Two hundred and sixteen 68-week-old Hy-Line brown laying hens were used in a 6-week feeding trial to compare the efficacy of phytases Optiphos (OPT) and Natuphos (NAT), which were isolated from Escherichia coli and Aspergillus niger, respectively. Hens were randomly allotted into six treatments with six replications (six layers in three adjacent cages) per treatment according to their initial BW. The hens were then subjected to one of the following dietary treatments: i) Positive Control (PC; available phosphorus (AP) 0.4%); ii) Negative control (NC; AP 0.2%); iii) NAT1 (NC+250 FTU/kg NAT); iv) NAT2 (NC+500 FTU/kg NAT); v) OPT1 (NC+250 FTU/kg OPT); vi) OPT2 (NC+500 FTU/kg OPT). Feed intake, egg production, egg quality, apparent nutrient digestibility and serum P and Ca concentration were evaluated to compare the effect of the two phytases. Feed intake and eggshell thickness were not affected by the treatments. Superior effects (p<0.05) of OPT were only observed in egg production and egg weight compared with NAT. Characteristics such as eggshell breaking strength, apparent digestibility of N, Ca and P and serum P concentration were equally increased with the supplementation of both phytases (p<0.05), where no significant difference was observed in those characteristics between PC and phytase supplementation at 500 FTU/kg. Equally effective improvements (p<0.05) were also observed in egg production and DM digestibility, where no improvements were observed (p<0.05) between the PC group and the groups with phytase supplementation at 500 FTU/kg. Equal increases in the serum Ca level were observed when the groups with phytase supplementation were compared to the PC group. Overall, the results of this study suggest that NAT and OPT are equally effective at liberating phytate-bound complexes when included in 0.2% available phosphorus diets for 68-week laying hens; either source of phytase can be fed to commercial 68-week laying hens at 500 FTU/kg to correct the negative effects associated with a 0.2% available phosphorus diet. In conclusion, either source of phytase can be fed to commercial first cycle laying hens at 500 FTU/kg to effectively replace inorganic phosphorus when economically justified.
The Debao pony ear marginal tissue fibroblast cell line (NDPEM 2/2) was uccessfully established using either primary explant technique or collagenase technique. The characterizations of the cell line were identified as following: the cells were adherent and of density limitation; population doubling time (PDT) of cells made with the two techniques were 35.9 h and 48 h, respectively; chromosome analysis showed that the frequency of cell chromosome number to be 2n=64 was 91.3%-92.8%. Confirmed by isoenzyme analysis, this cell line had no cross- contamination. Tests for microbial contamination from bacteria, fungi, virus or mycoplasma were negative. This newly established cell line meets all the standard quality controls of ATCC. It will provide a precious genetic resource for the conservation of the Debao pony breed, as well as effective experimental material for genetic studies on Debao ponies.
There are many methods of obtaining butadiene described in the literature. In the america it is produced largely from petroleum gases, i.e., by catalytic dehydrogenation of butene of butene-butane mixtures. Butadiene can be recovered from the $C_4$ residue of an olefin plant by distilling off a fraction containing most of the butadiene, catalytically hydrogenating the higher acetylenes to olefins and separating the product from other olefins and isobutane by extraction. Also it can be obtained by cracking naphtha and light oil. Among the individual dienes of commercial importance, 1, 3-butadiene is of first importance. It is used primarily for the production of polymers. In the present paper, it was investigated for a effect of the formation and the growth inhibition of popped corn polymer in butadiene extraction unit. As a result of study, inhibitors, $NaNO_2$ and TBC were good effective for inhibition of the formation and growth in popcorn polymer. The rational formula of popcorn polymer obtained was $(C_4H_6)_x$.
The present study was conducted to assess gene expression of bacterial lacZ driven by the SV40 promoter at early developmental stages of bovine embryos. The lacZ gene was linearized with BamHI digestion and introduced into the pronucleus by microinjection at 20 hrs after the commencement of in vitro fertilization. Intact bovine blastocysts were not stained with X-Gal, suggesting that there is no endogenous beta-galactosidase activity in these blastocysts. In contrast, the bovine blastocyst cells microinjected with the lacZ gene exerted a characteristic greenish-blue color originating from the bacterial beta-galactosidase activity, albeit at a low rate, i.e. 2.1% of the total fertilized oocytes injected. It was concluded, therefore, that the lacZ gene driven by the SV40 promoter could be used for an indirect screening method in which the presence of transgene is evaluated from the product of transgene expression.
This study was carried out to obtain the basic information about the sucker producing characteristics of certain burley tobacco varieties(Nicotiana tabacum L. var. burley) at Suwon and Chonju Experiment Station, Korea Ginseng & Tobacco Research Institute in 1999. The number and weight of suckers for 12 varieties or breeding lines were investigated three times, 45 days after transplanting for ground sucker, 10 days after topping, and 10 days after maleic hydrazide(MH) applying for upstalk sucker. KB 103 and KB 9416-1 produced less ground suckers and upstalk suckers than other entries did. KB 9210-8 produced less ground suckers and (Male sterile TC 612 x KB 108)F$_1$ also produced less upstalk suckers than other entries did. There was no differences among entries in sucker production at 10 days after MH treatment. Between the weight of total sucker and days to flower revealed the negative correlation, but not significant. The low sucker producing entries described above could be used as a parental line in the breeding program for the low sucker producing variety.
Environmental impact assessment survey reflecting farmers` opinion on the residence and production space in rural settlement area by ORD showed that more than 86% of respondents thought their reservoirs and waterways (small rivers) were getting seriously contaminated primarily by garbage and livestock manure. A typical rural settlement unit was taken to assess the impact of improper management of livestock manure in the farms on the water quality of small river flowing down along the villages where swine and dairy farms were situated in Daejook 2, 3-ri, Seolseong-myun, Icheon-gun. Nitrogen compounds such as NO$_3$-N, NO$_2$-N, NH$_3$-N, and phosphorus compound H$_x$PO$_4$, DO, BOD$_5$, COD, and microbial density were analyzed to evaluate water quality at five test sites designated along the water stream. Tests showed. for example, BOD$_5$ at site 4 was average 9.2mg/l which was about 3~8 times higher than that of observation site 2 and 3, at which most livestock houses were situated. This is a clear evidence that the nutrients of livestock manure illegally discharged to small river can lead to an eutrophication of the river at downstream. A soil absorption system with aeration could be one of alternatives to treat the contaminated wastewater by livestock manure. The place at downstream, inbetween observation site 1 and 2, could be the best construction site for the treatment facility from the standpoint of the overall treatment efficiency, An enclosed composting system can also be regarded as a good alternative for treatment of the sludge which is the by-product of the soil absorption system operation.

The present study was conducted to determine the best level of substitution of soybean meal by sunflower cake in diets for kids through the evaluation of quantitative carcass traits. Thirty-two Boer kids X 1/2 NDB (no defined breed), males, non-castrated, with 4 months of age and initial body weight of $15{\pm}3.2$ kg, were randomly assigned to individual pens. The treatments contained four substitution levels of soybean meal by sunflower cake (0, 33, 66 and 100% DM). At the end of the experimental period, the animals were slaughtered. There was no influence of the treatments on any of the mean values of the evaluated measures (p>0.05): 21.78 kg (body weight at slaughter), 8.65 kg (hot carcass weight), 8.59 kg (cold carcass weight), 40.27% (hot carcass yield), 39.20% (cold carcass yield), 7.73 $cm^2$ (rib eye area), 46.74 cm (carcass outer length), 45.68 cm (carcass internal length), 36.92 cm (leg length), 26.04 cm (leg perimeter), 48.66 cm (hind perimeter), 58.62 cm (thoracic perimeter), 0.20 (carcass compactness index), 68.48% (total muscle of the leg), 2.79% (total leg fat), 55.19% (subcutaneous leg fat), 28.82% (total bone), 81.66 g (femur weight), 14.88 cm (femur length), 0.38 (leg muscularity index), 2.53 (muscle:bone ratio) and 33.42 (muscle:fat ratio). The substitution of soybean meal by sunflower cake may be recommended up to a level of 100% without alterations to quantitative carcass traits.
In this study, we aim to describe kinematic characteristics of Korean /l/-flapping in two speech rates (fast vs. comfortable). Production data was collected from seven native speakers of Seoul Korean (four females and three males) using electromagnetic midsagittal articulometry (EMMA), which provided two dimensional data on the x-y plane. We examined kinematic properties of the vertical/horizontal tongue tip gesture, the vertical/horizontal (rear) tongue body gesture, and the jaw gesture in an /i/-/i/ context. Gestural landmarks of the vertical tongue tip gesture are directly measured. This serves as the actual anchoring time points to which relevant measures of other trajectories referred. The study focuses on velocity profiles, closing/opening spatiotemporal properties, constriction duration, and constriction minima were analyzed. The results are summarized as follows. First, gradiently distributed spatiotemporal values of the vertical tongue tip gesture were on a continuum. This shows more of a reduction in fast speech rate, but no single instance of categorical reduction (deletion). Second, Korean /l/-flapping predominantly exhibited a backward sliding tongue tip movement, in 83% of production, which is apparently distinguished from forward sliding movement in English. Lastly, there was an indication of vocalic reduction in fast rate, truncating spatial displacement of the jaw and the tongue body, although we did not observe positional variations with speech rate. The present study shows that Korean /l/-flapping is characterized by mixed articulatory properties with respect to flapping sounds of other languages such as English and Xiangxiang Chinese. Korean /l/ flapping demonstrates a language-universal property, such as the gradient nature of its flapping sounds that is compatible with other languages. On the other hand, Korean /l/-flapping also shows a language-particular property, particularly distinguished from English, in that a backward gliding movement occurs during the tongue tip closing movement. Although, there was no vocalic reduction in V2 observed in terms of jaw and tongue body height, spatial displacement of these articulators still suggests truncation in fast speech rate.

The ability of frozen-thawed fetal skin was examined to generate viable cell lines for nuclear transfer. Fetal skin frozen at -20$-20^{\circ}C$, $-30^{\circ}C$ or $-80^{\circ}C$ in the presence of 5% DMSO used as tissue explants to generate somatic cells. The resultant confluent cells were then used as donors for nuclear transfer (NT). Of the bovine NT embryos reconstracted from the somatic cells, 62.3%, 76.6% to 65% showed cleavage 70.5%, 81.9% to 78.5% reached the stage of morula formation and 39.7%, 43.2% or 47.6% reached the blastocyst stage. There was no significant difference in development when the NT embryos were compared with those reconstracted from fresh somatic cell derieved skin tissues (72%, 75.3%, and 45.2%, for cleavage, and development to morula and blastocyst stage, respectively). NT embryos were then placed in a portable $CO_2$ incubator and carried to China from Japan by air. After reaching to farm, two NT embryos were transferred to each of 5 recipients. We obtained 2 NT calves which birth weights is 30kg and 36kg female, and gestation periods is 281 and 284 days, respectively. There were no observation any abnormality from those calves. The results indicated that cell lines derieved from bovine fetal skin cryopreserved by a simple method could be used as donors in nuclear transfer using the portable $CO_2$ incubator.
To evaluate the effects of pulsed electric field (PEF) application on scalder and chiller water on Campylobacter contamination, four different treatments under three different water conditions including hard scalder water ($55^{\circ}C$), soft scalder water ($45^{\circ}C$) and chiller water, were applied as follows: i) a control treatment with no salt and no electric treatment, ii) a PEF only treatment, iii) a PEF treatment with 0.5% salt water, and iv) a PEF treatment with 1% salt water treatment. The use of PEF in hard scalding water showed an effect of reducing Campylobacter when compared to the control during the 200 s timeframe. With the addition of salt, the intervention caused at least 5.81 log CFU/ml reduction of Campylobacter counts after 200 s of PEF exposure. Similar effects were observed under soft scalding conditions. Campylobacter reductions were evident under chilling conditions with up to 2.00 log for PEF only, 5.77 log for PEF+0.5% salt and 2.69 log for PEF+1% salt treatment in water. Therefore, the current PEF setting for the scalder and chiller water can be successfully used to reduce pathogenic loads of Campylobacter on broiler chicken carcasses, and further research may be necessary to apply it in the poultry processing industry.
