Analysis of Nutritional Components and Physiological Activity of Butternut Squash (Cucurbita moschata) by Drying Methods (건조방법을 달리한 땅콩호박의 영양성분 분석 및 생리활성 평가)

  • The purpose of this study was to provide the basic data of butternut squash (Cucurbita moschata) according to the drying methods. The proximate composition and chromaticity of freeze-dried and hot air-dried Cucurbita moschata powders were evaluated, and the antioxidant effects of these ethanolic extracts were analyzed by DPPH, ABTS, FRAP, reducing power activity, total phenolic, and flavonoid content. As a result, Cucurbita moschata was revealed to have high carbohydrate and crude protein contents, while crude ash and crude fat contents were relatively low. Also, it was confirmed that the hot air-dried Cucurbita moschata had lower L, a, and b values than the freeze-dried Cucurbita moschata. Additionally, as a result of analyzing the antioxidant activity of Cucurbita moschata ethanolic extracts, hot air-dried Cucurbita moschata ethanolic extract showed significantly superior antioxidant activity than freeze-dried Cucurbita moschata ethanolic extract. Also, hot air-dried Cucurbita moschata ethanolic extract was revealed to have a higher polyphenol content, but slightly lower flavonoid content than freeze-dried Cucurbita moschata ethanolic extract. Based on the data from this study, further experiments on Cucurbita moschata material are necessary.

Experimental Study on Effects of Cucurbita moschata Duch. on Antioxidation (호박잎의 항산화 효과 연구)

  • In recent year, We are concerned about anti-aging, disease-protection, long-life, many methods are used in solving this problem. And Those are related with antioxidative ability. Recently, We heard that Cucurbita moschata Duch. has anti-hypertensive effect and good for health. So I let made a experiment for this result of the anti-oxidant effect of Cucurbita moschata Duch. used for 3 methods, those are DPPH radical scavenging activity, Nitric oxide(NO) radical scavenging activity and Bovine serum albumin(BSA). The results of this study were as follows: We measured level of DPPH radical scavenging activity. And we obtained results that the ability of DPPH radical's elimination was increased when concentration of Cucurbita moschata Duch. was to $20\;mg/m{\ell}$. We measured level of Nitric oxide(NO) radical scavenging activity. And we founded that the ability of NO radical's elimination was increased when concentration of Cucurbita moschata Duch. was to $10\;mg/m{\ell}$. When we inspected antioxidation with Bovine serum albumin(BSA). And we obtained that antioxidative ability was increased after$2.5\;mg/m{\ell}$ of Cucurbita moschata Duch.. So I guess that Cucurbita moschata Duch. has effect of antioxidative ability. Hereafter we need differential experimental methods for evidence of antioxidative effect on Cucurbita moschata Duch.

Effect of Solvent Extraction on the Anti-complementary Activities of Green and Ripe Cucurbita moschata Duch

  • The edible part of cucurbita moschata Duch, which is commonly used as a Korean traditional medicine as well as a popular food source, was studied to isolate anti-complementary substance. Extracts of Cucurbita moschata Duch showed significant anti-complementary activities on the classical pathway of the complement system. Especially, the ripe Cucurbita moschata Duch had more activity than that of the complement system. Especially, the ripe Cucurbita moschata Duch had more activity than that of the green one in terms of the overall anti-complementary activity. Among the extracts of various organic solvents of the ripe Cucurbita moschata Duch, chloroform and ethyl-acetate extracts, which are non-polar solvent extracts, showed the strongest activities. These results suggest that the major difference in the solvent extraction for the anti-complementary substances depends on the change in the chemical composition such as the fatty acid with the degree of ripening.

Isolation of Angiotensin Converting enzyme inhibitors from Ripe Cucurbita moschata Duch

  • Angiotensin converting enzyme (ACE) inhibitor acts on the inhibition of ACE and causes a decrease in blood pressure. There have been several reports on screening of ACE inhibitors from natural food products and protein hydrolysates of various food sources. Ripe Cucurbita moschata Duch has been used as an oriental medicine in Korea. To isolate ACE inhibitors, crude water extracts of the edible portion of ripe Cucurbita moschata Duch were obtained after heating in water at 95$^{\circ}C$ for 2 h. Crude extracts were then filtered using PM-10 and YM-1 membranes. The membrane-filtered solution was loaded onto Sephadex G-15 column equlibrated with a phosphate buffer. Among the four major fractions of gel permeation chromatography, the second fraction had the highest inhibitory activity of 65%. Further purification of the fraction using reversed-phase HPLC with a $C_{18}$ column produced ACE inhibitors, which were identified as a mixture having molecular mass of 222 and 273 by Tandem mass spectrometry.

  • The effects of the water extracts from autumn pumpkin (Cucurbita maxima Duchesne, Cucurbitaceae) were evaluated on inflammation induced by carrageenan and hyperlipidemia induced by lipid rich diet, respectively in rats. The water extracts, when administered orally showed significant anti-inflammatory activity in rats treated with 1%-carrageenan and also anti-hyperlipidemia in rats treated with lipid rich diet(15% cholesterol, 1% sodium cholate and 84% corn oil).

  • A new unsaturated hydroxy fatty acid was isolated from the leaves of Cucurbita moschata through repeated silica gel column chromatography and chemical methods. The structure of the new fatty acid was determined as 13-hydroxy-9, 11, 15-octadecatrienoic acid on the basis of several spectral data including 2D-NMR. The stererostructures of double bonds were determined to be 9Z, 11 E and 15E by coupling patterns of related proton signals in the $^1H-NMR$ and NOESY experiments.

  • Light membrane vesicles were isolated from the zucchini hypocotyl by floatation on ficoll density gradients and the proteins were solubilized with Triton X100. Three ATP-hydrolyzing enzymes were partially purified by ion-exchange and gel filtration chromatography and isoelectric focusing. There are plasma membrane-type ATPase whose activity was inhibited by vanadate but not by nitrate, tonoplast-type ATPase which was sensitive to nitrate but insensitive to vanadate and one having a phosphatase activity with a pI value different from that of an acid phosphatase. A fraction was obtained after DEAE-ion-exchange chromatography crossreacting with polyclonal antibodies against Ca2+ -ATPase from human erythrocytes.

Incidence of Watermelon Mosaic Virus in Cucurbits (박과 작물에 발생하는 Watermelon Mosaic Virus에 관한 연구)

  • Cucurbits including pumpkin (Cucurbita pepo), gourd (Lagenariaa siceraria), cucumber (Cucumis sativus), melon(Cucumis melo) and watermelon(Cucurbita anguria) were diseased with mosaic symptoms. The causal virus was identified as watermelon mosaic virus(WMV). The WMV was transmitted by Myzus persicae Sulzer, and no seed borne virus was found. The virus caused large local lesions on the inoculated leaves of the Chenopodium amaranticolor and mosaic symptom on the upper leaves of Cucumis melo, Cucumis sativus, Lagenaria siceraria, Cucurbita anguria and Cucurbita pepo. There were no symptoms on the inoculated leaves of the Nicotiana tabacum var. Bright yellow, Nicotiana glutinosa, Vigna unguiculata. Petunia hybrida and Datura stramonium. Thermal inactivation point was $55\~65^{\circ}C$, dilution end point was $10^{-4}\;10^{-5}$ and longevity in vitro of the virus was $7\~8$ days. The virus showed positive reaction against watermelon mosaic virus antiserum in microprecipitin tests. The virus particles were flexuous rods in size of 750 nm.

Physiological Activities of Cucurbita moschata Duch. Extracts with Different Extraction Conditions (추출조건에 따른 늙은 호박 추출물의 생리활성)

  • This study was to investigate physiological activities of Cucurbita moschata Duch. extracts by various extraction conditions. Electron donating ability of 50% ethanol extract was 52.37% at 50 mL/g, which was higher than those of 1.0% and 0.1% L-ascorbate solutions. SOD-like activities were the most effective in all of the samples at 50 mL/g, which were barely detectable. Total polyphenol contents were the highest at 50 mL/g for all fiber extracts. The nitrite scavenging effects were the most effective in pH 1.2 at more than 80%. Tyrosinase-inhibitory activities ranged at fewer than 19%. The results will be useful for understanding of the physiological activities of Cucurbita moschata Duch. extracts.
