Genome-wide survey and expression analysis of F-box genes in wheat

  • The ubiquitin-proteasome pathway is the major regulatory mechanism in a number of cellular processes for selective degradation of proteins and involves three steps: (1) ATP dependent activation of ubiquitin by E1 enzyme, (2) transfer of activated ubiquitin to E2 and (3) transfer of ubiquitin to the protein to be degraded by E3 complex. F-box proteins are subunit of SCF complex and involved in specificity for a target substrate to be degraded. F-box proteins regulate many important biological processes such as embryogenesis, floral development, plant growth and development, biotic and abiotic stress, hormonal responses and senescence. However, little is known about the F-box genes in wheat. The draft genome sequence of wheat (IWGSC Reference Sequence v1.0 assembly) used to analysis a genome-wide survey of the F-box gene family in wheat. The Hidden Markov Model (HMM) profiles of F-box (PF00646), F-box-like (PF12937), F-box-like 2 (PF13013), FBA (PF04300), FBA_1 (PF07734), FBA_2 (PF07735), FBA_3 (PF08268) and FBD (PF08387) domains were downloaded from Pfam database were searched against IWGSC Reference Sequence v1.0 assembly. RNA-seq paired-end libraries from different stages of wheat, such as stages of seedling, tillering, booting, day after flowering (DAF) 1, DAF 10, DAF 20, and DAF 30 were conducted and sequenced by Illumina HiSeq2000 for expression analysis of F-box protein genes. Basic analysis including Hisat, HTseq, DEseq, gene ontology analysis and KEGG mapping were conducted for differentially expressed gene analysis and their annotation mappings of DEGs from various stages. About 950 F-box domain proteins identified by Pfam were mapped to wheat reference genome sequence by blastX (e-value < 0.05). Among them, more than 140 putative F-box protein genes were selected by fold changes cut-offs of > 2, significance p-value < 0.01, and FDR<0.01. Expression profiling of selected F-box protein genes were shown by heatmap analysis, and average linkage and squared Euclidean distance of putative 144 F-box protein genes by expression patterns were calculated for clustering analysis. This work may provide valuable and basic information for further investigation of protein degradation mechanism by ubiquitin proteasome system using F-box proteins during wheat development stages.

  • This paper presents characterizations on the independence of the exponential and Pareto distributions by record values. Let ${X_{n},\;n {\ge1}$ be a sequence of independent and identically distributed(i.i.d) random variables with a continuous cumulative distribution function(cdf) F(x) and probability density function(pdf) f(x). $Let{\;}Y_{n} = max{X_1, X_2, \ldots, X_n}$ for n \ge 1. We say $X_{j}$ is an upper record value of ${X_{n},{\;}n\ge 1}, if Y_{j} > Y_{j-1}, j > 1$. The indices at which the upper record values occur are given by the record times {u(n)}, n \ge 1, where u(n) = $min{j|j > u(n-1), X_{j} > X_{u(n-1)}, n \ge 2}$ and u(l) = 1. Then F(x) = $1 - e^{-\frac{x}{a}}$, x > 0, ${\sigma} > 0$ if and only if $\frac {X_u(_n)}{X_u(_{n+1})} and X_u(_{n+1}), n \ge 1$, are independent. 최소 빈도수 문자 우선 선택 방법의 3-SAT 다항시간 알고리즘 (A 3-SAT Polynomial Time Algorithm Based on Minimum Frequency Literal-First Selection Method)

  • 본 논문은 NP-완전으로 알려진 3-SAT 문제에 대해 O(nm)의 다항시간 알고리즘을 제안하였다. 여기서 n은 3-SAT 수식 f에 포함된 문자수이며, m은 모든 문자의 총 발생 빈도수이다. 제안된 알고리즘은 우선순위롤 정하여 문자의 진리 값을 결정하였다. 하나의 절에 포함된 최소 문자의 수가 1개(k=1)인 문자, k≥2 이고 진리 값이 '0' 또는 '1'만 존재하는 문자, 발생 빈도수가 최소인 문자 우선순위로 진리 값을 결정하였다. 진리 값이 결정된 문자의 r인 절을 삭제하고, 남아 있는 절에서 해당 문자를 삭제하는 방법으로 식을 축소시킨다. 이 과정을 문자 수 l회를 수행하면 주어진 f의 충족 가능 여부를 결정할 수 있다. 이 방법을 적용한 결과 주어진 식 f의 충족 가능 여부를 판단할 수 있었으며, 모든 문자의 진리 값도 결정할 수 있었다. 제안된 알고리즘을 다양한 문제들에 적용한 결과 모든 문제들에서 충족가능 여부를 정확히 결정할 수 있었다. 결국, 제안된 알고리즘은 모든 SAT 문제를 선형시간으로 풀 수 있는 일반화된 알고리즘으로 확장 가능하며, 오랜 숙원이던 P vs. NP 문제를 풀 수 있는 기반을 제공하였다.

Hemoglobin Concentration and Hematocrit Value of Black Bengal Goats Infected with Fasciola gigantica

  • A total of 72 Black Bengal goats of 2.5 to 3.5 and 4.0 to 6.0 years old were used in this study. Equal number of animals were included in Fasciola gigantica infected and non-infected control groups. For each age and treatment groups 18 blood samples were collected in glass vials contained EDTA anticoagulant in summer and winter seasons before the animals were slaughtered at abottoir. Packed cell volume (PCV) was determined using microhematocrit and hemoglobin (Hb) concentration by cyanmethemoglobin methods. The PCV of F. gigantica infected animals were significantly lower than the non-infected animals. The average PCV values obtained were 26.60 and 32.20% for F. gigantica infected and non-infected animals, respectively. The Hb values of infected animals were significantly lower than the non-infected animals. The average Hb values obtained were 9.17 and 10.51 gm% for F. gigantica infected and non-infected goats, respectively. There was no significant effect of age and season on the values of PCV and Hb of infected and non-infected animals.

학교 급식 관리 영양사의 영양교사 제도에 대한 기대감에 따른 직무만족도, 근로가치관 및 이직의도의 차이 분석 (Comparison Analysis of School Foodservice Dietitians' Job Satisfaction, Work Value, and Turnover Intention Based on the Expectation to be a Nutrition Teacher)

  • The purpose of this study was to identify the difference of school food service dietitians' work value, job satisfaction, and turnover intention based on their expectation to be a nutrition teacher. Also, this study explored the differences in expectation to be a nutrition teacher, work value, job satisfaction, turnover intention depending on demographics and school foodservice operational characteristics. This study surveyed dietitians who worked at school foodservice in Kyunggii and Daegu Gyeoungbuk Province. A total of 509 responses were collected using on-site survey and online survey (59.3%). Dietitians expressed favorable work value ($3.88{\pm}0.34$) and mid level of a job satisfaction ($2.99{\pm}0.64$). The percentage of dietitians who had an intention to leave after the promulgation of a nutrition teacher system (13.2%) was lower than that of dietitians who had an intention to leave prior to it (23.2%). Most dietitians expected a salary rise (84.7%), more chance to teach nutrition education (76.0%), and the improvement of power not only as a teacher but as a dietitian (72.7%). Dietitians who had higher expectation levels to be a nutrition teacher (> 3 on the 5 point Likert scale) showed a higher tendency of significant turnover intention (F = 4.40, p < 0.01) and more confident work value (F = 15.292, p < 0.001) when comparing those who had lower expectations. There were significant differences in work value, job satisfaction, turnover intention, and expectation to be a nutrition teacher depending on career length and employment status of dietitians. This study revealed that school food service dietitians have favorable expectations to be nutrition teachers with high work value.

Genetic Diversity and Population Structure Analyses of SSIV-2 Gene in Rice

  • Soluble starch synthase (SS) IV-2 is one of the starch synthase gene family members and responsible for starch chain elongation interacting with other rice eating and cooking quality controlling genes (e.g., AGPlar and PUL). SSIV-2 is mainly expressed in leaves, especially at grain-filling stage and its alleles can significantly affect rice quality. Here, we investigated the genetic diversity and population structure analyses of SSIV-2 gene by using 374 rice accessions. This rice set was grouped into 320 cultivated bred (subsequently classified into temperate japonica, indica, tropical japonica, aus, aromatic and admixture) and 54 wild rice. Haplotyping of cultivated rice accessions provided a total of 7 haplotypes, and only three haplotypes are functional indicating four substituted SNPs in two exons of chromosome 5: T/A and G/T in exon 4, and C/G and G/A in exon 13. Including the wild, a highest diverse group (0.0041), nucleotide diversity analysis showed temperate japonica (0.0001) had a lowest diversity value indicating the origin information of this gene evolution. Higher and positive Tajima5s D value of indica (1.9755) indicate a selective signature under balancing selection while temperate japonica (-0.9018) was in lowest Tajima's D value due to a recent selective sweep by positive selection. We found the most diverse genetic components of the wild in PCA but shared in some portion with other cultivated groups. Fixation index (FST-values) and phylogenetic analysis indicate a closer relationship of the wild with indica (FST=0.256) than to its association to both of temperate japonica (FST=0.589). Structure analysis shows a clear separation of cultivated subpopulations at every K value, but genetic components were admixed within the wild illustrating the same genetic background with japonica and indica in some proportion.

해빈 단면의 지형변화 모의를 위한 Delft3D 내의 표사이동 관련 매개변수의 민감도 분석 (Sensitivity Analysis of Sediment Transport Scaling Factors on Cross-Shore Beach Profile Changes using Deflt3D)

  • 본 연구에서는 강원도 맹방해변을 대상으로 Delft3D 지형변화모듈 내의 표사이동 관련 매개변수(fBED, fBEDW, fSUSW)가 지형변화 모의 결과에 미치는 영향을 검토하였다. 맹방해변 해역에서 2018년도에 관측한 1년 간의 파랑자료를 외력으로 적용하고, 동년도 1월과 10월에 관측한 수심자료는 각각 초기 지형자료와 년간 지형변화 자료로 사용하였다. 모델의 지형변화 모의성능은 계산영역 내의 임의의 한 단면을 연안역과 외해역 구간으로 구분하고, 각 영역에 대해 Brier Skill Score 지표에 근거하여 평가하였다. 그 결과 fBED 변수가 지형변화 모의 결과에 미치는 영향은 미약하다는 것을 알 수 있었다. fBEDW와 fSUSW 변수는 그 값이 0.5 이하일 때는 연안역에 대해 좋은 모의 성능을 보이고, 반대의 경우에는 외해역에 대해 좋은 성능을 보이는 것으로 나타났다. 실험조건 중 연안역에 대해 가장 좋은 성능을 보인 변수 조합은 fBED = 1.0, fBEDW = 1.0, fSUSW = 0.1이고, 외해역에 대해서는 fBED = 1.0, fBEDW = 1.0, fSUSW = 0.5이었다. 그러나, 이 조합은 맹방해변 2018년도 자료에 근거하여 산출된 값이므로, 타해역에 적용할 때는 주의가 필요할 것으로 판단된다.

SOME RESULTS ON COMPLEX DIFFERENTIAL-DIFFERENCE ANALOGUE OF BRÜCK CONJECTURE

  • In this paper, we utilize the Nevanlinna theory and uniqueness theory of meromorphic function to investigate the differential-difference analogue of $Br{\ddot{u}}ck$ conjecture. In other words, we consider ${\Delta}_{\eta}f(z)=f(z+{\eta})-f(z)$ and f'(z) share one value or one small function, and then obtain the precise expression of transcendental entire function f(z) under certain conditions, where ${\eta}{\in}{\mathbb{C}}{\backslash}\{0\}$ is a constant such that $f(z+{\eta})-f(z){\not\equiv}0$.

The black hole mass-stellar velocity relation of the present-day active galaxies

  • To investigate whether the present-day active galaxies follow the same black hole mass vs. stellar velocity dispersion (MBH-$\sigma*$) relation as quiescent galaxies, we measured the velocity dispersions of a sample of local Seyfert 1 galaxies, for which black hole masses were measured via reverberation mapping. We measured stellar velocity dispersions from high S/N optical spectra centered on the Ca II triplet region (${\sim}8500^{\circ}A$), obtained at the Keck, Palomar, and Lick Observatories. For two objects, in which the Ca II triplet region was contaminated by nuclear emission, we used high-quality H-band spectra obtained with the OH-Suppressing Infrared Imaging Spectrograph and laser-guide star adaptive optics at the Keck-II Telescope. Combining our new measurements with data from the literature, we assemble a sample of 24 active galaxies with stellar velocity dispersions and reverberation MBH in the range of black hole mass 106< MBH /$M{\odot}$ < 109,toobtainthefirstreverberationmappingconstraintsontheslopeandintrinsicscatteroftheMBH- $\sigma*$ relation of active galaxies. Assuming a constant virial coefficient f for the reverberation MBH, we find a slope ${\beta}=3.55{\pm}0.60$ and the intrinsic scatter ${\sigma}int=0.43{\pm}0.08$ dex in the relation log (MBH/M${\odot}$)=$\alpha+\beta$ log(${\sigma}*$/200 km s-1), which are consistent with those found for quiescent galaxies. We derive an updated value of the virial coefficient f by finding the value which places the reverberation masses in best agreement with the MBH - $\sigma*$ relation of quiescent galaxies; using the quiescent MBH - $\sigma*$ relation determined by Gultekin et al. we find log f=0.72+0.09 (or $0.71{\pm}0.10$) with an intrinsic scatter of $0.44{\pm}0.07$ (or 0.46+0.07) dex. No correlations between f and parameters connected to the physics of accretion (such as the Eddington ratio or line-shape measurements) are found. The uncertainty of the virial coefficient remains one of the main sources of the uncertainty in black hole mass determination using reverberation mapping, and therefore also in single-epoch spectroscopic estimates of black hole masses in active galaxies.
