Levels of total, neutral and polar lipids from the seeds of eight species of the Cucurbitaceae f Cucurbita moschata, Lufa cylindrica, Citrullus vulgari, Cucumis melo var. makuwa, Cucumis satvus, Lag leucantha. Trichosanthes kirilowii and Momordica charantia, were determinded, and their fatty compositions were also analyzed by gas-liquid chromatography. The results were summarized as foll. Lipid contents of the seeds range from 21.9 to 50.7%, which contained 98% up of neutral lipi the fatty acid compositon of ottal lipids from the seeds of Cucurbita moschata, Lufa cylindrica, Ci vulgari, Cucumis melo var. makuwa, Cucumis sativus and Lagenaria leucantha, linoleic acid is the mos dominant component(56.8${\sim}$84.0%) followed by oleic acid(5.7${\sim}$22.2%) and palmitic acid(6.1${\sim}$1) with a trace amount of ${\alpha}-linolenic$ acid(below 0.6%). On the contrary, the seed oils of Tricho kirilowii and Momordica charantia are characterized by presence of considerable amounts of con trienoic acid such as punicic acid($_{9c.11t.13c-}C_{18:3}$) and ${\alpha}-eleostearic$ acid($_{9c.11t.13c-}C_{18:3}$). For example total lipids of T. kirilowii seeds were mainly composed of linoleic acid(40.5%) and punicic acid(3) in the fatty acid composition, while those of M. charantia seeds predominantly comprised ${\alpha}-eleos$ acid as a main component(66.9%), accompanied by oleic acid(11.7%) and linoleic acid(10.4%). oil ${\beta}-eleostearic$ acid($_{9t.11t.13c-}C_{18:3}$) was checked as a trace. Fatty acid profiles of neutral lipids close resemblance to those of total lipids in all the seed oils, but are different from those of polar In particular, conjugate trienoic acids including punicic acid and ${\alpha}-eleostearic$ acid which are oc as the most abundant component in both neutral lipids of T. kirilowii and M. charantia seed oils, ar ent in a extremely small amount in both polar lipids. The fatty acid distribution in the polar lipid the samples except for T. kirilowii and M. charantia seed oils, showed a tendency of consid increased level of saturated fatty acids(25.0${\sim}$29.4%) compared with that in the neutral lipids(9.9%). To investigate the effects of low dose gamma radiation and seed moisture content (SMC) on germination and early growth of vegetable crops, seeds of chinese cabbage (Brassica campestris L.), radish (Raphanus sativus L.), red pepper (Capcicum annuum L.), figleaf gourd (Cucurbita ficifolia Bouche) and bottle gourd (Lagenaria siceraria Standl), with different SMC were irradiated with different doses ($0{\sim}20\;Gy$) of gamma-ray by irradiator ($^{60}Co$, ca.150 TBq of capacity, AECL). Vegetable crops in which low dose gamma radiation was irradiated in seeds with different moisture content showed different response in seed germination and early growth to low dose gamma radiation. The germination rate of chinese cabbage, figleaf ground and bottle gourd irradiated with $2{\sim}8\;Gy$ showed interactive responses against relative SMC. Also, significant interactions occurred for the early growth between those factors. The stimulating effects of gamma radiation were more pronounced for hydrated seeds of chinese cabbage, radish, figleaf gourd and bottle gourd showing prominent responses with $2{\sim}10\;Gy$ irradiation, particularly for chinese cabbage and bottle gourd. These results suggest that radiation may promote germination and early growth of vegetable crops through interaction with SMC.
Ethanol extracts from 46 plants were tested for their fungicidal activity against six plant diseases consisting of Maynaporthe grisea, Rhizoctonia solani, Botrytis cinerea, Phytophthora infestans, Puccinia recondita, and Erysiphe graminis in the greenhouse studies. Strong activity at 5 and 10 mg/pot was produced from the extracts of Helianthus annuus flowers and Zea mays leaves against P. grisea. In a test with B. cineara, extracts of H. annuus leaves, H. annuus flowers, Chrysanthmum coronarium var. spatiosum, Cucurbita moschata seeds, Lycopersicon esculentum, Z. mays, and Z. mays leaves had strong activities at 5 mg/pot. In a test with P. recondita, strong activity was obtained from the extracts of Capsicum frutescens, C. moschata seeds, H. annuus seeds, L. esculentum, and Malva veticillata at 5 mg/pot. Against E. graminis, extracts of Cucumis sativus, H. annuus seeds, Salanum tuberosum, Z. mays, and Z. mays leaves produced strong activities at 10 mg/pot. All the extracts were ineffective against P. infestans and R. solani. Among seven extracts tested, the extracts of H. annuus leaves and flowers were highly effective against all the strains of B. cinerea resistant to carbendazim, procymidone, and diethofencarb. Furthermore, potent fungicidal activity was produced from the extracts of C. coronarium var. spatiosum and C. moschata seeds against the SSR, SRR, and RSR strains of B. cinerea, and Z. mays and Z. mays leaves against SSR and RSR. Extract of L. esculentum showed very strong activity only against RRS of B. cinerea.

Inhibitory effects of several edible plant extracts against 3-hydroxy-3-methylglutaryl coenzyme A(HMG-CoA) reductase, a rate-limiting enzyme in the biosynthesis of cholesterol, were screened. Inhibition rates of $10{\sim}15%$ were observed with hot water extracts of Allium fistulosum, Allium sativum and Cucurbita maxima. Methanol extracts of Aster scaber, Allium sativum, Zingiber officinale, Oenanthe javanica and Angelica keiskei effectively reduced the enzyme activity with inhibition rates of $29{\sim}51%$. The methanol extract of Angelica keiskei was fractionated sequentially with chloroform, ethyl acetate and n-butanol. Of the fractions ethyl acetate fraction showed the highest inhibition against the enzyme. $Luteolin-7-O-{\beta}-D-glucoside$ and hyperoside isolated from the ethyl acetate fraction of Angelica keiskei inhibited the enzyme activity by 65.5% and 14.8%, respectively, at the concentration of $30{\mu}M$.
To develop new high-quality Yakchobugak, features of Lactococcus lactis-fermented waxy rice paste after addition of some colored powdered agro-food products were investigated. Total and reducing sugars of waxy rice paste fermented by lactic acid bacteria were higher than those of control raw waxy rice paste. Total acidity gradually increased as powder concentration rose, being 1.02-1.56% and 0.96-1.87% in samples fermented with Cucurbita maxima and Capsicum annuum powders, respectively; these values were 3-4 times those in rice fermented with other powders. Fermented waxy rice paste viscosities were lower than those of non-fermented samples. The viscosities of samples fermented with Curcuma longa and Opuntia ficus powders were in the range $100-160{\times}10^4$ centipoise($mPa{\cdot}s$), and those of pastes fermented with Robus coreanus and Camellia sinensis extracts were under $40{\times}10^4mPa{\cdot}s$. Hunter color lightness(L) values decreased and yellowness(b) values rose after fermentation. Waxy rice paste fermented with Robus coreanus showed uniform particle size distribution, and many pores, by scanning electron micrography.

This virus was inactivated by heating at $50-65^{\circ}C$ for 10 min, by diluting at $10^{-4}-10^{-5}$, and aging at room temperature for 1-6 days. Preparations examined under the electron microscope by direct negative staining method(DN -method) always showed particles of flexuous filament bout 750nm in length and cytopasmic inclusions. Cytoplasmic inclusions and virus particles were also confirmed to present in the cytoplasm of a mesophyll cell by ultrathin sections of BICMV infected cowpea leaves. Cucumber mosaic virus (CMV) was transmitted by sap- inoculation on inoculated leaves of Chenopodium amaranticolor, C. quinoa producing local lesions, but non-inoculated upper leaves of Nicotiana glutinosa, Cucurbita pepo and Vigna sesquipedalis producting systemic mosaic symptoms. Electron microscopic examination of virus preparation by direct negative staining showed spherical particles of about 30nm in diameter. In ultrathin sections of CMV infected tissues, virus particles of crystalline array were found in the vacuole and a large number of virus particles were found in the cytoplasm and the plasmodesmata of mesophyll cells.
To determine the economic threshold level of Meloidogyne incognita on cucumber in greenhouse conditions, cucumber seedlings(Cucumis sativa L. cv. 'Super Manchon') grafted on 'Jangsushintozoa'(Cucurbita maxima x Cu. moschata) were planted in wooden boxes($30{\times}40{\times}15cm,\;L{\times}W{\times}D$) under a plastic house in August 01 and harvested from Sep. 01-Oct. 30, 2006. The initial nematode population densities(Pi) in the wooden boxes were adjusted to 0, 10, 30, 100, 300, and 600 second-stage juveniles(J2)/100 $cm^3$ soil. The relationship of total fruit yield to Pi level could be adequately described by a linear regression equation, $Y=0.82-0.04{\cdot}Log_{10}$(Pi+1). Initial nematode densities(Pi) before planting in excess of 5 J2/1,000 $cm^3$ soil caused in total yields loss that is equivalent to the costs of granular nematicide application; Pi level in excess of 25 J2/1,000 $cm^3$ soil caused in total yields loss that exceed the costs for application of fumigants at current control costs. We propose two different management strategies depending on nematode densities, (i) application of non-fumigant granular nematicides for M. incognita Pi level of 5 J2/1,000 $cm^3$ soil and (ii) fumigant treatment with Pi level over 25 J2/1,000 $cm^3$ soil. Soil samples to determine population density of Meloidogyne spp. for advisory purposes should be taken several months before planting time, which allows a period of time enough for implementing management procedures.
The aim of this study was to investigate the antioxidant and anti-cancer activities of squash leaf extract (SLE) in vitro. The total polyphenol and flavonoid levels of SLE were 263.4 mg gallic acid equivalent/100 g and 73.6 mg quercetin equivalent/100 g, respectively. The radical-scavenging activity of SLE at the concentration of 300 ${\mu}g/mL$ was 69.4%. SLE significantly inhibited human cancer cell growth (by 60.6-87.9% in HCT116 colon cancer cells and by 73.4-86.4% in H1299 lung cancer cells at the concentrations of 37.5, 75, and 150 ${\mu}g/mL$) and attachment (by 28.4% in HCT116 and by 16.8% in H1299 at the concentration of 150 ${\mu}g/mL$). SLE also altered nucleus morphology and increased nuclear staining intensity (by 42.8-58.2% in HCT116 and by 25.5-32.9% in H1299 at the concentrations of 37.5 and 75 ${\mu}g/mL$), indicating its apoptosis-inducing activity. These results demonstrate the antioxidant and anti-cancer activities of SLE in vitro.

A white powder-like substance, so called 'bloom', is formed on the surface of fruits of many varieties of cucumber. Although it is a natural phenomenon, bloomed fruits are accepted lower in quality compared to bloomless fruits by consumers. The experiments were conducted to obtain basic informations for breeding rootstocks, and to develop promising bloomless rootstocks from basic source materials collected and selected by seed company. The surfaces of bloomless fruits were appeared on the bladder cell of glandular trichome and the epicuticular waxes. Those of bloom fruits appeared with an injured bladder cell and many particles, which were not seen on those of bloomless fruits. The chromaticity was investigated on the surface of the bloom and the bloomless fruits. The 'a' and 'b' value of the bloom and the bloomless fruits did not show any significant difference. The 'L' value was significantly different and that of bloom fruits was higher than that of bloomless fruits. Fruit Si content was conspicuously lower in the fruits of cucumber plants grafted on the bloomless rootstock than in the fruits of those grafted on general bloom rootstocks.

Effects of low temperature ($8^{\circ}C$) on the hydraulic conductivity of young roots of a chilling-sensitive (cucumber; Cucumis sativus L.) and a chilling-resistant (figleaf gourd; Cucurbita ficifolia Bouche) crop have been measured at the levels of whole root systems (root hydraulic conductivity, $Lp_r$) and of individual cortical cells (cell hydraulic conductivity, Lp). In figleaf gourd, there was a reduction only in hydrostatic $Lp_r$ but not in osmotic $Lp_r$ suggesting that the activity of water channels was not much affected by low root temperature (LRT)treatment in this species. Changes in cell Lp in response to chilling and recovery were similar asroot level, although they were more intense at the root level. Roots of figleaf gourd recovered better from LRT treatment than those of cucumber. In figleaf gourd, recovery (both at the root and cell level) often resulted in Lp and $Lp_r$ values which were even bigger than the original, i.e. there was an overshoot in hydraulic conductivity. These effects were larger forosmotic (representing the cell-to-cell passage of water) than for hydrostatic $Lp_r$. After a short term (1 d) exposure to $8\;^{\circ}C$ followed by 1 d at $20\;^{\circ}C$, hydrostatic $Lp_r$ of cucumber nearly recovered and that of figleaf gourd still remained higher due to the overshoot. On the contrary, osmotic $Lp_r$ and cell Lp in both species remained high by a factor of 3 as compared to the control, possibly due to an increased activity of water channels. After pre-conditioning of roots at LRT, increased hydraulic conductivitywas completely inhibited by $HgCl_2$ at both the root and cell levels. Different from figleaf gourd, recovery from chilling was not complete in cucumber after longer exposure to LRT. It is concluded that at LRT, both changes in the activity of aquaporins and alterations of root anatomy determine the water uptake in both species. To better understand the aquaporin function in plants under various stress conditions, we examined the transgenic Arabidopsisand tobacco plants that constitutively overexpress ArabidopsisPIP1;4 or PIP2;5 under various abiotic stress conditions. No significant differences in growth rates were found between the transgenic and wild-type plants under favorable growth conditions. By contrast, overexpression of PIP1;4 or PIP2;5 had a negative effect on seed germination and seedling growth under drought stress, whereas it had a positive effect under cold stress and no effect under salt stress. Measurement of water transport by cell pressure probe revealed that these observed phenotypes under different stress conditions were closely correlated with the ability of water transport by each aquaporin in the transgenic plants. Together, our results demonstrate that PIP-type aquaporins play roles in seed germination, seedling growth, and stress response of Arabidopsis and tobacco plants under various stress conditions, and emphasize the importance of a single aquaporin-mediated water transport in these cellular processes.
