Degradation Mechanism of Cholesterol and Its Derivatives by Microorganisms. II Iso of-Hydronyandrost-4-ene-3,17-dione (미생물에 의한 cholesterol과 그유도체의 분해기구 II 19-Hydronyandrost-4-ene-3, 17-dione의 분리)

  • When 19-hydroxycholesterol acetate was added into CSD-10 in Nutrient Broth or in a mineral salts medium consisting of KH$_{2}$PO$_{4}$(0.1%), $K_{2}$HPO$_{4}$(0.1%), NH$_{4}$NO$_{3}$(0.1%), MgSO$_{4}$(0.02%), CaCl$_{2}$(0.002%), and FeCl$_{3}$(0.005%), a substantial amount of 19-hydroxyandrost-4-ene-3,17-dione was accumulated prior to accumulation of estrone. From this result and all of previous works, a tentative degradation pathway of 19-hydroxycholesterol acetate to estrone by CSD-10 was derived. 19-hydroxyandrost-4-ene-3,17-dione seems to be an attractive intermediate for the synthesis of 19-norsteroids.

  • Four strains of yeasts were chosen from those isolated previously, and a strain from 160 isolates collected in this year were examined for the treatment of pulp waste liquor. Experiments about optimum nutrient condition, composition of cells, and reduction of B.O.D. on the "S" pulp industry waste liquor were performed with 5 strains. 1. The isolates(strain 112) was identified as Candida utilis. 2. The optium concentration of 4 components of nutrients were ($NH_4$)$SO_2$lg/l, yeast extract 70mg/l, $KH_2PO_4$ 300mg/l, and $MgSO_4{\ddot}7H_2O$ 500mg/l. 3. Specific growth ratio of Candida utilis KYRI 112 was 0.48/hr at optimum nutrient media and the yield was 0.45%(V/V). 4. Endomycopsis capsularis KYRI 613 contained more crude protein than the most of commercial yeasts. 5. The B.O.D. of waste liquor was reduced ro 20% of its value by the culture.e culture.

Control Efficacy of Milk Concentration Against Powdery Mildew of Strawberry

  • The aim of this study was to determine the effect of milk as one of the environmental friendly materials that substitute chemical fungicides for control powdery mildew (Sphaerotheca aphanis) of strawberries $(Fragaria{\times}ananassa\;Duch.)$. 'Maehyang' and 'Akihime' varieties planted in greenhouses were evaluated for the control of powdery mildew. Applications of $5\%,\;10\%\;and\;20\%$ milk had much better effects on controlling powdery mildew. In particular, $10\%$ milk showed a higher efficacy than other concentrations applied onto straw­berry in greenhouse experiments. Foliar spray application of $10\%$ milk was effective for powdery mildew, whereas drench application was not. Also, foliar spray of $10\%$ milk was able to accelerate more firmness and calcium contents of strawberry fruits than the non-treated. The $10\%$ milk applied with fertilizer salts $(Ca(H_2PO_4)_2-H_2O\;and KH_2PO_4)$ showed there was a similar efficacy to $10\%$ milk alone in greenhouse experiments. White crystals and cracks on strawberry fruits appeared by $20\%$ milk. This result indicated that $10\%$ milk was a useful substitute for fungicides to control powdery mildew of strawberry.

Biosurfactant 생산 효모 Rhodotorula sp. G-1의 분리 및 Biosurfactant 생산

  • Some microorganisms including yeasts produce surface tension-decreasing biosurfactants. The strain G-1, the best producer of biosurfatants was isolated from the soil and identified as Rhodotorula sp., which was not discribed any report. The Rhodotorula sp. G-1 produced biosurfactant from vegetable oils, but failed to produce it from n-alkane or carbohydrate. Yeast extract was found to be more effective for the biosurfactant production as nitrogen source than any other inorganic nitrogen source. The composion of the optimal medium contained the following conponents: soybean oil 4%, glucose 2%, yeast extract 0.5%, KH$^{2}$PO$^{4}$ 0.1%, K$^{2}$HP0$^{4}$ 0.l%, MgSO$^{4}$ 5%, CaCl$^{2}$ 0.01%, NaCl 0.01%, pH 6.0. The surface tension activity was increased to 14% when, at first, the culture broth was fermented with only soybean oil as carbon sourse, and after 90 hours, feeded glucose, than that Of glucose and soybean oil added to it simultaneously. The maxium yield of the biosurfactant was about 15 g/l by after 90 hours, the feeding method of glucose.

  • Nine novel cyclodextrin glycosytransferase-producing bacteria were isolated from soil in a low acidic pH (3-4) medium at high temperature (45-60C). The isolated acidophilic bacteria were identified as Bacillus acidocaldarius. Highest yield of enzyme was obtained by using the following medium: 4% raw potato, 1% peptone, 0.1% yeast extract, 0.02% (NH4)2SO4, 0.05% MgSO4, 0.02% CaCl2, 0.3% KH2PO4. The crude enzyme showed a very broad pH-activity curve and had two optium pH ranges at 30 and 5.0-6.0. The crude enzyme was most active at 90C.

  • An interspecific fusant strain, Streptomyces MS1 was obtained by protoplast fusion between S. peucetius subsp. caesius and S. platensis. We studied on the fermentation characteristics of the fusant strain. The fermentation products of the fusant MS1 was identical with S. peucetius, but its production of anthracycline was more stable than S. peucetius under various fermentation conditions in regard to acidogenesis of fermentation broth. The optimal medium composition for anthracycline production by fusant MS1 as follows: sucrose 2.0%, glucose 1.0%, soytone 0.7%, $CaCO_3$ 0.2%, $KH_2PO_4$ 0.013%, casamino acids 0.01%, $K_2SO_4$ 0.025%, $MaCl_2\;6H_2O$ 1.024%, 5M $CaCl_2\;5H_2O$ 0.4%, 1N NaOH 0.7%, 20% L-proline 1.5%. In this condition, the productivity of anthracycline was $80{\sim}100\;{\mu}g/ml$.

  • The culture variables were optimized to increase 1,3-dihydroxyacetone (DHA) production by Gluconohacter oxydans ZJB09112 in shake flasks and bubble column bioreactors. After fermentation in the optimized medium (g/l: yeast extract 5, glycerol 2.5, mannitol 22.5, $K_2HPO_4$ 0.5, $KH_2PO_4$ 0.5, $MgSO_4{\cdot}7H_2O$ 0.1, $CaCO_3$ 2.0, pH 5.0), when five times of glycerol feeding were applied, $161.9{\pm}5.9\;g/l$ of DHA was attained at a $88.7{\pm}3.2%$ conversion rate of glycerol to DHA.

  • It was reported in our Previous paper that the fermented products from Gloeostereum incarnatum strongly inhibit the growth of six kinds of bacteria in human bodies. In this paper the appropriated conditions of immersing culture for the strain 8 903 of Gloeostereum incarnatum was analysed. And the output of the hypha and fermentative product was determined or compared. The prelimenaryresults showed that the appropriated conditions for the growth of Gloeostereum incarnatum are: (1)culture medium:glucose 3%; protein peoptne 0.2%; soybeancake power 1% yeast power 0.3%; KH2PO40.05%; MgSO4 0.03%; CaCO3 0.01%; vitamin Bl 0.001%; befor sterilization pH Value of six should be maintained; (2) temperature; 27f ~28f ; (3) time; about 200 hours; (4) ventilation; (30%∼50%)/min. The sigh of the end culture are: pH coming down about 4: remnant glucoses less 1%; amino nitrogens about 20%; time about eight days. In the aforementioned conditions, the output of fermentative product achieve to 2.5∼3g/L.

  • Over 90 of lysine producing auxotrophs were obtained from Corynebacterium sp. S-27-12, Brevibacterium flavum ATCC 15168 and Micrococcus glutamicus ATCC 13032 by UV light, $Co^{60}$ irradiation and N-methyl-N'-nitro-N-nitrosoguanidine treatment. One of the mutant, Brev. flavum U46-N59, was identified as a leucine auxotroph and accumulated lysine during flask (500 ml) cultivation (180 strokes/min.) up to 21.6 mg per ml of broth at pH 7.5 and $28^{\circ}C$ after 4 days. The medium consisted of glucose, 100; urea, 10; corn steep liquor, 40; $KH_2PO_4,\;2;\;K_2HPO_4,\;0.5;\; MgSO_4.\;7H_2O,\;0.4;\;antifoam\;S-57,\;1g;\;Fe_2(SO_4)_3.XH-2O,\;10;\; MnCl_2,\;4H_2O,\;10mg;\;biotin,\;30;\;thiamine-HCl,\;100{\mu}g$in 1l of distilled water, and 40 U/ml of penicillin was added after 36 hrs fermentation.

Improvement of Amidase Production by a Newly Isolated Delftia tsuruhatensis ZJB-05174 Through Optimization of Culture Medium

  • The R-amidase production by a newly isolated strain of Delftia tsuruhatensis ZJB-05174 was optimized in this paper. Effects of factors such as carbon sources, nitrogen sources, and inducers on amidase production were investigated. The medium composition was optimized using central composite designs and response surface analysis. The optimal medium components for enhanced amidase production were found to be as follows: glucose, 8.23 g/l; yeast extract, 11.59 g/l; 2,2-(R,S)-dimethylcyclopropane carboxamide, 1.76 g/l; NaCl, 1 g/l; ${KH_2}{PO_4}$ 1 g/l; and ${K_2}{HPO_4}$ 1 g/l. A maximum enzyme production of 528.21 U/l was obtained under the optimized conditions, which was 4.7 times higher than that obtained under initial conditions.
