Studies on toxicity of ochratoxin A to chromosomes of turmor cell-line (종양세포(腫瘍細胞)의 염색체(染色體)에 대한 오크라톡신 A의 독성(毒性)에 관한 연구(硏究))

  • This study was performed to investigate the toxicity of ochratoxin A (OA) to the chromosomes of $K_{562}$ tumor cell-line in vitro. The results of this experiment were as follows: 1) Chromosomes of $K_{562}$tumor cell-line resulted in pseudotriploidy on the control group. Chromosomes of $K_{562}$ tumor cell-line treated with OA resulted in heteroploidy compared with the control group. The mean number of chromosomes in the karyotype of the control group (60) were 7 in the A group, 5 in the B group, 20 in the C+X group, 7 in the D group, 9 in the E group, 6 in the F group, and 6 in the G+Y group respectively. The number of chromosomes were increased as follows: Treating with $0.7{\mu}M$ OA, the number of chromosomes were increased one in E and F group, two in G+Y group compared with control group. In treated with $1.5{\mu}M$ OA, the increasing number of chromosome was one in E and F group. In treated with $3{\mu}M$ OA, E and F group was increased one and G+Y group were increased two chromosomes compared with control group. But in treated with $6{\mu}M$ OA, the number of chromosome in G+Y group was decreased one. 2) $K_{562}$ tumor cell line treated with OA showed Philadelphia-Chromosome in the long arm of the G group karyotype chromosome. The rate of chromosome aberration in $K_{562}$ tumor cell-line treated with OA was 77% in $0.7{\mu}M$ OA group, 71% in $1.5{\mu}M$ OA group, 82% in $3{\mu}M$ OA group and 94% in $6{\mu}M$ OA group respectively. The rate of chromosome aberration of $K_{562}$ tumor cell-line treated with OA was high in the high dose level of OA, and chromosome aberration of $K_{562}$ tumor cell-line treated with OA showed deletion, minute, dicentric-chromosome and translocation in the long arm of the C-group karyotype. As a result of this study, the toxicity of OA showed deletion, minute, dicentric-chromosome and translocation in the long arm of the C-group karyotype, and then, the toxicity of OA resulted in the damage to RNA and protein synthesis in $K_{562}$ tumor cell-line, and the C-group karyotype of $K_{562}$ tumor cell-line was target of the toxicity of OA.

Oleanolic Acid Promotes Neuronal Differentiation and Histone Deacetylase 5 Phosphorylation in Rat Hippocampal Neurons

  • Oleanolic acid (OA) has neurotrophic effects on neurons, although its use as a neurological drug requires further research. In the present study, we investigated the effects of OA and OA derivatives on the neuronal differentiation of rat hippocampal neural progenitor cells. In addition, we investigated whether the class II histone deacetylase (HDAC) 5 mediates the gene expression induced by OA. We found that OA and OA derivatives induced the formation of neurite spines and the expression of synapse-related molecules. OA and OA derivatives stimulated HDAC5 phosphorylation, and concurrently the nuclear export of HDCA5 and the expression of HDAC5 target genes, indicating that OA and OA derivatives induce neural differentiation and synapse formation via a pathway that involves HDAC5 phosphorylation.

Content of Orotic Acid in Raw milk and Reduction of OA by Lactic Acid Bacteria during Fermentation (Orotic acid의 원유내 함량과 발효중 유산균에 의한 감소)

  • Changes in erotic acid (OA) contents of raw milk and during cultivation and storage at refrigerated temperature were determined. OA contents of raw and 10% reconstituted milk held at $121^{\circ}C$ for up to 15 min decreased by 17.6 and 22.4%, and those fermented at 40 and $37^{\circ}C$ for 72 hr with Lactobacillus helveticus 166 and Lactobacillus casei 955 decreased 37.8-43.2 and 41.8-76%, whereas when fermented at $40^{\circ}C$ for 72 hr with Streptococcus thermophilus ST-37, did not decrease significantly compared with fermentation by other lactobacilli. OA content did not change during storage at refrigeration temperature. OA reduction by Lactobacillus sp. was dramatically increased at the initiation of stationary phase during fermentation. OA reduction varied among different lactobacilli. These results show decrease in cultivation time, rapid cooling of yogurt, and proper selection of Lactobacillus sp. prevent OA content reduction.

Antimicrobial Effect of Ursolic Acid and Oleanolic Acid against Methicillin-Resistant Staphylococcus aureus (Ursolic Acid와 Oleanolic Acid의 메티실린 저항성 Staphylococcus aureus에 대한 항균작용)

  • The antimicrobial activity of ursolic acid (UA) and oleanolic acid (OA), both triterpenoid compounds, against methicillin-resistant Staphylococcus aureus (MRSA) is controversial. We examined the antimicrobial effects of UA and OA against 19 strains of MRSA isolated from Koreans by determining minimum inhibitory concentrations (MIC) and minimum bactericidal concentrations (MBC). The data showed that the methicillin-sensitive strain S. aureus KCTC $1621^T$ was more resistant to UA and OA than that of the MRSA strains. The MBC values of UA and OA against MRSA had broad ranges; 4 to 32 ${\mu}g/ml$ and 16 to >256 ${\mu}g/ml$, respectively. It was difficult to understand the different antimicrobial activities of UA and OA among the MRSA strains, because UA and OA antimicrobial mechanisms are unknown. These results indicate that the antimicrobial effects of UA and OA against MRSA are dependent on resistance to UA and OA in each strain.

Prevalence Of Radiographic Ankle Osteoarthritis (방사선적 족관절염의 유병율)

  • Objectives: This study was done to determine the prevalence of radiographic ankle osteoarthritis (OA) in Korean women with clinical primary ankle OA. Methods: This analysis involved 160 Korean women who visited our out patient clinic due to ankle joint pain. Ankle AP and lateral weight-bearing radiographs were evaluated retrospectively to check whether they have radiographic OA or not. Radiographs were read for features of ankle OA using School classification. Demographic factors and radiographic findings in both radiographic ankle OA group and non-radiographic ankle OA group were analyzed by the student t-test, the chi square test and the Pearson's analysis. A p value <0.05 was considered significant. Results: The prevalence of radiographic ankle OA in Korean women is 18.2 % in patient between 18 and 39 years and is 55.6 % in patient over 40. There was significant difference between two groups with age (P<0.001) and relatively strong correlation between radiographic OA and age (r=0.474, P<0.001). Conclusion: These results show that the prevalence of Korean women with radiographic changes of ankle OA was 45 % and there was relatively strong correlation between age and radiographic ankle OA.

Impact of Open Access Models on Citation Metrics

  • We report results of selection-bias-free approaches to the analysis of the impact of open access (OA) models on citation metrics. We studied reference groups of Gold and Green OA articles and the group of non-OA (Paywall) articles with the new functionality of the Web of Science Core Collection database, the InCites platform of Clarivate Analytics, and the Dimensions database of Digital Science. For each reference group we obtained the values of the percent of cited articles and citation impact and their dependence on the depth of the citation period. Different research fields were analyzed in two schemas of the InCites platform. We report the higher values and growth rates of the citation metrics: citation impact and %Cited, in the OA reference groups over the Paywall group. The Green OA articles demonstrate the highest values of citation metrics among all the OA models. Dependence of the value of citation impact on citation period follows linear law with R2 values close to 0.9-1.0. The overall annual growth rates of citation impact of the Green OA, Gold OA, and the Paywall articles, k equal, respectively, 3.6, 2.4, and 1.4 in Dimensions and 4.6, 3.6, and 2.3 in the Web of Science Core Collection. We suppose that earlier results reported for the articles in pure OA journals vs. articles in Paywall journals were affected by the high citation impact of the Green and Hybrid OA articles that could not be elucidated in the Paywall journals at that time.

Effects of Bee Venom on Papain-induced Osteoarthritis in an Animal Model (봉독이 Papain으로 유도된 골관절염 동물 모델에 미치는 영향)

  • Osteoarthritis (OA) is an age-related joint disease characterized by degradation of articular cartilage and its association with chronic pain. Purified Bee venom (PBV) has been traditionally used to the treatment for inflammatory diseases. The purpose of the current study is to examine whether PBV regulates the pro-inflammation against a mouse model of knee OA induced by papain. We studied the effect of PBV on papain-induced OA in the knee joints of mice. Mice were split into following groups: normal control, papain induced OA, OA treated with PBV, OA treated with meloxicam as positive control. Proteoglycan deposition was analyzed by safranin O-fast green staining and H&E staining. Papain injection significantly degraded the proteoglycan in OA mice at 42 days. Cartilage proteoglycan density was significantly higher in PBV treated OA group than those of the positive control groups. These results demonstrate that PBV efficiently suppresses pathological processes in an OA model. Thus, PBV could be a potential therapeutic strategy for the treatment of OA.

Anticoagulant activities of oleanolic acid via inhibition of tissue factor expressions

  • Oleanolic acid (OA), a triterpenoid known for its anti-inflammatory and anti-cancer properties, is commonly present in several medicinal plants but its anticoagulant activities have not been studied. Here, the anticoagulant properties of OA were determined by monitoring activated partial thromboplastin time (aPTT), prothrombin time (PT), fibrin polymerization as well as cell-based thrombin and activated factor X (FXa) generation activities. Data showed OA prolonged aPTT and PT significantly and inhibited thrombin catalyzed fibrin polymerization. In addition, OA inhibited the activities of thrombin and FXa and inhibited the generation of thrombin or FXa in human endothelial cells. OA also inhibited TNF-${\alpha}$-induced tissue factor expression on human endothelial cells. In accordance with these anticoagulant activities, OA showed an anticoagulant effect in vivo. These results indicate that OA possesses antithrombotic activities and suggest that daily consumption of a herb containing OA may be preventing thrombosis in pathological states.

Antigenicity Changes of Ovomucoid and Ovalbumin in Chicken Egg White by NaOH, Heat and Protease Tratments (NaOH, 열, 및 효소 처리에 의한 계란 난백 중 ovomucoid와 ovalbumin의 항원성 변화)

  • Antigenicities of ovomucoid (OM) and ovalbumin (OA) in chicken egg white (EW) before and after NaOH, heat, and pretense treatments were examined by competitive indirect enzyme-linked immunosorbent assay (ciELISA), using rabbit anti-OM and-OA antibodies, Enzymatic hydrolysis of EW did not effectively reduce antigenicity of OM, whereas that of OA was decreased to 1/5,000-1/100,000 by treatment of plant-derived or microbial pretenses. Heat treatment below $100^{\circ}C$ for 30min did not decrease antigenicity of OM, whereas that of OA in heated EW increased maximally to 100 times, Antigenicity of OM in EW effectively decreased by NaOH treatment, disappearing at over 1% NaOH, whereas that of OA increased. Additional heat treatment of NaOH-treated EW at $70^{\circ}C$ for 15min slightly reduced antigenicities of OM and OA.

An Experimental Study of Temperature Profiles in Mixing Zone of AHU with an Air Mixer (에어믹서가 설치된 공조기 혼합실 내의 온도분포에 관한 실험적 연구)

  • A study of temperature profiles in mixing zone of AHU (air handling unit) can contribute greatly to enhance performance of AHU system, so the study on the temperature distribution between RA (return air) and OA (outdoor air) is important to analyze the mixing characteristics in a mixing zone of AHU. Accordingly, the temperature profiles during RA (return air) and OA (outdoor air) supply process into mixing zone of AHU with an air mixer are studied experimentally. The effect of air mixer, OA temperature and RA/OA flow rate are studied in detail. In this study, the results show that the mixing efficiency is all high for installed the air mixer. The more OA temperature increase and OA flow rate decrease, the more mixing efficiency is high.
