Setting and Hydroxyapatite Formation of Bioactive Glass Bone Cement (생체활성 유리 골 시멘트의 응결 및 수산화 아파타이트 형성)

  • Hardening and hydroxyapatite(HAp) formation behavior of the bioactive cements in the system of $CaO-SiO_{2}-P_{2}O_{5}$ glasses and the corresponding glass-ceramics were studied. DCPD (Dicalcium Phosphate Dihydrate: $CaHPO_4{\cdot}2H_2O$) and DCPA (Dicalcium Phosphate Anhydrous: $CaHPO_4$) were developed when the prepared glass and glass-ceramic powders were mixed with three different solutions. The DCPD and DCPA transformed to HAp when the cement was soaked in Simulated Body Fluid (SBF), and this HAp formation strongly depended on the releasing capacity of $Ca^{2+}$ ions from the cements. The glass-ceramic containing apatite showed fast setting, but no HAp formation was observed because no $Ca^{2+}$ ions were released from this glass-ceramics. The compressive strength of the cements increased with reaction time in SBF until all DCPD and DCPA transformed to HAp.

pH Variance Model Depending on Phosphate Ion Form (인산염 이온 형태에 의한 pH 변량 모형)

  • This experiment was performed to develop a model for nutrition ion concentration and EC in regard to change in pH from 4.0 to 8.0 in nutrient solution. The pH changes according to the variation of $HPO_4{^{-2}}$ and $H_2PO_4{^-}$ in the nutrient solution while variation of EC increased from pH 4.0 to 5.0, stabilized from pH 5.0 to 7.0 and increased again from pH 7.0 to 8.0. For the variance of major elements in the nutrient solution, K, Ca, N and P increased while pH was higher, especially the variables for K and P were increased largely. On the other hand, variables of Mg and S were stable. Based on analysis of the ion balance model of nutrient solution, the cation increased than anion over rising the variation of pH while balance point of ion moved from a-side to d-side. In addition, the imbalance increased while it moved away from the EC centerline as variance of pH increased. It was larger than effect of EC variance to correction values of equivalence ratios of K and Ca about variation of $HPO_4{^{-2}}$ and $H_2PO_4{^-}$ while as variance of pH increased, K decreased but Ca increased. These showed the result that variance of pH about correction values of equivalence ratios of K and Ca gave a second-degree polynomial model rating of 0.97. Through this research, it was identified the pH variable model about variance among pH, ion and EC according to gradient of phosphate.

  • The synthesis of Ca,,(PO,),(OH), by adding (NFIa)J),HPO, to lhe solution of Ca(NO,), dlssolvad CaO in HNO, and contmlled pH with NH,OH was carried out for certain time at room temperahire and atmosphere. Ca,,(PO,),(OH), was rorrned at the range from pN 10 to pH 13. The particle s~zeof Ca,,(PO,),(OH)i was 0.1-0.5 &In. Thc optimum reaction lime was 30 min, and the temperature was 40-70$^{\circ}$C. the shape also was not changed in spite of heating to iDVC hr 1 hour the c~ystalliratian temperature was 90$^{\circ}$C Ca,,(PO,),(OH), was calcinatcd and the shape also was not changzd, in spile of hcaling to 500$^{\circ}$C for 1 hour. But Ca,,(PO,),(OH), calcinated for lhour al 800$^{\circ}$C was changed to the spheric particle of Ca,,,(PO,),(OH),, CaO and Ca,(PO,),.

Formation of Brushite (CaHPO4 · 2H2O) in the Oyster Extracts (굴 엑기스에서 결정물질 Brushite(CaHPO4 · 2H2O)의 생성)

  • Small yellowish brown crystals were found in some concentrated oyster extracts which prepared by heating with the drip and washed water of boiled oyster in oyster processing factories. We collected those crystals by filtering the oyster extracts and analysed X ray diffraction (XRD), scanning electron microscopy (SEM) and minerals by ICP. Those were composed of moisture (8.1%), organic materials (21.6%) and inorganic materials (70.2%). Those major inorganic materials were determined as Ca (53.8%) and P (43.8%), from the ICP and elucidated those crystal as brushite $(CaHPO_4{\cdot}2H_2O)$ by XRD and SEM. Organic materials contained crude protein (33.7%) consisting of 7 amino acids. It is presumed that brushite may be formed by the reaction of excess Ca with P in the oyster extracts during concentrating process under higher temperature.

Production of $\beta$-Glucosidase from Aspergillus niger (Aspergillus niger에 의한 $\beta$-Glucosidase 생산)

  • This study was designed to reveal the conditions for $\beta$-glucosidase production from Aspergillus niger. The maximal enzyme production was obtained when the fungus was cultured at $30^{\circ}C$ for 5~6 days in the optimal medium containing 0.8% CMC, 0.5% beef extract, 0.3% Ca(NO3)2, 0.03% K2HPO4, 0.03% FeSO4, 0.05% Li2SO4, 0.2% tween 80, trace solution 1.0ml and initial pH 4.0, and then final enzyme activity under above conditions was 8.5-9.8 unit/ml culture filtrate.

  • The process of coprecipitation of biocomposite hydroxyapatite/chitosan from aqueous solution at low temperature in alkali environnement was examined. We have shown that initially we have the formation of amorphous octocalcium phosphates $(Ca_8(HPO_4)(PO_4)_5,\;nH_2O:\;OCP)$ and the transferring from OCP to amorphous calcium phosphate $(Ca_9(PO_4)_3,\;nH_2O:\;TCP)$, and then from TCP to calcium-deficient hydroxyapatite $(Ca_{10-X}\;(HPO_4)_X(PO_4)_{6-x}(OH)_{2-X}\;:\;ACP)$ and hydroxyapatite $(Ca_{10}(PO_4)_6(OH)_2\;:\;HAP)$. The transformation of ACP to HAP was inhibited in the presence of chitosan. The result suggests that there is an affinity binding between ACP and chitosan and subsequently blocking the active growth site of ACP.

  • A slimy non-spore-forming bacterium strain SAF-C isolated from bean stem and root was motile with flagella and identified to one of Agrobacterium radiobacter. Studies were made on the conditions necessary for maximal production of this acidic succinoglucan polysaccharide by this strain in shaken culture. Much production was observed with yeast extract,$(NH_4)_2\;HPO_4$, distillers' dried solubles(D.D.S.), as nitrogen source in the medium composed of 4% glucose, 0.5% nitrogen source, 0.3% $CaCO_3$. The yield was greatest with yeast extract and decreased in order with the above nitrogen source from 22.9% to 9.6 percent. A polysaccharide was produced in a yield of about 25% in a medium composed of 3% glucose, 0.4% D.D.S., 0.5% $K_2\;HPO_4$, 0.01% $MgSO_4{\cdot}7H_2O$.

  • The most active strain for the amylase activity from the native Youngkwang koji, was isolated and identified as Aspergjllus awamori. The optimal conditions for the production of ${\alpha}-amylase$ and glucoamylase were investigated and the properties of these enzymes were also examined. Maximum yields of ${\alpha}-amylase$ and glucoamylase were obtained at $30^{\circ}C$, pH 5.0 for days. The production of these two enzymes were increased with the addition of maltose, urea, $K_2HPO_4$, $MgSO_4{\cdot}7H_2O$, and $CaCl_2{\cdot}2H_2O$. The activities of these enzymes were maximized at $50^{\circ}C$, $pH\;5{\sim}6$. The heat stabilites of ${\alpha}-amylase$ and glucoamylase were maintained at $50^{\circ}C$ for 20min and 40min, respectively. Also, the addtion of $MgSO_4{\cdot}7H_2O$, $K_2HPO_4$, and $CaCl_2{\cdot}2H_2O$ salt increased the heat stabilities of these enzymes.

  • This study was concentrated on the variation of cordycepin production from liquid culture of Paecilomyces japonica.. Cordycepin production was measured from mycelium and culture broth of P. japoniaz. Optimum media composition was studied on inorganic N-sources, inorganic salts, trace elements and C/N ratio. In this results, cordycepin production in medium containing $(NH_4)_2HPO_4$, histidine and $CaCO_3$ was higher than control in culture broth.
