Effect of Dietary Antimicrobials on Immune Status in Broiler Chickens

  • This study evaluated the effects of dietary anticoccidial drugs plus antibiotic growth promoters (AGPs) on parameters of immunity in commercial broiler chickens. Day-old chicks were raised on used litter from a farm with endemic gangrenous dermatitis to simulate natural pathogen exposure and provided with diets containing decoquinate (DECX) or monensin (COBN) as anticoccidials plus bacitracin methylene disalicylate and roxarsone as AGPs. As a negative control, the chickens were fed with a non-supplemented diet. Immune parameters examined were concanavalin A (ConA)-stimulated spleen cell proliferation, intestine intraepithelial lymphocyte (IEL) and spleen cell subpopulations, and cytokine/chemokine mRNA levels in IELs and spleen cells. ConA-induced proliferation was decreased at 14 d post-hatch in DECX-treated chickens, and increased at 25 and 43 d in COBN-treated animals, compared with untreated controls. In DECX-treated birds, increased percentages of $MHC2^+$ and $CD4^+$ IELS were detected at 14 d, but decreased percentages of these cells were seen at 43 d, compared with untreated controls, while increased $TCR2^+$ IELs were evident at the latter time. Dietary COBN was associated with decreased fractions of $MHC2^+$ and $CD4^+$ IELs and reduced percentages of $MHC2^+$, $BU1^+$, and $TCR1^+$ spleen cells compared with controls. The levels of transcripts for interleukin-4 (IL-4), IL-6, IL-17F, IL-13, CXCLi2, interferon-${\gamma}$ (IFN-${\gamma}$), and transforming growth factor${\beta}$4 were elevated in IELs, and those for IL-13, IL-17D, CXCLi2, and IFN-${\gamma}$ were increased in spleen cells, of DECX- and/or COBN-treated chickens compared with untreated controls. By contrast, IL-2 and IL-12 mRNAs in IELs, and IL-4, IL-12, and IL-17F transcripts in spleen cells, were decreased in DECX- and/or COBN-treated chickens compared with controls. These results suggest that DECX or COBN, in combination with bacitracin and roxarsone, modulate the development of the chicken post-hatch immune system.

Modulation of Nrf2/ARE and Inflammatory Signaling Pathways by Hericium erinaceus Mycelia Extract

  • Hericium erinaceus is an edible mushroom used as a medicinal food in Asian countries. In this study, the chemopreventive effects of H. erinaceus mycelia hot water extract (HEW) were evaluated. HEW remarkably induced the luciferase activity of the antioxidant response element (ARE), located in the promoter region of phase 2 and antioxidant genes and regulated by nuclear factor E2-related factor 2 (Nrf2). The up-regulation of ARE activity by HEW corresponded with the induction of Nrf2 and the antioxidant enzyme, hemeoxygenase-1. The inhibition of cyclooxygenase-2 (COX-2) activity is a promising effective approach in cancer chemoprevention, and HEW prominently suppressed COX-2 protein expression in HepG2 cells. Furthermore, HEW showed anti-inflammatory activity by modulating inflammatory mediators such as nitric oxide (NO), inducible NO synthase, tumor necrosis factor-${\alpha}$, interleukin-$1{\beta}$, and the transcription factor, nuclear factor-${\kappa}B$, in lipopolysaccharide-stimulated RAW 264.7 cells. These results suggest that H. erinaceus possessed anti-tumor and anti-inflammatory effects via the modulation of Nrf2/ARE and inflammatory signaling pathways, and may therefore have potential use as a natural chemopreventive agent.

Black ginseng-enriched Chong-Myung-Tang extracts improve spatial learning behavior in rats and elicit anti-inflammatory effects in vitro

  • Background: Chong-Myung-Tang (CMT) extract is widely used in Korea as a traditional herbal tonic for increasing memory capacity in high-school students and also for numerous body ailments since centuries. The use of CMT to improve the learning capacity has been attributed to various plant constituents, especially black ginseng, in it. Therefore, in this study, we have first investigated whether black ginseng-enriched CMT extracts affected spatial learning using the Morris water maze (MWM) test. Their molecular mechanism of action underlying improvement of learning and memory was examined in vitro. Methods: We used two types of black ginseng-enriched CMT extracts, designated as CM-1 and CM-2, and evaluated their efficacy in the MWM test for spatial learning behavior and their anti-inflammatory effects in BV2 microglial cells. Results: Our results show that both black ginseng-enriched CMT extracts improved the learning behavior in scopolamine-induced impairment in the water maze test. Moreover, these extracts also inhibited nitric oxide production in BV2 cells, with significant suppression of expression of proinflammatory cytokines, especially inducible nitric oxide synthase, cyclooxygenase-2, and $interleukin-1{\beta}$. The protein expression of mitogen-activated protein kinase and nuclear $factor-{\kappa}B$ pathway factors was also diminished by black ginseng-enriched CMT extracts, indicating that it not only improves the memory impairment, but also acts a potent anti-inflammatory agent for neuroinflammatory diseases. Conclusion: Our research for the first time provides the scientific evidence that consumption of black ginseng-enriched CMT extract as a brain tonic improves memory impairment. Thus, our study results can be taken as a reference for future neurobehavioral studies.

Abrogation of the Circadian Nuclear Receptor REV-ERBα Exacerbates 6-Hydroxydopamine-Induced Dopaminergic Neurodegeneration

  • Parkinson's disease (PD) is a neurodegenerative disease characterized by progressive degeneration of dopaminergic (DAergic) neurons, particularly in the substantia nigra (SN). Although circadian dysfunction has been suggested as one of the pathophysiological risk factors for PD, the exact molecular link between the circadian clock and PD remains largely unclear. We have recently demonstrated that $REV-ERB{\alpha}$, a circadian nuclear receptor, serves as a key molecular link between the circadian and DAergic systems. It competitively cooperates with NURR1, another nuclear receptor required for the optimal development and function of DA neurons, to control DAergic gene transcription. Considering our previous findings, we hypothesize that $REV-ERB{\alpha}$ may have a role in the onset and/or progression of PD. In the present study, we therefore aimed to elucidate whether genetic abrogation of $REV-ERB{\alpha}$ affects PD-related phenotypes in a mouse model of PD produced by a unilateral injection of 6-hydroxydopamine (6-OHDA) into the dorsal striatum. $REV-ERB{\alpha}$ deficiency significantly exacerbated 6-OHDA-induced motor deficits as well as DAergic neuronal loss in the vertebral midbrain including the SN and the ventral tegmental area. The exacerbated DAergic degeneration likely involves neuroinflammation-mediated neurotoxicity. The $REV-erb{\alpha}$ knockout mice showed prolonged microglial activation in the SN along with the over-production of interleukin $1{\beta}$, a pro-inflammatory cytokine, in response to 6-OHDA. In conclusion, the present study demonstrates for the first time that genetic abrogation of $REV-ERB{\alpha}$ can increase vulnerability of DAergic neurons to neurotoxic insults, such as 6-OHDA, thereby implying that its normal function may be beneficial for maintaining DAergic neuron populations during PD progression.

Saprolegnia parasitica Isolated from Rainbow Trout in Korea: Characterization, Anti-Saprolegnia Activity and Host Pathogen Interaction in Zebrafish Disease Model

  • Saprolegniasis is one of the most devastating oomycete diseases in freshwater fish which is caused by species in the genus Saprolegnia including Saprolegnia parasitica. In this study, we isolated the strain of S. parasitica from diseased rainbow trout in Korea. Morphological and molecular based identification confirmed that isolated oomycete belongs to the member of S. parasitica, supported by its typical features including cotton-like mycelium, zoospores and phylogenetic analysis with internal transcribed spacer region. Pathogenicity of isolated S. parasitica was developed in embryo, juvenile, and adult zebrafish as a disease model. Host-pathogen interaction in adult zebrafish was investigated at transcriptional level. Upon infection with S. parasitica, pathogen/antigen recognition and signaling (TLR2, TLR4b, TLR5b, NOD1, and major histocompatibility complex class I), pro/anti-inflammatory cytokines (interleukin $[IL]-1{\beta}$, tumor necrosis factor ${\alpha}$, IL-6, IL-8, interferon ${\gamma}$, IL-12, and IL-10), matrix metalloproteinase (MMP9 and MMP13), cell surface molecules ($CD8^+$ and $CD4^+$) and antioxidant enzymes (superoxide dismutase, catalase) related genes were differentially modulated at 3- and 12-hr post infection. As an anti-Saprolegnia agent, plant based lawsone was applied to investigate on the susceptibility of S. parasitica showing the minimum inhibitory concentration and percentage inhibition of radial growth as $200{\mu}g/mL$ and 31.8%, respectively. Moreover, natural lawsone changed the membrane permeability of S. parasitica mycelium and caused irreversible damage and disintegration to the cellular membranes of S. parasitica. Transcriptional responses of the genes of S. parasitica mycelium exposed to lawsone were altered, indicating that lawsone could be a potential anti-S. parasitica agent for controlling S. parasitica infection.

강황(薑黃) 약침이 Monosodium Iodoacetate 유도 골관절염 흰쥐에 미치는 영향 (Effects of Curcumae Longae Rhizoma Pharmacopuncture on Monosodium Iodoacetate-induced Osteoarthritis Rats)

  • Objectives The purpose of this study was to evaluate the effects of Curcumae Longae Rhizoma pharmacopuncture on the monosodium iodoacetate (MIA)-induced osteoarthritis rats. Methods Osteoarthritis was induced by injection of MIA ($50{\mu}L$ with 80 mg/mL) into knee joint cavity of rats. Rats were divided into 6 groups. Normal group was injected by normal saline into knee joint cavity only. Control group was induced for osteoarthritis by MIA and orally administered with distilled water. Normal Saline group was induced for osteoarthritis by MIA and injected with normal saline $100{\mu}L$. Positive comparison group was injected with MIA and orally administered with indomethacin 5 mg/kg. Curcumae Longae Rhizoma pharmacopuncture low concentration (CL) group was induced for osteoarthritis by MIA and injected with Curcumae Longae Rhizoma pharmacopuncture low concentration $100{\mu}L$. Curcumae Longae Rhizoma pharmacopuncture high concentration (CH) group was induced for osteoarthritis by MIA and injected with Curcumae Longae Rhizoma pharmacopuncture high concentration $100{\mu}L$. Curcumae Longae Rhizoma pharmacopuncture was injected at ST35 and EX-LE4 each group (CL, CH). After that, hind paw weight distribution was measured and oxidative stress biomarker in serum, liver function biomarker in serum, western blot analysis were measured. Histological analysis of knee joint tissue was performed by hematoxylin and eosin staining, Safranin-O staining and Masson's trichrome staining. Results Hind paw weight distribution was significantly improved in both group. alanine aminotransferanse and aspartate aminotransferase were decreased significantly in CH group compare with Indomethacin threated group. Antioxidant enzyme glutathione peroxidase, Catalase and heme oxygenase-1 were increased in CH group compare with control group. Inflammatory cytokine cyclooxygenase-2, inducible nitric oxide synthase and interleukin-1 beta were decreased significantly in CH group. Histological analysis result shows that protective effects of joint and cartilage were observed in both CH and CL groups in a concentration-dependent. Conclusions The result suggest that Curcumae Longae Rhizoma pharmacopuncture has anti-oxidation effect, anti-inflammatory effect and also can prevent progression of osteoarthritis and protect joint cartilage.

RAW264.7 대식세포에서 MAPK 및 NF-κB 신호전달 억제를 통한 rebaudioside A의 항염 효과 (Anti-inflammation effect of rebaudioside A by inhibition of the MAPK and NF-κB signal pathway in RAW264.7 macrophage)

  • 리바우디오사이드 A는 Stevia rebaudiana Bertoni에서 분리된 천연감미료로 널리 알려진 스테비올배당체 중 하나이다. 최근 연구에서 LPS 자극에 의해 활성화된 RAW264.7 마우스 대식 세포에서 리바우디오사이드 A가 인터루킨-$1{\alpha}/1{\beta}$ 같은 염증성 사이토카인 분비를 억제하는 기능이 확인되었다. 그러나 LPS처리 시 리바우디오사이드 A의 항염 활성에 대한염증 억제기작은 정확히 제시하지 못하였다. 따라서 본 연구에서는 리바우디오사이드 A의 LPS 신호전달 메카니즘에서의 항염증 효능을 단백질 수준에서 규명하고자 하였다. NO 생성에 관여하는 iNOS 단백질 발현양을 분석한 결과 리바우디오사이드 A의 $250{\mu}M$ 처리군에서 농도 의존적으로 단백질 발현이 감소하는 것을 확인하였다. 또한 염증 신호에 의한 대표적 핵 전사 인자인 $NF{\kappa}B$의 mRNA 발현량 분석 결과에서도 LPS 처리군에 비해 그 발현양이 감소하였다. 또한 세포질에 존재하는 $NF-{\kappa}B$$I-{\kappa}B$ 복합체는 LPS신호에 의한 $I-{\kappa}B$의 인산화 및 ubiquitination로 인해 $NF-{\kappa}B$가 이탈되기 때문에, 리바우디오사이드 A에 의한 $pNF-{\kappa}B$, $pI-{\kappa}B$의 단백질 발현을 비교 분석한 결과 $NF-{\kappa}B$ 단백질의 인산화가 농도 의존적으로 감소하였고, $I-{\kappa}B$의 인산화 또한 저해되는 것을 확인 하였다. 최종적으로 리바우디오사이드 A는 LPS처리 조건에서 MAPK중 특이적으로 extracellular signal-regulated kinase (ERK1/2)의 인산화를 농도 의존 방식으로 감소시킴으로써 $NF-{\kappa}B$ 조절 기작에 관여함을 알 수 있었다. 따라서 본 연구 결과들을 통해 우리는 리바우디오사이드 A가 RAW264.7 세포에서 LPS에 의해 활성화 되는 MAPK 및 $NF{\kappa}B$의 발현 억제를 통해 염증이 억제될 수 있음을 확인하였다.

대봉감의 항산화, 항염증 및 면역증강 효과 (Beneficial Effects of Daebong Persimmon against Oxidative Stress, Inflammation, and Immunity in vivo)

  • 본 연구는 대표적인 떫은 감 품종인 대봉감과 반시의 항산화, 항염증 및 면역증강 효과를 탐색, 비교하였다. 쥐 모델을 대상으로 4주간 대봉감과 반시의 섭취는 식이섭취량과 체중 변화에 영향을 미치지 않았다. B지역의 대봉감 섭취는 고지방식이 대조군 대비 유의적으로 카탈레이즈 활성 수준을 증가시켰으며, 간 조직의 지질과산화물인 말론디알데하이드 수준은 고지방식이 대조군 대비 A지역 대봉감을 섭취한 군에서 유의적으로 감소하였다. 또한 A지역 대봉감의 섭취는 염증인자인 IL-1 beta의 수준을 고지방식이군 대비 유의적으로 감소시켰다. 또 다른 대표적인 염증인자인 IL-6는 고지방식이 대조군과 비교하여 A지역과 B지역의 대봉감을 섭취한 군에서 유의적으로 감소시켰다. TNF-alpha 수준은 A지역의 대봉감을 섭취한 군에서 고지방식이대조군 대비 유의적으로 감소되었다. A지역 대봉감 섭취는 IgG의 수준을 고지방식이 대조군 대비 유의적으로 증가시켰다. IGF-1의 수준은 고지방식이 대조군과 비교하여 A, B, C 지역 대봉감 섭취군에서 증가하였다. 이상의 결과로부터 대봉감의 섭취는 우수한 항염증 효과와 면역증강 효과가 있을 것으로 사료된다.

비소세포 폐암세포에서 Uteroglobin의 면역 조절 기능에 대한 연구 (The Role of Uteroglobin in the Immunomodulation of Nonsmall Cell Lung Cancer Cells)

  • 연구배경 : 폐암에 대한 거의 대부분의 면역 치료는 실패하였는데, 이는 폐암 자체에 존재하는 면역 억제 기전을 극복하지 못한데 기인하는 것으로 판단된다. Uteroglobin (UG, CCSP)은 항염증 등의 활성을 보인다. 방 법 : A549에 Ad-UG을 처리하고, 상층액의 $PGE_2$ 농도를 측정하였다. RPMI 1640, A549 배양액과 UG 혹은 COX-2 억제제인 NS-398을 처리 후 얻은 폐암세포주 상층액으로 PBMC를 배양 후 Th 1 type cytokines과 Th 2 type cytokines의 농도를 측정하였다. 결 과 : $PGE_2$는 UG이 발현되는 세포주에서 감소하였다. 폐암 세포 배양 배지로 키운 면역 세포의 cytokines가 증가하는 양상을 보였으나, UG등을 처리한 비소세포 폐암주의 배양액은 PBMC의 면역 반응을 정상적으로 유도하였다. 결 론 : 비소세포폐암주 배양액은 PBMC의 면역 반응을 비정상적으로 유도하지만, UG은 $PGE_2$의 분비를 억제함으로써, PBMC의 면역 반응을 강화시킨다.

천궁 및 당귀를 함유한 한방처방제 휘발성 향기추출물의 항염증 효과 (Anti-Inflammatory Effects of Volatile Flavor Extract from Herbal Medicinal Prescriptions Including Cnidium officinale Makino and Angelica gigas Nakai)

  • 본 연구는 항염증 효능을 가지는 한방처방제 휘발성 향기추출물을 이용하여 염증질환 치료 한방화장품 기능성 소재를 개발하고자 하였다. 먼저 전보에서 항염증 효능이 있는 것으로 알려진 천궁, 당귀, 박하 및 애엽을 비롯하여 5가지 생약(적작약, 숙지황, 황금, 인삼 및 감초)으로 구성된 4가지 한방처방제(HH-1: 천궁, 당귀, 적작약, 숙지황, HH-2: 천궁, 당귀, 적작약, 숙지황, 황금, 인삼, 감초, HH-3: 천궁, 당귀, 박하, 애엽, HH-4: 천궁, 당귀, 박하, 애엽, 황금, 인삼, 감초)를 선정한 후 연속수증기증류법(simultaneous steam distillation extraction, SDE)을 이용하여 추출한 휘발성 향기추출물의 항산화 및 항염증 활성을 1,1-diphenyl-2-picrylhydrazyl (DPPH) 및 soybean lipoxygenase (SLO)를 이용하여 각각 측정한 결과, HH-2가 다소 높은 항산화 활성뿐 아니라 SLO 저해활성이 가장 높게 나타났다. 따라서 항염증 활성이 강한 HH-2의 항염증 효능을 보다 상세하게 확인하기 위해 lipopolysaccharide (LPS) 유발한 RAW 264.7 세포를 이용하여 염증 유발매개인자인 nitric oxide (NO)와 prostaglandin $E_2$ ($PGE_2$) 및 interleukin-6 (IL-6) 생성 억제효과를 측정한 결과, HH-2가 강한 NO 생성 억제 효과뿐 아니라 $PGE_2$ 및 IL-6를 강하게 억제하여 우수한 항염증 활성을 나타내었다. 항염증 활성이 높은 HH-2의 휘발성향기성분을 GC-MS로 분석한 결과, eugenol, paeonol, butyl phthalide, ${\beta}$-eudesmol 및 butylidene dihydrophthalide로 확인되었다. 이러한 연구결과로부터 항산화뿐만 아니라 항염증활성이 높은 HH-2를 피부노화 및 염증질환 치료용 한방화장품의 기능성 소재 개발 가능성을 확인하였다.
