in Larix leptolepis, Pinus koraiensis, and Quercus mongolica stands. Soil solution samples were collected from September 1996 through August 1998 and analyzed for $K^+$, $NA^+$, $NH_4^+$, $Mg^{2+}$, $Ca^{2+}$, $Al^{3+}$, $Cl^-$, $SO_4^{2-}$, $NO_3^-$, and $PO_4^{3-}$. The experimental site had high nitrogen loading from the atmosphere, and $NO_3^-$ was positively correlated with $K^+$ and $Mg^{2+}$. However, $NO_3^-$ and $NH_4^+$ showed a positive correlation only in the O horizon of Q. mongolica stand. Mg^(2+) deficit in the soil was predicted owing to the positive relationship of $Mg^{2+}$ with $NO_3^-$. Concentrations of $K^+$ and $Ca^{2+}$in soil solution were estimated higher in L. leptolepis than in other stands because of high leaching from the plant. Concentration of $Al^{3+}$ in soil solution was negatively correlated with soil solution pH. Mean soil solution pH of A and B horizons in P. koraiensis was lower than 4.7, however the $Al^{3+}$ concentration was lower than the toxic level to plants.

  • 1. Production of the artificial zeolite from coal ash Coal fly ash is mainly composed of several oxides including $SiO_2$ and $Al_2O_3$ derived from inorganic compounds remained after burning. As minor components, $Fe_2O_3$ and oxides of Mg, Ca, P, Ti (trace) are also contained in the ash. These components are presented as glass form resulting from fusion in the process of the combustion of coal. In other word, coal ash may refer to a kind of aluminosilicate glass that is known to easily change to zeolite-like materials by hydrothermal reaction. Lots of hot seawater is disposing near thermal power plants after cooling turbine generator periodically. Using seawater in the hydrothermal reaction caused to produce low price artificial zeolite by reduction of sodium hydroxide consumption, heating energy and water cost. As coal ash were reacted hydrothermally, peaks of quartz and mullite in the ash were weakened and disappeared, and new Na-Pl peaks were appeared strengthily. Si-O-Si bonding of the bituminous coal ash was changed to Si-O-Al (and $Fe^{3+}$) bonding by the reaction. Therefore the produced Na-Pl type zeolite had high CEC of 276.7 $cmol^+{\cdot}kg^{-1}$ and well developed molecular sieve structure with low concentration of heavy metals. 2. Utilization of the artificial zeolite in agro-environment The artificial zeolite(1g) could remove 123.5 mg of zinc, 164.7 mg copper, 184.4 mg cadmium and 350.6 mg lead in the synthetic wastewater. The removability is higher 2.8 times in zinc, 3.3 times in copper, 4.7 times in cadmium and 4.8 times in lead than natural zeolite and charcoal powder. When the heavy metals were treated at the ratio of 150 $kg{\cdot}ha^{-1}$ to the rice plant, various growth inhibition were observed; brownish discoloration and death of leaf sheath, growth inhibition in culm length, number of panicles and grains, grain ripening and rice yield. But these growth inhibition was greatly alleviated by the application of artificial zeolite, therefore, rice yield increased $1.1{\sim}3.2$ times according to the metal kind. In addition, the concentration of heavy metals in the brown rice also lowered by $27{\sim}75%$. Artificial Granular Zeolites (AGZ) was developed for the purification of wastewater. Canon exchange capacity was 126.8 $cmol^+{\cdot}kg^{-1}$. AGZ had Na-Pl peaks mainly with some minor $C_3S$ peaks in X-ray diffractogram. In addition, AGZs had various pore structure that may be adhere the suspended solid and offer microbiological niche to decompose organic pollutants. AGZ could remove ammonium, orthophosphate and heavy metals simultaneously. Mixing ratio of artificial zeolite in AGZs was related positively with removal efficiency of $NH_4\;^+$ and negatively with that of $PO_4\;^{3-}$. Root growth of rice seedling was inhibited severely in the mine wastewater because of strong acidity and high concentration of heavy metals. As AGZ(1 kg) stayed in the wastewater(100L) for 4days, water quality turned into safely for agricultural usage and rice seedlings grew normally.

Chemical Characteristics of Ground Water for Hydroponics and Waste Nutrient Solution after Hydroponics in Chungbuk Area (충북지역 양액 재배용 지하수 및 폐양액의 화학적 특징)

  • This survey has been conducted to obtain basic data of the quality of ground water for hydroponics and waste nutrient solution after hydroponics in hydroponic farms in Chungbuk area. Ground water samples were collected and analyzed at 19 sites of hydroponic farms. Waste nutrient solution samples were analyzed at 15 sites selected of them. The values of several components in ground water for hydroponics were as follows. pH range was shown from 6.2 to 7.7 and the average was 6.8. EC range was shown from 0.10 to 0.45 dS $m^{-1}$ and the average 0.23 dS $m^{-1}$. $NO_3-N$ concentrations was ranged from 0.12 to 13.77 mg $L^{-1}$, $SO_4^{2-}$ concentrations was ranged from 1.84 to 63.01 mg $L^{-1}$ and $Cl^-$ concentrations were ranged from 10.46 to 72.09 mg $L^{-1}$. Average values of $NO_3-N$, $SO_4^{2-}$ and $Cl^-$ were 4.00 mg $L^{-1}$, 12.70 mg $L^{-1}$ and 27.57 mg $L^{-1}$, respectively. $Ca^{2+}$, $Mg^{2+}$ and $Na^+$ concentrations were ranged from 3.24 to 36.99 mg $L^{-1}$, 1.44 to 14.93 mg $L^{-1}$ and 6.12 to 25.25 mg $L^{-1}$, respectively. Average concentrations were 13.06 mg $L^{-1}$ in $Ca^{2+}$, 6.02 mg $L^{-1}$ $Mg^{2+}$ and 12.08 mg $L^{-1}$ in $Na^+$. In waste nutrient solution after hydroponics, pH range was shown from 4.3 to 8.8 and the average was 6.7. EC range was shown from 0.44 to 2.37 dS $m^{-1}$ and the average 1.15 dS $m^{-1}$. Range of $NO_3-N$, $PO_4-P$, $K^+$, $Ca^{2+}$, $Mg^{2+}$ and $Na^+$ in waste nutrient solution were $10{\sim}212$, $0.56{\sim}26.1$, $10{\sim}295$, $16{\sim}215$, $9{\sim}54$ and $10{\sim}53$ mg $L^{-1}$ respectively. Average concentration were 100 mg $L^{-1}$ in $NO_3-N$, 12.15 mg $L^{-1}$ in $PO_4-P$, 99 mg $L^{-1}$ in $K^+$, 78 mg $L^{-1}$ in $Ca^{2+}$, 26 mg $L^{-1}$ in $Mg^{2+}$ and 26 mg $L^{-1}$ in $Na^+$. Inorganic matters in waste nutrient solution after hydroponics was higher than that of ground water for hydroponics.

Degradation rate of several types of Calcium Polyphosphate;Long term results (다양한 형태의 다공질 Calcium Polyphosphate의 생분해성에 관한 장기적인 연구)

  • The purpose of this study is to evaluate the biocompatibility and the biorsorbability of several types of calcium polyphosphate made through change of manufacturing process for 12 month. To solve limitation of calcium phosphate, we developed a new ceramic, Calcium Polyphosphate(CPP), and report the biologic response to CPP in extraction sites of beagle dog. Porous CPP blocks were prepared by condensation of anhydrous $Ca(H_2PO_4)_2$ to form non-crystalline $Ca(PO_3)_2$ and then milled to produce CPP powder. CPP powder, CPP block, and CPP granules added with $Na_2O$ were implanted in extraction sockets and histologic observation were performed at 12 months later. Like 3 months results, histologic observation at 12 months revealed that CPP matrix were mingled with and directly apposed to new bone without any adverse tissue reaction, CPP powder show direct bony contact, but new bone formation and fibrous tissue encapsulation showed in CPP block. 10% $Na_2O$ CPP granules show more inflammatory cells infiltration around graft materials compared at 3 month, but 15% $Na_2O$ CPP granules show less. This result revealed that regardless of addition of $Na_2O$, CPP had a high affinity for bone and had been resorbed slowly. From this results, it was suggested that CPP is promising ceramic as a bone substitute and addition of $Na_2O$ help biodegradation but optimal concentration of $Na_2O$ and other additive component to increase degradation rate should be determined in further study.

Effects of Fouling and Scaling on the Retention of Explosives in Surface Water by NF-the Role of Cake Enhanced Concentration Polarisation (지표수 조건의 나노여과공정에서 파울링 및 스케일링이 화약류 물질 잔류에 미치는 영향 연구 - 케익층 형성 및 농도분극 영향 분석)

  • The combined impact of Dissolved Organic Matter (DOM) fouling and inorganic ($CaSO_4,Ca_3(PO_4)_2$) scaling on the retention of TNT (2, 4, 6-Trinitrotoluene), RDX (Hexahydro-1, 3, 5-trinitro-1, 3, 5-triazine) and HMX (1, 3, 5, 7-Tetranitro-1, 3, 5, 7-tetrazocane) explosive contaminants by nano-filtration membrane were studied, since organic fouling and salt scaling are the major limitations for membrane filtration. Results reported here indicate that DOM fouling layer with a humic acid does not necessarily lead to an immediate loss of permeate flux but can result in a severe impact on the flux loss when both humic acid and inorganic scaltants were presented simultaneously. The $Ca_3(PO_4)_2$ mixed with humic acid showd most sever flux loss (42%) compared to that of only humic acid presence (8%). It could be a result that the scaling formation of the NF membrane was dominated by cake layer formation of DOM and it was along with pore blocking by the formation of crystals inside the porous active matrix of the NF membrane. In addition, these results indicated that the membrane selectivity of the explosives retention trended correlated with respect to increasing explosives size (listed by MW) based on greater steric interactions and followed the order (MW, g $mol^{-1}$; removal, %): HMX (296.15; 83%) ${\gg}$ RDX (222.12; 49%) ≋ TNT (227.13; 32%). Because the scaling and fouling layer could lead to a additional cake-enhanced concentration polarisation effect, the retention of explosives with the presence of humic acid in the feed solution and inorganic scaling formation on top of an organic fouling layer do not differ substantially retention from that of pure DI feed and NaCl solution.

  • The thick film type sensor having Pt/Na Super Ionic Conductor(NASICON) solid electrolyte/Pt/$Na_2$$SO_4$/Pt catalyst system for $SO_2$gas was fabricated by screen-print method. The phase of Na Super Ionic Conductor solid electrolyte sintered at different temperature of 1050, 1150,$ 1250^{\circ}C$ and for different time of 1.5, 2.5, 3.5 hr were investigated by XRD. The Electromotive Force variation of the sensor with $SO_2$concentrations and operating temperatures were investigated. The major phase of Na Super Ionic Conductor film sintered at 115$0^{\circ}C$ for 3.5 hr was sodium zirconium silicon phosphate($Na_3$Zr$_2$$Si_2$PO$_{12}$). The Nernst's slope of Na Super Ionic Conductor sensor for $SO_2$gas with the variation of concentration from 10 to 100 ppm was 167.14 ㎷/decade at the operating temperature of $500 ^{\circ}C$. The increase of oxygen partial pressure was not affected to the variation of Nernst's slope.e.

Effect of Seed Priming on the Enhancement of Seed Germination in Cool Season Turfgrass (Priming 처리가 한지형 잔디류의 발아 증진에 미치는 영향)

  • This study was carried out to identify the optimum condition for priming, as a series of experiments for the enhancement of germination percentage and germination synchronization in cool-season turfgrass seeds. The optimum priming conditions to maximize the germinability in both Kentucky bluegrass and Tall fescue was a 1 day treatment of 100 mM $K_3PO_4$ at $25^{\circ}C$. The seeds treated with the optimum priming condition enhanced the germinability compared to control, and shortened the time for germination with reduced $T_{50}$ and mean daily germination (MDG). However, as the concentration and treatment period of NaOH or KOH increased, significantly decrease in germination percentage was observed. Germination percentage in Creeping bentgrass and Bentgrass was below 10%, regardless of priming treatment. However, the priming treatment with 200 mM $KNO_3$ for three days improved the germination up to 6%, which was not a statistically significant level. The most effective priming period for Kentucky bluegrass and Tall fescue was one day treatment, but the germinability was suppressed as the treatment period was extended until six days. For priming chemical treatment, germinability was improved with the concentration of 100 mM, while it was reduced at higher concentrations of 200 mM and 300 mM.

Effect of Nitrate Concentration in Culture Solution on the Growth and the Uptake of Inorganic Elements of Tomato Plants(Lycopersicum esculentum Mill) (배양액(培養液)의 질산태질소농도가 토마토(Lycopersicum esculentum Mill)의 생육 및 무기원소(無機元素) 흡수에 미치는 영향)

  • This experiment was carried out to investigate the effects of nitrate concentration in culture solution on the growth and the uptake of inorganic elements in Tomato plant in the greenhouse. Tomato plants(cv. TVR-2) were grown with nitrate concentrations 8, 16, 24, 32cmol/l, based on Japan ENSI standard solution. Dry weights of lamina and petiole increased with the nitrate concentration. However, the dry weight of fruit was the highest in the treatment of nitrate concentration of 16cmol/l. The proportion of dry weights of vegitative organ to reproductive organ was the lowest in the treatments of nitrate concentrations of 16cmol/l and it increased with the nitrate concentration. The fruit yield was the highest at the treatment of nitrate concentration of 16cmol/l. With the increase of nitrate level the concentrations of N, $NO_3-N$, Ca and Na increased in lamina and petioles. The concentrations of K, P, S and Cl tended to decline in the nitrate concentration of 16 and 32cmol/l. These results indicate that optimum nitrate concentrations in a tomato grown by hydroponics change with growth stage, and the optimum concentrations for vegitative and reproductive stage were 8 and 16cmol/l, respectively. It also was proved that the nitrate concentrations in the culture solution affected antagonistically the uptake of inorganic anion in tomato : In low nitrate level $Cl^-$ uptake was affected much, while $SO_4{^{2-}}$ and $H_2PO_4{^-}$ uptake were affected in high nitrate level.

유류 오염지역으로부터 분리된 균주를 이용한 항공유의 분해

  • Bioremediation has been showing promise as an alternative to conventional environmental cleanup technologies. The objective of this study is to maximize the degradability of jet fuel in the soil system. The cells isolated from petroleum contaminated site was used for the degradation of jet fuel. When this strain was cultured in the MSM(minimal salt media) containing jet fuel for ten days, the degradability of jet fuel was almost 100%. The concentration of jet fuel did not affect the degradability much and the increased inoculution of strain and addition of nitrogen source decreased the time for complete degradation of jet fuel in the liquid culture. Inoculation of this strain increased the jet fuel degradability in the soil column by 15% and the aeration(50ml/min) and the addition of nutrients($NaNO_3$, $KH_2PO_4$) enhanced the jet fuel degradability(about 90%).
