Developmental Delay Effect of Harpacticoid Copepod, Tigriopus japonicus s.l. Exposure to 4-tert-octylphenol (4-tert -octylphenol에 노출된 저서성 요각류 Tigriopus japonicus s.l.의 발생지연 현상)

  • The ecotoxicological effects of 4-tert-octylphenol were observed on Harpacticoid copepoda Tigriopus japonicus s.l. gathered and cultured from tidal pool of Korean coast. There were no significant differences in survival rate (except 10 $\mu$g/L; 70.00%) and sex ratio (except 30 $\mu$g/L) on T. japonicus s.l. exposed to 4-tert-octylphenol. However, 4-tert-octylphenol induced developmental delay (copepodite emergence day and adult male mergence day) and retardation of first brooding day of adult female. Moreover the body size and biomass decreased at 4-tert-octylphenol exposure. As a result, detailed life-cycle research of T. japonicus s.l. may yield potential bioindicators for environmental monitoring and assessment.

Effect of food concentration on grazing, growth and fecundity of cyclopoid copepod Paracyclopina nana (기수산 요각류 Paracyclopina nana 의 섭식, 성장 및 생산력에 관한 먹이농도의 영향)

  • We studied the effect of food concentration on grazing, growth and fecundity of cyclopoid copepod Paracyclopina nana. Marine phytoplankton Tetraselmis suecica was used as a livefood for the copepod. Six stage compositions were used and food concentrations for the experiment were 0.5, 1, 2, 5, 10, 20, 40, 60 and $80{\times}10^4$ cells/mL. Range of food concentrations with 0.2, 0.5, 1, 2, 3 and $4{\times}10^4$ cells/mL were used for nauplii production experiment. Grazing rates of P. nana in all developmental stages on the different concentrations were increased with increasing diet concentration. While the growth of nauplius was not affected by increase of food concentration, food concentration outside of $1{\times}10^4$ to $5{\times}10^4$ cells/mL range negatively affected that of copepodite. Daily nauplii production was increased with increasing food concentration but $3{\times}10^4$ and $4{\times}10^4$ cells/mL treatments were not significantly different with $2{\times}10^4$ cells/mL treatment. As a result, optimum concentration of T. suecica for mass culture of P. nana was considered to be 5,000 cells/mL for nauplius stage, 10,000 cells/mL for copepodite stage and adult male, 20,000 cells/mL for adult female, respectively.

Immunomodulatory and Anti-Allergic Effects of Orally Administered Lactobacillus Species in Ovalbumin-Sensitized Mice

  • We investigated the effects of orally administered probiotic bacteria (Lactobacillus species) as allergic immune modulators in ovalbumin (OVA)-sensitized mice. BALB/c mice were intraperitoneally injected with OVA twice at a 2-week interval for allergy sensitization. The mice were then orally administered Lactobacillus casei YIT9029 (L1), L. casei HY7201 (L2), L. brevis HY7401 (L3), or L. plantarum HY20301 (L4) every 2 days for 3 weeks. Total IgE levels significantly decreased in sera of L3-administered mice but increased in the other groups. OVA-specific IgE levels decreased slightly in sera of mice administered L1, L3, and L4 but increased significantly in L2-administered mice. In passive cutaneous anaphylaxis (PCA) using sera from administered mice, only the L3-administered group showed reaction inhibition. High expression of TLR-2 with interferon (IFN)-${\gamma}$ stimulation on peripheral blood mononuclear cells occurred in L3- or L4-administered mice. Th1 cytokines, including IFN-${\gamma}$ and interleukin (IL)-12, increased in splenocytes of L3-administered mice; however, IL-4 decreased in L1- and L4-administered groups; IL-5 decreased in all experimental groups. IL-6 decreased in the L3-administered group; and IL-10 decreased in L1-, L2-, and L3-administered groups. L3 induced antiallergic effects by increasing Th1 cytokines, decreasing Th2 cytokines, and inhibiting the PCA reaction, whereas L2 administration increased allergic effects.

  • The structures of dioscin(1) and saponin P-d(2), isolated from the subterranean part of Allium fistulosum, were determined as diosgenin 3-O-$\alpha$-L-rhamnopyranosyl (1longrightarrow2)-[$\alpha$-L-rhamnopyranosyl (1longrightarrow4)]-$\beta$-D-glucopyranoside (1, ) and diosgenin 3-O-$\alpha$-L-rhamnopyranosyl (1longrightarrow4)-$\alpha$-L-rhamnopyranosyl(1longrightarrow4)[$\alpha$-L-rhamno-pyranosyl (1longrightarrow2)]-$\beta$-D-glucopyranoside (2) by spectroscopic and chemical degradational methods. The prosa-pogenin(4), which was provided on partial hydrolysis of 2, was not reported in previous literature.

Functional Analysis of BCTV ORF L4 by Site-directed Mutagenesis (Site-directed mutagenesis를 이용한 BCTV ORF L4의 기능 분석)

  • Beet curly top virus (BCTV) mutant has been constructed in vitro that contain G-to-T transversions at nucleotide 2727 within overlapping open reading frames (ORFs) L1 and L4. The mutations introduce termination codon in ORF L4 without affecting the amino acid encoded by ORF L1. When agroinoculated into Arabidopsis thaliana the mutant caused mild stunting and stem curling, but not the callus induction and hyperlasia on infected tissues of Sei-O ecotype. However, this mutant was not infectious on Col-O. Levels of single stranded DNA forms were similar in mutant and wild type BCTV infections. The DNA quantitation data showed that the DNA of BCTV-L4 mutant virus was accumulated in shoot tips, infection origin and roots with similar levels to those of wild type virus infected. Three tissues of asymptomatic ecotype Col-O also had as much as virus DNA from wild type virus infections. In both ecotypes infected with BCTV-Logan and BCTV-L4 mutant, root tissues contained more virus DNA than any other tissues by the Southern hybridization data. The results suggest that ORF L4 encodes a functional protein that is a major determinant of pathogenesis that might affect the hyperplastic response of the host to BCTV infection.

  • New synthetic method for diethyl N-[4-{[(2,4-diamino-6-yl)methyl]-amino}benzoyl]-L-glutamate(10) which is an intermediate of methotrexate is described. p-Nitrobenzoyl-L-glutamate was obtained via a two-step sequence which involves condensation of p-nitrobenzoyl chloride with diethyl-L-glutamate and Fischer esterification reaction with ethanol. Reductive methylation of diethyl-p-nitrobenzoyl-L-glutamate were carried out by reaction with formic acid and paraformaldehyde in the presence of $PtO_2$ catalyst and yielded diethyl N-(4-methylaminobenzoyl)-L-glutamate(7). It was followed by allylation and iodoazidozation to give the diethyl-p-[N-(2-azido-3-iodopropyl)-N-methyl]aminobenzoyl-L-glutamate(9). The cyclization reaction of compound(9) with 2,4,5,6-tetraaminopyrimidine was carried out by intermolecular nucleophilic substitution to give the desired methotrexate diethylester.

  • The plate under shear loading umformly distributed on the end portions of its side boundary was considered. Infinite hyperbolic serieses and Fourier serieses were combined as a stress function and from which exact solutions for the 15 cases for the parameters of b/L=0.25, 0.5, 1.0 and l/L=0.2, 0.4, 0.6, 0.8, 1.0 are obtained. In each cases the first 5 terms of the infinite series at the 36 points as shown in Fig. 3. The results are presented in Fig. 4-1, 4-2, and 4-3. The conclusions are as follows: 1) The stresses ${\sigma}_x$ increase very slightly as $\chi$ increases in the range of 0<x<L-l 2) When the parameters satisfy the conditions b/L<0.25 and l/L<0.2, the stresses in the region of 0<x<L-l can be obtained by replacing the uniform shear loading by the equivalent uniform shear loading by the equivalent uniform tensile force and pure bending moment at x=l. 3) The stress ${\sigma}_y$ is negligible throughout the region. 4) When the parameter b/L varies, the stresses ${\sigma}_x$ and u vary as L/b, while strain $\upsilon$ varies as $(L/b)^2$.

Effect of Plant Growth Regulator on Antioxidant Activity of Rosmarinus officinalis L. and Lavandula spica L. (로즈마리(Rosmarinus officinalis L.)와 라벤더(Lavandula spica L.)의 항산화 활성에 미치는 식물생장물질의 영향)

  • This experiment was conducted to investigate the induction rate of callus and antioxidative activity of rosemary and lavender. Callus induction rate of rosemary was 92% and 94% at 0.5mg/L and 1.0mg/L of 2,4-D in the media, respectively. The antioxidative activities of rosemary appeared differently according to the callus cultured under various concentrations of plant growth regulators. The strongest effect was observed at 0.1mg/L of 2,4-D extract, with $43{\pm}1.5{\mu}g/mL$ of an RC50 value. Callus induction rate of lavender was 98% and 99% at 1.0mg/L of 2,4-D and 1.0mg/L of NAA in the media, respectively. The regeneration rate of roots lavender was 85% at 1.0mg/L of NAA in the media. Antioxidative activities of lavender appeared diffently according to the callus cultured under plant growth regulators. The strongest effect was observed in 0.1mg/L of 2,4-D extract, with $37.6{\pm}0.9{\mu}g/mL$ of an RC50 value, which could be compared with value of leaf.

Relation between Tuffier's Line and Spondylolisthesis (척추전방전위증과 Tuffier's line 높이와의 상관관계)

  • Objectives : One of the most important cause of spondylolisthesis is it's epidemiological structure and L5's stability takes a significant role. The purpose of this study is to assess the difference of the Tuffier's line among of Spondylolythesis patients. Methods : We analyzed the Anterior view of lumbar spine checked at erect position on 60 patients who had been diagnosed as spondylolythesis. We investigated the Tuffier's Line on X-ray film. Results and Conclusions : Age distribution was 15 to 66 and the average was 47.81. Men were 19 cases(31.6%) and women were 41 cases(68.4%). Compared to men, women were more exposed to spondylolisthesis and for the age distribution, 50's showed the biggest portion. Group of normal people showed proportion of 14.3% at L4, 44% at L4/5 and 41.7% at L5 each. And group of patients showed proportion of 48.4% at L4, 35% at L4/5 and 51.6% at L5 each. Comparing these results, significance probability was 0.004(<0.05) which was significant. As the results of comparing the relation between L4/5 and L5/S1 patients, Normal group showed average of 3.33 which were close to L4/5. When there are spondylolisthesis at L4/5, averege was 3.33, placed at lower part of the vertebra body. When there are spondylolisthesis at L5/S1, averege was 3.566, placed between lower part of the veterbra body of L4 and L4/5. The p value of Normal group and L4/5 spondylolisthesis patient group was 0.022(p<0.05) which was significant. But the p value of Normal group and L5/S1 spondylolisthesis patient group was 0.0239 which was not significant. Also p value of L4/5 spcndylolisthesis patient group and L5/S1 spondylolisthesis patient group was 0.721 which was also not significant.

Identification and Fermentation Characteristics of Lactic Acid Bacteria Isolated from Hahyangju Nuruk (하향주 누룩으로부터 분리한 젖산균의 동정 및 발효 특성)

  • The purpose of this study was to isolate lactic acid bacteria, useful in the fermentation industry from Hahyangju Nuruk. Five strains were isolated, and identified as Lactobacillus based on growth inhibition by 10% (v/v) alcohol at pH 4.0. Isolated strains were identified to species, and named Lactobacillus plantarum L-3, L. sakei L-10, and L. curvatus strains L-8, L-11, and L-12. Morphological characteristics, physiological data, carbohydrate fermentation patterns, and 16S rRNA sequence data, were all used to characterize the bacterial isolates. L. plantarum L-3 showed the highest lactic acid productivity of all isolates, but grew only poony in the presence of 10% (v/v) alcohol at pH 4.0. The other strains exhibited lower lactic acid productivity than did L. plantarum L-3 and did not grow in the presence of 10% (v/v) alcohol at pH 4.0. The optimal temperature and pH for lactic acid production were $30^{\circ}C$ and pH 6.0 7.0, respectively. The lactic acid productivity of L. plantarum L-3, L. sakei L-10 and the three L. curvatus strains L-8, L-11, and L-12 were (% v/v of culture supematant) 1.55, 1.0, 1.06, 1.0, and 0.99, respectively, at $30^{\circ}C$ and pH 6.0. While L. plantarum L-3 suffered growth inhibition in the presence of 10% (v/v) alcohol, growth of the other strains was inhibited at 8% (v/v) alcohol.
