THE EFFECTS OF BIO-$OSS^{(R)}$ AS A SCAFFOLDS DURING SINUS BONE GRAFT USING MESENCHYMAL STEM CELLS IN RABBIT (가토에서 자가유래 골아줄기세포를 이용한 상악동 골 이식술시 비계체로서 Bio-$Oss^{(R)}$의 효과에 관한 연구)

  • Mesenchymal stem cells(MSCs) have been though to be multipotent cells that can replicate that have the potential to differentiate into lineages of mesenchymal tissue including the bone, cartilage, fat, tendon, muscle, and marrow stroma. Especially, scaffolds to support cell-based tissue engineering are critical determinants of clinical efforts to regenerate and repair the body. Selection of a matrix carrier imvolves consideration of the matrix's role as a scaffold for physical support and host tissue integration as well as its ability to support of synergize the osteoinductive program of the implanted mesenchymal stem cell. The aim of this study is to evaluate the effect of autobone and Bio-$Oss^{(R)}$ to adherent mesenchymal stem cells as scaffolds on sinus augmentation with fibrin glue mixture in a rabbit model. 16 New Zealand White rabbits were divided randomly into 4 groups based on their time of sacrifice(1, 2, 4 and 8 weeks). First, mesenchymal stem cells were isolated from iliac crest marrow of rabbits and expanded in vitro. Cell culture was performed in accordance with the technique described by Tsutsumi et al. In the present study, the animals were sacrificed at 1, 2, 4 and 8 weeks after transplantation, and the bone formation ability of each sides was evaluated clinically, radiologically, histologically and histomorphologically. According to the histological observations, autobone scaffolds group showed integrated graft bone with host bone from sinus wall. At 2 and 4 weeks, it showed active newly formed bone and neovascularization. At 8 weeks, lamellae bone was observed in sinus graft material area. Radiologically, autobone with stem cell showed more radiopaque than Bio-$Oss^{(R)}$ scaffolds group. there were significant differences in bone volume between 4 and 8 weeks(p<0.05).

  • 국내에서 대부분 생산되는 사료(사일리지) 수확 작업은 베일 생산 및 비닐 래핑 작업이 독립적으로 수행되고 있어서 비효율적이다. 본 연구에서 개발한 조사료 수확기는 수집, 롤링, 베일 네트 생성, 래핑, 래핑 종료 및 베일 방출작업을 통합적으로 수행하도록 설계-제작하였다. 통합형 다목적 조사료수확기의 설계는 3D 디자인 툴 (CATIA V5R18)을 이용하여 실시하였으며, 기구부 23 파트 어셈블리, 전기제어 어셈블리, 유압요소기술을 통합하여 통합시작기를 제작하였다. 기초 프레임, 오거장치파트, 픽업장치파트, 하부롤러파트, 상부롤러파트, 하부 프레임 및 주행부 파트, 래핑회전파트, 롤러부 구동 동력부, 유압파트, 전기제어파트, 드로우바파트, 체결 및 컨트롤러 파트 등 25개 파트로 구성되어 있다. 본 연구에서 개발한 시작기의 견인력 및 소요동력 분석은 선행 연구에서 사용한 Brixius (1987) 제안 모델을 체택하여 분석하였다. 이 Brixius 제안 모델은 견인력 예측에서 토양변수 및 토양강도 특성을 나타내는 원추지수 (Cone Index, CI)를 이용하여 트랙터의 견인력 예측에 사용하였다. 또한 트랙터-조사료수확기 시스템의 소요 견인력을 예측하는데 있어, 트랙터-조사료수확기 시스템이 운용되는 토양조건과 트랙터의 마력에 따른 소요 견인력 특성을 분석하기 위해 대표적으로 3수준의 토양조건 (CI: 356 kPa, 543 kPa, 1,429 kPa)을 적용하였으며, 베일의 개당 최대무게는 최고 수준인 옥수수 기준으로 800 kgf를 적용하였다. 본 연구에서 적용된 3수준의 CI 조건은 연구팀에서 선행연구과정 (토양특성에 따른 최적 경운작업 시스템 개발, 2006)에서 분석한 전국 10개 지역의 33개 지점의 경반층 CI지수의 측정범위인 1,050-3,170 kPa에 대해 견인력이 많이 소요되는 열악한 조건 수준을 적용하였다. 각 작업에 사용된 소요동력은 베일 작업시 (ASABE D497.7, 2011) 그리고 래핑작업시 (Zhortuylov et al., 2013)를 사용하였으며 두 소요마력을 트랙터-조사료 수확기 시스템의 필요 소요마력의 합계로 계산하였다. 트랙터-조사료수확기 시스템의 최소 소요 동력, 차축 소요 동력과 PTO 소요 동력을 Zoz and Grisso (2003)을 이용하여 계산하였다. 연구에서는 기본적으로 ASAE의 작업속도 및 작업효율을 적용하였는데, 적용된 조사료수확기의 현장 작업효율은 60-86%의 범위이고 일반적으로 70%를 적용하고 이때 작업속도는 2.5-8.0 km/h이며 전형적으로 5.0 km/h를 기준하고 있다. 자주식 (SP; sief-propelled machine) 조사료수확기의 경우 작업속도가 2.5-10.0 km/h의 범위에서 작업효율은 60-85% 범위이다. 적용되는 조사료 수확기의 작업효율인 60-85% 범위에서 일반적으로 적용되는 작업효율인 70%를 적용하면 트랙터의 소요동력은 95hp를 적정 작업환경으로 하였다.

  • The experiments were carried out to investigate the selection effects on the drug resistance to Agrepto in Xanthomonas oryzae, the causal bacteria of rice bacterial leaf blight. The results obtained were as follows. 1, The Agrepto was stable at the heat treatment of $105^{\circ}C$ for 20 minutes, when the drug was added in the media. 2. The local isolates of the bacteria, 75-6 and 75-9, showed the different resistante reaction, when they were selected by the Agrepto contained media which concentrations of 10r9/m1 and 100ug/ml. 3. The individuals shelving high degree of resistance, which can grow on the media contained 10,000ug/m1 of Agrepto, could selected by the concentrations as low as 10ug/ml and 100ug/m1, in one generation. 4. The highly resistant isolates which selected by 3-stepwise selections such as 100ug/ml, 3,000ug/ml and 10,000/ml plots, showed nearly normal growth at the media contained 100ug/ml of Agrepto 5. When the isolate 75-9 was selected at the 100ug/m1 of concentration, showed various degrees of resistance, indicating that the isolate may be composed of resistance groups that lower than 500ug/ml, between 500-1,000ug/ml and 1,000-3,000ug/ml, to the Agrept.

COMPARISON OF THE PHYSICAL PROPERTIES OF FOUR KINDS OF ACRYLIC RESIN DENTURE TEETH (4종 아크릴릭 레진 인공치의 물리적 성질에 관한 비교)

  • To compare the wear resistance of four kinds of commercial acrylic resin teeth [SR-Orthosit PosterioresR(Ivoclar Co., Liechtenstein), Endura PosteriorR(Shofu Inc. Japan), trubyte IPN teethR(Dentsply International Inc., York,), Trubyte BiotoneR(dentsply Inermational Inc. Brazil) by means of the toothbrush abrasion method, the artificial resin teeth were embedded in epoxy resin with the occlusal surfaces aligned in one plane for a total of 40 blocks. There after, each lock was mounted in the arm of the toothbrush abrasion machine(K 236, Japan). Wear measurements were made on the three preconditioned states. Those were as follows : no treatment specimens, thermocycled specimens, and thermocycled specimens which were immeresed applied load of 400g during the buring the brushing cycle. At the end of the 30,000-stroke cycle, each specimen was removed, and weighed. The microhardness of four kinds of commercial resin teeth were determined by means of microhardness tester. Microhardness tests were performed on te no treatment specimens, thermocycled specimens, and thermocycled specimens with immersion in the denture cleansing solution. Finally, the comparison of thermal properties were perfomed using differential scanning calorimeter(DSC-1500). The following results were obtained : 1. In the case of no treatment teeth, the wear amount of endura Posterior(EN) was the greatest among the others(p<0.01), and the wear amount of three kinds of artificial resin teeth was increased in the order of Trubyte IPN teeth(IN), Trubyte Biotone(BN), SR-Orthosit Posteriores(SN) but there was no statistic significance(p>0.01). 2. The wear amount of Trubyte IPN teeth(IT) and Trubyte Biotone(BT), was increased due to thermocycling effect, but that of Endura Posteriores(ET) was decreased conversely(p<0.01). 3. Except for the SR-Orthosit Posteriores(STC), the wear amount of three kinds of artificial resing teeth(that is, ETC, ITC, BTC) was increased due to denture cleansing solution$(Cledent^R)$, but there was no statistic significance(p>0.01). But the wear amount of the SR-Orthosit Posteriores(STC) was the greatest among the others(p<0.01). 4. The wear amount of toothbrush was the greatest in case of contact with occlusal surface of SROrthosit Posteriores resin teeth(p<0.01). 5. the microhardness values(KHN) of the SR-Orthosit Posteriores was the highest among the experimental artificial resin teeth(p<0.01). 6. There was no relationship between microhardness valuse(KHN) and wear amount of four kinds of experimental artificial resin teeth(p<0.01). 7. The differential canning calorimetric property of four kinds of artificial resin teeth did not show endothermal or exothermal peak in the range of $100^{\circ}C$

  • Interstitial pneumonitis associated with interferon alpha therapy for chronic hepatitis C was first describe6 in 1994 by Kazoo et al In Japan. The mechanism of interstitial pneumonitis developed by interferon alpha was still unknown but immunologic, allergic of direct lung toxicity were suggested. We experienced a case of interstitial pneumonitis developed during interferon alpha therapy for chronic hepatitis C in a 52-year-old male patient. He was treated with 6 million units of interferon alpha intramuscularly 3 times per week for 4 weeks and noted progressive dyspnea and cough. These symptoms were subsided after 6 weeks' discontinuation of interferon alpha therapy. And so, he was retreated with 3 million units of interferon alpha 3 times per week for 8 weeks and felt dyspnea again. He was admitted to our hospital for further evaluation of progressive dyspnea. Arterial blood gas(ABG) values were $PaO_2$ 90.7 mmHg and $PaCO_2$ 31.9 mmHg, and antinuclear antibody(ANA) was negative. A chest X-ray film revealed diffuse reticulo-nodular shadows in bilateral lung fields, suggesting a diagnosis of interstitial pneumonitis. A marked increase in lymphocyte count and suppressor T cell were observed in bronchoalveolar lavage(BAL) fluid. Lymphocyte stimulation test with interferon alpha was positive. Interstitial pneumonitis was confirmed by transbronchial lung biopsy. After discontinuation of interferon alpha, we gave oral steroid in the condition that clinical symptoms were being improved gradually.

Genetic Organization of the hrp Genes Cluster in Erwinia pyrifoliae and Characterization of HR Active Domains in HrpNEp Protein by Mutational Analysis

  • The disease-specific (dsp) region and the hypersensitive response and pathogenicity (hrp) genes, including the hrpW, $hrpN_{Ep}$, and hrpC operons have previously been sequenced in Erwinia pyrifoliae WT3 [Shrestha et al. (2005a)]. In this study, the remaining hrp genes, including the hrpC, hrpA, hrpS, hrpXY, hrpL and hrpJ operons, were determined. The hrp genes cluster (ca. 38 kb) was comprised of eight transcriptional units and contained nine hrc (hrp conserved) genes. The genetic organization of the hrp/hrc genes and their orientation for the transcriptions were also similar to and collinear with those of E. amylovora, showing ${\geq}80%$ homologies. However, ORFU1 and ORFU2 of unknown functions, present between the hrpA and hrpS operons of E. amylovora, were absent in E. pyrifoliae. To determine the HR active domains, several proteins were prepared from truncated fragments of the N-terminal and the C-terminal regions of $HrpN_{Ep}$ protein of E. pyrifoliae. The proteins prepared from the N-terminal region elicited HR, but not from those of the C-terminal region indicating that HR active domains are located in only N-terminal region of the $HrpN_{Ep}$ protein. Two synthetic oligopeptides produced HR on tobacco confirming presence of two HR active domains in the $HrpN_{Ep}$. The HR positive N-terminal fragment ($HN{\Delta}C187$) was further narrowed down by deleting C-terminal amino acids and internal amino acids to investigate whether amino acid insertion region have role in faster and stronger HR activity in $HrpN_{Ep}$ than $HrpN_{Ea}$. The $HrpN_{Ep}$ mutant proteins $HN{\Delta}C187$ (D1AIR), $HN{\Delta}C187$ (D2AIR) and $HN{\Delta}C187$ (DM41) retained similar HR activation to that of wild-type $HrpN_{Ep}$. However, the $HrpN_{Ep}$ mutant protein $HN{\Delta}C187$ (D3AIR) lacking third amino acid insertion region (102 to 113 aa) reduced HR when compared to that of wild-type $HrpN_{Ep}$. Reduction in HR elicitation could not be observed when single amino acids at different positions were substituted at third amino acids insertion region. But, substitution of amino acids at L103R, L106K and L110R showed reduction in HR activity on tobacco suggesting their importance in activation of HR faster in the $HrpN_{Ep}$ although it requires further detailed analysis.

Effects of Organic Fertilizers on Growth and Yield in Liriope platyphylla WANG et TANG (유기질 비료의 사용에 따른 맥문동 생육 및 수량)

  • This experiment was carried out to investigate the effect of several organic fertilizers on the growth and root yield of Liriope platyphylla was planted 30 cm ${\time}$ 10 cm in field from 1998 to 1999. Six treatments of organic fertilizers applied : N.P.K, NPKC (N,P,K+Compost), NPKCO (N.P.K+Compost+Oilcake),NPKCOF (N.P.K+Compost+Oilcake+Fowldropping), NPKCOFP (N.P.K+Compost+ Oilcake+Fowldropping+plant ash), CO (Compost+Oilcake), COF (Compost+Oilcake+fowldropping). Organic matter, available phosphate, and MgO are NaO of soil increased in all of plots with the addition of organic fertilizers. The leaf length, number of leaves and of tillers were increased by the use of organic fertilizers compared to N.P.K fertilizers compared with N.P.K fertilization. The number of tubers were increased by using the organic fertilization N.P.K fertilization and showed higher in the soil depth of 0${\sim}$20 cm. The dry root yield was showed higher in organic fertilizers than in N.P.K fertilizer and highest dry root yield resulted from NPKOF 39% and NPKCOFP 49%.

Increased Available Phosphate by Shell Meal Fertilizer Application in Upland Soil (밭 토양에서 패화석비료 시용에 따른 유효인산의 증대)

  • Previous studies showed that shell meal fertilizer from the oyster farming industry could be a potential inorganic soil amendment to increase Chinese cabbage productivity and to restore the soil nutrient balance in upland soil (Lee et al., 2004). Herein, shell meal fertilizer was applied at rates of 0, 4, 8, 12, and $16Mg\;ha^{-1}$ to upland soil (Pyeontaeg series, Fine silty, Typic Endoaquepts) for Chinese cabbage cultivation. We found available phosphate increased significantly with shell meal fertilizer application, due to high content of phosphate ($1.5g\;P_2O_5\;kg^{-1}$) in the applied shell meal fertilizer. In addition, high pH of shell meal fertilizer contributed to increase available phosphate content by neutralization of acidic soil. Total and residual P contents increased significantly with increasing shell meal fertilizer application, but we could not find any tendency in organic and inorganic P fraction. Of extractable P fraction, water-soluble phosphorus (W-P) and calcium-bound P (Ca-P) contents increased significantly with increasing application level. By contrast, aluminum and iron-bound P (Al-P and Fe-P) decreased slightly with shell meal application. The present experiment indicated that shell meal fertilizer had a positive benefit on increasing available phosphate content in arable soil. And so the increased available phosphate by shell meal fertilizer may decrease phosphate application level and then reduce phosphorus loss in arable soil.

  • In Korea, none of the design methods, which can consider the properties of hard soil, weathered rock and the condition of construction, are suggested. Therefore, the properties of geomaterial are simply classified into three categories such as sand, clay and rock for the resistance estimation of axially loaded drilled shafts in Korea. However, in America, O'Neill et al.(1996) presented design methods for a new category of geomaterial which is between soil and rock termed "intermediate geomaterials, IGM's". And FHWA(1999) adapted above most complete classification of geomaterials in its design manual. However, in Korea, these properties are depended on the engineer's judgement, the weathered rocks may be counted as soils, although they may be referred to as IGM's in America. In this study, the applicability of IGM method was investigated through the two construction sites in Korea. For the comparison, two geomaterial properties are applied, respectively. The one was sound soil condition and the other was IGM condition and classical design method and IGM method were applied, respectively. The results showed that the predicted bearing capacities of drilled shafts with IGM's were larger than the predicted values by the classical design method with sound soil condition.

Evaluations of Si based ternary anode materials by using RF/DC magnetron sputtering for lithium ion batteries

  • Generally, the high energy lithium ion batteries depend intimately on the high capacity of electrode materials. For anode materials, the capacity of commercial graphite is unlike to increase much further due to its lower theoretical capacity of 372 mAhg-1. To improve upon graphite-based negative electrode materials for Li-ion rechargeable batteries, alternative anode materials with higher capacity are needed. Therefore, some metal anodes with high theoretic capacity, such as Si, Sn, Ge, Al, and Sb have been studied extensively. This work focuses on ternary Si-M1-M2 composite system, where M1 is Ge that alloys with Li, which has good cyclability and high specific capacity and M2 is Mo that does not alloy with Li. The Si shows the highest gravimetric capacity (up to 4000mAhg-1 for Li21Si5). Although Si is the most promising of the next generation anodes, it undergoes a large volume change during lithium insertion and extraction. It results in pulverization of the Si and loss of electrical contact between the Si and the current collector during the lithiation and delithiation. Thus, its capacity fades rapidly during cycling. Si thin film is more resistant to fracture than bulk Si because the film is firmly attached to the substrate. Thus, Si film could achieve good cycleability as well as high capacity. To improve the cycle performance of Si, Suzuki et al. prepared two components active (Si)-active(Sn, like Ge) elements film by vacuum deposition, where Sn particles dispersed homogeneously in the Si matrix. This film showed excellent rate capability than pure Si thin film. In this work, second element, Ge shows also high capacity (about 2500mAhg-1 for Li21Ge5) and has good cyclability although it undergoes a large volume change likewise Si. But only Ge does not use the anode due to its costs. Therefore, the electrode should be consisted of moderately Ge contents. Third element, Mo is an element that does not alloys with Li such as Co, Cr, Fe, Mn, Ni, V, Zr. In our previous research work, we have fabricated Si-Mo (active-inactive elements) composite negative electrodes by using RF/DC magnetron sputtering method. The electrodes showed excellent cycle characteristics. The Mo-silicide (inert matrix) dispersed homogeneously in the Si matrix and prevents the active material from aggregating. However, the thicker film than $3\;{\mu}m$ with high Mo contents showed poor cycling performance, which was attributed to the internal stress related to thickness. In order to deal with the large volume expansion of Si anode, great efforts were paid on material design. One of the effective ways is to find suitably three-elements (Si-Ge-Mo) contents. In this study, the Si based composites of 45~65 Si at.% and 23~43 Ge at.%, and 12~32 Mo at.% are evaluated the electrochemical characteristics and cycle performances as an anode. Results from six different compositions of Si-Ge-Mo are presented compared to only the Si and Ge negative electrodes.
