Investigation on favorable conditions for mycelial growth of Paecilomyces tenuipes (눈꽃동충하초(Paecilomyces tenuipes)의 균사생장조건 구명)

  • Distribution and in vitro growth characteristics of entomopathogenic Paecilomyces species were studied based on the specimens and isolates deposited in Entomopathogenic Fungal Culture Collection (EFCC), Kangwon National University, Chuncheon, Korea. Paecilomyces species were frequently collected from different mountains and valleys and were almost distributed uniformly. Collection period of Paecilomyces species ranged from July to September, with most of the collections at the end of September. P. tenuipes was mostly collected one among different Paecilomyces species. The optimum medium for mycelial growth of P. tenuipes was PDA, whereas in other media such as SDAY and HMA also produced similar growth. The optimum temperature and pH levels were $25^{\circ}C$ and 6.0~7.0, respectively. Best carbon and nitrogen sources were dextrose and tryptone, respectively, while $KH_2PO_4$ was the best mineral source for mycelial growth. Highest mycelial growth was observed when the C/N ratio was 20:1.

  • Various methods for seed treatments have been devised to improve the rate and uniformity of seed germination of gourd (Lagenaria siceraria Standl.). Hydropriming and priming, the imbibition of seeds in distilled water and osmotic solutions respectively, are useful techniques for early establishment of uniform seedlings. In this study, the optimum conditions for hydropriming and priming were determined in gourd (Lagenaria siceraria Standl.) seeds. Effect of hydropriming was compared with those of osmotic priming. Seeds, hydroprimed or primed with 50 mM solution of $KH_2PO_4$ and $KNO_3$ at $30^{\circ}C$ for 2 days, showed shorter $T_{50}$ (number of days to 50% of the final germination percentage) and MDG (mean number of days to germination) than those unprimed. The relative effectiveness of hydropriming and priming in improving the germinability of gourd seeds was more evident when germinated at $20^{\circ}C$ than at $25^{\circ}C$ and $35^{\circ}C$.

  • Preparative HPLC (High-Performance Liquid Chromatography) is mainly used for separating useful component from biological samples. By reversed-phase HPLC packed with preparative packings ($15{\mu}m$), the adsorption characteristics with sample size were investigated. Sample was 5'-GMP, a flavor enhancer, and the composition of mobile phase was 20mM $KH_2PO_4$ solution:methanol (97:3 vol.%). From the experimental results, the effect of sample size on retention factor was negligible, but the peak was asymmetrical above $1{\mu}g$ of sample. In addition, the increase in sample size deteriorated the number of theoretical plates, and at small concentration, the number of theoretical plates was less because of large peak width. In the experimental condition, the adsorption isotherm of 5'-GMP was relatively well represented by Freundlich equation.

Effects of Foliar Application of Potassium Phosphate $(KH_2PO_4)$ on the Enhancement of Marketable Fruit Yields of Fresh Tomato (칼륨 엽면처리가 시설토마토 과실의 상품성 향상에 미치는 영향)

  • This study was performed to determine the optimum concentration of potassium for the foliar application, and its application stage to increase the production of marketable sized tomato. For this purpose, 0.25, 0.5 and 1.0 percent of potassium phosphate $(KH_2PO_4)$ were applied in the leaves of tomatoes by the stage of flowering, fruit setting and fruit development. The control plants were applied with distilled water. The results showed that foliar application with 0.5 percent of potassium phosphate increased the proportion of medium sized fruit of the products by 29.5 percent compared to the controlled plants, which is highest in the treatments. The foliar application of 0.5 percent of potassium phosphate at the fruit setting stage increased the yield of marketable sized fruit by 5.9 percent compared to the controlled plants.

  • In order to elucidate the possibility of artificial production of p. ferulae by solid-state culture, the optimization of culture conditions was carried out. When $NH_4H_2PO_4$ and $CaCO_3$ were used in the cultures using test tube with 30 g of Populus sawdust at $25^{\circ}C{\pm}1$ in the dark, the favored mycelial growth was observed with $1\%$ of $NH_4H_2PO_4$ and the production of polysaccharide was 7.85 mg/100 mg of mycelium with $1\%$ of $CaCO_3$. The mixtures of $80\%$ of Populus Sawdust and $20\%$ of rice bran at $60\%$ of water content were determined to be optimal for the production of fruiting bodies in the sawdust culture. When three treatments containing various ratios of garlic powder were conducted, yields of fruiting bodies were drasti[ally higher than those of Synthetic mixture without garlic powder The highest yield (143 g/bag) was obtained with $7\%$ garlic powder. The yield of synthetic mixture containing $7\%$ of garlic powder was $83\%$ higher than that of Sawdust culture. The reason why garlic powder did support growth was not clear but it is possible that garlic powder might contain effective components for the formation of fruiting body. The optimal synthetic mixture composition consisted of cotton seed $77\%$, lime $6.4\%,\;K_2HPO_4\;0.2\%,\;KH_2PO_4\;0.2\%,\;CaHPO_4\;0.2\%$, corn flour $4\%$, wheat flour $5\%$, and garlic pow-der $7\%$.

  • Biotin 요구성 glutamic acid 생산능을 가진 Br-evibacterium sp.의 isoleucine 영양요구성 변이주가 다량의 L-leucine을 생성함을 발견하였다. 본균주에 관한 탄소원, 질소원, 무기염, vitamin, amino acid 등의 영양물질과 온도, 공기주입량, pH등에 관한 실험으로 다음과 같은 결과를 얻었다. 본균주는 탄소원, 질소원으로서 10% glucose와 4% $(NH_4)_2SO_4를$ 공합하여 $30^{\circ}C,$ 88시간 배양시킨 결과, 액중에 21 mg/ml의 L-leucine이 축적되었다. Fe$^{2+}$$Mn^{2+}$의 동시첨가는 L-leucine생성에 효과적이었으며 thiamine 역시 L-leucine 생성에 효과적이었다. 30ι jar fermentor 에서 폐당밀을 주원료로 한 실험결과, 폐당밀, $(NH_4)/SO_4,$ $KH_2PO_4,$ $MgSO_4ㆍ7H_2O로$ 구성된 액체배지에서 $30^{\circ}C,$ 36시간 배양시킨 것이, 27mg/ml의 L-leucine이 축적되었으며 이때의 대당수율은 16%이었다.

Cultural Conditions for Pretense Production by a feather-Degrading Bacterium, Bacillus megaterium F7-1 (우모분해세균 Bacillus megaterium F7-1에 의한 단백질 분해효소 생산에 영향을 미치는 배양조건)

  • The effects of inorganic salts and feather concentrations on pretense production by Bacillus megaterium F7-1 were investigated. Pretense production was dependent on the presence of phosphates in the medium. Supplementation of medium with calcium ion slightly increased protease production. The highest protease production was obtained at $1.4\%$ feather. The optimal medium contained $2.0\%$ glucose, $0.8\%$ skim milk, $0.06\%\;K_{2}HPO_{4}\%,\;0.04\%\;KH_{2}PO{4},\;0.06\%\;NaCl,\;0.03\%\;MgCl_{2}\cdot6H_{2}O,\;0.002\%\;CaCl_{2}\cdot2H_{2}O,\;and\;1.4\%$ whole feather. By using this optimized medium, increased production of the protease was achieved compared with the cases of using basal medium.

  • A gram(+) bacteria that produced microbial flocculant was isolated from soil and classified as a Bacillus species and named as Bacillus sp. A56. The culture conditions of the strain for fluocculant production were studied in a shake flask. Optimum temperature and initial pH for flcculant production were 30C and 6.5, respectively. The optimized medium has gollowing composition: glucose 20g/l, NH4NO3 0.5g/l, K2HPO4 1.0g/l, KH2PO4 0.8g/l, MgSO4.7H2O 0.2g/l, MnSO4.4-6H2O 0.3g/l, CaCO3 0.5g/l, yeast extract 0.3g/l, tryptone 0.3g/l in tap water.

Control Effect of Phosphate Compounds on Phytophthora Blight and Anthracnose of Pepper (고추 역병과 탄저병에 대한 인산염 화합물의 방제 효과)

  • Antimicrobial activity of 6 phosphate compounds as $H_3PO_3$, $H_3PO_4$, $K_3PO_4$, $K_2HPO_4$, $KH_2PO_4$ and $NH_4H_2PO_4$ against Phytophthora capsici JHAW 1-2 and Colletotrichum acutatum JC24 was investigated in this study. Inhibitory effect on zoospore release, zoosporangia germination and zoospore germination was superior than mycelial growth. Among 6 compounds, $H_3PO_3$ and $H_3PO_4$ showed the best antimicrobial activity against P. capsici JHAW 1-2. Diseases controlling activity of the phosphate compounds tested on seedling and fruit of pepper against Phytophthora blight was also better than those against anthracnose. When $H_3PO_3$ was applied to the pepper seedlings at a concentration of $100{\mu}g\;mL^{-1}$, severe phytotoxicity was occurred. However, with applying $10{\mu}g\;mL^{-1}$ of $H_3PO_3$ showed 100% the disease control efficacy. In case of $100{\mu}g\;mL^{-1}$ $H_3PO_4$ application showed excellent antimicrobial activity against P. capsici JHAW 1-2, and 56.7% of the disease control efficacy with no phytotoxicity. To investigate the control efficacy against anthracnose, conidia suspension was inoculated with non-wound and wound inoculation method on pepper fruit. Among 6 compounds, only $100{\mu}g\;mL^{-1}$ of $H_3PO_3$ and $H_3PO_4$ had a activity of more than 70%, but the control activity on other treatments was minimal or unacceptable.

Optimization of Submerged Culture Conditions for Exo-biopolymer Production by Paecilomyces japonica

  • Optimization of submerged culture conditions for the production of exo-biopolymer from Paecilomyces japonica was studied. Maltose, yeast extract and potassium phosphate were the most suitable sources of carbon, nitrogen, and inorganic salt, respectively, for both production of the exo-biopolymer and mycelial growth. The optimal culture conditions in flask culture were pH 5.0, $25^{\circ}C$ and 150 rpm in a meidum containing of 30 g maltose, 6 g yeast extract, 2 g polypeptone, 0.5 g $K_2HPO_4$, 0.2 g $KH_2PO_4$, 0.2 g $MnSo_4\;{\cdot}\;5H_2O$, 0.2 g $MgSO_4\;{\cdot}\;7H_2O$ in 1-L distilled water. Exo-biopolymer production and mycelial growth in the suggested medium were significantly increased in a 2.5-L jar fermentor, where the maximum biopolymer concentration was 8 g/1.
