한국 동해안 영일만 표층 퇴적물의 금속 함량과 공간 변화 특성 (Spatial Variability and Contents of Metals in the Surficial Sediments of Youngil Bay, East Coast of Korea)

  • 한국 동해안 영일만 해역에서 퇴적물의 금속 함량과 공간 변화 특성을 연구하기 위해 총 27개의 표층 퇴적물을 채취하여, 입자조직, 유기탄소, 금속 원소들의 함량을 분석하였다. 연구해역애서 표층 퇴적물의 입도는 전반적으로 해안선 부근에서 조립하고 만의 중앙부에서 세립하며, 이러한 공간 변화는 해저지형과 반시계방향의 해류 패턴에 의한 것으로 해석된다. 유기탄소와 금속 원소들의 함량은 대부분 퇴적물의 입도 변화에 수반되어 니질 함량이 높은 만의 중앙부에서 높고 사질 함량이 높은 해안선 쪽으로 갈수록 낮아진다. 그러나 칼슘, 스트론튬, 칼륨의 함량은 조립한 퇴적물이 우세한 해안선 부근 퇴적물에서 높고, 몇몇 중금속(구리, 아연, 카드뮴) 함량 또한 유기물 함량이 매우 높은 구항과 형산강 입구의 퇴적물에서 높은 특징을 보인다. 상관관계 및 요인분석 결과에 의하면, 대부분의 금속 원소들의 함량과 공간 변화는 퇴적물의 입도에 의해 조절되는 것으로 보이나, 카드뮴, 구리 아연, 주석 등은 퇴적층의 무산소 환경에서 일어나는 지화학적 메카니즘이 이들의 함량과 공간 변화를 조절하는 일차적인 요인으로 제시된다. 약산추출 부분(labile fraction)의 함량과 농축비(concentration enrichment ratio) 연구결과에 의하면, 중금속 원소들의 존재 형태가 대부분 "sulfide minerals"와 관련된 것으로써, 이들의 형성과 축적이 일부 연구해역에서 중금속 오염의 주원인으로 제시된다. 한편, 칼슘과 스트론튬 경우에는 퇴적물에 함유된 패각 함량에 의해, 그리고 칼륨은 사질 퇴적물에 우세한 장석(feldspar) 광물에 의해 조절되는 것으로 해석된다.

홍도원추리(Hemerocallis hongdoensis Makino) 개체군의 식생구조와 토양특성 (Vegetation Structure and Soil Properties of Hemerocallis hongdoensis Population)

  • 본 연구는 홍도원추리개체군을 식물사회학적 방법으로 분류하고, 식생과 환경과의 상관관계를 밝히고자 분포서열법을 사용하여 분석하였다. 홍도원추리개체군은 예덕나무 우점개체군, 산가막살나무 우점개체군, 애기며느리밥풀 우점개체군으로 분류되었다. 홍도원추리 개체군은 주로 한반도 남서해안 도서에 분포하고 해발고는 6~362m의 해안가의 낮은 사면과 섬의 능선의 평균 $36.7^{\circ}$의 경사지에 위치하고 있다. 토양의 이화학적 특성을 분석한 결과 유기물함량은 16.18~21.70%, 전질소함량 0.56~0.97%, 치환성 K $0.42{\sim}0.88cmol^+/kg$, 치환성 Ca $3.38{\sim}5.65cmol^+/kg$, 치환성 Mg $1.12{\sim}2.38cmol^+/kg$, 양이온치환용량 $25.93{\sim}41.45cmol^+/kg$이며, 토양 pH는 4.45~4.86인 것으로 조사되었다. 예덕나무 우점개체군은 사면경사가 급하고 양료 중 CEC와 전질소가 높은 지역에 분포하였고, 애기며느리밥풀 우점개체군은 사면경사가 중간이고 CEC와 전질소의 양료가 중간인 입지에 분포하였다. 그리고 산가막살나무 우점개체군은 사면경사가 완만하고, CEC와 전질소의 양료가 적은 입지에 분포하는 것으로 나타났다. 천연보호구역으로 지정된 홍도에 분포하는 홍도원추리개체군의 생육특성을 파악하고 개발로 인한 훼손으로부터 자생지의 보전을 위한 구체적인 대책 마련이 요구된다.

Identification of Muscle Proteins Related to Objective Meat Quality in Korean Native Black Pig

  • This study examined the effects of pre-slaughter fasting, chasing stress and chiller ageing on objective meat quality, and their relations to the proteome profile of longissimus muscle using 20 male Korean native black pigs. Treatments were composed of two levels of pre-slaughter feed withdrawal, two levels of pre-slaughter stress and four chiller ageing times. A 15 min chasing stress immediately prior to slaughter significantly (p<0.05) decreased detectable levels of $\mu$-calpain activity during rigor development and chiller ageing, but did not have any direct effect on objective meat quality. On the other hand, pigs fed until the morning of slaughter resulted in significantly (p<0.05) higher hunter L* value and cooking loss than those which received an 18 h feed withdrawal prior to slaughter. Cooking loss and hunter L* value were constant during 7 d of chiller ageing, followed by significant increases at 14 d. The fed animals showed a significantly (p<0.05) higher hunter a* value at both 3 and 7 d, while the other group maintained a stable redness for 7 d. WB-shear force was not affected by the pre-slaughter treatments, but had significant (p<0.05) linear reduction from 1 to 7 d. A gelbased proteome analysis was performed on selected animals for low and high hunter L* values at 1 d. Ten and five spots had greater than two-fold spot densities for the low and high hunter L* groups, respectively. The ten spots included chain A, deoxyribounclease I complex with actin, heat shock protein 27 kDa, a protein similar to cardiac $Ca^{2+}$ release channel, and myosin heavy chain, while the five spots included chain A aldehyde dehydrogenase, glycerol-3 phosphate dehydrogenase, and hemoglobin alpha chain. In general, feeding until the morning of slaughter resulted in more desirable meat color, but appeared to reduce palatability due to increased cooking loss. Proteome analysis demonstrated that various proteins were concomitantly involved in the determination of final meat color. The most noticeable observation in the current study was that various isoforms for a particular protein differed in degradation and/or expression rate depending on meat quality.

Ultrafine Particle Events in the Ambient Atmosphere in Korea

  • In this study, real time measurements of particle number size distribution in urban Gwangju, coastal Taean, and industrial Yeosu in Korea were conducted in 2008 to understand the occurrence of ultrafine particle (UFP) (<100 nm) events, the variation of its concentration among different sampling sites, and UFP formation pathways. Also, to investigate seasonal and long-term variation of the UFP number concentration, data were collected for the period of 5 years (2007, 2008, 2010, 2011, and 2012) in urban Gwangju. Photochemical and combustion events were found to be responsible for the formation of UFP in the urban Gwangju site, whereas only photochemical event led to the formation of UFP in the coastal Taean site. The highest UFP concentration was found in industrial Yeosu (the average UFP number fractions were 79, 59 and 58% in Yeosu, Gwangju, and Taean, respectively), suggesting that high amount of gas pollutants (e.g., $NO_2$, $SO_2$, and volatile organic carbon (VOC)) emitted from industries and their photochemical reaction contributed for the elevated UFP concentration in the industrial Yeosu site. The UFP fraction also showed a seasonal variation with the peak value in spring (61.5, 54.5, 50.5, and 40.7% in spring, fall, summer, and winter, respectively) at urban Gwangju. Annual average UFP number concentrations in urban Gwangju were $5.53{\times}10^3\;cm^{-3}$, $4.68{\times}10^3\;cm^{-3}$, $5.32{\times}10^3\;cm^{-3}$, $3.99{\times}10^3\;cm^{-3}$, and $2.16{\times}10^3\;cm^{-3}$ in the year 2007, 2008, 2010, 2011, and 2012, respectively. Comparison of the annual average UFP number concentration with urban sites in other countries showed that the UFP concentrations of the Korean sites were lower than those in other urban cities, probably due to lower source strength in the current site. TEM/EDS analysis for the size-selected UFPs showed that the UFPs were classified into various types having different chemical species. Carbonaceous particles were observed in both combustion (soot and organics) and photochemical events (sulfate and organics). In the photochemical event, an internal mixture of organic species and ammonium sulfate/bisulfate was identified. Also, internal mixtures of aged Na-rich and organic species, aged Ca-rich particles, and doughnut shaped K-containing particles with elemental composition of a strong C with minor O, S, and K-likely to be originated from biomass burning nearby agricultural area, were observed. In addition, fly ash particles were also observed in the combustion event, not in the photochemical event.

The Effects of Vitamin D Supplementation to Peak-producing Hens Fed Diets Differing in Fat Source and Level on Laying Performance, Metabolic Profile, and Egg Quality

  • This experiment was designed to examine the effects of supplemental vitamin D on laying performance, metabolic profile and egg quality of hens fed diets containing different fat sources and levels. Lohman strains (n = 480) were assigned to one of 10 diets: basal diet (BD), BD plus 2.5 and 5.0% sunflower oil (SO) or tallow (T) at vitamin D provided $1{\times}$ and $3{\times}$ of the current recommendation. The experiment lasted from week 30 to 44 of age. Each diet was tested in 12 replicate cages of 4 hens. Production, metabolism, and egg quality data were subjected to three-way ANOVA. Both fats decreased feed intake (FI) as compared to BD. Increasing SO and T levels linearly decreased and quadratically increased FI, respectively. The dietary factors did not affect egg production (EP) and egg weight. Vitamin D supplementation increased and decreased EP when diets contained SO and T, respectively. Feed conversion efficiency (FCE) for hens fed SO was lower than for hens fed T. However, increasing T level improved FCE, whereas increasing SO level worsened FCR. Vitamin D supplementation increased serum vitamin D and glucose concentrations. Vitamin D supplementation also caused a decrease and an increase in serum vitamin D concentration when diets contained SO and T, respectively. Serum glucose concentration for hens fed SO was lower than hens fed T. Increasing fat level linearly increased serum triglyceride and VLDL concentrations, regardless of the fat type. Increasing SO level linearly decreased serum cholesterol concentration. Vitamin D supplementation did not alter lipid metabolites. The dietary factors did not affect serum total protein, Ca, and P concentrations. As compared with BD, feeding SO decreased dry tibia and ash weights more than feeding T. Vitamin D supplementation tended to increase dry tibia weight and decrease tibia ash weight. Eggshell strength and thickness, yolk and albumen indexes, and Haugh unit were not responsive to the dietary factors. Eggshell strength quadratically increased with increasing T level. Yolk color for hens fed SO was lower than for hens fed T. The dietary factors did not affect most of yolk fatty acids. Increasing SO level quadratically decreased yolk $C_{18:2}$ concentration. Vitamin D supplementation increased and decreased yolk $C_{18:2}$ concentration when diets contained SO and T, respectively. In conclusion, increasing fat level improved laying performance without altering metabolic profile and egg quality. Vitamin D supplementation had minor alteration effects on laying performance, metabolic profile, and egg quality in response to fat feeding.

춘계 한국 동해 남부해역에서의 식물 수문학적 수역과 질산염약층의 수직양상 (Phytohydrography and the Vertical Pattern of Nitracline in the Southern Waters of the Korean East Sea in Early Spring)

  • 한국 동해 남부해역에서 봄철의 정량적 식물플랑크톤 시료, 수온, 염분, 용존산소 및 영양소의 자료를 분석하여 나타난 식물플랑크톤 군집과 수문학적 조건과의 밀접한 상관관계로 보아 연구해역이 3개의 식물 수분학적 해역, 즉 1) 한국동부 난수해역(대마해류의 지류), 2) 북한한류 해역, 3) 위 두 해역수의 영향을 받지 않는 외해역으로 구분될 수 있다. 식물플랑크톤의 수직분포는 수층의 안정성과 영양소 농도에 좌우된다. 영양소 농도는 수괴에 따라 특징적인 분포를 보인다. 표층에서 N/P ratio 는 약 3으로서 연구 해역에서의 주 한정 영양소가 질소임을 나타낸다. N/P removal ratio 는 12.54 ($r^2$ = 0.96) 이며 redfield ratio 와 근사하다. 질소약층 중 prirnary nitrite maximum은 식물플랑크톤의 분비로 형성되고 secondary nitrite maximum은 용존산소의 값이 5.2ml/l 이상인 연안 수역에서 나타냈다. 이 secondary maximum 형성의 기작은 용존산소의 값이 0.25 이하인 경우에 나타나는 다른 해역의 경우와는 달리 5.2ml/l 이상인 수역에서 관찰되어, 박테리아 활동에 의한 유기물질의 분해에 기인하는 것으로 보이나, 결론을 내리기에는 더 연구가 요망된다.

한우 체외성숙란의 단위발생에 대한 전기자극의 효과 (Effect of Electric Stimulation on Parthenogenesis of In Vitro Matured Oocytes from Korean Native Cows)

  • The suitable electric stimulation is essential for activation and fusion of oocytes before or after nuclear transplantation The present study was undertaken to determine the optirnal condition for the parthenogenetic activation of in vitro rnatured(IVM) bovine oocytes by electric stimulation. Different direct current(DC) electric voltage of 1.0, 1.5 and 2.0 kV/cm and pulse duration of 30, 60 and 120 $\mu$sec were applied to the JVM nocytes in 0.3 M mannitol solution containing each 100 $\mu$M CaCl$_2$ and MgCl$_2$. IVM occytes at 24, 28 and 32 hours Post-maturation(hpm) were also electrically stimulated at 1.5 kV /cm, for 60 $\mu$ sec. The stimulated nocytes were then co-cultured in TCM-199 solution containing 10% fetal calf serum with bovine oviductal epithelial cells for 7~9 days in a 5% $CO_2$ incubator at 39$^{\circ}C$ ~ Their activation and in vitro development to morula and blastocyst were assessed under an inverted microscope. The higher activation rates 62.8 and 63.4% and in vitro de- velopment rates to morula and blastocyst 5.1 and 10.9% were shown in the oocytes stimulated at the voltage of 1.0 and 1.5 kV/cm than 2.0 kV/cm, respectively. No signifi- cantly(P<0.05) different activation rate was shown in JVM oocytes stimulated for 30, 60 and 120 $\mu$sec, but developmental rates to morula and blastocyst was significantly(P<0.05) higher in the oocytes stimulated for 30 $\mu$sec(6~3%) and 60 $\mu$sec(10~0%) than 120 $\mu$sec(0~ 0%). The aged oocytes at 28 and 30 hpm showed significantly(P<0.05) higher activation rates(72~7 and 79.7%) than the oocytes at 24 hpm(50~9%)~ Also, their developmental rates to morula and blastocyst were significantly(P<0.05) higher in the nocytes at 28(14.3%) and 32 hpm(15.9%) than 24 hpm(3.6%). From these results, it can be suggested that the optimal electric stimulation for IVM bovine occytes is a DC voltage between 1.0 and 1.5 kV/cm, pulse duration of 30 or 60 $\mu$sec, and the optimal age of IVM oocytes for electric activation is at 32 hpm.

cAMP-Dependent Signalling is Involved in Adenosine-Stimulated $Cl^-$ Secretion in Rabbit Colon Mucosa

  • An important property of the intestine is the ability to secrete fluid. The intestinal secretion is regulated by a number of substances including vasoactive intestinal peptide (VIP), ATP and different inflammatory mediators. One of the most important secretagogues is adenosine during inflammation. However, the controversy concerning the underlying mechanism of adenosine-stimulated $Cl^-$ secretion in intestinal epithelial cells still continues. To investigate the effect of adenosine on $Cl^-$ secretion and its underlying mechanism in the rabbit colon mucosa, we measured short circuit current ($I_{SC}$) under automatic voltage clamp with DVC-1000 in a modified Ussing chamber. Adenosine, when added to the basolateral side of the muocsa, increased $I_{SC}$ in a dose-dependent manner. The adenosine-stimulated $I_{SC}$ response was abolished when $Cl^-$ in the bath solution was replaced completely with gluconate. In addition, the $I_{SC}$ response was inhibited by a basolateral Na-K-Cl cotransporter blocker, bumetanide, and by apical $Cl^-$ channel blockers, dephenylamine-2-carboxylate (DPC), 5-nitro-2-(3-phenyl-propylamino)-benzoate (NPPB), glibenclamide. Amiloride, an epithelial $Na^+$ channel blocker, and 4,4-diisothiocyanato-stilbene-2,2-disulphonate (DIDS), a $Ca^{2+}-activated$ $Cl^-$ channel blocker, had no effect. In the mucosa pre-stimulated with forskolin, adenosine did not show any additive effect, whereas carbachol resulted in a synergistic potentiation of the $I_{SC}$ response. The adenosine response was inhibited by 10 ${\mu}M$ H-89, an inhibitor of protein kinase A. These results suggest that the adenosine-stimulated $I_{SC}$ response is mediated by basolateral to apical $Cl^-$ secretion through a cAMP-dependent $Cl^-$ channel. The rank order of potencies of adenosine receptor agonists was $5'-(N-ethylcarboxamino)adenosine(NECA)>N^6-(R-phenylisopropyl)adenosine(R-$ PIA)>2-[p-(2-carbonylethyl)-phenyl-ethylamino]-5'-N-ethylcarboxaminoadenosine(CGS21680). From the above results, it can be concluded that adenosine interacts with the $A_{2b}$ adenosine receptor in the rabbit colon mucosa and a cAMP-dependent signalling mechanism underlies the stimulation of $Cl^-$ secretion.

비지박 첨가 먹이원 급여에 따른 흰점박이꽃무지 유충의 생육과 영양성분 변화 (Growth Performance and Nutrient Composition in the White-spotted Flower Chafer, Protaetia brevitarsis (Coleoptera: Scarabaeidae) Fed Agricultural By-product, Soybean Curd Cake)

  • 세계적으로 곤충시장이 매년 성장하는 가운데 우리나라에서도 곤충산업이 미래 유망 산업으로 주목 받고 있다. 뿐만 아니라, 최근 갈색거저리와 쌍별귀뚜라미, 흰점박이꽃무지 및 장수풍뎅이가 일반식품원료로 등록됨에 따라 식용곤충에 대한 관심이 늘고 곤충의 대량사육이 전국적으로 이루어지고 있다. 이에 안전하고 고영양성분이 포함된 식용곤충 전용 먹이원 개발 연구가 필요하다. 최근 들어 각종 부존자원의 사료화는 부존자원의 활용, 사료비 절감, 환경보존 등의 효과를 거두고 있는 것으로 나타나고 있다. 따라서 식용곤충으로 많이 사육되고 있는 흰점박이꽃무지의 생육을 촉진하면서 경제적이고 안정적인 사육을 도모하기 위해 감귤박, 비지박, 대두박, 맥주박 등 4종의 부존자원을 이용하여 효과를 조사하였다. 10% 비지박을 급여한 유충의 무게가 대조구 대비 약 3.5배 증가하였고, 유충기간 또한 40% 이상 단축되었다. 또한, 10% 비지박 첨가 실험구에서 일반조성분과 미네랄 및 아미노산의 함량이 전체적으로 높은 것으로 나타났다. 중금속 함량 분석 결과, 납과 카드뮴 및 비소는 모든 실험구에서 검출되지 않았다. 본 연구결과는 흰점박이꽃무지 유충의 생육 촉진 효과를 가지는 비지박이 곤충 먹이원의 소재로 활용될 수 있음을 보여준다.

Current Status of the Synchrotron Small-Angle X-ray Scattering Station BL4C1 at the Pohang Accelerator Laboratory

  • The small-angle X-ray scattering (SAXS) beamline BL4C1 at the 2.5 GeV storage ring of the Pohang Accelerator Laboratory (PAL) has been in its first you of operation since August 2000. During this first stage it could meet the basic requirements of the rapidly growing domestic SAXS user community, which has been carrying out measurements mainly on various polymer systems. The X-ray source is a bending magnet which produces white radiation with a critical energy of 5.5 keV. A synthetic double multilayer monochromator selects quasi-monochromatic radiation with a bandwidth of ca. 1.5%. This relatively low degree of monochromatization is sufficient for most SAXS measurements and allows a considerably higher flux at the sample as compared to monochromators using single crystals. Higher harmonics from the monochromator are rejected by reflection from a flat mirror, and a slit system is installed for collimation. A charge-coupled device (CCD) system, two one-dimensional photodiode arrays (PDA) and imaging plates (IP) are available its detectors. The overall performance of the beamline optics and of the detector systems has been checked using various standard samples. While the CCD and PDA detectors are well-suited for diffraction measurements, they give unsatisfactory data from weakly scattering samples, due to their high intrinsic noise. By using the IP system smooth scattering curves could be obtained in a wide dynamic range. In the second stage, stating from August 2001, the beamline will be upgraded with additional slits, focusing optics and gas-filled proportional detectors.
