Background: Depression is one of the most commonly diagnosed neuropsychiatric diseases, but the underlying mechanism and medicine are not well-known. Although Panax ginseng has been reported to exert protective effects in various neurological studies, little information is available regarding its antidepressant effects. Methods: Here, we examined the antidepressant effect and underlying mechanism of P. ginseng extract (PGE) in a chronic restraint stress (CRS)-induced depression model in mice. Results: Oral administration of PGE for 14 d decreased immobility (depression-like behaviors) time in forced swim and tail suspended tests after CRS induction, which corresponded with attenuation of the levels of serum adrenocorticotropic hormone and corticosterone, as well as attenuated c-Fos expression in the amygdala. PGE enhanced messenger RNA expression level of brain-derived neurotrophic factor but ameliorated microglial activation and neuroinflammation (the level of messenger RNA and protein expression of cyclooxygenase-2 and inducible nitric oxide synthase) in the amygdala of mice after CRS induction. Interestingly, 14-d treatment with celecoxib, a selective cyclooxygenase-2 inhibitor, and $N_{\omega}$-nitro-L-arginine methyl ester hydrochloride, a selective inducible nitric oxide synthase inhibitor, attenuated depression-like behaviors after CRS induction. Additionally, PGE inhibited the upregulation of the nuclear factor erythroid 2 related factor 2 and heme oxygenase-1 pathways. Conclusion: Taken together, our findings suggest that PGE exerts antidepressant-like effect of CRS-induced depression by antineuroinflammatory and antioxidant (nuclear factor erythroid 2 related factor 2/heme oxygenase-1 activation) activities by inhibiting the hypothalamo-pituitary-adrenal axis mechanism. Further studies are needed to evaluate the potential of components of P. ginseng as an alternative treatment of depression, including clinical trial evaluation.
최근 국내 기후 변화로 인해 열대 과일 재배가 가능해지게 되었다. 특히 대표적으로 제주도에서 애플망고의 재배가 활발히 이루어지고 있으며 이로 인해 소비자들의 수요가 증가하고 있다. 그러나 애플망고 과육을 제외한 껍질과 같은 부산물은 활용도가 떨어져 폐기되고 있는 실정이다. 이에 본 연구에서는 부산물로서 폐기되어지는 애플망고 껍질을 활용하기 위해 이를 이용하여 항산화 및 항염증 효과를 검증하고자 열수 및 70% 에탄올로 추출하여 실험을 진행하였다. 먼저 애플망고 껍질의 열수 및 70% 에탄올 추출물의 총 폴리페놀 함량을 확인하였으며, 그 결과 66.08 ± 0.12 mg TAE/100 g, 100.13 ± 0.23 mg TAE/100 g의 함량을 나타내었다. 항산화 활성을 측정하기 위해 전자공여능과 2,2'- azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) 라디칼 소거능을 측정하였다. 전자공여능 측정결과, 1,000 ㎍/ml 농도에서 열수 추출물은 86.89%가 나타났고, 70% 에탄올 추출물은 94.06%를 확인하였다. ABTS 라디칼 소거능을 측정 결과로 1,000 ㎍/ml 농도에서 열수 추출물은 80.62% 를 나타내었고, 70% 에탄올 추출물은 98.66%의 소거능을 확인하였다. 이로 인해 애플망고 껍질 추출물의 항산화 활성은 대조군인 비타민 C와 유사한 효과를 가진 것을 확인하였다. 애플망고 껍질 추출물의 세포 생존율을 측정한 결과, 대식세포인 RAW 264.7 세포에서는 500 ㎍/ml 농도에서 99% 이상의 세포 생존율을 확인하였다. 이후 세포실험은 세포생존율이 99% 이상인 농도구간을 설정하여 진행하였다. 염증성 매개 물질로 알려진 nitric oxide (NO)에 대한 생성 억제 효과를 확인한 결과, 애플망고 껍질의 열수 추출물은 46%의 억제 효과가 나타났고, 70% 에탄올 추출물은 40%의 억제 효과를 확인하여 애플망고 추출물의 NO 생성량 저해 효과를 가진 것을 확인하였다. 염증 매개 인자의 억제 효과를 확인하기 위해 RAW 264.7 세포를 이용하여 iNOS와 COX-2의 단백질 및 mRNA 발현량을 측정하였다. 그 결과, LPS 단독 처리군에 비해 애플망고 껍질 추출물을 처리하였을 때 iNOS와 COX-2의 단백질 발현량은 최종 농도인 500 ㎍/ml에서 열수 추출물은 12%, 97% 억제하는 것을 확인하였고, 70% 에탄올 추출물은 62%, 91% 억제하는 것을 확인하였다. 또한 LPS 단독 처리군에 비해 iNOS와 COX-2의 mRNA 발현량 결과, 500 ㎍/ml 농도에서 열수 추출물은 3%, 33%의 억제효과를 확인하였고, 70% 에탄올 추출물은 29%, 36%의 억제효과가 나타나는 것을 확인하였다. 본 연구 결과들을 토대로 애플망고 껍질이 항산화 및 항염증 활성을 갖고 있는 것을 검증하여 천연 소재로서의 활용 가치가 높을 것으로 사료된다.
높은 레이놀즈수를 갖는 공학적인 흐름을 예측하는 가장 일반적인 방법은 여전히 벽함수를 이용하는 난류모형에 근거한 RANS 수치모의이다. 최근 벽근처의 점성영역 관계식과 벽에서 떨어진 대수영역 관계식을 혼합하여 개발된 일반화된 벽함수들은 두 영역사이의 난류량과 유속이 부드럽게 천이하도록 한다. 이 연구는 난류운동에너지(TKE), 에너지 소산율, 비소산율, 와점성에 대해서 적용 가능한 벽함수들을 조합하여 일련을 수치 모의를 수행하여 널리 이용되고 있는 난류모형들의 성능과 수렴 특성을 분석하였다. 이 연구 결과는 RNG k-𝜖 모형의 경우 첫번째 계산격자가 완충층에 놓이게 될 때는 반복 계산시 작은 허용오차를 이용하여 주의 깊게 적용을 하여야 안정된 해를 구할 수 있음을 보여준다. 표준 k-𝜖과 RNG k-𝜖 모형은 TKE와 와점성에 대해서 적용 가능한 벽함수들 중 어느 것을 선택하여 적용하더라도 수치모의 결과가 민감하게 반응하지 않는 것으로 나타났다. 한편, k-ω SST 모형의 경우 TKE에 대해서는 kL-벽함수 그리고 와점성에 대해서는 nutUB-벽함수를 이용하여야 정확하고 안정된 경계 조건 설정을 보장할 수 있다. 레이놀즈수 155,000조건에서 적용한 후방계산흐름 수치모의 결과 격자 해상도에 상관없이 약 13% 정도 재부착 거리를 과소평가하는 모형을 제외하고 나머지 적용한 난류모형들 모두 적절히 세밀한 해상도의 격자에서 양호하게 재부착거리를 잘 예측하는 것으로 나타났다.
Background: Ginseng contains three active components: ginsenosides, gintonin, and polysaccharides. After the separation of 1 of the 3 ingredient fractions, other fractions are usually discarded as waste. In this study, we developed a simple and effective method, called the ginpolin protocol, to separate gintonin-enriched fraction (GEF), ginseng polysaccharide fraction (GPF), and crude ginseng saponin fraction (cGSF). Methods: Dried ginseng (1 kg) was extracted using 70% ethanol (EtOH). The extract was water fractionated to obtain a water-insoluble precipitate (GEF). The upper layer after GEF separation was precipitated with 80% EtOH for GPF preparation, and the remaining upper layer was vacuum dried to obtain cGSF. Results: The yields of GEF, GPF, and cGSF were 14.8, 54.2, and 185.3 g, respectively, from 333 g EtOH extract. We quantified the active ingredients of 3 fractions: L-arginine, galacturonic acid, ginsenosides, glucuronic acid, lysophosphatidic acid (LPA), phosphatidic acid (PA), and polyphenols. The order of the LPA, PA, and polyphenol content was GEF > cGSF > GPF. The order of L-arginine and galacturonic acid was GPF >> GEF = cGSF. Interestingly, GEF contained a high amount of ginsenoside Rb1, whereas cGSF contained more ginsenoside Rg1. GEF and cGSF, but not GPF, induced intracellular [Ca2+]i transient with antiplatelet activity. The order of antioxidant activity was GPF > GEF = cGSF. Immunological activities (related to nitric oxide production, phagocytosis, and IL-6 and TNF-α release) were, in order, GPF > GEF = cGSF. The neuroprotective ability (against reactive oxygen species) order was GEF > cGSP > GPF. Conclusion: We developed a novel ginpolin protocol to isolate 3 fractions in batches and determined that each fraction has distinct biological effects.
Purpose : To examine the relative absolute reliability and validity of step test (ST) scores in subjects with chronic stroke. Method : A total of 27 stroke patients, participated in the study. A relative reliability index (intraclass correlation coefficient, ICC) was used to examine the level of agreement of inter-rater test-retest reliability for ST score. Absolute reliability indices, including the standard error of measurement(SEM) and the minimal detectable change (MDC), and limits of agreement by Bland and Altman analysis. The validity was demonstrated by spearman correlation of ST score with 10 m Walk Test (10mWT), Fugl-Meyer Assessment-Lower/Extremity (FMA-L/E)-total score, Berg Balance Scale (BBS)-total score. Result : An excellent inter-rater reliability in ST scores was found (paretic, ICC=0.993~0.996; nonparetic, ICC=0.982~0.991). In addition, excellent test-retest reliability was found (paretic, ICC=0.992; nonparetic, ICC=0.967). It all showed acceptable SEM of the ST score as paretic and nonparetic were 0.22 and 0.46 respectively (average score <10 %), and the MDC of the paretic and nonparetic were 0.61 and 1.27 respectively (possible highest score <20 %). indicating that measures had a small and acceptable measurement error. The ST score of paretic and nonparetic were also found to be significantly associated with 10MWT (r=0.77~0.79), FMA-LE scores (r=0.73~0.81) and BBS scores (r=0.72~0.76). Conclusion : The ST showed highly sufficient Inter-rater test-retest agreement and validity and acceptable measurement errors caused by due to chance variation in measurement. It also can be used by clinicians and researchers to assess the balance and mobility performance and monitor functional change in chronic stroke patients.
Torrefaction is considered as a promising pre-treatment for thermochemical utilization of biomass. Torrefaction temperature and time are the critical operation parameters. In this study, investigated were the effects of reaction temperature and time on product composition of torrefaction. scanning electron microscope (SEM) images and thermo gravimetric analyzer (TGA) results were also compared for the effects of the operating parameters. SEM images showed that the pores were observed at the temperature of $250^{\circ}C$ for 30 minutes. Rapid decreases in weight were observed the temperature between 200 and$400^{\circ}C$. Higher heating value of the torrefied biomass was over 5,000 kcal/kg at the temperature of $250^{\circ}C$ for 45 minutes. Energy density, which is defined as the ratio of the energy yield over the mass yield was 1.36 at the temperature of $250^{\circ}C$ for 45 minutes. The energy density was higher up to 1.6 at the temperature of $280^{\circ}C$, which indicates greater loss in mass. The major components of the gas produced in the torrefaction were $CO_2$ and CO, with traces of methane. The total amount of gas was 31.54 l/kg and the calorific value of the gas was $1,164.4Kcal/Nm^3$ at the temperature of $250^{\circ}C$ for 30 minute reaction time. Based on the results of this study, the temperature of effective torrefaction is about $250^{\circ}C$ for 30 to 45 minutes of reaction time. Considering the heating value, it is desirable to utilize the gas for efficient process of torrefaction.

There is an increasing interest in using inactivated probiotics to modulate the host immune system and protect against pathogens. As the immunomodulatory function of heat-killed Lactobacillus brevis KCTC 12777BP (LBB) and its mechanism is unclear, we investigated the effect of LBB on immune response based on the hypothesis that LBB might exert stimulatory effects on immunity. In the current study, we demonstrate that administration of LBB can exert immune-stimulatory effects and promote clearance of foreign matters through enhancing phagocytosis. Treatment with LBB induced the production of TNF-α, IL-6, and nitric oxide in macrophages. Importantly, LBB directly increased the phagocytic activity of macrophages against bacterial particles. LBB was able to promote the production of TNF-α in bone marrow-derived macrophages and splenocytes and also increase the proliferation rate of splenocytes, suggesting that the immune-stimulating activity of LBB can be observed in primary immune cells. Investigation into the molecular mechanism responsible revealed that LBB upregulates TAK1 activity and its downstream ERK, p38, and JNK signaling pathways. To further confirm the immunomodulatory capability of LBB in vivo, we orally administered LBB to mice and assessed the effect on primary splenocytes. Splenocytes isolated from LBB-treated mice exhibited higher TNF-α expression and proliferative capacity. These results show that heat-killed L. brevis, a wildly consumed probiotic, may provide protection against pathogens through enhancing host immunity.
Although probiotics have been shown to improve health when consumed, recent studies have reported that they can cause unwanted side effects due to bacterial-human interactions. Therefore, the importance of prebiotics that can form beneficial microbiome in the gut has been emphasized. This study isolated and identified bacteria capable of producing biopoymer as a candidate prebiotic from traditional fermented foods. The isolated and identified strain was named WCYSK01 (Wissella sp. strain YSK01). The composition of the medium for culturing this strain was prepared by dissolving 3 g K2HPO4, 0.2 g MgSO4, 0.05 g CaCl2, 0.1 g NaCl in 1 L of distilled water. The LMBP(low molecular weight biopoymers) produced when fermentation was performed with sucrose and maltose as substrates were mainly consisted of DP3 (degree of polymer; isomaltotriose), DP4 (isomaltotetraose), DP5 (isomaltopentaose), and DP6 (isomaltoheptaose). The optimization of LMBP (low molecular weight of biopolymer) production was performed using the response surface methodology. The fermentation process temperature range of 18 to 32℃, the fermentation medium pH in the range of 5.1 to 7.9. The yield of LMBP production by the strain was found to be significantly affected by q fermentation temperature and pH. The optimal fermentation conditions were found at the normal point, and the production yield was more than 75% at pH 7.5 and temperature of 23℃.

Species distribution models are a useful tool for predicting future distribution and establishing a preemptive response of invasive species. However, few studies considered the possibility of habitat for the aquatic organism and the number of target sites was relatively small compared to the area. Environmental DNA (eDNA) is the emerging tool as the methodology obtaining the bulk of species presence data with high detectability. Thus, this study applied eDNA survey results of Micropterus salmoides and Lepomis macrochirus to species distribution modeling by seasons in the Anyang stream network. Maximum Entropy (MaxEnt) model evaluated that both species extended potential distribution area in October compared to July from 89.1% (12,110,675 m2) to 99.3% (13,625,525 m2) for M. salmoides and 76.6% (10,407,350 m2) to 100% (13,724,225 m2) for L. macrochirus. The prediction value by streams was varied according to species and seasons. Also, models elucidate the significant environmental variables which affect the distribution by seasons and species. Our results identified the potential of eDNA methodology as a way to retrieve species data effectively and use data for building a model.

경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
