  • Free surface impinging jet on a moving plate, which is applicable to cooling of hot metals in a steel-making process, is investigated numerically by solving the Navier-Stokes equations in the liquid and gas phases. The free surface of liquid-gas interface is tracked by a level-set method which is improved by incorporating the ghost fluid approach based on a sharp-interface representation. The method is further improved by employing a nonequilibrium $\kappa-\varepsilon$ turbulence model including the effect of low Reynolds number. The computations are made to investigate the effects of the nozzle pitch, moving velocity of plate and jet velocity on the interfacial motion and the associated flow and temperature fields.

Application of Carrageenan for Sustained Drug Release (약물의 서방출을 위한 ${\kappa}-Carrageenan$의 응용)

  • ${\kappa}-Carrageenan$, an anionic polysaccharide, was employed in tablet formulations and its function as a drug release sustaining agent was investigated. Tablets composed of ${\kappa}-carrageenan$ and hydroxypropyl methylcellulose were fabricated by using direct compression method. Lactose and sodium alginate were utilized as controls for ${\kappa}-carrageenan$. Drug release experiments performed at pHs 1.2 and 7.4 revealed that ${\kappa}-carrageenan$ retains pH-dependent sustained release effects due to its anionic characteristics. Also, the ionic interaction between ${\kappa}-carrageenan$ and drugs exerted significant affects on drug release kinetics. ${\kappa}-Carrageenan$ was found out to be a useful additive for sustained release tablet formulations.

  • Background: NF-$\kappa$B is a characteristic transcriptional factor whose functional activity is determined by post-translational modification of protein and subsequent change of subcellular localization. The involvement of the NF-$\kappa$B family of the transcription factors in the control of such vital cellular functions as immune response, acute phase reaction, replication of certain viruses and development and differentiation of cells has been clearly documented in many previous studies. Several recent observations have suggested that the NF-$\kappa$B might also be involved in the carcinogenesis of some hematological and solid tumors. Investigating the possibility that members of the NF-$\kappa$B family participate in the molecular control of malignant cell transformation could provide invaluable information on both molecular pathogenesis and cancer-related gene therapy. Method: To determine the expression patterns and functional roles of NF-$\kappa$B family transcription factors in human lung cancer cell lines NCI-H792, NCI-H709, NCI-H226 and NCI-H157 were analysed by western blot, using their respective antibodies. The nuclear and the cytoplasmic fraction of protein extract of these cell lines were subsequently obtained and NF-$\kappa$B expression in each fraction was again determined by western blot analysis. The type of NF-$\kappa$B complex present in the cells was determined by immunoprecipitation. To detect the binding ability of cell-line nuclear extracts to the KB consensus oligonucleotide, electrophoretic mobility shift assay(EMSA) was performed. Results: In the cultured human lung cancer cell lines tested, transcription factors of the NF-$\kappa$B family, namely the p50 and p65 subunit were expressed and localized in the nuclear fraction of the cellular extract by western blot analysis and immunocytochemistry. Immunoprecipitation assay showed that in the cell, the p50 and p65 subunits made NF-$\kappa$B complex. Finally it was shown by Electrophoretic Mobility Shift Assay(EMSA) that nuclear extracts of lung cancer cell lines are able to bind to NF-$\kappa$B consensus DNA sequences. Conclusion: These data suggest that in human lung cancer cell lines the NF-$\kappa$B p50/p65 complex might be activated. and strengthen the hypothesis that NF-$\kappa$B family transcription factors might be involved in the carcinogenesis of human lung cancer.

Lipid Peroxidation, $NF-_{\kappa}B$ Activation and Cytokine Production in Neutrophil-Stimulated Pancreatic Acinar Cells

  • Reactive oxygen species (ROS), generated by infiltrating neutrophils, are considered as an important regulator in the pathogenesis and deveolpment of pancreatitis. The present study aims to investigate whether neutrophils primed by $4{\beta}-phorbol\;12{\beta}-myristate\;13{\alpha}-acetate$ (PMA) affect the productions $H_2O_2$ and lipid peroxide (LPO), $NF-_{\kappa}B$ activation and cytokine production in pancreatic acinar cells, and whether these alterations were inhibited by an antioxidant, N-acetylcysteine (NAC) and superoxide dismutase (SOD). $H_2O_2$ (ferrithiocyanate method), LPO (as thiobarbiturate reactive substances), and cytokines $(IL-l{\bata},\; IL-6,\;TNF-{\alpha};\;enzyme-linked\;immunosorbent\;assay)$ and $NF-_{\kappa}B$ activation (electrophoretic mobility shift assay) were analyzed in acinar cells treated with or without PMA-primed neutrophils in the absence or presence of NAC (10 mM) or SOD (300 U/ml). As a result, the productions of H2O2, LPO and $TNF-{\alpha}$ were increased with the ratio of PMA-primed neutrophils to acinar cells while the productions of LPO, $IL-l{\beta},\;IL-6\;and\;TNF-{\alpha}$ were increased with time. PMA-primed neutrophils resulted in the activation of $NF-_{\kappa}B.$ Both NAC and SOD inhibited neutrophil-induced alterations in acinar cells. In conclusion, ROS, generated by neutrophils, activates $NF-_{\kappa}B,$ resulting in upregulation of inflammatary cytokines in acinar cells. Antioxidants might be clinically useful antiinflammatory agents by inhibiting oxidant-mediated activation of $NF-_{\kappa}B$ and decreasing cytokine production.

  • A numerical study of underexpanded jet and impingement on a wall mounted at various distances from the nozzle exit is presented. The 3-dimensional Navier-Stokes equations and $\kappa-\omega$ turbulence equations are solved. The grids are constructed as overlapped grid systems to examine the distance effect. The DADI method is applied to obtain steady-state solutions. To avoid numerical instability such as the carbuncle that sometimes accompany approximate Riemann solver, the HLLE+ scheme is employed for the inviscid flux at the cell interfaces. A goal of this work is to apply a number of two-equation turbulence models based on the $\omega$ equation to the impinging jet problem.

Shear Wave Velocity Structure Beneath White Island Volcano, New Zealand, from Receiver Function Inversion and H-κ Stacking Methods (수신함수 역산 및 H-κ 중합법을 이용한 뉴질랜드 White Island 화산 하부의 S파 속도구조)

  • To estimate the shear-velocity ($v_s$) structure beneath the WIZ station on White Island in New Zealand, we applied receiver function (RF) inversion and H-${\kappa}$ stacking methods to 362 teleseismic events (Mw > 5.5) recorded during April 20, 2007 to September 6, 2013. Using 71 RFs with errors less than 20% after 200 iterative computations, we determined that the depth to Moho of $v_s$ = 4.35 km/s is $24{\pm}1km$ within a 15 km radius of the station. In an 1-d $v_s$ model derived by RF inversions, a 4-km thick low-velocity layer (LVL) at depths of 18 ~ 22 km was identified in the lower crust. This LVL, which is 0.15 km/s slower than the rocks above and below it, may indicate the presence of a deep magma reservoir. The H-${\kappa}$ stacking method yielded an estimate of the depth to the Moho of 24.5 km, which agrees well with the depth determined by RF inversions. The low $v_p/v_s$ ratio of 1.64 may be due to the presence of gas-filled rock or hot crystallizing magma.

  • For vibration analysis of stiffened plates the orthotropic plate analogy is commonly accepted. As to stiffened plates in contact with water, however, there is still much uncertainty in estimation of the added mass because of the lack of direct methods. The authors, considering that for added mass of plates there are many reliable data derived theoretically or experimentally available, suggest a method to estimate the added mass of a stiffened plate by combining the mass increase factor, $\beta$, of an equivalent orthotropic plate and the correction factor, $\kappa$, for the effects of stiffeners. The latter is to be derived from systematic experimental investigations. Then, the natural frequency in water, f', can be calculated from that in air, f, by the equation $f'=f/\sqrt{1+\kappa\beta}$. To investigate practical applicability of this method, a systematic experiment was carried out with five uniaxially stiffened plates. Each of them had a plate of same size, $600mm{\times}600mm{\times}3.2mm$, but stiffeners of different size in the web-depth, 41.6mm, 51.2mm or 66.8mm and of different spacing 75mm, 100mm, or 150mm. Natural frequencies were measured under simply supported-edge conditions in both air and water, and corresponding $\kappa$ values derived. In spite of wide variations of web-depth and spcae of stiffeners, the experimental results show that the diversity of $\kappa$ values is not remarkable; mean values of $\kappa$ are 1.31 with standard deviation of 0.025 for the first modes and 1.27 with that 0.077 for the second modes. Hence, the authors concluded that the above $\kappa$ values can be used generally for the cases of uniaxially stiffened plates both sides of which contact with water, and that $\kappa$ values of general use for the cases of cross-stiffened plates may also be obtainable from similar experiments.

DFA IV를 생산하는 levan fructotransferase의 포괄고정화

  • The condition of immobilization of the partially purified levan fructotransferase and the properties of the immobilized enzyme was investigated. Levan fructotransferase was immobilized on ${\kappa}\;-carrageenan$ beads by entrapment method. The optimal ${\kappa}\;-carrageenan$ concentration was obtained 2%(w/v) (or the matrix. At that time, immobilized enzymes(0.81 units) have relative low activity compare with soluble enzyme(7.7 units). To immobilized and soluble enzyme, optimal activity temperature and pH were measured $55^{\circ}C$, 6.0 in sodium phosphate buffer 20mM solution. If crosslinking agent was added, proper concentration was 0.5%(v/v). At $37^{\circ}C$, immobilized and soluble enzyme converted levan to oligofructose and DFA IV, and the conversion ratio was 32% and 61% at 60 hr.

Mixed-mode fatigue crack growth behaviors in 5083-H115 aluminum alloy (5083-H115 알루미늄 합금의 혼합 모우드 피로 균열성장 특성)

  • For the mixed-mode crack problems the direction of crack growth, the crack path and the rational representation of fatigue crack growth rates should be studied to predict fatigue life and safety of structures. In this study, a round specimen which produce nearly identical effects in all loading directions is proposed to make an easy measurement of initial direction of crack growth. The mode I and mode II stress intensity factors of the specimen were calculated using finite element method, in which the square root singular stresses at the crack tip are modeled by means of four rectangular quarter-point eight-noded elements surrounding the crack tip. Experimental results for high strength aluminum alloy showed that the direction of mixed-mode crack growth agree well with maximum principal stress criterion as well as minimum strain energy density criterion, but not with maximum shear stress criterion. From data of fatigue crack growth rates using crack geometry projected on the line perpendicular to the loading direction it is easily established that mixed-mode fatigue crack growth in 5083-H115 aluminum alloy goes predominantly with mode I crack growth behaviors.
