Production of lactic acid by Lactobacillus paracasei isolated from button mushroom bed

  • A galactose fermentation bacterium producing lactose from red seaweed, which was known well to compromise the galactose as main reducing sugar, was isolated from button mushroom bed in Buyeo-Gun, Chungchugnamdo province. The lactic acid bacteria MONGB-2 was identified as Lactobacillus paracasei subsp. tolerans by analysis of 16S rRNA gene sequence. When the production of lactic acid and acetic acid by L. paracasei MONGB-2 was investigated by HPLC analysis with various carbohydrates, the strain MONGB-2 efficiently convert the glucose and galactose to lactic acid with the yield of 18.86 g/L and 18.23 g/L, respectively and the ratio of lactic acid to total organic acids was 1.0 and 0.91 g/g for both substrates. However, in the case of acetic acid fermentation, other carbohydrates besides galactose and red seaweed hydrolysate could not be totally utilized as carbon sources for acetic acid production by the strain. The lactic acid production from glucose and galactose in the fermentation time courses was gradually enhanced upto 60 h fermentation and the maximal concentration reached to be 16-18 g/L from both substrates after 48 h of fermentation. The initial concentration of glucose and galactose were completely consumed within 36 h of fermentation, of which the growth of cell also was maximum level. In addition, the bioconversion of lactic acid from the red seaweed hydrolysate by L. paracasei MONGB-2 appeared to be about 20% levels of the initial substrates concentration and this results were entirely lower than those of galactose and glucose showed about 60% of conversion. The apparent results showed that L. paracasei MONGB-2 could produce the lactic acid with glucose as well as galactose by the homofermentation through EMP pathway.

  • In this study, an ultrasound-assisted micellar extraction process was developed to efficiently purify the anticancer substance paclitaxel from the plant cell Taxus chinensis. The problem of many extraction steps and long phase separation time in the traditional micellar process could be dramatically improved. The highest paclitaxel yield (~96%, extracted twice) was obtained at 180 W of ultrasonic power and 1.5 h of ultrasonic irradiation time, which was 24.7% higher than that of the traditional method. In addition, the partition coefficient (K) showed a maximum value (24.0) at 180 W of ultrasonic power and 1.5 h of irradiation time. There was no significant difference in the purity of paclitaxel, and the purity of initial paclitaxel (6.81%) increased to 22.0% after purification. Compared to the traditional method, the phase separation time of the back extraction decreased by 40.7-56.2% (ultrasonic power 80 W), 46.3-67.6% (ultrasonic power 180 W), and 51.9-67.6% (ultrasonic power 250 W), respectively. The phase separation time decreased as the ultrasonic power (80-250 W) and irradiation time (0.5-2.5 h) increased.

Effect of Different Rumen-degradable Carbohydrates on Rumen Fermentation, Nitrogen Metabolism and Lactation Performance of Holstein Dairy Cows

  • Four multiparous lactating Holstein cows fitted with rumen cannulae were fed diets varying in the amount and source of rumen-degradable carbohydrates (starch vs. sucrose) to examine their effects on rumen fermentation, nitrogen metabolism and lactation performance. A $4{\times}4$ Latin square with four diets and four periods of 28 days each was employed. Corn starch and sucrose were added to diets and corn starch was replaced with sucrose at 0 (0 S), 2.5 (2.5 S), 5.0 (5.0 S) 7.5% (7.5 S) of diet dry matter in a total mixed ration (TMR) containing 60% concentrate and 40% forage (DM basis). Replacing corn starch with sucrose did not affect (p>0.05) ruminal pH which averaged 6.41, but the ruminal pH for 7.5 S decreased more rapidly at 2 h after morning feeding compared with other treatments. Sucrose reduced ($p{\leq}0.05$) ruminal $NH_3-N$ concentration (13.90 vs. 17.09 mg/dl) but did not affect peptide-N concentration. There was no dietary effect on total volatile fatty acids (110.53 mmol/L) or the acetate to propionate ratio (2.72). No differences (p>0.05) in molar proportion of most of the individual VFA were found among diets, except for the molar proportion of butyrate that was increased ($p{\leq}0.05$) with the inclusion of sucrose. Total branched chain volatile fatty acids tended to increase ($p{\geq}0.051$) for the control treatment (0 S) compared with the 7.5 S treatment. Dry matter intake, body weight changes and digestibility of DM, OM, CP, NDF and ADF were not affected by treatments. Sucrose inclusion in the total mixed ration did not affect milk yield, but increased milk fat and total solid percentage ($p{\leq}0.05$). Sucrose tended ($p{\geq}0.063$) to increase milk protein percentage (3.28 vs. 3.05) and reduced ($p{\leq}0.05$) milk urea nitrogen concentration (12.75 vs. 15.48 mg/dl), suggesting a more efficient utilization of the rapidly available nitrogen components in the diet and hence improving nitrogen metabolism in the rumen.

Effects of LED(Light Emitting Diode) Photoperiod and Light Intensity on Growth and Yield of Taraxacum coreanum Nakai in a Plant Factory (식물공장 내 광주기 및 광도가 흰민들레의 생육과 수량에 미치는 영향)

  • The objective of this study was to examine the effect of photoperiod and light intensity of RBW LED (red:blue:white = 2:1:1) on the growth of Taraxacum coreanum Nakai in a fully artificial light type plant factory. 3 photoperiods and 4 light intensity were used respectively in a fully artificial light type plant production system. Plants were cultured with three photoperiods and four light intensity regimes (conditions) for 270 and 120 days, respectively, using nutrient film technique (NFT) or aeroponics culture methods. For each photoperiod, the total leaves per plant harvested 8 times in all cultivation period was 224 in the 16/8(day/light) photoperiod that had no significant difference from 220 in the 12/12 photoperiod and the lowest number of leaves was 151 occurred in the 8/16 photoperiod, which means that the longer photoperiod, the more leaves harvest. Total fresh weight of above ground was the high in order of in 16/8 photoperiod as 125g, 12/12 photoperiod as 91g, 8/16 photoperiod as 56g. For each light intensity, the total leaves per plant harvested 4 times in all cultivation period was the great in order of $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 123, $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 107, $100{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 95, $50{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 56 which was the smallest number of total leaves harvest. Total fresh weight of above ground per plant was the high in order of $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 43.6g, $100{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 34.6g, $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 32.2g, $50{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ as 18.2g. From these results, it was concluded that photoperiod of 16/8 and light intensity at $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ can be used as the light condition of RBW LED (red:blue:white = 2:1:1) for optimal growth of Taraxacum coreanum Nakai in a fully artificial light type plant factory.

Composition of Nutrient Solution According to Soil Texture in Fertigation Culture of Cucumber (Cucumis sativus L.) (오이 관비재배 시 토성에 따른 적정 배양액 조성)

  • To determine suitable composition of nutrient solution according to soil textures in fertigation culture of cucumber using three strengths (S) of Yamasaki cucumber recipe, chemical changes of soil, growth characteristics and yield of cucumber were investigated. Electric conductivity of drainage solution was risen in all treatments, pH of loam soil was generally optimum level and that of sandy soil was high level. Photosynthetic rate in loam soil supplied the 1/2 S and transpiration rate in sandy soil supplied the 1.0 S were most low. Diffusive resistance in sandy soil supplied the 1.0 S was high. Chlorophyll contents was higher concentrations by the kinds of soil. Amount of drainage solution in sandy loam soil supplied the 1/2 S and loam soil supplied the 1.0 S were most much and little, respectively. Water absorption rate was the opposition to amount of drainage solution. Nutrient contents in soil except calcium were most high in the 1.0 S by the kinds of soil. Nutrient contents in leaves, nitrogen in sandy soil supplied 1.0 S, phosphorus in loam soil supplied 1/2 S, potassium in two soil supplied 1.0 S, calcium in loam supplied 1/2 S and sandy loam soil supplied 1.0 S, magnesium in loam soil supplied the 1/2 S was high. The growth and yield of fruit were more in loam than in sandy loam soil. Therefore, the suitable compositions of macro-nutrients for fertigation culture of cucumber were determined in loam and sandy loam soils as follows: In loam soil, they were $NO_3$-N 12.3, $NH_4$-N 1.0, P 3.0, K 5.9, Ca 5.7, Mg $3.5\;me{\cdot}L^{-1}$, whereas in sandy loam soil $NO_3$-N 11.7, $NH_4$-N 1.0, P 3.0, K 5.9, Ca 4.9, Mg $3.2\;me\;L^{-1}$.

Studies on the Rice Yield Decreased by Ground Water Irrigation and Its Preventive Methods (지하수 관개에 의한 수도의 멸준양상과 그 방지책에 관한 연구)

  • The purposes of this thesis are to clarify experimentally the variation of ground water temperature in tube wells during the irrigation period of paddy rice, and the effect of ground water irrigation on the growth, grain yield and yield components of the rice plant, and, furthermore, when and why the plant is most liable to be damaged by ground water, and also to find out the effective ground water irrigation methods. The results obtained in this experiment are as follows; 1. The temperature of ground water in tube wells varies according to the location, year, and the depth of the well. The average temperatures of ground water in a tubewells, 6.3m, 8.0m deep are $14.5^{\circ}C$ and $13.1^{\circ}C$, respercively, during the irrigation period of paddy rice (From the middle of June to the end of September). In the former the temperature rises continuously from $12.3^{\circ}C$ to 16.4$^{\circ}C$ and in the latter from $12.4^{\circ}C$ to $13.8^{\circ}C$ during the same period. These temperatures are approximately the same value as the estimated temperatures. The temperature difference between the ground water and the surface water is approximately $11^{\circ}C$. 2. The results obtained from the analysis of the water quality of the "Seoho" reservoir and that of water from the tube well show that the pH values of the ground water and the surface water are 6.35 and 6.00, respectively, and inorganic components such as N, PO4, Na, Cl, SiO2 and Ca are contained more in the ground water than in the surface water while K, SO4, Fe and Mg are contained less in the ground water. 3. The response of growth, yield and yield components of paddy rice to ground water irrigation are as follows; (l) Using ground water irrigation during the watered rice nursery period(seeding date: 30 April, 1970), the chracteristics of a young rice plant, such as plant height, number of leaves, and number of tillers are inferior to those of young rice plants irrigated with surface water during the same period. (2) In cases where ground water and surface water are supplied separately by the gravity flow method, it is found that ground water irrigation to the rice plant delays the stage at which there is a maximum increase in the number of tillers by 6 days. (3) At the tillering stage of rice plant just after transplanting, the effect of ground water irrigation on the increase in the number of tillers is better, compared with the method of supplying surface water throughout the whole irrigation period. Conversely, the number of tillers is decreased by ground water irrigation at the reproductive stage. Plant height is extremely restrained by ground water irrigation. (4) Heading date is clearly delayed by the ground water irrigation when it is practised during the growth stages or at the reproductive stage only. (5) The heading date of rice plants is slightly delayed by irrigation with the gravity flow method as compared with the standing water method. (6) The response of yield and of yield components of rice to ground water irrigation are as follows: \circled1 When ground water irrigation is practised during the growth stages and the reproductive stage, the culm length of the rice plant is reduced by 11 percent and 8 percent, respectively, when compared with the surface water irrigation used throughout all the growth stages. \circled2 Panicle length is found to be the longest on the test plot in which ground water irrigation is practised at the tillering stage. A similar tendency as that seen in the culm length is observed on other test plots. \circled3 The number of panicles is found to be the least on the plot in which ground water irrigation is practised by the gravity flow method throughout all the growth stages of the rice plant. No significant difference is found between the other plots. \circled4 The number of spikelets per panicle at the various stages of rice growth at which_ surface or ground water is supplied by gravity flow method are as follows; surface water at all growth stages‥‥‥‥‥ 98.5. Ground water at all growth stages‥‥‥‥‥‥62.2 Ground water at the tillering stage‥‥‥‥‥ 82.6. Ground water at the reproductive stage ‥‥‥‥‥ 74.1. \circled5 Ripening percentage is about 70 percent on the test plot in which ground water irrigation is practised during all the growth stages and at the tillering stage only. However, when ground water irrigation is practised, at the reproductive stage, the ripening percentage is reduced to 50 percent. This means that 20 percent reduction in the ripening percentage by using ground water irrigation at the reproductive stage. \circled6 The weight of 1,000 kernels is found to show a similar tendency as in the case of ripening percentage i. e. the ground water irrigation during all the growth stages and at the reproductive stage results in a decreased weight of the 1,000 kernels. \circled7 The yield of brown rice from the various treatments are as follows; Gravity flow; Surface water at all growth stages‥‥‥‥‥‥514kg/10a. Ground water at all growth stages‥‥‥‥‥‥428kg/10a. Ground water at the reproductive stage‥‥‥‥‥‥430kg/10a. Standing water; Surface water at all growh stages‥‥‥‥‥‥556kg/10a. Ground water at all growth stages‥‥‥‥‥‥441kg/10a. Ground water at the reproductive stage‥‥‥‥‥‥450kg/10a. The above figures show that ground water irrigation by the gravity flow and by the standing water method during all the growth stages resulted in an 18 percent and a 21 percent decrease in the yield of brown rice, respectively, when compared with surface water irrigation. Also ground water irrigation by gravity flow and by standing water resulted in respective decreases in yield of 16 percent and 19 percent, compared with the surface irrigation method. 4. Results obtained from the experiments on the improvement of ground water irrigation efficiency to paddy rice are as follows; (1) When the standing water irrigation with surface water is practised, the daily average water temperature in a paddy field is 25.2$^{\circ}C$, but, when the gravity flow method is practised with the same irrigation water, the daily average water temperature is 24.5$^{\circ}C$. This means that the former is 0.7$^{\circ}C$ higher than the latter. On the other hand, when ground water is used, the daily water temperatures in a paddy field are respectively 21.$0^{\circ}C$ and 19.3$^{\circ}C$ by practising standing water and the gravity flow method. It can be seen that the former is approximately 1.$0^{\circ}C$ higher than the latter. (2) When the non-water-logged cultivation is practised, the yield of brown rice is 516.3kg/10a, while the yield of brown rice from ground water irrigation plot throughout the whole irrigation period and surface water irrigation plot are 446.3kg/10a and 556.4kg/10a, respectivelely. This means that there is no significant difference in yields between surface water irrigation practice and non-water-logged cultivation, and also means that non-water-logged cultivation results in a 12.6 percent increase in yield compared with the yield from the ground water irrigation plot. (3) The black and white coloring on the inside surface of the water warming ponds has no substantial effect on the temperature of the water. The average daily water temperatures of the various water warming ponds, having different depths, are expressed as Y=aX+b, while the daily average water temperatures at various depths in a water warming pond are expressed as Y=a(b)x (where Y: the daily average water temperature, a,b: constants depending on the type of water warming pond, X; water depth). As the depth of water warning pond is increased, the diurnal difference of the highest and the lowest water temperature is decreased, and also, the time at which the highest water temperature occurs, is delayed. (4) The degree of warming by using a polyethylene tube, 100m in length and 10cm in diameter, is 4~9$^{\circ}C$. Heat exchange rate of a polyethylene tube is 1.5 times higher than that or a water warming channel. Effects of Expeller Cake Fertilizer on Soil Properties and Tah Tasai Chinese Cabbage Yield in Organic Greenhouse Farm (유기농 시설하우스 토양에서 유박 시용이 토양특성 및 다채 생육에 미치는 영향)

  • BACKGROUND: To evaluate the application level of expeller cake fertilizer(ECF), we have investigated soil chemical properties, leaf mineral contents and yield of tah tasai Chinese cabbage in a plastic film greenhouse. METHODS AND RESULTS: Five levels of fertilizer were applied as 0%(ECF 0), 50%(ECF 50), 75%(ECF 75), 100% (ECF 100) and 150%(ECF 150) by base 1,848 kg/ha of ECF. In 2012, tah tasai Chinese cabbage was planted on April 28 in a silt loam soil and harvested on July 12. Commercial yields were measured 10 times from May 10 to July 12. Electrical conductivity (2.24~3.09 dS/m), available $P_2O_5$(484~581 mg/kg) and exchangeable cations($K^+$, $Ca^{2+}$ and $Mg^{2+}$) were tended to increase by the application of ECF. However, the range of those was not significant. The contents of T-N, K, Ca and P of tah tasai Chinese cabbage leaves were 62.2~66.5 g/kg, 44.3~48.7 g/kg, 5.1~5.9 g/kg and 5.6~6.2 g/kg, respectively. The nitrogen utilization rate of tah tasai Chinese cabbage was 39.4~51.6%, and it was decreased with increased application amount of ECF. The yield of tah tasai Chinese cabbage was 9,806 to 12,730 kg/ha on the basis of application amount of ECF and it was not increased in spite of increased ECF. CONCLUSION: The optimum dose of application of ECF for cultivation of tah tasai Chinese cabbage was ranged from 924 kg/ha(as ECF 50) to 1,386 kg/ha(as ECF 75). Environment-friendly and economical amount of applied fertilizer is more important than yield for cultivation of tah tasai Chinese cabbage.

  • This experiment has been carried out to determine the pH dependent cation exchange capacity concerning the sorption phenomenon of long-lived radionuclides contained in low-level liquid radioactive waste on various clay minerals. The pH dependent cation exchange capacity determined by Sawhney's method are used to the analysis of sorption phenomenon. About 70 percent of the total cation exchange capacity is contributed by the pH dependent CEC due to the negative charge originated naturally in clays in case of clinoptilolite, vermiculite and sodalite. It is sugested in this test that the high neutral salt CEC, that is, highly charged clays would show good fixation yield. The removal of radionuclides at the pH range more than pH 9 is considered the hydroxide precipitation of metal ion rather than the cation exchange. The Na-clay prepared by the method of successive isomorphic substitution with electrolyte showed a considerable improvement in removal efficiency for the decontamination.

Quality characteristics of high-acidity vinegar prepared with grape juice (포도과즙을 이용하여 제조한 고산도 식초의 품질특성)

  • This study was conducted to develop a high-acidity vinegar production (over TA 10%) technology using grape juice to reduce the importation of high-acidity vinegar. The manufacturing condition's effect on the quality of high-acidity vinegar using pure grape juice without the addition of other nutrients for fermentation was investigated. Twelve percent acidity in vinegar was obtained from grape wine with 6% alcohol content. The acetic-acid yield from grape wine decreased when the wine's initial alcohol content was high, which extended the induction time. The pH value was similar in all the treatment groups. The sugar content of the 1st-stage fermentation (1st AAF) was proportional to the initial alcohol content whereas in the 2nd-stage fermentation (2nd AAF), the sugar content was highest in the 6%-alcohol treatment. The major organic acids of the high-acidity grape vinegar included tartaric acid, malic acid, and citric acid. The acid content of the high-acidity initial alcohol group was higher than that of the low-acidity initial alcohol group due to the alcohol content added by the fed-batch and acetic-acid yield difference. The ethyl alcohol content was 364~6,091 ppm (the main alcohol while the others had only traces in all the groups). In conclusion, it was possible to manufacture 12% high-acidity vinegar without the addition of an external nutrient source to grape wine containing 6% initial alcohol content. Finally, a complementary study will be required to shorten the fermentation period through the fed-batch-style addition of alcohol for the purpose of industrialization.

The synthesis of the first target compound, 4,5-dihydro-6-(4-methoxy-3-methylphenyl)-3(2H)-pyridazinone (1), was achieved by Friedel-Crafts acylation of o-cresyl methyl ether with succinic anhydride and subsequent cyclization of the intermediary g-keto acid with hydrazine hydrate. Condensation of compound 1 with aromatic aldehydes in the presence of sodium ethoxide affords the corresponding 4-substituted benzyl pyridazinones (3a-d). The dihydropyridazinone 1 underwent dehydrogenation upon treatment with bromine/acetic acid mixture to give (4). Pyridazine (5) has been synthesized upon the reaction of pyridazinone (1) with 1,3-diphenyl-2-propen-1-one under the Michael addition reaction. N-dialkylaminomethyl derivatives 6a-b have been obtained from the reaction of pyridazinone 1 with formaldehyde and secondary amine, whereas reaction of 1 with formaldehyde gives N-hydroxymethyl derivative (7). This study also includes the synthesis of the 3-chloropyridazine derivative 8 in excellent yield by heating pyridazinone 3b in phosphorus oxychloride. The behaviour of the chloro derivative toward sodium azide, benzyl amine and anthranilic acid was also studied. The proposed structures of the products were confirmed by elemental analysis, spectral data and chemical evidence.
