The aim of the present study was to evaluate the bioequivalence of two domperidone maleate tablets, Motilium-$M^{(R)}$ Tablet (Janssen Korea Ltd., reference product) and $Toriem^{(R)}$ Tablet (Daewon Pharm. Co., Ltd., test product). Domperidone was extracted by liquid-liquid extraction using tert-butyl methyl ether and separated in less than 3 min on $C_{18}$ reverse-phase column using an isocratic elution. A tandem mass spectrometer, as detector, was used for quantitative analysis in positive mode by a multiple reaction monitoring mode to monitor the m/z $426.1{\rightarrow}119.1$ and the m/z $837.4{\rightarrow}158.2$ transitions for domperidone and the internal standard (roxithromycin), respectively. Calibration curves, from $0.05{\sim}50$ ng/mL of domperidone, showed correlation coefficients (r) higher than 0.9941. Intra day and inter day precision (C.V. %) for quality control were ranged from 10.04 to 16.09% and from 10.87 to 18.69%, respectively. The lower limit of quantification (LLOQ) of domperidone was 0.05 ng/mL. The method described is precise and sensitive and has been successfully applied to the study of bioequivalence of domperidone in 24 healthy Korean volunteers. Twenty-four healthy male Korean volunteers received a single dose of each medicine ($2{\times}12.72\;mg$ domperidone maleate) in a $2{\times}2$ crossover study. There was a one-week washout period between the doses. Plasma concentrations of domperidone were monitored for over a period of 24 hr after the administration. $AUC_{0-t}$ (the area under the plasma concentration-time curve) was calculated by the linear trapezoidal rule. $C_{max}$ (maximum plasma drug concentration) and $T_{max}$ (time to reach $C_{max}$) were compiled from the plasma concentration-time data. The 90% confidence intervals for the log transformed data were within acceptable range of log 0.8 to log 1.25 (e.g., $log\;0.92{\sim}log\;1.05$ for $AUC_{0-t}$, $log\;0.81{\sim}log\;1.05$ for $C_{max}$). The major parameters, $AUC_{0-t}$ and $C_{max}$ met the criteria of KFDA for bioequivalence indicating that $Toriem^{(R)}$ tablet is bioequivalent to Motilium-$M^{(R)}$ tablet.
The bioequivalence of two nilvadipine products was evaluated in 16 normal male volunteers (age 22-32 yr, body weight 57-80 kg) following sidle oral dose. Test product was Overca $l_{R}$ tablet (Choong-Wae Pharm. Corp., Korea) and reference product was Nivadi $l_{R}$ tablet (Hyundai Pharm. Corp., Korea). Both products contain 4 mg of nilvadipine. One tablet of the test or the reference product was administered to the volunteers, respectively, by randomized two period cross-over study (2$\times$2 Latin square method). The determination of nilvadipine was accomplished using a validated capillary column GC with electron-capture detection. As a result of the assay validation, the quantiflcation of nilvadipine in human plasma by this technique was possible down to 0.5 ng/ml using 1 ml of plasma. Absolute overall recovery from five replicate analyses of nilvadipine-spiked sample were 88.4$\pm$ 10.24% (mean$\pm$ 5.D.) for human plasma of 10 ng/ml. The coefficients of variation (C.V.) were less than 20% and the actual concentration of nilvadipine measured by GC ranged from 80 to 99% in all plasma. Average drug concentrations at each sampling time and pharmacokinetic parameters calculated were not significantly different between two products (p>0.05); the area under the curve from time zero to 8 hr (AUCo-$_{8 hr}$) (22.8$\pm$5.90 vs 22.2$\pm$6.10 ng . hr/ml), maximum plasma concentration ( $C_{max}$) (10.0$\pm$2.85 vs 9.3$\pm$3.28 ng/ml) and time to reach maximum plasma concentration ( $T_{max}$) (1.2$\pm$0.31 vs 1.3 $\pm$0.47 hr). The differences of mean AU $Co_{8hr}$$C_{max}$, and $T_{max}$ between the two products (2.25, 7.65, and 10.30%, respectively) were less than 20%. The power (1-$\beta$) and treaeent difference (7) for AU $Co_{8hr}$, and $C_{max}$ were more than 0.8 and less than 0.2, respectively. Although the power for Tmax was under 0.8, Tm\ulcorner of the two products was not significantly different from each other (p>0. 05). These results suggest that the bioavailability of Overeat tablet is not significantly different from that of Nivadil tablet. Maximal cardiac output and oxygen uptake $(VO_{2max})$ were measured during treadmill exercise for seven top-class marathoners and nine non-athletes using impedance cardiograph developed by one of the authors (DW Kim). Results of this study are summarized as belows. 1) New shoes with sponge and silicon rubber attached to the soles were developed to reduce motion artifact during treadmill exercise. Ensemble everaging techneque with the developed shoes was also used to improve the measurement of stroke volume using impedance cardiography. 2) Maximal cardiac output of the athletes, 14.98 L/min, was significantly higher than that of the non-athletes, 13.46 L/min. As maximal heart rate of the marathoners is lower than that of non-athletes, stroke volume of the former is significantly larger than that of the latter. 3) $VO_{2max}$ of the marathoners, 59.38 ml/kg/min, was higher than that of the non-athletes, 40.22 ml/kg/min. At the anaerobic threshold. $VO_{2max}$ of the former was 62.3% of $VO_{2max}$ and this was significantly higher than that of the non-athletes, 57.2%, This results indicates that the marathoners have higher aerobic capacity than the non-athletes. 4) The marathoners showed larger $VO_2$ than the non-athletes at the same cardiac output, indicating that a-v $O_2$ of the former is higher than that of the latter. 5) Maximal systolic pressure of the marathoners was higher than that of the non-athletes, and so was maximal rate-pressure products. These results indicate that heart oxygen consumption rate $(hVO_2)$ of the marathoner is higher than that of the non-athletes is mainly due to higher stroke volume. And higher oxygen consumption of the marathoners is due to higher stroke volume. And higher oxygen consumption of the marathoners is due to their larger a-v $O_2$. The marathoners show both higher threshold and $VO_{2max}$. Especially, measurement of cardiac output during treadmill exercise by improved impedance cardiography is expected to contribute in study of cardiac function of athletes.
Soybean (Glycine max L. (Merr) is commonly consumed and found in major foods including soymilk, soy sauce, tofu, and soy sprout in Korea and east Asia. In addition, it is common to cook the whole seeds with rice. Soybean is known to have ranges of health benefits including antiaging, anticancer, neuroprotective and antidiabetic taken either as supplement or dietary food. Anthocyanins and flavonoids in G. max are found to be the main contributors to such wide arrays of health benefits. Due to increasing economic values of soybean, development of specialty soybean cultivars is becoming an area of interest worldwide. In this study, 746 black soybean accessions from National Agrobiodiversity Center were characterized as part of an attempt to identify important germplasms of G. max. Seed coats of each accession were analyzed for their total anthocyanin, cyanidin 3-O-Glucoside (C-3-O-G), delphinidin 3-O-glucoside (D-3-O-G), petunidin-3-O-glucoside (Pt-3-O-G), and their whole seeds for crude protein contents. HPLC was used to determine and quantify the anthocyanin compositions while crude protein was determined using Kjeldahl method by Kjeltec auto-analyzer (Kjeltec 8400, Foss, Sweden). Accessions were grouped according to their anthocyanins and protein contents; the mean content of which were correlated to agronomic traits including maturity date, one hundred seed weight, cotyledon color and seed lust color. The results indicated that the total anthocyanin content (TAC) ranged from 273.77 to 6250.52 mg/100 g, with mean value of 1853.03 mg/100 g while the crude protein content (CPC) being between 33.43 and 47.51%, with mean value of 40.81%. The highest number of accessions (45.97%) showed TAC between 1000~1900 mg/100 g while 30.96% of accessions showed CPC between 41~43%. Among the 746 accessions considered, 11 (IT142935, 175818, 175855, 177191, 177209, 177211, 177214, 177216, 177218, 177220, 177274) of them showed TAC above 4000 mg/100 g. C-3-O-G was found to be the major contributor to TAC showing strong correlation. Accessions with green cotyledon color showed high mean TAC compared to those having yellow cotyledon color, and accessions with dull seed lust color showed high mean TAC than those having shiny seed lust color. One hundred seeds weight and maturity date showed positive correlation with all anthocyanin contents, except for Pt-3-O-G in the latter case. The overall result of the present study could be used as background for developing new black soybean cultivars and breeds with high anthocyanin and protein contents. The result depicted that many of the accessions could be used as potential parental lines.
밀의 영양체 저장탄수화물인 fructan의 대사에 관련된 fructan exohydrolase(FEH) 효소의 특성을 구명하기 위해 FEH를 밀의 줄기와 엽초에서 정제, 실험한 결과를 요약하면 다음과 같다. 1. FEH의 분자량은 63.7kD이었으며, pH 5.5와 3$0^{\circ}C$에서 최고의 활성을 보였다. 2. FEH의 $K_{m}$ 과 $V_{max}$는 fructan의 크기에 따라 각각 10~37mM, 5g~179mM로 변이를 보였으며, tetrasaccharide가 본 실험에서 사용된 기질 중에서 가장 낮은 $K_{m}$ 과 $V_{max}$를 보였다. 3. 밀의 FEH는 $eta$(2 $\longrightarrow$1)로 연결된 fructan에 대하여 기질 특이성을 보였으며 $\beta$(2 $\longrightarrow$6) fructan과 sucrose는 분해할 수 없었다.
In Korea, a local soybean (Glycine max) genotype 56l. was found to be strongly resistant to a virulent bacterial strain of a Pseudomonas sp. SN239. Specific genes involved in the resistance of the soybean genotype 561 were identified and the pattern of gene expression against the Pseudomonas infection was analyzed using differential-display reverse transcription PCR (DDRT-PCR). More than 126 cDNA fragments representing mRNAs were induced within 48 hours of bacteria inoculation. Among them, 28 cDNA fragments were cloned and sequenced. Twelve differentially displayed clones with open reading frames had unknown functions. Sixteen selected cDNA clones were homologous to known genes of other organisms. Some of the identified cDNAs were pathogenesis-related (PR) genes and PR-like genes. These cDNAs included a putative calmodulin-binding protein; an endo-l,3-1,4-$\bate$-D-glucanase; a $\bate$-1,3-endoglucanase; a $\bate$-1,3-exoglucanase; a phytochelatin synthetase-like gene; a thiol protease; a cycloartenol synthase; and a putative receptor-like serine/threonine protein kinase. Among them, four genes were found to be putative PR genes induced significantly by the Pseudomonas infection. These included a calmodulin-binding protein gene, a $\bate$-1,3-endoglucanase gene, a receptor-like serine/threonine protein kinase gene, and pS321 (unknown function). These results suggest that the differentially expressed genes may mediate the strong resistance of soybean 561 to the strain SN239 of Pseudomonas sp.
After a mixed carbohydrate diet, inhibition of ${\alpha}$-amylase and ${\alpha}$-glucosidase involved in the digestion and absorption of carbohydrates can significantly decrease the postprandial increase of blood glucose level. In the course of screening these useful enzyme inhibitors, we selected five kinds of bean, using an in-vitro enzyme inhibition assay method. To evaluate the effect of germination process on the functionality of the bean, we investigated the inhibitory activities of the water extracts of non-germinated bean and germinated bean against ${\alpha}$-amylase and ${\alpha}$-glucosidase, relevant to postprandial hyperglycemia. We also investigated the oxygen radical absorbance capacity(ORAC), total phenolics content, and postprandial blood glucose lowering effect in rats(Sprague-Dawley rat model). Most germinated beans showed significantly higher ${\alpha}$-glucosidase inhibitory activity, compared with non-germinated beans. Among germinated beans, Glycine max had the highest ${\alpha}$-glucosidase inhibitory activity(53.3%). The water extract of germinated Phaseolus vulgaris L. had the highest ${\alpha}$-amylase inhibitory activity(95.1%), followed by Glycine max(58.7%), and Glycine max L. Merr(54.1%). Furthermore, the five germinated beans also showed high antioxidant activities in ORAC assay. Results suggested that the germination process may improve and enhance the anti-hyperglycemia potential and antioxidant activity of the bean.
Six volunteers (mean $age=25.7{\pm}1.7$, $height=173{\pm}1.9$ and $weight=63.4{\pm}2.3{\;}kg$) participated in a graded exercise test and one hour of basic form of ChunDoSunBup (CDSB) Qi-training to investigate the cardiorespiratory responses and exercise intensity of Qi-training, a Korean traditional psychosomatic training. In the maximal exercise, the trainee showed $96.2{\pm}8.89{\;}l/min$ in ventilation (VE), $46.0{\pm}4.4$ in breath frequency (BF), $1.31{\pm}0.05$ in respiratory exchange ratio (RER), $180.7{\pm}3.0$ in heart rate (HR), and $2.6{\pm}1.1{\;}l/min$ or $40.7{\pm}2.3{\;}ml/kg/ml$ in oxygen consumption $(VO_2)$. Qi-training induced significant changes in BF, RER, HR, and $VO_2$. The exercise intensity of Qi-training were 42.3%, 46.9% and 38.7% of $HR_{max}$ during the sound exercise, slow motion (haeng-gong) and meditation respectively and the average was 46.2% of $HR_{max}$. We conclude that Qi-training is an aerobic exercise of a light (mild) intensity exercise, and it leads to decrease the metabolic rate in the trainee by breathing efficiently and relaxing them. In addition, Qi-training may affect cardiorespiratory function of BF, RER, HR and $VO_{2max}$ in trainees.
