Determination of Selenium and Tellurium by HG-AAS in Foods

  • A method has been investigated for the determination of selenium and tellurium in Ginseng, Ganoderma and Garlic using hydride generation atomic absorption spectrometry(HG-AAS). The concentration effects of hydrochloric acid and sodium tetrahydroborate on the hydride generation for the determination of selenium and tellurium were investigated. The method of sample decomposition was also investigated using various mineral acids, such as nitric, perchloric and sulfuric acid in the closed system, and foreign ion effects containing in the samples were studied. The calibration curves of selenium and tellurium were obtained in the range of 0~40 ppb. The detection limits(S/N=2) of selenium and tellurium are 0.1 and 0.2 ppb. Analytical data of selenium and tellurium in Garlic, Ganoderma and Ginseng are 289, 296 and 198 ng/g for selenium and 146, 127 and 110 ng/g for tellurium, respectively.

Biological Assay of Mercury and Cadmium Ions Using DNA Immobilized on a Nanotube Paste Electrodes

  • Bio assay of mercury and cadmium ions were searched using voltammetric analysis using DNA doped carbon nanotube paste electrodes (DCP). The square-wave stripping voltammetryic optimized results indicated working ranges of 1-10.0 $ngL^{-1}$ and 20-100 $ugL^{-1}$, Hg(II) Cd(II) within an accumulation time of 120 seconds, in 0.1-M phosphate buffer solutions of pH 6.3. The relative standard deviations of 5 $ngL^{-1}$ Hg(II) and Cd(II) that observed were 0.14 and 0.22% (n=12), respectively, using optimum conditions. The low detection limit (S/N) was pegged at 0.1 $ngL^{-1}$ ($4.9{\times}10^{-11}M$) Hg(II) and 0.2 $ngL^{-1}$ ($1.77{\times}10^{-10}M$) Cd(II). The developed methods can be applied to assays in biological fish kidneys and water samples.

Electrochemical Detection of Pesticide in Living Plant and Fish Brain Cell

  • The three electrode system was used to detect the pesticide fenitrothion ($C_9H_{12}NO_5PS$. MW=277.24) using cyclic voltammetry (CV) and square wave anodic stripping voltammetry (SWASV). The working electrode was mercury immobilized on a carbon nanotube paste electrode (Hg-CNTPE). At the optimized condition, the limit of detection (LoD) was 0.6 ppt ($2.16{\times}10^{-12}\;M$), and the relative standard deviation was 0.035% (n=15). And there is more sensitive in detecting fenitrothion than common type carbon nanotube paste electrode. When it was implanted into the brain of live fish (carp), the existence of fenitrothion was measured without any destruction or damage of tissue.

The Effect of Vacuum Pressure in Membrane Filtration Systems for the Efficient Detection of Bacteria from Natural Mineral Water

  • The procedures currently used for determining microbiological quality of natural mineral water recommend filtration through membrane filters. In this study, we evaluated the effect of vacuum pressure for the accurate detection of bacteria from water samples seeded with Escherichia coli. We observed that the number of E. coli detected increased with increasing vacuum pressure. In order to examine the retention rate of bacteria in the holes of the membranes under the different pressures, the membrane filters were removed after filtration, washed with sterile water by vortexing, and placed on m-Endo agar plates. With all the filters tested, the number of E. coli retained within the filters at negative 600 mmHg was approximately 10 to $20\%$ higher than that obtained with 100 mmHg. These results demonstrate that the vacuum pressure exerted during the filtration procedure may affect the fixation of bacteria into some portions of openings in the membrane filter.

Residues of Heavy metals in Culture Environment of Ginseng at Gyeongbuk, Korea (경북지역 인삼 재배 환경 중 중금속의 잔류)

  • To obtain the safety evaluation of the ginseng, residues of heavy metals in culture environment of ginseng on Punggi and Sangju, Kyeongbuk are surveyed. The concentration for component of ginseng on Punggi and Sangju were 14.12mg/kg and 15.74mg/kg, respectively. The concentration for general component such as crude fiber, ash, crude lipid, crude protein, carbohydrate, of ginseng were coincided between Punggi and Sangju. The concentration for As, Pb, Cd, and Hg in soil on Punggi were 14.24 ppb, 43.13 ppb, 8.73 ppb and 0.82 ppb, respectively. The concentration for As, Pb, Cd, and Hg in soil on Sangju were 19.20 ppb, 54.82 ppb, 15.90 ppb and 1.04, respectively. Residual heavy metals are not polluted in the soil with culture ginseng on Punggi and Sangju. The concentration for As, Pb, Cd, and Hg with ginseng on Punggi were 29.30ppb, 21.78 ppb, 1.32 ppb and 2.72 ppb, respectively. The concentration for As, Pb, Cd, and Hg with ginseng on Sangju were 3.22 ppb, 24.43 ppb, 1.44 ppb and 4.74 ppb, respectively. Also the detection concentration for As, Pb, Cd, and Hg in ginseng were also lower than the Korea Food & Drug Administration advisory level for heavy metal in herbal medicines. Residual heavy metals are not polluted in the ginseng on Punggi and Sangju at Kyeongbuk, Korea.

Syntheses and Ion Selectivities of Dimeric Rhodamine 6G Chemosensors

  • Novel rhodamine 6G fluorescent chemosensors 1 and 2 for the detection of transition metal cations were synthesized through the condensation of rhodamine 6G ethylenediamine with each of 2-hydroxy-1-naphthaldehyde and 2,6-pyridinedicarbaldehyde, respectively. 1 and 2 were characterized using $^{13}C$ NMR, $^1H$ NMR and mass spectroscopy. Fluorometric and colorimetric measurements involving various metal ions revealed the ring opening of the rhodamine 6G spirocycle framework. In the absence of metal cations, 2 was colorless and non-fluorescent, whereas the addition of metal cations ($Hg^{2+}$ and others) changed the color to pink, accompanied by the appearance of an orange fluorescence. The chemosensors exhibited high selectivity for $Hg^{2+}$ over other divalent first-row transition metals. The complexes of $Hg^{2+}$ with 1 and 2 were successfully isolated. A huge enhancement in the fluorescence for both one- and two-photon excitations makes these compounds suitable candidates to be used for fluorescent labeling of biological systems.

Association of Hypercapnia in the First Week of Life with Severe Intraventricular Hemorrhage in the Ventilated Preterm Infants (기계적 환기 요법을 시행 받은 미숙아에서 고탄산혈증과 뇌실내 출혈의 발생과의 관계)

  • Purpose : The aim of this study was to examine whether hypercapnia during the first seven days of life was associated with severe intraventricular hemorrhage (IVH) in preterm infants requiring mechanical ventilation. Methods : A matched pair analysis was performed for 19 preterm infants with severe IVH(grade$\geq$3) and 38 infants with no severe IVH (normal or grade 1), who required mechanical ventilation for more than seven days. The univariate and multivariate analysis of severe IVH with maximal and minimal $PaCO_2$, averag $PaCO_2$, SD of $PaCO_2$, and difference in the $PaCO_2$ were assessed. The major perinatal factors and maximal ventilator index (VI) were also compared. Results : Infants with severe IVH had a higher maximal $PaCO_2$ (86.1$\pm$18.4 mmHg vs. 60.1$\pm$ 11.6 mmHg, P <0.001) and mean $PaCO_2$ (47.5$\pm$5.6 mmHg vs. 41.2$\pm$6.3 mmHg, P=0.004) and a larger SD or difference in $PaCO_2$ (14.0$\pm$4.4 mmHg vs. 9.0$\pm$2.4 mmHg; 60.3$\pm$20.9 mmHg vs. 35.5$\pm$11.8 mmHg, P <0.001). However the minimal $PaCO_2$ values did not differ between the groups. Disseminated intravascular coagulation, pulmonary hemorrhage, and the air leak syndrome were more frequent in the IVH group than in the controls. The maximal VI on each day was higher in the IVH group. The multivariate logistic regression analysis after controlling for bleeding tendency showed that the air leak syndrome, maximal VI, and maximal $PaCO_2$ were independently associated with severe IVH [OR, 1.324 (95% CI, 1.011-1.733; P=0.041)]. Conclusion : Extreme hypercapnia was significantly associated with severe IVH in preterm infants, after adjustment for major perinatal risk factors. Frequent monitoring of the $PaCO_2$ may be important for early detection of inadvertent hypercapnia and prompt correction of high PaCOS levels.

Quantitative Analysis of Heavy Metals in Packaging Papers

  • This study was carried out to investigate various heavy metal contents in packaging papers by pre-treatments for ICP-ES (Inductively Coupled Plasma Emission Spectrometry) analysis. Pre-treatment methods of heavy metals in this study include extraction, migration and decomposition methods (dry ashing, $HNO_3-HClO_4-HF,\;HNO_3,\;and\;H_2SO_4-HNO_3$). Test results were compared with conventional extraction (water) and migration (3% acetic acid) methods. The five representative heavy metals (Cd, As, Pb, Cr and Hg) were analyzed. For Cd, Hg, and As, the results were below detection limit of the instrumental technique. It was considered that the migration test was a better method compared to extraction test, but all the decomposition methods showed much higher detection values than the extraction or migration test. In case of recycled corrugated containers, 3% acetic acid solution extracted about 25% of chromium and 30% of lead compared to the content by decomposition methods. Among all decomposition methods, the nitric acid - perchloric acid - hydrofluoric acid treatment brought a slightly higher detection value than others, but there was no significant difference among them except sulfuric acid - nitric acid method.

A study on electrical response property of photoconductor film for x-ray imaging sensor (X선 영상센서 적용을 위한 광도전체 필름의 전기적 응답특성 연구)

  • Recently, the compound materials(a-Se, $HgI_2$, PbO, CdTe, $PbI_2$, etc.) that are used in flat panel x-ray imager have been studied for digital x-ray imaging. In this paper, the signal detection properties of $HgI_2$ and a-Se conversion layer, are compared. The thick $HgI_2$ film is fabricated by special particle-in-binder method and the conventional vacuum thermal evaporation is used for a deposition of a-Se film. And an electrical characteristic measurements were investigated about leakage current, signal response property and x-ray sensitivity. From the experimental results show that the $HgI_2$ film has a low operation voltage and high signal generation than that of a-Se.

Study on the Applicability of Semiconductor Compounds for Dose Measurement in Electron Beam Treatment (전자선 치료 분야의 선량 측정을 위한 반도체 화합물의 적용가능성 연구)

  • In this study, it was intended to replace the existing plane parallel ionization chamber, which requires cross-calibration in electron beam treatment. The semiconductor compounds HgI2 was fabricated as detector, and the characteristics of HgI2 detector for the 6, 9 and 12 MeV electron beam was analyzed in the linear accelerator. It was also intended to evaluate the possibility of substitution with existing detectors and their applicability as electron beam dosimetry and to use them as a basic study of the development of electronic beam dosimeter. As a result of reproducibility, RSD was 0.4246%, 0.5054%, and 0.8640% at 6, 9, and 12 MeV energy, respectively, indicating that the output signal was stable. As a result of the linearity, the R2 was 0.9999 at 6 MeV, 0.9996 at 9 MeV, and 0.9997 at 12 MeV showed that the output signal is proportional to HgI2 as the dose is increased. The HgI2 detector of this study is highly applicable to electron beam measurement, and it may be used as a basic research on electron beam detection.
