Flavonol Glycosides from the Aerial Parts of Aceriphyllum rossii and Their Antioxidant Activities

  • The methanol extract obtained from the aerial parts of Aceriphyllum rossii (Saxifragaceae) was fractionated into ethyl acetate (EtOAc), n-BuOH and $H_2O$ layers through solvent fractionation. Repeated silica gel column chromatography of EtOAc and n-BuOH layers afforded six flavonol glycosides. They were identified as kaempferol 3-O-$\beta$-D-glucopyranoside (astragalin, 1), quercetin 3-O-$\beta$-D-glucopyranoside (isoquercitrin, 2), kaempferol 3-O-$\alpha$-L-rhamnopyranosyl $(1{\to}6)-\beta$-D-glucopyranoside (3), quercetin 3-O$\alpha$-L-rharnnopyranosyl $(1{\to}6)-\beta$-D-qlucopyrano-side (rutin, 4), kaempferol 3-O-[$\alpha$-L-rharnnopyranosyl $(1{\to}4)-\alpha$-L-rhamnopyranosyl $(1{\to}6)-\beta$-D-glucopyranoside] (5) and quercetin 3-O-[$\alpha$-L-rhamnopyranosyl $(1{\to}4)\alpha$-L-rhamnopyranosyl $(1{\to}6)\beta$-D-glucopyranoside] (6) on the basis of several spectral data. The antioxidant activity of the six compounds was investigated using two free radicals such as the ABTS free radical and superoxide anion radical. Compound 1 exhibited the highest antioxidant activity in the ABTS $\{2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid)\}$ radical scavenging method. 100 mg/L of compound 1 was equivalent to $72.1\pm1.4\;mg/L$ of vitamin C, and those of compounds 3 and 5 were equivalent to $62.7\pm0.5\;mg/L$ and $54.3\pm1.3\;mg/L$ of vitamin C, respectively. And in the superoxide anion radical scavenging method, compound 5 exhibited the highest activity with an $IC_{50}$ value of $17.6{\pm}0.3{\mu}M$. In addition, some physical and spectral data of the flavonoids were confirmed.

Effect of Porcine Placenta Extract from Subcritical Water Extraction on Photodamage in Human Keratinocytes

  • The objective of this study was to evaluated the photoprotective effects of porcine placenta extract (PPE) on ultraviolet B (UVB)-induced oxidative stress in human keratinocytes (HaCaT) to evaluate its functional activities as a skin food ingredient. PPE prepared by subcritical water extraction was termed SPE, and subsequently digested by enzymes to prepare E-SPE. Increased intracellular reactive oxygen species (ROS) levels (192.0%) induced by UVB were decreased by SPE and E-SPE. SPE had more effective ROS scavenging activity than E-SPE treatment. UVB treatment increased expression of tissue inhibitor of metalloproteinase 1 (TIMP-1), and this elevated expression was decreased by E-SPE treatment. High-dose treatment with E-SPE (50 and 100 µg/mL) reduced TIMP-1 expression levels of UVB-C (control) to 33.5 and 34.6%, respectively. In contrast, at low SPE doses (1 and 10 µg/mL), the treatment slightly decreased TIMP- 1 expression levels to 73.3% and 71.3% of UVB-C, respectively. In conclusion, the present study demonstrated the protective effect of SPE and E-SPE against UVB damage in keratinocytes via ROS scavenging, down-regulating MMP-2 expression and up-regulating TIMP- 1 expression. This highlights the potential for SPE as an ingredient in the preparation of functional food against photoaging.

한라봉 추출액이 첨가된 커피 분말의 제조 및 품질 특성에 관한 연구 (Manufacturing and Quality Characteristics Analysis of Coffee Powder with Added Hallabong Extract)

  • In this study, Korean Hallabong produced in Jeju Island and coffee were grafted to prepare coffee containing Hallabong extract and the nutritional components were analyzed. As the amount of Hallabong extract increased, the water content and total polyphenol content increased. However, the crude flour, crude protein, and total flavonoid content decreased significantly. The selenium content per 100 g was 91.28 mg in the 1% Hallabong group, and the iron content was 6.84 mg in the 3% Hallabong group. As the content of Hallabong extract in coffee increased, the L-value (brightness) and b-value (yellowness) increased, but the a-value (redness) showed a tendency to decrease. In the case of DPPH(2,2-Diphenyl-1-picrylhydrazyl) radical scavenging activity, the group containing 9% of Hallabong extract showed the highest value at 47.20 μmol/g of TEAC. In particular, the ABTS(2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonate)) and DPPH radical scavenging activity were significantly increased from coffee powder containing 6% or more of Hallabong extract(p<0.05). The caffeine content decreased as the amount of Hallabong extract added to coffee increased. Therefore, when making powder coffee with Hallabong extract added, it is recommended to set the content of Hallabong extract to 6%.

2차 발효 후 로제 스파클링 와인의 품질 특성 및 기능성 (Quality Characteristics and Functionality of Rose Sparkling Wines by Secondary Fermentation)

  • This study investigated quality characteristics and functionality of rose sparkling wine that were manufactured by secondary fermentation after inoculation of yeast strains. The pH of the samples ranged from 3.96 to 4.05 and total acidity ranged from 0.23 to 0.32%. The alcohol contents of wines ranged from 6.4 to 6.6% and the CO2 pressure ranged from 2.0 to 2.6 bar. The brightness of rose wines ranged from 84.72 to 87.36, the redness from 9.28 to 14.15, and the yellowness from 9.50 to 19.20. The hue value of wines ranged from 1.137 to 1.513 and color intensity ranged from 0.724 to 0.882. Aroma analysis identified 14 alcohols, 22 esters, 4 ketones, 4 acids, and 17 miscellaneous compounds. The total tannin contents and total polyphenol contents of wines were 11.28~12.43 mg% and 24.79~28.20 mg%, respectively. The DPPH radical scavenging activity of wines ranged 63.33 to 67.89% and the ABTS radical scavenging activity of wines 82.16~86.06%. The results of this study provide a basis for establishing the brewing process of rose sparkling wines yeast strains.

두과작물 새싹의 폴리페놀 함량 및 항산화성 비교 (Phenolics Content and Antioxidant Activity of Sprouts in Several Legume Crops)

  • 대두, 녹두, 및 동부 종자로 7일간 재배된 새싹나물의 생육, 폴리페놀 함량, 플라보노이드 함량, 항산화성 및 항산화효소 활성 차이를 검토하였다. 작물별 새싹나물의 총 신장은 녹두와 콩이 동부보다 유의적으로 컸으며 생체중은 오히려 콩과 동부가 녹두보다 유의적으로 높게 나타났다. Folin-Denis방법에 따른 총 페놀 함량은 콩나물의 메탄올 추출물(82.2 mg $kg^{-1}$)이 가장 높았으며, 그 다음이 동부나물(32.2 mg $kg^{-1}$), 녹두나물(24.5 mg $kg^{-1}$) 순으로 나타났다(p < 0.05). 한편, 총 플라보노이드 함량은 총 페놀 함량과 같은 경향을 보였으나 더 낮은 함량이 검출되었다. DPPH 라디컬 소거능은 추출물 농도가 증가할수록 높은 활성을 보였으며 전체적으로 낮은 활성이었으나 동부와 녹두 추출물(44와 42%)이 콩나물(25%)보다 비교적 높은 활성을 보였다. 항산화효소 활성은 APX와 POX활성은 동부가 가장 높았고 그 다음이 녹두, 콩 순으로 나타났고, CAT와 SOD 활성은 콩나물이 동부와 녹두나물보다 높게 나타났다. 따라서 본 연구는 총 페놀 함량($r^2$ = 0.53 ~ 0.90)과 총 플라보노이드 함량($r^2$ = 0.47 ~ 0.94)은 항산화성과 항산화효소 활성에 높은 연관성이 있으며, 그 함량과 활성은 작물별로 다르게 나타남을 확인하였다.

Bioconverted Jeju Hallabong tangor (Citrus kiyomi × ponkan) peel extracts by cytolase enhance antioxidant and anti-inflammatory capacity in RAW 264.7 cells

  • BACKGROUND/OBJECTIVES: Citrus and its peels have been used in Asian folk medicine due to abundant flavonoids and usage of citrus peels, which are byproducts from juice and/or jam processing, may be a good strategy. Therefore, the aim of this study was to examine antioxidant and anti-inflammatory effects of bioconversion of Jeju Hallabong tangor (Citrus kiyomi ${\times}$ ponkan; CKP) peels with cytolase (CKP-C) in RAW 264.7 cells. MATERIALS/METHODS: Glycosides of CKP were converted into aglycosides with cytolase treatment. RAW 264.7 cells were pre-treated with 0, 100, or $200{\mu}g/ml$ of citrus peel extracts for 4 h, followed by stimulation with $1{\mu}g/ml$ lipopolysaccharide (LPS) for 8 h. Cell viability, DPPH radical scavenging activity, nitric oxide (NO), and prostagladin $E_2$ ($PGE_2$) production were examined. Real time-PCR and western immunoblotting assay were performed for detection of mRNA and/or protein expression of pro-inflammatory mediators and cytokines, respectively. RESULTS: HPLC analysis showed that treatment of CKP with cytolase resulted in decreased flavanone rutinoside forms (narirutin and hesperidin) and increased flavanone aglycoside forms (naringenin and hesperetin). DPPH scavenging activities were observed in a dose-dependent manner for all of the citrus peel extracts and CKP-C was more potent than intact CKP. All of the citrus peel extracts decreased NO production by inducible nitric oxide synthase (iNOS) activity and $PGE_2$ production by COX-2. Higher dose of CKP and all CKP-C groups significantly decreased mRNA and protein expression of LPS-stimulated iNOS. Only $200{\mu}g/ml$ of CKP-C markedly decreased mRNA and protein expression of cyclooxygenase-2 in LPS-stimulated RAW 264.7 cells. Both 100 and $200{\mu}g/ml$ of CKP-C notably inhibited mRNA levels of $interleukin-1{\beta}$ ($IL-1{\beta}$) and IL-6, whereas $200{\mu}g/ml$ CKP-C significantly inhibited mRNA levels of $TNF-{\alpha}$. CONCLUSIONS: This result suggests that bioconversion of citrus peels with cytolase may enrich aglycoside flavanones of citrus peels and provide more potent functional food materials for prevention of chronic diseases attributable to oxidation and inflammation by increasing radical scavenging activity and suppressing pro-inflammatory mediators and cytokines.

양릉천 시호 약침이 고지혈증에 미치는 영향 (The Effect of Bupleuri Radix Herbal-acupuncture at GB34(Yangneungcheon) on Hyperlipidemia in Rats)

  • Objectives & Methods : The purpose of this study is to observe the effects of Bupleuri Radix Herbal-acupuncture at GB34(Yangneungcheon) of hyperlipidemic rats. The author performed several items to analyze body-weight of the rats, weight of its liver, serum as well as urine changes in each group. Results : 1. Bupleuri Radix herbal-acupuncture(BR-HA) infused solution (MCR-HAS) increased both DPPH radical scavenging activity and HMG-CoA reductase inhibition rate in rat liver cells. 2. BR-HA at GB34 significantly decreased the level of serum total cholesterol and atherogenic index(AI), while significantly increased the ratios of high-density lipoprotein (HDL)/total cholesterol and phospholipid/total cholesterol in hyperlipidemic rats. 3. BR-HA at GB34 significantly decreased urine glucose in hyperlipidemic rats. Conclusions : From the results above, it is suggested that Bupleuri Radix herbal -acupuncture at GB34 has a therapeutic effect on hyperlipidemia.

병이소초 추출물의 항산화 및 MMP 발현 저해 효과 (Inhibitory Effect of Ophioglossum vulgatum on Free Radical and MMP Expression in UV-irradiated Human Dermal Fibroblasts)

  • 자외선에 의한 활성산소 종의 생성과 피부 광노화 촉진은 피부표면 조직의 파괴와 피부염증유발, 홍반 등을 수반하며, 일상생활에서 항상 생활 자외선에 노출되어 있는 피부는 이러한 위험 요인과 직면하고 있다. 병이소초추출물의 유해 라디칼 소거 효과 실험 결과 $IC_50$ values가 $18.2\;{\mu}g/mL$로 superoxide 소거 효과가 우수하게 나타났으며, 세포 내에서 ROS에 의해 형광을 띠는 물질로 전환되는 CM-DCFDA를 이용하여 ROS의 양을 측정한 결과 자외선에 의해 증가된 세포 내 ROS의 양이 병이소초 추출물을 처리함으로써 $100\;{\mu}g/mL$ 농도에서 30 % 이상의 우수한 소거효과를 나타내었다. 섬유아세포에 자외선(UVA) $6.3\;J/cm^2$으로 조사하고 병이소초 추출물을 처리한 결과, 자외선에 의해 높아졌던 MMP-1 단백질 발현량이 농도의존적으로 감소하였으며, $50\;{\mu}g/mL$의 농도에서 37.7 % 정도의 MMP-1 발현 억제효과를 나타냈다. RT-PCR 실험에서는 병이소초 추출물을 $10\;{\sim}\;50\;{\mu}g/mL$ 농도로 처리한 결과, 자외선에 의해 높아졌던 MMP-1 mRNA의 발현이 농도 의존적으로 확연히 감소함을 확인할 수 있었다. 따라서 병이소초추출물은 자외선에 의해서 발생할 수 있는 피부손상에 대하여 효과적으로 보호할 수 있는 우수한 항산화 및 피부세포 보호소재로 적용될 수 있다.

약재 추출물을 첨가한 과립의 품질 특성 및 항산화 효과 (Quality and Antioxidant Activity of Granules Containing Herbal Medicine Extracts)

  • 원료를 배합비율에 맞게 칭량한 후 granule type으로 과립을 제조한 다음 유동층 건조기에 과립물을 넣고 건조하여 과립을 제조하였다. 과립의 항산화 활성을 평가한 결과 항산화 활성을 나타내는 DPPH radical 소거능, ABTS radical 소거능, 환원력에서 모두 약재 추출물을 첨가한 과립의 항산화 활성이 가장 높았으며, 폴리페놀 함량도 126.44 mg/100g으로 가장 많았다(P<0.05). 기호도를 조사한 결과 약재 추출물을 첨가하여 제조한 과립이 맛과 전반적인 기호도에서 좋은 점수를 얻어 통계적으로 유의적인 차이를 보였으나 색상, 향미, 조직감에서는 약재 추출물을 첨가하지 않은 과립과 통계적으로 유의적인 차이를 보이지 않았다. 따라서 약재 추출물을 첨가한 과립이 기호도를 만족시키면서 기능성식품으로의 가능성이 있다고 판단된다.

유산균 발효에 의한 톳 (Hizikia fusiforme) 추출액의 항산화 및 항염증 활성 증가 (Enhancement of the Antioxidant and Anti-inflammatory Activity of Hizikia fusiforme Water Extract by Lactic Acid Bacteria Fermentation)

  • We previously reported that fermentation by Lactobacillus brevis LB-20 isolated from Kimchi resulted in improvement of the sensory quality of Hizikia fusiforme water extract. This study was conducted to evaluate the possible application of lactic acid bacteria fermentation to improve the functional qualities of H. fusiforme extract. L. brevis LB-20 was inoculated and cultivated in H. fusiforme extract. The antioxidant and anti-inflammatory activities of extract were then assayed both before and following fermentation for two days. Antioxidant activity was determined by assaying levels of radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl, superoxide, and alkyl radical. Lactic acid bacterial fermentation of H. fusiforme extract resulted in enhancement of antioxidant activity. The greatest enhancement of antioxidant activity was seen in the hydroxyl radical scavenging assay that incorporated 0.5 mg/mL of raw and fermented H. fusiforme extract. Fermented extract exhibited greater (21.95%) inhibition of nitric oxide synthesis than did raw extract (14.66%) at a concentration of 1 mg/mL. The fermented extract exerted its potent anti-inflammatory activity via attenuation of expression of inflammation-related cytokine proteins (TNF-${\alpha}$ and iNOS).
