To develop a natural antioxidant from three Chrysanthemum species, flower and shoot extracts of Chrysanthemum frutescens, Chrysanthemum morifolium and Chrysanthemum zawadskii ssp. naktongense were obtained and their phenolic compound contents, scavenging effects on DPPH and ABTS radicals, ferrous ion chelating effects and inhibition activity on lipid peroxidation of linoleic acid were studied. Shoots of C. morifolium showed the highest levels in all above mentioned analyses. Especially, shoot extract of C. morifolium had high scavenging activities on ABTS radicals, similar to ascorbic acid or BHT. Ferrous ion chelating effect was the lowest in a C. morifolium shoot extract, but the highest in a C. morifolium flower extract. Inhibition activity on lipid peroxidation of linoleic acid was the highest with C. frutescens and C. morifolium shoots, but activity was lower than BHT. From present study, a shoot extract of C. morifolium is demonstrated as a valuable source for the development of a natural antioxidant. However, due to its low levels of ferrous ion chelating effects and inhibition activity on lipid peroxidation, a combination of other antioxidants with C. morifolium extract is recommended for the development of a new antioxidant.
Reactive oxygen species (ROS) damage DNA and cause cancer. Therefore, the research is being conducted on the development of antioxidants for the removal of ROS. This study was performed to investigate antioxidant activity and protective effect against oxidative DNA damage using ethyl acetate fractions from the cone of Pinus rigida × taeda (ERT). The antioxidant activity was evaluated using the DPPH, ABTS radical scavenging assay, reducing power assay, and Fe2+ chelating assay. Also, the contents of phenolic compounds and vitamin C related to antioxidant activity were analyzed to confirm phytochemicals. The DNA protective effect against oxidative stress was confirmed by the φX-174 RF I plasmid DNA cleavage assay. As a result, ERT showed DPPH and ABTS radical scavenging activities in a concentration-dependent manner. The results of reducing power and Fe2+ chelating activities were 77.32 ± 2.28% and 64.09 ± 1.01% at 200 ㎍/㎖. Also, ERT showed a DNA protective effect against oxidative stress.
Methanol extract and solvent fractions of Aster scaber Thunb. were tested to elucidate antioxidative characteristics in vitro. Methanol extract showed strong inhibition on linoleic acid autoxidation and conjugated diene production. Hexane and diethyl ether fractions showed stronger chelating effect on $10^{-4}M\;FeCl_3$ than the other fractions. Butanol fraction had the strongest capacity on $10^{-3}M\;CuSO_4$ similar to ${\alpha}$-tocopherol. Synergistic effect of butanol fraction with histidine, methionine, and lysine on linoleic acid autoxidation had been observed. Contents of total phenol, vitamin E, vitamin C, and carotenoid were evaluated. From the results, Aster scaber Thunb. could be expected to play a role as an antioxidant in vivo.
In order to screen the functional constituents from nature resource, we studied the bioactivities of methanol extract of the Liriodendron tulipifera branch(MLT). The total phenolic and flavonoid contents, DPPH radical scavenging capacity, reducing power, $Fe^{2+}$ chelating ability, inhibition of lipid peroxidation and cell toxicity of MLT were investigated in this study. We found that the total phenolic and flavonoid content of MLT is 75.34 mg gallic acid/g and 20.15 mg quercetin/g respectively. MLT exhibited the antioxidant activity on DPPH radical with a $EC_{50}$ value of $289.68\;{\mu}g$/mL, the absorbance is 0.388 at $100\;{\mu}g$/mL in reducing power assay, MLT prevented 38.56% lipid peroxidation at $200\;{\mu}g$/mL. Furthermore, MLT exhibits the potent anti-proliferative activity which inhibited 56.94%, 35.73% growth of HT-29 and Hela cell at $200\;{\mu}g$/mL respectively. It showed that the antioxidant activities of MLT were correlated with its total phenolic and flavonoid contents. However further study need to be exploring in the future.
The anti-oxidant properties of $Lactuca$$indica$ were determined using in-vitro assay systems. The vitamin C contents of the leaf and root extracts were 24.14 and 0.38 mg/100 g, respectively. The total polyphenol contents of the leaf and root extracts were 42.8 and 7.66 mg/g, and their flavonoid contents were 23.09 and 0.77 mg/g. The leaf extract showed higher DPPH and ABTS radical scavenging ability than the root extract at all the extract concentrations. Especially, the ABTS radical scavenging ability of the leaf extract was 92.3% at a concentration of 5 mg/mL. The reducing power was increased with the increase in the concentration of extracts, and the leaf extract had a higher reducing power than the root. The $Fe^{2+}$-chelating ability of the leaf and root were 97.2% and 34.3% at 14 mg/mL, respectively. The $IC_{50}$ values of the leaf for DPPH, its ABTS radical scavenging ability, and its $Fe^{2+}$-chelating ability were 0.19, 2.7, and 6.27 mg/mL, respectively, and the leaf extract showed lower $IC_{50}$ values than root extract. These results show that the $L.$$indica$ leaf extract contained high amounts of anti-oxidative compounds and had higher anti-oxidant activity levels than the root extract. It is suggested that Lactuca indica is very high in availability as a functional food and in its materials.

Objectives : Reactive oxygen species (ROS) have been associated with pathogenic processes including carcinogenesis through direct effect on DNA directly and by acting as a tumor promoter. Therefore, it has been regarded that ROS may be a major target for cancer prevention. The root of Paeonia lactiflora pall (PL), a traditional Chinese herb, has been a component of effective prescriptions for treatment of liver disease. Also, there are some reports about the antioxidant activities of the extracts from PL. However, little has been known about the effects of PL against oxidative damage. This work aimed to elucidate the anti-oxidant effects of Paeonia lactiflora pall (PL) in the non-cellular system and cellular system. Methods : Antioxidant activities of PL were evaluated by hydroxyl radical scavenging assay and $Fe^{2+}$ chelating assay. Anti-oxidative effect of PL was evaluated by ${\varphi}X$-174 RF I plasmid DNA cleavage assay in non-cellular system. In addition, DNA migration assay, expression level of phospho-H2AX, MTT assay and lipid peroxidation assay were performed for evaluate the anti-oxidative effect of PL in cellular system. Results : PL had a dose-dependent hydroxyl radical scavenging and $Fe^{2+}$ chelating capacity. In addition, PL inhibited oxidative DNA and cell damage induced by hydroxyl radical in non-cellular system and cellular system. Conclusion : Taken together, P. lactiflora pall may be possible for the application to a potential drug for treating the oxidative diseases such as cancer.

Abalone protein was hydrolyzed by enzymatic hydrolysis and the optimal enzyme/substrate (E/S) ratios were determined. Abalone protein hydrolysates (APH) produced by Protamex at E/S ratio of 1:100 showed angiotensin I converting enzyme inhibitory activity with $IC_{50}$ of 0.46 mg/mL, and APH obtained by Flavourzyme at E/S ratio of 1:100 possessed the oxygen radical absorbance capacity value of $457.6{\mu}M$ trolox equivalent/mg sample. Flavourzyme abalone protein hydrolysates (FAPH) also exhibited $H_2O_2$ scavenging activity with $IC_{50}$ of 0.48 mg/mL and $Fe^{2+}$+ chelating activity with $IC_{50}$ of 2.26 mg/mL as well as high reducing power. FAPH significantly (P<0.05) protected $H_2O_2$-induced hepatic cell damage in cultured hepatocytes, and the cell viability was restored to 90.27% in the presence of FAPH. FAPH exhibited 46.20% intracellular ROS scavenging activity and 57.89% lipid peroxidation inhibition activity in cultured hepatocytes. Overall, APH may be useful as an ingredient for functional foods.

This study investigated the antioxidant activities of processed Deoduck (Codonopsis lanceolata) extracts treated through high-pressure extraction and steaming with fermentation. The antioxidant activities were determined for DPPH and ABTS radical-scavenging activity, SOD-like activity, ferric reducing antioxidant power (FRAP), and $Fe^{2+}$ chelating. Total phenolic and flavonoid contents were also measured. Among eight Deoduck extracts, the S5FDW extract had the highest total phenolic and flavonoid content, 73.9 mg GAE/g and 50.9 mg QUE/g, respectively. The S5FDW extract had the highest DPPH radical-scavenging activity (27%) at a 1.0 mg/mL concentration. The ABTS radical-scavenging activity was highest for S5FDW extract (82.1%) at a 10 mg/mL concentration. The HFDE extract showed the highest SOD-like activity (29.7%) at a 1.0 mg/mL concentration. FRAP was highest in S5FDW extract (140.8 ${\mu}M$) at a 1.0 mg/mL concentration. The DE extract showed the highest $Fe^{2+}$ chelating (46%) at a 1.0 mg/mL concentration. The phenolic and flavonoid contents significantly correlated with the antioxidant activity of several processed Deoduck extracts and was higher in the processed Deoduck extracts compared to the raw Deoduck extracts. Therefore, processing techniques can be useful methods for making Deoduck a more potent and natural antioxidant.

Krill byproduct was hydrolyzed with Alcalase 2.4L to produce functional ingredients for high antioxidative activities against 1,1-dimethyl-2-picryl-hydrazyl (DPPH) radical and Fe. The objective of this study was to investigate the optimum condition for degree of hydrolysis and antioxidative activity of enzymatic hydrolysate produced with the commercial Alcalase using response surface methodology (RSM) with a central composite rotatable design (CCRD). The ranges of independent variables were pH 7.6~10.4 for initial pH and $50.9{\sim}79.1^{\circ}C$ for hydrolysis temperature and their dependent variables were degree of hydrolysis, Brix, amount of phenolic compounds, DPPH-scavenging activity and Fe-chelating activity. RSM with CCRD was well designed to investigate the optimum condition for functional ingredients with high antioxidative activities using Alcalase 2.4L because of their high $R^2$ values of the range of 0.93~0.99 except the $R^2$ value of 0.50 for the amount of total phenolic compounds. The optimum hydrolysis conditions were pH 9.5 and $62^{\circ}C$ for degree of hydrolysis (DH) and pH 9.1 and $64^{\circ}C$ for DPPH-scavenging activity by response surface methodology. The yield of DH and DPPH-scavenging activity were $14.1{\pm}0.5%$ and $10.5{\pm}0.2%$, respectively. It is advantageous to determine the optimum hydrolysis conditions of krill and its by-products for the creation of different kinds of food products, as well as to increase the usage of marine protein sources.

Five extraction conditions (AE, autoclave extraction; OE, oven extraction; HWSE, hot water and sonication extraction; HWASE, hot water acidified with 0.5% (v/v) acetic acid and sonication extraction; and BE, boiling extraction) were examined to compare the effects of different hot water extraction methods on the antioxidant properties of blueberries. The extraction yields of the AE, OE, HWSE, HWASE, and BE were 7.94%, 8.35%, 8.55% 9.15%, and 8.50%, respectively. The polyphenol and flavonoid contents of AE were 3.47 mg GAE/g and 1.59 mg RE/g, respectively, which were highest centents among others. Those of OE were ranked second to the highest. The total anthocyanin content of HWSE (5.29 mg/g) was significantly higher than that of others whereas that of AE showed the lowest content (0.96 mg/g). The order of ABTS radical and alkyl radical scavenging activities was as follows: AE > BE > OE > HWSE > HWASE. The antioxidant properties were considerably correspondent with the total polyphenol and flavonoid content. DPPH radical scavenging activity was quite high in HWSE, AE, and BE extraction, however, there were no significant differences among the five extraction methods in the aspect of $Fe^{2+}$ ion chelating activities. Moreover, AE showed the highest SOD activity, and protected the dermal fibroblast the best against $H_2O_2$-induced cytotoxicity. In conclusion, it was suggested that the autoclave extraction (AE) would be the most effective method for preparing blueberry hot water extracts with relatively high antioxidant activities.
