Distribution and Cycling of Elements in a Pinus resinosa Plantation-Ecosystem, Wisconsin (Wisconsin의 Pinus resinosa 조림지(造林地) 생태계(生態系) 내(內)의 양료분포(養料分布) 및 순환(循環))

  • Biogeochemistry of a 34-year-old red pine plantation on a Typic Udipsamment was investigated in the central sand plains of Wisconsin Biomass and net primary production were $106Mg\;ha^{-1}$ and $11.0Mg\;ha^{-1}y^{-1}$ respectively. Concentrations of elements in the vegetation were greatest in the 2- to 3-year-old needles, followed by current needles, current twigs, bole bark, live branches, dead branches, bolewood production, and bolewood. For a given tree component (excluding the current twigs), elemental concentrations were greatest in the upper one- third and least in the lower one- third of the tree. Total elements in the vegetation are ranked: N($222kg\;ha^{-1}$), Ca(217), K (85), Mg(38), S(31), and p. The soil contained from 60 to 89% of the readily available P, K, Ca, Mg, and S in the ecosystem. Whereas precipitation was the major source of N and S, weathering supplied the bulk of the Ca, Mg, P, and K. Output of a given element by leaching was less than $7kg\;ha^{-1}y^{-1}$ except for S which was $12kg\;ha^{-1}y^{-1}$ Whereas throughfall returned the greatest amounts of N, K, and S to the forest floor, litterfall accounted for the greatest returns of Ca, Mg, and P. Because of foliar leaching, net loading of all elements studied in the throughfall exceeded that of the precipitation. Ammonium in net precipitation was reduced by a factor of three within the upper 7.5cm due to uptake by fine roots.

  • 삼차신경절의 뉴런이 구강악안면영역에서의 촉각, 입각, 온도각 및 통각 등 다양한 감각을 중추신경계로 전달하는 역할을 하는 것은 주지의 사실이다. 이러한 신경전달에 있어서 이온통로는 감각정보를 전달하는데 핵심적인 역할을 수행하며 특히 소디움 통로는 활동전위의 발생에 중요하다. 소디움 통로는 tetrodotoxin-sensitive(TTX-s) 및 tetrodotoxin-resistant(TTX-r) 통로로 나누어지는 데 이 중 TTX-r 통로에 발생되는 tetrodotoxin-resistant sodium current(TTX-r $I_{Na}$)는 capsaicin에 민감한 일차구심신경세포에서 유해자극에 의해 통각신호를 발생시키고 전달하는데 중요하다. 또한 칼슘 통로는 시냅스 전도에 있어서 필수적인 역할을 수행하고 있다 한편 치과영역에서 치수의 진정 목적으로 eugenol이 흔히 사용되고 있다. 그러나 eugenol의 그 작용 기전에 대해서 현재까지 이온 통로에 대한 상세한 결과가 없는 실정이며 최근의 보고에 의하면 eugenol이 capsaicin 수용기를 통하여 감각신경에 대한 억제작용을 나타낸다고 한다. 따라서 본 실험은 eugenol과 capsaicin이 흰쥐의 삼차신경절의 TTX-r $I_{Na}$와 칼슘통로에 어떠한 영향을 미치는지를 알아보고 eugenol이 capsaicin 수용기를 통하여 작용하는지를 검증하고자 시행되었다. 삼차신경절 뉴런은 100~150g의 흰쥐의 삼차신경절로부터 외과적으로 절제하여 통법의 화학적 및 기계적 처리를 통해 단일세포로 분리하였고 이를 whole-cell patch clamp 방법을 이용하여 시행한 바 다음과 같은 결론을 얻었다. 1. 1mM의 dugenol은 흰쥐 삼차신경절 뉴런의 TTX-r $I_{Na}$와 HVA $I_{Ca}$를 억제하였다. 2. $1{\mu}m$의 capsaicin은 흰쥐 삼차신경절 뉴런의 TTX-r $I_{Na}$와 HVA $I_{Ca}$를 억제하였다. 3. Capsazepine은 capsaicin의 HVA $I_{Ca}$에 대한 억제작용을 차단하였다. 4. Capsazepine은 capsaicin의 HVA $I_{Ca}$에 대한 억제작용을 차단하지 못하였다. 결론적으로 eugenol과 capsaicin은 tetrodotoxin-resistant sodium current(TTX-r $I_{Na}$)와 high voltage-activated calcium current(HVA $I_{Ca}$)를 모두 억제하는 것으로 나타났으며, 이러한 작용이 통각의 발생과 시냅스 전달과정을 차단하여 치수 진정 목적으로 많이 사용하는 eugenol의 작용기전으로 판단된다. 한편 capsaicin의 길항제인 capsazepine을 전처치하였을 때에도 eugenol의 HVA $I_{Ca}$에 대한 억제효과는 변화가 없었다. 이와같은 결과로 보아 HVA $I_{Ca}$에 관한 한 eugenol은 capsaicin 수용기를 통하여 나타나지 않는 것으로 사료된다.

Mechanism of $Ca^{2+}$ -activated $Cl^-$ Channel Activation by Ginsenosides in Xenopus Oocytes

  • Relatively little is known about the signaling mechanism of ginseng saponins (ginsenosides), active ingredients of ginseng, in non-neuronal cells. Here, we describe that ginsenosides utilize a common pathway of receptor-mediated signaling pathway in Xenopus oocytes: increase in intracellular $Ca^{2+}$ concentration via phospholipase C (PLC) and $Ca^{2+}$ mobilization. Ginsenosides induced a marked and robust artivation of $Ca^{2+}$-activated Cl- channels in Xenopus oocytes. The effect of ginsenosides was completely reversible, in a dose-dependent manner with EC$_{50}$ of 4.4 $\mu\textrm{g}$/mi, and specifically blocked by niflumic acid, an inhibitor of $Ca^{2+}$-activated Cl- channel. Intracellular injection of BAPIA abolished the effect of ginsenosides. Intracellular injection of GTP${\gamma}$S also abolished the effect of ginsenosides. The effect of gin senosides on $Ca^{2+}$-activated Cl- currents was greatly reduced by the intracellular injection of heparin, an IP$_3$ receptorantagonist or the pretreatment of PLC inhibitor. These results indicate that ginsenosides activate endogenous $Ca^{2+}$-activated Cl- channels via the activation of PLC and the release of $Ca^{2+}$ from the IP$_3$-sensitive intracellular store following the initial interaction with membrane component(s) from extracellular side. This signaling pathway of ginsenosides may be one of the action mechanisms for the pharmacological effects of ginseng.ts of ginseng.

Inhibitory Effect of $Mg^{2+}$ on the Release of $Ca^{2+}$ from Ryanodine Receptor of the Sarcoplasmic Reticulum in the Skeletal Muscle (골격근 망상체 $Ca^{2+}$유리 Channel[Raynodine receptor]의 $Mg^{2+}$에 의한 유리 억제)

  • The precise mechanism of the Excitation-Contraction Coupling is still uncertain. But the concept that Ca2+ induced Ca2+ release [CICR] from the Ryanodine receptor in the sarcoplasmic reticulum [foot structure] may play a major role in E-C coupling has been widely accepted since 1970`s. It is believed that increased cytosolic Ca2+ followed by CICR is main contributor for E-C coupling of striated muscle. Resulting phenomena of ischemic /post-reperfusion myocyte is increased cytosolic Ca2+, even to the absence of Ca2+ in reperfusate. So intracellular inhibitor to CICR might prevent the ischemic and reperfusion damage of myocardial cells. The relatively purified foot protein, especially heavy sarcoplasmic reticulum rich, of the skeletal muscle was incorporated into the black lipid bilayer [Phosphatidyl ethanolamine: Phosphatidyl serine=l: 1]. Under the steady state of membrane potential [+20 mV], ionic current through Ryanodine receptor was measured with Cs+ as charge carrier. In the cis chamber [Cytoplasmic side], Mg2+ strongly inhibited CICR of Ryanodine receptor[Kd=6.2 nM]. In conclusion, naturally existing intracellular free Mg2+ can inhibit CICR from intracellular Ca2+ reservior [heavy SR]. So post-ischemic or post-reperfusing myocardium could be preserved using additional free Mg2+ in cardioplegic solution or reperfusate, otherwise the optimal concentration is undetermined.

  • Calcium(Ca) ion plays an important role to trigger the secretion of important neurotransmitters. Since Ca ion flows into the cell thru the ion selective channel, the conductance of which depends on the transmembrane potential, the voltage-dependent characteristic of Ca ion channel is crucial to elucidate the stimulus-secretion coupling of exocytosis. The present study measured the Ca ion currents thru a whole-cell configuration patch at the transmembrane potential clamped at various desired levels in the rat chromaffin cell. The resultant current-voltage relationship was differentiated to obtain dynamic conductance at each clamped voltage. Based on these measured data, five numerical parameters were extracted to reveal electrical properties of Ca ion inflow process thru the voltage-gated channel. The present study can be applied to comparing the electrical characteristics of Ca channel under different experimental conditions. Also, further study is warranted to model the conformational changes of the channel molecules.

Ryanodine Receptor-mediated Calcium Release Regulates Neuronal Excitability in Rat Spinal Substantia Gelatinosa Neurons

  • Nitric Oxide (NO) is an important signaling molecule in the nociceptive process. Our previous study suggested that high concentrations of sodium nitroprusside (SNP), a NO donor, induce a membrane hyperpolarization and outward current through large conductances calcium-activated potassium ($BK_{ca}$) channels in substantia gelatinosa (SG) neurons. In this study, patch clamp recording in spinal slices was used to investigate the sources of $Ca^{2+}$ that induces $Ca^{2+}$-activated potassium currents. Application of SNP induced a membrane hyperpolarization, which was significantly inhibited by hemoglobin and 2-(4-carboxyphenyl) -4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide potassium salt (c-PTIO), NO scavengers. SNP-induced hyperpolarization was decreased in the presence of charybdotoxin, a selective $BK_{Ca}$ channel blocker. In addition, SNP-induced response was significantly blocked by pretreatment of thapsigargin which can remove $Ca^{2+}$ in endoplasmic reticulum, and decreased by pretreatment of dentrolene, a ryanodine receptors (RyR) blocker. These data suggested that NO induces a membrane hyperpolarization through $BK_{ca}$ channels, which are activated by intracellular $Ca^{2+}$ increase via activation of RyR of $Ca^{2+}$ stores.

Flux Loss and Neutron Diffraction Measurement Ag-sheathed Bi-2223 Tapes in terms of Flux Creep

  • Alternating current (AC) losses of two Bi-2223 ([Bi, Pb]: Sr: Ca: Cu: O = 2:2:2:3) tapes [(Tape I, un-twist-pitch) and the other with a twist-pitch of 10 mm (Tape II)] were measured and compared. These samples, produced by the powder-in-(Ag) tube (PIT) method, are multi-filamentary. Also, it's produced by non-twist and different twist pitch (8, 10, 13, 30, 50 and 70 mm). The critical current measurement was carried out under the environment in liquid Nitrogen and in zero-field by 4-probe method. Susceptibility measurements were conducted while cooling in a magnetic field. Flux loss measurements were conducted as a function of ramping rate, frequency and field direction. The AC flux loss increases as the twist-pitch of the tapes decreased, in agreement with the Norris Equation. Neutron-diffraction measurements have been carried out investigate the crystal structure, magnetic structures, and magnetic phase transitions in Bi-2223([Bi, Pb]:Sr:Ca:Cu:O)

Voltage-Current Characteristics of (Sr.Ca)$TiO_3$-based Ceramics with contents of Calcite (Ca 변화량에 따른 (Sr.Ca)$TiO_3$계 세라믹의 전압-전류 특성)

  • In this paper, the structural and electrical properties of the $Sr_{1-x}Ca_xTiO_3$($0{\leq}x{\leq}0.2$)-based grain boundary layer ceramics were investigated by X-ray, SEM and V-I system. Increasing content of Ca, the average gram size and the lattice constant were decreased. The relative density of all specimens was > 96%. The 2nd phase formed by thermal diffusing from the surface lead to a very excellent electrical properties, that is, ${\varepsilon}_r$ > 50000, tan${\delta}$ < 0.05, $\Delta$C < ${\pm}$ 10%.

The Action and Pacemaker Potential in the Frog Truncus Arteriosus (개구리 대동맥의 활동전압 및 Pacemaker 전압에 관한 연구)

  • The frog truncus arterious were studied with conventional glass microelectrode technique in order to elucidate the underlying mechanism of spontaneous pacemaker activity. The analyses were focussed on the ionic nature of pacemaker current by changing the concentrations of extracellular $K^+$ and, $Na^+$, or by using blockers of K- and Ca-current and chronotropic transmitters. 1) The action potential of the spontaneously active truncus arteriosus has some characteristic feature of maximal distolic potential ranged from -65 to -75 mV, resting potential from -45 to -50 mV and overshoot voltage about +30 mV, respectively. Duration of the action potential taken from rapid upstroke to maximal diastolic potential was about 600 msec. Usual discharge rate was $25{\sim}30/min$ at room temperature $(18{\sim}20^{\circ}C)$. 2) The sensitivity of the resting membrane potential to change extracellular potassium concentrations $(0{\sim}12\;mM)$ was relatively low. Transient hyperpolarization was appeared in the 12 mM K Ringer after 10 min exposure to 0 mM K and it could be related to Na-pump reactivation by high potassium. 3) Reduction of extracellular sodium concetrations diminished the amplitude and frequency of the action potential. In Ringer solution containing 30% Na (substituted by equimolar Tris), spontaneous activity stopped but reappeared as very slow and small action potential. There was no spotaneous activity in zero Na Ringer solution. 4) Caesium(10 mM), K-current blocker decreased the frequency of the action potential and also pacemaker depolarization. Manganese (2 mM) known to be Ca-current antagonist, blocked spontaneous activity completely. 5) Adrenaline and acetylcholine had no chronotropic effect. But adrenaline increased the duration of plateau phase and the magnitude of the action potential in the follower cell. It is concluded that K-, Na-and Ca-current components are involved in the genesis of spontaneous activity of the frog truncus arteriosus like cardiac pacemaker tissues. But the insensitivity of truncus arteriosus to adrenaline and acetylcholine indicates that there are some different control mechanisms of spontaneous rhythm in two tissues.

Effect of pH on Calcium-Activated Potassium Channels in Pulmonary Arterial Smooth Muscle Cells of the Rabbit

  • Single smooth muscle cells of the rabbit pulmonary artery were isolated by treatment with collagenase and elastase. Using the patch clamp technique, potassium channel activity was recorded from the inside-out membrane patch. The channel had a sin히e channel conductance of about 360 pS in symmetrical concentration of K on both sides of the patch, 150 mM, and had a linear current-voltage relationship. During the application of 10 mM tetraethylammonium (TEA) to the intracellular membrane surface, the amplitude of single channel current was reduced and very rapid flickering appeared. The open probability $(P_0)$ of this channel was increased by increasing positivity of the potential across the patch membrane, with e-fold increase by 20 mV depolarization, and by increasing the internal $Ca^{2+}$ concentration. These findings are consistent with those of large conductance Ca-activated K channels reported in other tissues. But the shortening of the mean open time by increasing $[Ca^{2+}]_i$, was an unexpected result and one additional closed state which might be arisen from a block of the open channel by Ca binding was suggested. The $P_0-membrane$ potential relationship was modulated by internal pH. Decreasing pH reduced $P_0$. Increasing pH not only increased $P_0$ but also weakened the voltage dependency of the channel opening. The modulation of Ca-activated K channel by pH was thought to be related to the mechanism of regulation of vascular tone by the pH change.
