근관세척액의 항균효과에 관한 연구 (A STUDY ON THE ANTIMICROBIAL EFFECT OF IRRIGATION SOLUTIONS)

  • The purpose of this study was to investigate antimicrobial effect of several irrigation solutions on 7 anaerobes and 2 aerobes, which are found frequently in infected root canals. The antimicrobial effects of normal saline, 3% $H_2O_2$, 0.5% & 3.5% NaOCl, 10% & 50% citric acid and mixed solutions of 3% $H_2O_2$ plus 3.5% NaOCl were compared. No. 80 paper points dipped in bacterial broth were soaked in each irrigation solutions and moved into thioglycolate broth, subcultured in agar plate for bacterial growth. The results were as follows: 1. Normal saline had no antimicrobial effect. 2. Mixed solutions of 3% $H_2O_2$ plus 3.5% NaOCl, 10% citric acid had relatively weak antimicrobial effect. 3.3% $H_2O_2$, 50% citric acid, 0.5% NaOCl showed relatively strong antimicrobial effect. 4. 3.5% NaOCl had the strongest antimicrobial effect among used 7 irrigation solutions.

티타니아에 담지된 귀금속촉매의 H2O-H2 전처리에 따른 휘발성유기화합물 산화에 미치는 영향 (A Effect of H2O-H2 Pretreatment on VOCs Oxidation over Noble Catalysts on Titania)

  • 연구에서는 $TiO_2$ 담체에 귀금속(Pd, Ru, Ir)을 담지시켜 촉매를 제조하였다. 금속분산의 향상을 위해 $H_2O-H_2$ 전처리 기술이 이용되었고, 반응물로서 xylene, toluene 그리고 MEK를 사용하였다. 단일금속과 합금금속의 촉매들은 함침법에 의해 준비하였고, XRD, XPS 분석을 통하여 특성 분석하였다. Pd-Ru, Pd-Ir 촉매는 다양한 활성점을 나타내었고, 그것은 Pd의 metal 영역를 강화시켰다. 합금금속으로 이루어진 촉매가 단일 금속으로 이루어진 촉매에 비해 VOCs 전환율이 더 높았다. $H_2O-H_2$ 전처리 기술은 Pd 입자를 고르게 분산시켰다. 이는 촉매의 효율을 증진시키는 효과를 보여주었다. 본 연구에서 Pd에 소량의 Ru, Ir 첨가는 VOCs의 산화반응을 증진시켰고, $TiO_2$ 담체상에서 $H_2O-H_2$ 전처리는 VOCs의 제거효율을 증진시켰다.

  • DNA damage induced by reactive oxygen species (ROS) seems to play an important role in the induction of mutation and cancer. Hydrogen peroxide $(H_2O_2)$ has been shown to induce a variety of genetic alterations, probably by the generation of hydroxyl radicals via Fenton reaction. In this study, we examined the ability of medicinal herbs in the suppression of $H_2O_2$-induced mutagenesis. Human fibroblast GM00637 cells were treated with $H_2O_2$ in the presence or absence of medicinal herbs, and $H_2O_2$-induced mutant frequency was measured at the hypoxanthine guanine phosphoribosyl transferase (HPRT) locus. Treatment of cells with various doses of $H_2O_2$ caused a significant increase of the HPRT mutant frequency. However, pretreatment of cells with several medicinal herbs reduced $H_2O_2$-induced mutant frequency. The strong antimutagenic effects were observed from the methylene chloride and ethyl acetate fractions of Selaginella tamariscina, Panax ginseng, and Angelica acutiloba; ethyl acetate fractions of Rehmania glutinosa, Leonurus sibiricus, Curcuma zedoaria and Commiphora molmol; butanol fractions of Scutellaria barbata, Tribulus terrestris, Curcuma zedoaria, Cyperus rotundus and Carthamus tinctorius, which were more than 60% inhibition of $H_2O_2$-induced mutant frequency at the HPRT locus.

$H_2O_2$로 유발된 Neuro2A 신경세포고사에 대한 줄풀의 억제 효과 (Inhibition Effect of Zizania latifolia on Apoptosis Induced by $H_2O_2$ in Neuro2A Cell)

  • The purpose of this study was to examine the inhibition effect of Zizania latifolia that has been used heart disease, Diabetes Mellitus and Skin disease for a long time on apoptosis induced by $H_2O_2$ in Neuro2A cell. Neuro2A cells were cultivated in RPMI(GibcoBRL) with $5\%$, FBS and treated with $H_2O_2$, and Zizania latifolia. We measured the cell viability and analyzed DNA fragmentation. Activity of PARP, Cytochrome C, caspase-9, caspase-3, p53, p21, Bax and Bcl-2 in the cell was examined by using western blot. The cell viability in Zizania latifolia treatment (60ug/ml<) decreased significantly compared with that of none treatment. (p<0.001) Zizania latifolia increased cell viability about twice as much as that being injury by $H_2O_2$. (Zizania Latifolia 20ug/ml, $H_2O_2$ 200uM, P<0.001) DNA fragmentation developed by $H_2O_2$, but was not developed in Zizania latifolia treatment. PARP, Cytochrome C, caspase-9 and caspase-3 activated all by $H_2O_2$ but were not activated in Zizania latifolia treatment.. P53, P2l and Bu activated by $H_2O_2$, and Bcl-2 got into inactivation. But the opposite results appeared in Zizania latifolia treatment. In conclusion, these results suggest that Zizania latifolia inhibit the development of DNA fragmentation and apoptosis by $H_2O_2$ and the antioxidant action of Zizania latifolia is effective. More researches about effect of Zizania latifolia are considered to need.

  • Further improvement of existing $H_{2}O/LiBr$ absorption refrigeration machine is absolutely neces-sary to promote the utilization of gas-cooling system Among various methods to improve the per-formance of the absorption refrigeration machine this research has focused on the use of a new working fluid that has better properties than the existing $H_{2}O/LiBr$ working fluid. In the series of the research, $H_{2}O/LiBr+HO{(CH_{2})}_{3}OH$ system was selected as the most promisable candidate. The absorption refrigeration machine is water-cooled double-effect, $H_{2}O/LiBr+HO{(CH_{2})}_{3}OH$ sys-tem with series flow type. In this study we found out the characteristic of new working solution through the cycle simulation and compared the result with that of LiBr solution to evaluate. Theoptimum designs and operating conditions were determined based on the operating constraints and the coefficient of performance. Results demonstrate that new working fluid subsrantially increases COP by as much as 10% and has a wider working range with 8% higher crystallization limits compared to the conventional $H_{2}O/LiBr$.

Vanadate의 혈소판 응집작용과 Vanadium Yeast의 억제효과 (Vanadate-induced Platelet Aggregation and Inhibition Effect of Vanadium Yeast)

  • It has been well known that vanadium shows various physiological and pharmacological properties such as an insulin-mimetic effect. In view of the reported toxic effects there is the problems that the safety margin is narrow because of its strong toxicity, Vanadate was tested for its ability to cause blood aggregation. Although vanadate or $H_2O$$_2$ alone had little effect on platelet aggregation, treatment of vanadate and $H_2O$$_2$ together induced platelet aggregation indicated that it was occurred by pervandate or hydroxyl radical produced from the reaction of vanadate and $H_2O$$_2$. It was dependent on extracellular $Ca^{2+}$ion. Platelet aggregation caused by vanadate and $H_2O$$_2$ was inhibited by ascorbic acid, tocopherol, catalase, mannitol, and Tiron. In contrast to vanadate, vanadium yeast prepared by uptaking vanadate in yeast cells did not induce platelet aggregation in the presence of $H_2O$$_2$.>.

배양 혈관 내피세포에서 Hydrogen Peroxide에 의한 투과성 증가에 미치는 성향정기산의 효과 (Protection by Sunghyangchungisan against Hydrogen Peroxide-induced Increase in Endothelial Permeability)

  • Objectives : Hindered barrier function of vascular endothelium has been implicated in the initiation and progression of degenerative vascular diseases such as atherosclerosis. In this study, the effect of Sunghyangchungisan(SHCS) as a protectant against oxidant-induced destruction of endothelial barrier function was assessed. Methods : Toward this end, endothelial cells derived from the human umbilical vein were cultured as monolayers on permeable membrane filters. Endothelial permeability was monitored by measuring transendothelial electrical resistance and movement of low density lipoprotein (LDL) across the endothelial monolayer. Results : Along with increased movement of LDL, $H_2O_2$-induced increase in endothelial permeability was paralleled by a decrease in transendotheliaI electrical resistance. The effect of $H_2O_2$ was mimicked by phorbol 12-myristate 13-acetate (PMA), a potent activator of proteinkinase C. Calphostin-C, a protein kinase C inhibitor, effectively blocked the increase in endothelial permeability induced by $H_2O_2$ or PMA, indicating that activation of protein kinase C is associated with the $H_2O_2-induced$ permeability change. SHCS effectively protected the endothelial monolayer against $H_2O_2-induced$ increase in permeability, whereas, it did not affect PMA-induced change. Forskolin, a potent activator of adenylyl cyclase, antagonized $H_2O_2$ to increase endothelial permeability. In addition, in ${H_2O_2}-treated$ cens, intracenular cAMP concentration was significantly decreased, indicating that impaired cAMP production as well as activation of proteinkinase C is a mechanism underlying ${H_2O_2}>-induced$$H_2O_2$ with regard to its effect on intracellular cAMP content. However, SHCS itself did not affect resting cAMP concentration in endothelial cells. Conclusions : These results suggest that SHCS might operate as an effective protectant against oxidant-induced destruction of endothelial barrier function. The mechanism does not appear to involve direct interaction with protein kinase C- or cAMP-associated signaling mechanism.

연꽃수술추출물이 과산화수소로 손상된 배양 인체피부흑색종세포에 대한 항산화효과 및 멜라닌화에 미치는 영향 (Antioxidative Effect and Melanogenesis of Nelumbo nucifera Stamen Extract on Cultured Human Skin Melanoma Cells Injured by Hydrogen Peroxide)

  • 활성산소의 하나인 과산화수소(hydrogen peroxide, $H_2O_2$)에 대한 세포독성과 이에 대한 연꽃수술(Nelumbo nucifera stamen, NNS)추출물의 항산화효과 및 멜라닌화에 미치는 영향을 알아보기 위하여 세포부착율 (cell adhesion activity, CAA)을 비롯한 티로시나제활성저해능 및 총멜라닌합성량을 분석하였다. 본 실험 결과 hydrogen peroxide($H_2O_2$)는 배양 인체피부흑색종세포 (SK-MEL-3)의 CAA를 유의하게 감소시킴으로서 세포독성효과를 나타냈다. 이에 대하여 NNS추출물은 $H_2O_2$의 산화적 손상에 의하여 감소된 CAA를 유의하게 증가시킴으로서 항산화효과를 나타냈다. 한편, $H_2O_2$에 대한 NNS추출물의 멜라닌화에 대한 영향에 있어서 NNS추출물은 $H_2O_2$에 의하여 활성화된 티로시나제활성과 총멜라닌합성량을 감소시킴으로서 멜라닌화에 대하여 억제효과를 나타냈다. 이상의 결과로 부터 $H_2O_2$는 배양 인체피부흑색종세포에 고독성을 나타냈으며 NNS와 같은 식물추출물은 $H_2O_2$와 같은 활성산소에 의한 산화적 손상 및 멜라닌화를 방어하는데 효과적인 것으로 나타났다.

  • This study was done to evaluate the antioxidant effect of Crataegi Fructus (CF) extract on the oxidative stress induced by reactive oxygen species (ROS), The human skin fibroblasts (Detroit 551) were cultured with various concentrations of hydrogen peroxide $(H_2O_2)$. The cytotoxicity of $H_2O_2-induced$ oxidative stress was performed by XTT assay for the cell viability according to the dose- and time-dependent treatment. For the protective effect of CF extract on $H_2O_2-mediated$ oxidative stress, cell viability, lactate dehydroganase (LDH) activity, and ferric thiocyanate (FTC) assay for the inhibitive activity of lipid peroxidation on CF extract were carried out. In this study, $H_2O_2-mediated$ oxidative stress was decreased cell viability dose-, and time-dependent manner and increased LDH activity compared with the control in these cultures. In the protective effect, CF extract increased cell viability and decreased LDH activity on $H_2O_2-mediated$ oxidative stress, especially, CF extract has antioxidant effect by the showing the inhibitive activity of lipid peroxidation by FTC assay. From these results, It is suggested that $H_2O_2-mediated$ oxidative stress was highly toxic, and also, CF extract showed the protective effect on $H_2O_2-mediated$ oxidative stress by showing the increased cell viability, decreased LDH activity and lipid peroxidation inhibition in these cultures.

Ab Initio Studies of Lithium Bonded Complexes with H$_2$O Molecule

  • Lithium bonded complexes with $H_2O$ molecule were investigated theoretically by varying the substituent of lithium compound as follows; LiH, LiLi, $LiCH_3,\;LiNH_2$, LiOH, LiF, and LiCl. Some hydrogen bonded complexes with $H_2O$ molecule were also investigated to be compared with lithium bonded analogues. Electron correlation effect on the structures and energies of lithium bond was also investigated through MP2 and MP4 corrections. Unlike hydrogen bond with $H_2O$ molecule, lithium bonded complexes with $H_2O$ molecule were found to be interacting linearly with $H_2O$ molecule. Electron correlation effect was very small for lithium bonded complexes. The lithium bond energies were found to be less affected by the choice of substituent of lithium compound.
